Physicochemical metabolic characteristics for calcium oxalate stone formation in patients with gouty diathesis.

PURPOSE: We determined why calcium oxalate stones instead of uric acid stones form in some patients with gouty diathesis. MATERIALS AND METHODS: Gouty diathesis was diagnosed from absence of secondary causes of uric acid stones or low urinary pH, and reduced fractional excretion of urate with discriminant score of the relationship between urinary pH and fractional excretion of urate less than 80. From the stone registry 163 patients with gouty diathesis were identified, including 62 with uric acid stones (GD + UA) and 101 patients with calcium oxalate stones (GD + Ca). Metabolic data and 24-h

Secondary cutaneous oxalosis: cutaneous deposition of calcium oxalate dihydrate after long-term hemodialysis.

A 48-year-old Japanese man had miliary calcified papules on the distal fingers after long-term hemodialysis. The crystals in the dermis consisted of calcium oxalate, as revealed by light microscopy and scanning electron microscopy with energy-dispersive x-ray microanalysis. The calcium oxalate was in the form of a dihydrate crystal. Therefore a diagnosis of secondary cutaneous oxalosis was made. In cutaneous oxalosis that occurs as a result of hemodialysis, miliary deposits occur in the fingers, but not the toes, and are present more frequently on the palmar aspects of the fingers. This locali

Cementum Attachment Protein-Derived Peptides Modulate Brushite and Calcium Oxalate Crystallization In Vitro.

Pathological mineralization involves the uncontrolled crystallization of calcium phosphate (brushite) and calcium oxalate, leading to renal calculi and ectopic calcifications. Peptides enriched in acidic or phosphorylated residues are potential crystal growth modulators due to their ability to interact with calcium-rich surfaces. This study investigated the in vitro effects of cementum attachment protein-derived peptides, CAP-pi and its phosphorylated analog CAP-pip, on brushite and calcium oxalate crystallization. By isolating these highly anionic motifs, this work introduces a novel biomimet

Inhibition of NLRP3 alleviates calcium oxalate crystal-induced renal fibrosis and crystal adhesion.

Kidney stones are gaining attention as one of the most common urological diseases. In this study, we first constructed a mouse model of calcium oxalate (CaOx) crystal deposition by intraperitoneal injection of glyoxalate (Gly) and found that the levels of NLRP3, CASP1 and ASC, which constitute the NLRP3 inflammasome, as well as the level of its downstream product, IL-1β, were elevated in the kidneys of the model group of mice, as determined by RNA-seq. We then examined NLRP3 expression via immunohistochemistry, immunofluorescence, qPCR, and Western blotting in human samples, calcium oxalate mo

Cats and calcium oxalate: strategies for managing lower and upper tract stone disease.

PRACTICAL RELEVANCE: Calcium oxalate (CaOx) containing stones are among the most common of the urinary tract stones identified in cats. RISK FACTORS: Risk factors for CaOx stone formation include such things as breed, gender and diet; stress and obesity have also been hypothesized to be risk factors for this disease. MANAGEMENT APPROACH: A tailored, individual management strategy for preventing CaOx stone recurrence is important and should include addressing the diet, environment and any other comorbid conditions present. Increasing the cat's moisture intake is one of the key mechanisms for

Developmental aspects of calcium oxalate tubular deposits and calculi induced in rat kidneys.

We employed an experimental hyperoxaluric protocol involving the administration of 1 per cent ethylene glycol drinking water to rats to determine (i) the location and mechanism of retention of renal tubular calcium oxalate crystals, and (ii) how this tubular deposition is modified by magnesium deficiency and contributes to stone formation. Calcium oxalate monohydrate deposition was found predominantly within the lumina of proximal tubules, and was markedly accelerated by magnesium deficiency. Sheet- and strandlike mucoid appearing material attached the crystals to the luminal surface of the re

Measurement of precipitated calcium citrate in 24-hour urine samples is clinically useful for ruling out calcium oxalate urolithiasis.

Objective: Combining calcium with citrate clinically prevents the formation of calcium oxalate (CaOx) stones. In this study, we developed a new method for measuring calcium citrate (CalCit) precipitated out of urine samples. Method: CalCit in urine was formed by challenging with excessive calcium and precipitated out by ethanol. The precipitated CalCit (pCalCit) was redissolved and measured by an indicator displacement assay (IDA) using murexide as an indicator. This method is called the murexide-based IDA for measurement of precipitated CalCit (miDA-pCalCit). Results: Urea, creatinine, ammoni

Effect of biomolecules from human renal matrix of calcium oxalate monohydrate (CaOx) stones on in vitro calcium phosphate crystallization.

PURPOSE: Investigate the activity of high and low molecular weight biomolecules present in the matrix of human calcium oxalate (CaOx) stones not only on the initial mineral phase formation of calcium and phosphate (CaP) but also on its growth and demineralization of the preformed mineral phase. MATERIALS AND METHODS: Surgically removed renal stones were analyzed by Fourier Transform Infra Red (FTIR) spectroscopy and only CaOx stones were extracted with 0.05M EGTA, 1 mM PMSF and 1% Β-mercaptoethanol. Renal CaOx stone extract was separated into > 10 kDa and < 10 kDa fractions by dialysis. Activ

Apoptosis, ferroptosis, necrosis, necroptosis and pyroptosis in the formation of calcium oxalate kidney stones.

