Ca(2+) -independent effects of BAPTA and EGTA on single-channel Cl(-) currents in brown adipocytes.

The Cl(-) channels of brown adipocytes electrophysiologically resemble outwardly rectifying Cl(-) channels (ORCC). To study tentative Ca(2+) regulation of these channels, we attempted to control Ca(2+) levels at the cytoplasmic side of the inside-out membrane patches with Ca(2+)-chelating agents. However, we found that the commonly used Ca(2+)-chelators EGTA and BAPTA by themselves influenced the Cl(-) channel currents, unrelated to their calcium chelating effects. Consequently, in this report we delineate effects of Ca(2+)-chelators (acting from the cytoplasmic side) on the single Cl(-) chann

Ammonium tetrathiomolybdate attenuates acetaminophen-induced acute liver failure by inhibiting the TRPV4/Calcium/NF-κB signaling pathway.

Acute liver failure (ALF), characterized by fulminant hepatic necrosis and excessive inflammatory-oxidative cascades, remains a critical clinical challenge with limited therapeutic options. This study investigates the therapeutic potential of ammonium tetrathiomolybdate (ATTM)-a copper-chelating agent with multimodal anti-inflammatory and antioxidant properties-in acetaminophen (APAP)-induced ALF. Utilizing APAP-challenged C57BL/6J mice, we demonstrated that ATTM administration, whether prophylactic or delayed by 2 h post-exposure, significantly attenuated hepatotoxicity, as evidenced by reduc

Antioxidant mechanism of milk mineral-high-affinity iron binding.

Milk mineral (MM), a by-product of whey processing, is an effective antioxidant in meat systems, but the antioxidant mechanism has not been established. MM has been postulated to chelate iron and prevent iron-catalysis of lipid oxidation. The objective of this research was to examine this putative mechanism. MM was compared to sodium tripolyphosphate (STPP), calcium phosphate monobasic (CPM), and calcium pyrophosphate (CPP) to determine iron-binding capacity, sample solubility, and eluate soluble phosphorus after treating samples with a ferrous chloride standard. Moringa oleifera Lam. Leaf Peptide-Calcium Chelate (KTFQGPPHG-Ca): Screening, In Vitro Digestive Stability, Cellular Transport, and Potential Calcium Absorption Mechanisms.

This study aimed to address the poor bioavailability of traditional calcium supplements by investigating the Moringa oleifera Lam. leaf-derived peptide KTFQGPPHG and its peptide-calcium chelate. Virtual screening predicted potential interactions of KTFQGPPHG with Ca2+ and calcium transport-related proteins, including TRPV6, Cav1.3, and PepT1. During simulated digestion, KTFQGPPHG-Ca showed high intestinal-phase stability, with a calcium retention rate of 94.13 ± 1.47%, while its major digestive fragment, TFQGPPHG, retained substantial calcium-binding activity. In Caco-2 cell monolayers, both K

Sodium thiosulfate: an emerging treatment for calciphylaxis in dialysis patients.

Calciphylaxis is a rare but life-threatening complication seen in dialysis patients. We report a case with calciphylaxis and severe secondary hyperparathyroidism. Sodium thiosulfate (STS) was used for treatment in this case. The painful skin ulcers improved significantly soon after treatment, with a slight reduction in the calcium-phosphorus product. Total parathyroidectomy was carried out 6 months after the STS therapy, which successfully solved hyperparathyroidism and the high calcium-phosphorus profiles. Until now, no recurrence of calciphylaxis has been observed during the more than 7 year

ST1926, a novel and orally active retinoid-related molecule inducing apoptosis in myeloid leukemia cells: modulation of intracellular calcium homeostasis.

Retinoid-related molecules (RRMs) are derivatives of retinoic acid and promising antileukemic agents with a mechanism of action different from that of other common chemotherapeutics. Here, we describe a novel chemical series designed against the RRM prototype, CD437. This includes molecules with apoptotic effects in acute promyelocytic leukemia and other myelogenous leukemia cell lines, as well as ST2065, an RRM with antagonistic properties. The most interesting apoptotic agent is ST1926, a compound more powerful than CD437 in vitro and orally active in vivo on severe combined immunodeficiency

Nanoparticles reduce nickel allergy by capturing metal ions.

