Pharmacological treatment facilitating locomotor expression will also have some effects on reflex expression through the modulation of spinal circuitry. Buspirone, a partial serotonin receptor agonist (5-HT1 A), was recently shown to facilitate and even trigger locomotor movements in mice after complete spinal lesion (Tx). Here, we studied its effect on the H-reflex after acute Tx in adult mice. To avoid possible impacts of anesthetics on H-reflex depression, experiments were performed after decerebration in un-anesthetized mice (N = 20). The H-reflex in plantar muscles of the hind paw was rec

Effects of a single dose of the anxiolytic buspirone (15 mg) on memory and psychomotor performance were studied in healthy volunteers and compared to those of the classic benzodiazepine anxiolytic diazepam (15 mg). The study was performed in a double-blind, placebo-controlled way. Three groups of 12 subjects were exposed to an extended test battery before and after intake of drug or placebo. Next to this, an evaluation session took place 1 week later. Immediately after intake, diazepam exerted major effects on memory, impaired psychomotor performance and decreased alertness. In particular, lon

1. Metabolism of the antianxiety drug buspirone was studied by in vitro incubations with rat liver microsomes and hepatocytes. Metabolites were isolated and purified by h.p.l.c. The purified metabolites were identified by co-elution on h.p.l.c. with authentic standards and by g.l.c.-electron impact mass spectrometry of their trimethylsilyl (TMS) derivatives. 2. Five metabolites of buspirone were identified in the microsomal incubates and seven in the hepatocyte incubates. The major metabolites arose from aromatic hydroxylation at C-5, N-dealkylation of the butyl chain, and hydroxylation at C-6

Buspirone is a novel anti-anxiety drug which has neither a benzodiazepine structure nor any other benzodiazepine-like properties. Since buspirone is known to block dopamine autoreceptors and increase dopamine metabolism, it was compared to classical antipsychotic drugs for subchronic and chronic effects on dopaminergic function. When continuously infused into rats at 1.0 mg/kg/day s.c. for 2 weeks there was a small diminution of striatal dopamine metabolism response to acute injection of buspirone, but haloperidol produced a normal increase in DOPAC concentrations. In contrast, infusion of 0.3

Buspirone, a 5-hydroxytryptamine 1A (5-HT1A) receptor agonist, has been investigated for its use in various diseases. However, knowledge about its side effects and potential cognitive benefits in different conditions is limited. Cognitive impairment is also a prevalent symptom in many diseases, yet effective treatments are still lacking. Therefore, to explore the potential side effects of buspirone and the possible cognitive benefits of buspirone, we conducted a comprehensive search of several databases, including PubMed, Embase, Web of Science, Cochrane Review, Cochrane Trial, and ClinicalTri

Study Design: Prospective open-label trial. Objectives: The objective of this study was to determine whether buspirone showed preliminary evidence of effectiveness, safety, and tolerability in individuals with Williams syndrome (WS). Methods: This is a 16-week, prospective, flexibly dosed, open-label trial of buspirone in 20 individuals with WS aged 5-65 years. The primary outcome measure was the Pediatric Anxiety Rating Scale (PARS). Results: Buspirone use (mean dose, 22.6 mg per day) was associated with a reduction in anxiety severity, with Cohen's d estimate of -4.02 for the PARS. All 18 pa

Co-morbid anxiety disorders, including generalized anxiety disorder (GAD), are highly prevalent among individuals with Williams syndrome (WS). However, reports of the pharmacologic treatment of only a limited number of previous anxiety disorders in WS have appeared in the literature. Here, we review the case histories of three adolescents/young adults with WS and the treatment course of co-morbid GAD with buspirone. Treatment with buspirone was well-tolerated and resulted in sustained response in all three cases. Common medical disorders in WS are highlighted with regards to safe and appropria

The study examined the possibility that propranolol, buspirone, pCPA, and reserpine have antianxiety effects by comparing their effects with those of chlordiazepoxide on an open-field test of emotionality in rats. The effects of intraperitoneal injections of d,l, propranolol (5, 10, 20 mg/kg), buspirone (1.25, 2.5, 5 mg/kg), reserpine (0.5 mg/kg), parachlorophenylalanine (pCPA) (100 mg/kg), and chlordiazepoxide (CDP) (2.5, 5, 10 mg/kg) were compared with performance of rats under saline or water using an open-field test on 5 successive days. Significant effects were found on peripheral movemen

