BACKGROUND: Telomere dysfunction has been reported to be directly involved in carcinogenesis owing to chromosomal instability and immortalization; however, the clinicopathological significance of telomeres remains controversial. We have shown that telomere shortening occurs in normal-appearing duct cells at initiation and then continues during the progression of pancreatic cancer. In this study, we determined the clinicopathological and prognostic value of telomere length (TL) in cancer progression.
METHODS: TL in both cancer cells and cancer-associated fibroblasts (CAFs) was analyzed by high

PURPOSE: The role of angiopoietins (ANGs) in cancer progression is a new field of research. We examined the patterns of expression of ANGs in human renal cancer tissues (n =12) and investigated their roles in cancer progression in vitro and in vivo.
METHODS AND RESULTS: In normal renal tissue, the expression of ANG-1 was apparent in the glomerular capillaries and podocytes as well as in the endothelial cells of vessels, whereas we observed no expression of ANG-2. In contrast, in tumor tissue, dense, diffuse expression of ANG-1 was apparent in the fibroblasts, but not in the cancer cells. Inte

Immune checkpoint inhibitors (ICIs) have achieved breakthroughs in various solid tumors; however, their efficacy remains limited in prostate cancer (PCa), which is closely associated with its highly immunosuppressive tumor microenvironment (TME). As the most abundant stromal component in the TME, cancer-associated fibroblasts (CAFs) exhibit remarkable heterogeneity and functional plasticity, playing a central role in orchestrating an immunologically "cold" tumor phenotype and driving resistance to immunotherapy. We have moved beyond simple marker-based classification of cancer-associated fibro

Despite extensive research, the 5-year survival rate of pancreatic cancer (PDAC) patients remains at only 9%. Patients often show poor treatment response, due partly to a highly complex tumor microenvironment (TME). Cancer-associated fibroblast (CAF) heterogeneity is characteristic of the pancreatic TME, where several CAF subpopulations have been identified, such as myofibroblastic CAFs (myCAFs), inflammatory CAFs (iCAFs), and antigen presenting CAFs (apCAFs). In PDAC, cancer cells continuously adapt their metabolism (metabolic switch) to environmental changes in pH, oxygenation, and nutrient

Complex interactions take place during cancer formation and progression. In this regard, there has been increasing focus on the non-malignant cells that make up the tumour microenvironment (TME), and how they interact with malignant tumour cells. TME is highly heterogeneous and has a major influence on tumour behaviour and therapy response. Cancer-associated fibroblasts (CAFs), one of the main components of the TME, establish dangerous liaisons with cancer cells and other components of the TME to shape a tumour-supportive environment in many types of cancer. Head and neck squamous cell carcino

Hypoxia is pervasive within the solid tumor microenvironment (TME), reshaping it through exosome release. As the main component of the tumor stroma, fibroblasts influence TME remodeling and tumor progression. Recent advances in targeting tumor-derived exosomes offer promising opportunities for innovative colorectal cancer (CRC) therapies. Here, we found that exosomes induced by hypoxia in CRC cells (H-Exo) can promote the activation of normal tissue-associated fibroblasts (NAFs) into cancer-associated fibroblasts (CAFs) phenotype, with a stronger effect compared to normoxic exosomes (N-Exo). UNLABELLED: The use of immunotherapy in solid tumors has significantly improved the outcomes and quality of life for patients with cancer. However, a substantial fraction of patients remains unresponsive, highlighting the urgent need to identify mechanisms of resistance to enhance immune-centric, chemo-free treatment regimens. In this study, we investigated associations with immunotherapy resistance by integrating RNA sequencing data from 2,800 patients with bladder cancer enrolled in the IMvigor trials, focusing on resistance to the anti-PD-L1 therapy atezolizumab, along with single-cell data

BACKGROUND: Cancer-associated fibroblasts (CAFs) drive breast cancer (BC) progression. While N6-methyladenosine (m6A) regulates tumorigenesis, the mechanisms by which CAF-derived exosomes transmit oncogenic m6A signals remain unclear.
MATERIALS AND METHODS: Primary CAFs and normal fibroblasts were isolated and exosomes purified via ultracentrifugation. Exosomal impact on BC cell malignancy and T-cell cytotoxicity was evaluated in vitro. Molecular targets were validated using methylated RNA immunoprecipitation and dual-luciferase assays. In vivo tumorigenesis was assessed using xenograft model

