Pleuritis caused by Campylobacter jejuni subspecies jejuni in a patient undergoing long-term hemodialysis.

A 73-year-old female hemodialysis patient experienced fever, shortness of breath on effort, and chest discomfort. A decrease in breath sounds in the right lung field, leukocytosis, elevated CRP level, and a right massive pleural effusion were observed. The patient was diagnosed with bacterial pleuritis based on leukocyte-predominant exudative pleural effusion, and treated with ceftriaxone. Her symptoms, however, were not improved, so thoracic drainage was attempted. Campylobacter species were isolated from cultured pleural fluid samples, and Campylobacter jejuni subspecies jejuni was detected

Campylobacter jejuni/Campylobacter coli-associated Guillain-Barré syndrome. Immunoblot confirmation of the serological response.

Immunoblot (Western blot) examination of the sera of 45 patients with the Guillain-Barré syndrome demonstrated that between three and five immunoreactive bands that were characteristic of a recent Campylobacter jejuni/Campylobacter coli infection were present in 22 of these patients. The immunoblots paralleled the serological response that is found in campylobacter enteritis and confirmed the specificity of our previous enzyme-linked immunosorbent assays (ELISAs), which indicates that antecedent C. jejuni/C. coli infection is common in patients with the Guillain-Barré syndrome. We postulate th

Unusual Campylobacter jejuni infection causing life- threatening cardiac tamponade.

The incidence of Campylobacter jejuni enterocolitis is increasing, with this bacterium being the leading cause of acute bacterial diarrhea. However, cardiac complications associated with C. jejuni remain rare. A 47-year-old man presented with sudden chest pain and shortness of breath. Echocardiography revealed pericardial tamponade and pleural effusion over time. Despite the patient having no prior gastrointestinal complaints, Campylobacter jejuni was identified in pericardial aspirates and stool samples by microbiological analysis. Exclusive pericarditis caused by C. jejuni remains an extreme

cj0371: A Novel Virulence-Associated Gene of Campylobacter jejuni.

Campylobacter jejuni is the major cause of human bacterial diarrhea worldwide. Its pathogenic mechanism remains poorly understood. cj0371 is a novel gene that was uncovered using immunoscreening. There have been no previous reports regarding its function. In this study, we constructed an insertion mutant and complement of this gene in C. jejuni and examined changes in virulence. We observed that the cj0371 mutant showed significantly increased invasion and colonization ability. We also investigated the role of cj0371 in motility, chemotaxis, and growth kinetics to further study its function. W

Selective biochemical labeling of Campylobacter jejuni cell-surface glycoconjugates.

The display of cell-surface glycolipids and glycoproteins is essential for the motility, adhesion and colonization of pathogenic bacteria such as Campylobacter jejuni. Recently, the cell-surface display of C. jejuni glycoconjugates has been the focus of considerable attention; however, our understanding of the roles that glycosylation plays in bacteria still pales in comparison with our understanding of mammalian glycosylation. One of the reasons for this is that carbohydrate metabolic labeling, a powerful tool for studying mammalian glycans, is difficult to establish in bacterial systems and

Phylogenetic relationships of Campylobacter jejuni based on porA sequences.

Campylobacter porins are the dominant major outer membrane protein (MOMP) of these bacteria. They are composed of hypervariable, surface-exposed, peptide loops and membrane-embedded, conserved peptide regions. Porins are functionally important and may also be useful for molecular subtyping methods but have not yet been well characterized. We therefore sequenced the porA gene from 39 Campylobacter isolates, including multilocus sequence type (MLST) reference strains, isolates from patients with the Guillain-Barré syndrome, other clinical isolates, and serotyping reference strains. These were co

Campylobacter jejuni colonization of mice with limited enteric flora.

We have developed experimental murine Campylobacter infection models which demonstrate efficient establishment and reproducible, high-level colonization. Following oral inoculation, wild-type C3H mice with normal enteric flora were colonized inconsistently and inefficiently by C. jejuni strain 81-176. However, C3H mice with a limited gut flora (LF) were efficiently colonized at high levels (10(8) CFU/g of stool or large intestine tissue) followed by clearance after several weeks. Large intestine tissue showed minimal to mild inflammation at days 7 and 28 postinoculation. In striking contrast,

Guillain-Barre syndrome with meningoencephalitis after Campylobacter jejuni infection.

A 14-year-old boy presented with progressive ascending muscle weakness, urinary retention and disturbed consciousness. Initially his cerebrospinal fluid showed pleocytosis, and protein-cellular dissociation developed later. Campylobacter jejuni was isolated from his stool and serum anti-ganglioside antibodies were positive. Our case suggests that coexistence of meningoencephalitis at an early stage of illness does not necessarily exclude the diagnosis of Guillain-Barre syndrome.

Effectiveness of chemical sanitizers against Campylobacter jejuni-containing biofilms.

Survival of Campylobacter jejuni in mixed-culture biofilms was determined after treatment with chemical sanitizers including chlorine, quaternary ammonia, peracetic acid (PAA), and a PAA/peroctanoic acid mixture (PAA/POA). Biofilm-producing bacteria (gram-positive rods, Y1 and W1) were isolated from chicken house nipple drinkers. A meat plant isolate (Pseudomonas sp.) was also included as a biofilm producer. Two-day-old biofilms grown on polyvinyl chloride (PVC) plastic coupons in R2A broth at 12 degrees C were incubated with 10(6) CFU/ml C jejuni for 6 h to allow attachment. The coupons were

Explosive Campylobacter jejuni diarrhea in immunoproliferative small intestinal disease.