Kidney stones are one of the most common and debilitating urological disorders, putting substantial financial burden on healthcare services. Most common kidney stones are comprised of calcium oxalate often mixed with some calcium phosphate. Pathogenesis involves crystallization and retention of crystals within the kidneys, which is achieved either through the formation of crystalline plugs in the terminal collecting ducts blocking their openings into the renal pelvis, or formation of plaques of calcium phosphate on the renal papillary surface. The plugs are termed Randall's plugs and the plaqu

The potential role of alkaline diets in prevention of calcium oxalate kidney stone formation.

Formation of kidney stones is considered a major global problem. Diet plays an important role in the management of kidney stone formation. The main goal of the present research was to evaluate the protective role of fruit and vegetable mixtures as models of an alkaline diet on formation of kidney stones in rats and to conduct molecular docking study. The chemical compositions, phenolic compound profile, β-carotene content, vitamin C and antioxidant activity of both mixtures were assessed. Fruit (-42.419 ) and vegetable (-11.13) mixtures recorded a negative potential renal acid load in the pres

Endoscopic and histologic findings in a cohort of uric acid and calcium oxalate stone formers.

OBJECTIVE: To characterize the endoscopic and histologic renal papillary lesions in a cohort of uric acid (UA) stone formers (SF). METHODS: Data were prospectively obtained during percutaneous nephrolithotomy between 2009 and 2013. Renal papillae were endoscopically analyzed to quantitate surface area occupied by plaque or plug, and biopsies were obtained. UA SF were compared with non-SF controls and patients with >50% calcium oxalate (CaOx) in the absence of UA. RESULTS: There were 23 UA SF; of which 19 stones (83%) were admixed with CaOx and 4 (17%) were pure. Compared with CaOx SF and con

Macropinocytosis is the major mechanism for endocytosis of calcium oxalate crystals into renal tubular cells.

During an initial phase of kidney stone formation, the internalization of calcium oxalate (CaOx) crystals by renal tubular cells has been thought to occur via endocytosis. However, the precise mechanism of CaOx crystal endocytosis remained unclear. In the present study, MDCK renal tubular cells were pretreated with inhibitors specific to individual endocytic pathways, including nystatin (lipid raft/caveolae-mediated), cytochalasin D (actin-dependent or macropinocytosis), and chlorpromazine (CPZ; clathrin-mediated) before exposure to plain (non-labeled), or fluorescence-labeled CaOx monohydrate

Urinary response to an oxalic acid load is influenced by the timing of calcium loading in rats.

PURPOSE: Dietary intake of calcium or dairy products has been shown to decrease urinary oxalate excretion by limiting its intestinal absorption. However, not enough attention has been given to whether there is any benefit from altering the schedule of ingesting calcium and oxalate. Therefore, we investigated the effects of changes in the timing of calcium and oxalate loading on urinary oxalate excretion. MATERIALS AND METHODS: Male Wistar rats weighing 180 to 200 gm were fasted and randomized into several groups. They were then administered normal saline or oxalic acid with or without calcium

Calcium oxalate crystal formation in patients with hyperparathyroidism and hyperthyroidism and related metabolic disturbances.

The crystallization of calcium oxalate in the urine of patients with hyperparathyroidism and hyperthyroidism was studied using a mixed suspension mixed product removal (MSMPR) system. In addition, calcium metabolism in hyperthyroidism and its relationship to urolithiasis was investigated. The urines from all the three groups (normal subjects, hyperparathyroid and hyperthyroid patients) showed reduced nucleation rates and increased growth rates in comparison with the control synthetic urine. The nucleation rate was not significantly different between the three human urine groups, while the grow

Arthritis associated with calcium oxalate crystals in an anephric patient treated with peritoneal dialysis.

We report a case of calcium oxalate arthropathy in a woman undergoing intermittent peritoneal dialysis who was not receiving pharmacologic doses of ascorbic acid. She developed acute arthritis, with calcium oxalate crystals in Heberden's and Bouchard's nodes, a phenomenon previously described in gout. Intermittent peritoneal dialysis may be less efficient than hemodialysis in clearing oxalate, and physicians should now consider calcium oxalate-associated arthritis in patients undergoing peritoneal dialysis who are not receiving large doses of ascorbic acid.

RESTORATIVE EFFECT OF SALTS OF MAGNESIUM AND CALCIUM AFTER LETHAL DOSES OF SODIUM OXALATE.

The experiments presented in this paper brought out the following facts. Intramuscular injections of sodium oxalate into rabbits in doses of 0.18 and 0.2 gm. proved to be invariably fatal, death generally occurring in a comparatively short time. The symptoms consisted in excitation and tonic and clonic convulsions of diminishing strength if death was delayed. Some animals succumbed in the first convulsion. From the experiments in which magnesium or calcium was added, it was evident that massage of the site of the oxalate injection is unmistakably an aggravating factor. In the experiments with

Effects of calcium oxalate crystals on neutrophil cellular proteome and functions: implications for nephrolithiasis.

BACKGROUND: The majority of stone formers (87.5-95.9%) exhibit mild to moderate interstitial inflammation surrounding the stone. Neutrophils and neutrophil-derived genes/proteins have been found in renal papillae, stone matrix and urine of calcium oxalate monohydrate (COM) stone formers. However, neutrophil-crystal interactions, especially responses of neutrophils to COM crystals, remained unknown. METHODS: This study addressed the effects of COM crystals on neutrophil cellular proteome and functions, including phagocytosis, activation/degranulation, neutrophil extracellular traps (NETs) form