Approximately 10% of the population in the USA suffer from nickel allergy, and many are unable to wear jewellery or handle coins and other objects that contain nickel. Many agents have been developed to reduce the penetration of nickel through skin, but few formulations are safe and effective. Here, we show that applying a thin layer of glycerine emollient containing nanoparticles of either calcium carbonate or calcium phosphate on an isolated piece of pig skin (in vitro) and on the skin of mice (in vivo) prevents the penetration of nickel ions into the skin. The nanoparticles capture nickel i

Stimulation of deoxycytidine kinase results in prolonged maintenance of the enzyme activity.

A number of genotoxic and antiproliferative agents such as 2-chlorodeoxyadenosine (Cladribine; CdA) and aphidicolin (APC) have been shown to stimulate the activity of deoxycytidine kinase, the main deoxynucleoside salvage enzyme in lymphocytes. Here we show that enzyme activation could be prevented by treating cells with the membrane-permeant calcium chelator BAPTA-AM. Long-term incubations demonstrated that CdA and APC not only stimulated but also sustained deoxycytidine kinase activity in the cellular context, as compared to the control and BAPTA-AM treated enzyme activities.

Calcium is essential for fructan synthesis induction mediated by sucrose in wheat.

The role of Ca(2+) in the induction of enzymes involved in fructan synthesis (FSS) mediated by sucrose was studied in wheat (Triticum aestivum). Increase of FSS enzyme activity and induction of the expression of their coding genes by sucrose were inhibited in leaf blades treated with chelating agents (EDTA, EGTA and BAPTA). Ca(2+) channel blockers (lanthanum chloride and ruthenium red) also inhibited the FSS response to sucrose, suggesting the participation of Ca(2+) from both extra- and intra- cellular stores. Sucrose induced a rapid Ca(2+) influx into the cytosol in wheat leaf and root tissu

Synergistic Antifungal Effect of a Combination of Iron Deficiency and Calcium Supplementation.

Fungal diseases have become a major public health issue worldwide. Increasing drug resistance and the limited number of available antifungals result in high morbidity and mortality. Metal-based drugs have been reported to be therapeutic agents against major protozoan diseases, but knowledge of their ability to function as antifungals is limited. In this study, we found that calcium supplementation combined with iron deficiency causes dramatic growth inhibition of the human fungal pathogens Aspergillus fumigatus, Candida albicans, and Cryptococcus neoformans. Calcium induces the downregulation

Effects of salicylate on rectal absorption of theophylline.

The rectal absorption of theophylline in rats was facilitated by concurrent administration of salicylic acid or sodium salicylate. The absorption of theophylline depended on the simultaneous absorption of salicylate and increased with an increasing concentration of salicylate. Calcium and magnesium ions inhibited the effects of salicylate at pH 7.4 but not at pH 4.5. Sodium lauryl sulfate caused lasting changes in rectal absorption, whereas the effects of salicylate on absorption were observed only when salicylate was present Strong chelating agents, such as ethylenediaminetetraacetic acid and

Protection of mature oligodendrocytes by inhibitors of caspases and calpains.

Mature mouse oligodendrocytes (OLs) are susceptible to death in demyelinating diseases such as multiple sclerosis and in brain injury following neurotrauma, ischemia, or stroke. To understand mechanisms leading to death of mature OLs and develop strategies for protection, we utilized cultures of mature mouse OLs to investigate the role of caspases and calpains in OL cell death mediated by different mechanisms. The agents used were (i) staurosporine, which induces apoptotic death via inhibition of protein kinases; (ii) kainate, which activates non-NMDA glutamate receptors; (iii) thapsigargin, w

Caspase-independent apoptosis induced by differentiation-inducing factor of Dicytostelium discoideum in INS-1 cells.