Campylobacter jejuni is an infrequent cause of self limiting acute diarrheal disease in adults in the Indian subcontinent. We report the occurrence of a life threatening diarrhea due to C jejuni infection in a patient with immunoproliferative small intestinal disease. We postulate that immunosuppression due to malignancy, malnutrition and cancer chemotherapy was responsible for the unusually severe diarrhea.

Identification and characterization of Campylobacter jejuni outer membrane proteins.

Outer membrane proteins from isolates of Campylobacter jejuni were examined by sodium dodecyl sulfate-polyacrylamide gel electrophoresis. Sarcosinate-insoluble membrane preparations were outer membrane enriched based on increased ketodeoxyoctonate concentrations, the presence of surface-exposed 125I-labeled proteins that were hydrophobic, and similarity to membrane vesicle (bleb) sodium dodecyl sulfate-polyacrylamide gel electrophoresis profiles. Most isolates contained a single major band with molecular weight of 41,000 to 45,000. Profiles of C. jejuni and Campylobacter coli isolates were ind

Pathogenicity of Campylobacter jejuni isolates from animals and humans.

Fourteen isolates of Campylobacter jejuni of different serotypes and one Campylobacter coli isolate, from various human and animal sources, were tested for potential pathogenic mechanisms. Enterotoxin production was not detected in the infant mouse test or by calf and piglet ligated intestinal loop studies. Isolates were not invasive by the Sereny test. All isolates associated with and penetrated HeLa cells, although both actions occurred generally in a minor way under the conditions of our study. The C. coli isolate showed extensive HeLa cell association, but three other C. coli isolated test

Midtrimester abortion associated with septicaemia caused by Campylobacter jejuni.

The clinical record of a patient who suffered a mid-trimester missed abortion during an acute febrile illness is presented. Campylobacter jejuni was isolated from her blood cultures. Complete clinical recovery followed evacuation of the uterus and antibiotic therapy. Evidence of placental infection was found on histological examination. The possible role of campylobacter and other infections in fetal and perinatal death is discussed.

Serological studies in two outbreaks of Campylobacter jejuni infection.

Two outbreaks of campylobacter enteritis in residential student populations are described. In the first outbreak, 34 of 240 students in one hall of residence became ill over the course of 4 or 5 days. A single serotype of C. jejuni was isolated from sample of faeces. Sera collected demonstrated that the students had little pre-existing antibody and antibody responses were demonstrated in those who were infected. About 30% of students with serological evidence for infection with C. jejuni did not have gastro-intestinal symptoms. In the second outbreak, 77 of 300 agricultural students developed

Isolation of Campylobacter fetus subsp jejuni from zoo animals.

Over a 1-year period, 619 fecal specimens from animals at the Denver Zoo were cultured for Campylobacter fetus subsp jejuni. The organism was isolated from 35 animals, including 12 primates, 2 felids, a red panda, 13 hooved animals, 6 birds, and 1 reptile. Of 44 cultured fecal specimens from diarrheal animals, 31.8% were positive for Campylobacter, whereas only 5.6% of 575 specimens from animals without diarrhea were positive (P less than 0.001). Among 25 isolates tested, 12 serotypes were represented; several of these serotypes are commonly associated with Campylobacter enteritis in human bei

Comparison of basal broth media for the optimal laboratory recovery of Campylobacter jejuni and Campylobacter coli.

BACKGROUND: Campylobacter species are the most common known bacterial cause of acute diarrhoeal disease and incidences are increasing yearly in Ireland. Concern is expressed that this increase may be due to poor laboratory techniques in recognising these organisms as they are fastidious and technically demanding to culture. AIMS: To evaluate and compare the efficacy of seven commercially available basal broth media for the laboratory cultivation of thermophilic C. jejuni and C. coli isolated from foods and clinical specimens, as well as for optimal laboratory passage. METHODS: Four strains o

Production of monoclonal antibody specific to Campylobacter jejuni and its potential in diagnosis of Campylobacter enteritis.

A monoclonal antibody (MAb 3G6) specific for Campylobacter jejuni and Campylobacter coli was produced by immunizing BALB/c mice with a local strain of C.jejuni (28.1). No cross-reactivity was observed with Enterobacteriaceae controls. By immunoprecipitation, MAb 3G6 identified a major protein band of molecular weight 45 kDa and also gave a slight reactivity with 30 and 55 kDa proteins. Using an indirect enzyme-linked immunosorbent assay, MAb 3G6 was able to detect C.jejuni suspended in stool without cross-reactivity to 14 other enteropathogenic bacteria suspended, normal flora in fecal suspens

Evolution and Role of Proteases in Campylobacter jejuni Lifestyle and Pathogenesis.

Infection with the main human food-borne pathogen Campylobacter jejuni causes campylobacteriosis that accounts for a substantial percentage of gastrointestinal infections. The disease usually manifests as diarrhea that lasts for up to two weeks. C. jejuni possesses an array of peptidases and proteases that are critical for its lifestyle and pathogenesis. These include serine proteases Cj1365c, Cj0511 and HtrA; AAA+ group proteases ClpP, Lon and FtsH; and zinc-dependent protease PqqE, proline aminopeptidase PepP, oligopeptidase PepF and peptidase C26. Here, we review the numerous critical roles