The effects of the clinically available 5-HT1A agonist, buspirone, were investigated on food intake in food-deprived rats in both nonoperant and operant feeding paradigms. Buspirone (0.5-2.0 mg/kg), administered subcutaneously (s.c.) 15 min prior to presentation of food, produced a dose-related inhibition of food intake in the nonoperant feeding paradigm. The main inhibitory effects of buspirone were apparent during the first 30 min after food presentation. Likewise, in the operant feeding paradigm, buspirone (0.25-1.0 mg/kg, s.c.) produced a dose-related suppression of food intake in the firs

OBJECTIVE: The efficacy of buspirone in controlling self-injurious behavior was examined in five individuals with mental retardation. Buspirone was used alone in two individuals and as an adjunct to thioridazine in the other three.
METHOD: Standard behavioral observation methods were used to collect data on the number of self-injurious responses of the individuals during baseline and several doses of buspirone in an open trial.
RESULTS: When compared with baseline levels, all five individuals showed some response to buspirone, with reductions in self-injury ranging from 13% to 72%, depending

Hostility is considered to be a risk factor for coronary artery disease. Despite the findings of reduced serotonergic function and effectiveness of treatment with serotonergic agents in similar symptom complexes, there have been no attempts at using serotonergic psychopharmacologic agents to treat this population. We conducted an 8-week open trial of 10 type A, irritable men with coronary artery disease and no diagnosable psychiatric (axis I) condition to examine the effects of buspirone on these subjects. Buspirone appears to significantly reduce type A behavior, hostility, anxiety, impatienc

Buspirone and gepirone were evaluated as potential pharmacotherapies for cocaine abuse by studying the effects of acute and repeated treatment on IV cocaine self-administration in rhesus monkeys. Chlorpromazine was also evaluated as a positive control. Effects of IV drug pretreatments were tested during daily 60-min sessions with lever-pressing reinforced under a fixed-ratio 10 schedule of 0.02 or 0.05 mg/kg cocaine infusions. Acute pretreatment with buspirone (0.1 and 0.3 mg/kg, IV) increased rates of cocaine self-administration without disrupting food pellet consumption. Some doses of buspir

Buspirone hydrochloride (HCl)1 is a new anxiolytic with a unique chemical structure. Its mechanism of action remains to be elucidated. Unlike the benzodiazepines, buspirone lacks hypnotic, anticonvulsant and muscle relaxant properties, and hence has been termed 'anxioselective'. As evidenced by a few double-blind clinical trials, buspirone 15 to 30 mg/day improves symptoms of anxiety assessed by standard rating scales similarly to diazepam, clorazepate, alprazolam and lorazepam. Like diazepam, buspirone is effective in patients with mixed anxiety/depression, although the number of patients stu

Spinal cord injury (SCI) is a risk factor for central sleep apnea (CSA). Previous studies in animal models with SCI have demonstrated a promising recovery in respiratory and phrenic nerve activity post-injury induced by the systemic and local administration of serotonin receptor agonists such as Buspirone and Trazodone. Human trials must be performed to determine whether individuals with SCI respond similarly. We hypothesized that Buspirone and Trazodone would decrease the propensity to hypocapnic CSA during sleep. We studied eight males with chronic SCI and sleep-disordered breathing (SDB) [a

This study compared behavioral and subjective effects of two anxiolytics, the benzodiazepine lorazepam and the azaspirodecanedione buspirone, in healthy male volunteers with histories of sedative drug abuse. Placebo, lorazepam (1, 2, 4, 8mg/70kg) and buspirone (15, 30, 60, 120mg/70kg) were administered p.o. in a mixed sequence in a double-blind, cross-over design. Lorazepam, but not buspirone, decreased psychomotor/cognitive performance. Both drugs produced similar increases in ratings of drug strength, however the onset and offset times for lorazepam were later than for buspirone. Lorazepam i

The anxiolytic drug buspirone and its structural analogues gepirone and ipsapirone produce behavioural effects which differ from those seen with benzodiazepines and barbiturates. Buspirone has been reported to disrupt active avoidance responding without interfering with the ability to escape the aversive stimuli, an effect usually associated with antipsychotic drugs. In the present study, buspirone, gepirone and ipsapirone produced dose-related decreases in one-way, active avoidance responding of rats. However, these drugs did not consistently give rise to a within-session decline in avoidance