Differentiation-inducing factor (DIF) is a lipophilic hormone of Dicytostelium discoideum and has been shown to exert diverse effects in mammalian cells. We investigated the effect of DIF on cell viability in insulin-secreting INS-1 cells. DIF induced cell death in a dose-dependent manner. In DIF-treated cells, nuclear condensation and shrinkage of the cell body were observed. After 6 h of DIF treatment, cells became Tdt-mediated dUTP-biotin nick end-labeling-positive, and DNA ladder formation was detected, indicating that DIF induced apoptosis in these cells. DIF did not activate caspase-3, a

Effect of intracellular calcium modulation on sulfur mustard cytotoxicity in cultured human neonatal keratinocytes.

Previous studies in human skin keratinocyte cultures have shown that sulfur mustard (HD) induces an immediate and irreversible increase in internal free calcium levels that was independent of external calcium concentrations. These findings suggested a role for calcium in the aetiology of HD-induced cell death and that modulation of intracellular calcium concentrations may assist in providing protection against this agent. In the current work, actively proliferating and confluent cultures of first passage neonatal human skin keratinocytes were used to assess the effect of altered intra- and ext

Zinc ions cause the thimerosal-induced signal of fluorescent calcium probes in lymphocytes.

Most fluorescent probes for the investigation of calcium signaling also detect zinc ions. Consequently, changes in the intracellular zinc concentration could be mistaken for calcium signals. Thimerosal (TMS) is used as a calcium-mobilizing agent and we analyzed the contribution of zinc ions to the signal observed with fluorescent calcium probes after TMS stimulation. Our findings show that the fluorescent signal in lymphocytes is entirely due to zinc release. Experiments in the T lymphocyte cell line Jurkat and primary human lymphocytes show that TMS and its active metabolite, ethyl mercury, c

Metal ion chelation of poly(aspartic acid): From scale inhibition to therapeutic potentials.

Poly(aspartic acid) (PASP) is a biodegradable, biocompatible water-soluble synthetic anionic polypeptide. PASP has shown a strong affinity and thus robust complexation with heavy and alkaline earth metal ions, from which several applications are currently benefiting, and several more could also originate. This paper discusses different areas where the ion chelation ability of PASP has thus far been exploited. Due to its calcium chelation ability, PASP prevents precipitation of calcium salts and hence is widely used as an effective scale inhibitor in industry. Due to potassium chelation, PASP p

The experimental and clinical pharmacology of propofol, an anesthetic agent with neuroprotective properties.

Propofol (2,6-diisopropylphenol) is a versatile, short-acting, intravenous (i.v.) sedative-hypnotic agent initially marketed as an anesthetic, and now also widely used for the sedation of patients in the intensive care unit (ICU). At the room temperature propofol is an oil and is insoluble in water. It has a remarkable safety profile. Its most common side effects are dose-dependent hypotension and cardiorespiratory depression. Propofol is a global central nervous system (CNS) depressant. It activates gamma-aminobutyric acid (GABA A) receptors directly, inhibits the N-methyl-d-aspartate (NMDA)

Investigation of Ca2+ signaling induced by the flavonoid diosmin in glioblastoma cells and its potential implications in the treatment using the Ca2+ chelating agent BAPTA-AM.

Flavonoids, found in fruits and vegetables, can potentially prevent brain diseases. Diosmin (diosmetin-7-O-rutinoside), a flavonoid, exhibits various pharmacological activities, but its impact on calcium ion (Ca2+) signaling and the associated mechanisms in human glioblastoma cells remain unclear. This study investigated the effect of diosmin on intracellular Ca2+ levels ([Ca2+]i), cell viability, and the participation of Ca2+-related pathways in DBTRG-05MG human glioblastoma cells. It also investigated the connection between Ca2+ signaling and toxicity in cells treated with diosmin and the Ca

Desferrioxamine therapy in hemodialysis patients with aluminum-associated bone disease.

Aluminum toxicity in dialysis patients is associated with decreased bone turnover and a relative parathyroid hormone (PTH) deficiency. Desferrioxamine (DFO), a chelating agent, has been reported to improve bone histology in aluminum associated, low turnover bone disease in dialysis patients not subjected to parathyroidectomy. Information on the effect of DFO therapy on parathyroid gland function is lacking. In the present study, in addition to changes in bone histology, parathyroid gland function was evaluated in 18 hemodialysis patients with aluminum associated, low turnover bone disease (ost