UNLABELLED: Perineural invasion (PNI) is a pivotal prognostic factor in pancreatic cancer, associated with aggressive tumor behavior and adverse patient outcomes. The recognized clinical impact of PNI highlights the need to better understand the molecular mechanisms underlying PNI-induced phenotypes. In this study, we isolated PNI-associated cancer-associated fibroblasts (pCAF), which demonstrated a markedly enhanced capacity to promote neural invasion in pancreatic cancer compared with non-PNI-associated CAFs. Single-cell, high-throughput sequencing and metabolomics data showed a significant

BACKGROUND: Cancer-associated fibroblasts (CAFs) are recruited to the tumor microenvironment (TME) and are critical drivers of breast cancer (BC) malignancy. Circulating tumor cells (CTCs) travel through hematogenous routes to establish metastases. CTCs circulate both individually and, more rarely, in clusters with other cell types. Clusters of CTCs have higher metastatic potential than single CTCs. Previously, we identified circulating CAFs (cCAFs) in patients with BC and found that while healthy donors had no CTCs or cCAFs, both were present in most Stage IV patients. cCAFs circulate individ

BACKGROUND: Lung cancer remains the leading cause of cancer-related mortality worldwide, and therapeutic outcomes are frequently compromised by drug resistance and the complex tumor microenvironment. Although malignant progression is fundamentally driven by genetic alterations, cancer-associated fibroblasts (CAFs) have emerged as critical regulators of tumor progression, immune evasion, and therapeutic resistance.
MAIN BODY: This review systematically summarizes the multidimensional roles of CAFs in lung cancer pathogenesis. CAFs remodel the extracellular matrix to generate high-stiffness str

Immune checkpoint blockade (ICB) therapy has demonstrated promising efficacy in patients with mismatch repair-deficient (dMMR) or microsatellite instability-high (MSI-H) colorectal cancer (CRC). However, a substantial proportion of these patients remain resistant to treatment. This study aims to investigate the mechanisms underlying ICB resistance in dMMR/MSI-H CRCs. We analyzed single-cell transcriptomic data from 34 CRC patients treated with ICB and integrated ten spatial transcriptomic datasets to compare the tumor microenvironment (TME) features between responders and non-responders. Our a

Cancer-associated fibroblasts (CAFs) are a major component of the tumor microenvironment and represent a potential therapeutic target for fibrotic breast cancer. Myofibroblastic CAFs (myCAFs), the major subtype, form a tumor barrier by generating a dense extracellular matrix (ECM) that greatly limits the penetration of immune cells and promotes an immunosuppressive microenvironment. Reprogramming the immunosuppressed myCAFs into the immune activated antigen presenting CAF (apCAFs), rather than directly eliminating CAFs, is a better treatment modality. We developed a spatiotemporally ultrasound

Transgelin is an actin-binding protein that promotes cancer progression via activation of cancer-associated fibroblasts and has been identified as a prognostic marker. However, its distribution and functional role in colon cancer remain unclear. In this study, we aimed to elucidate the mechanistic role of transgelin in colon cancer progression by focusing on its functional impact in cancer-associated fibroblasts. Tissue microarrays from 359 human colon cancer tissues were investigated to elucidate the clinical importance of transgelin expression in cancer stroma. We focused on transgelin in fi

Chemoresistance remains a critical challenge in breast cancer (BC) treatment. By integrating multi-omics (single-cell, spatial, and bulk transcriptomics) with clinical validation, we identified a specific COL3Ahigh CAF subset that drives BC chemoresistance. Mechanistically, these CAFs undergo lipid metabolic reprogramming, secreting excess oleic acid via SCD. This oleic acid binds to ENO1 on tumor cells, activating the PI3K/Akt pathway and inhibiting chemotherapy-induced apoptosis. Simultaneously, COL3Ahigh CAFs orchestrate an immunosuppressive niche by recruiting regulatory T cells and impair

Lactate in the tumor microenvironment (TME) is typically generated by cells exhibiting high glycolytic flux, exemplified by tumor cells. However, in glycolysis-low malignancies such as prostate cancer, stroma-derived lactate may drive noncanonical signaling and functions that remain unclear. Here, we identified APCDD1+ cancer-associated fibroblasts (CAFs) as a distinct stromal population that secretes lactate into the TME in response to androgen deprivation therapy (ADT). Lactate uptake by prostate cancer cells induces androgen receptor variant 7 expression, thereby conferring resistance to AD

Prostate cancer (PCa) is the second most common malignancy in men worldwide. Advanced stages are characterized by tumor heterogeneity, metastasis, and resistance to androgen deprivation therapy and chemotherapy. Cancer-associated fibroblasts (CAFs), the predominant stromal cells in the PCa tumor microenvironment (TME), critically drive tumor progression, metastasis, and therapeutic resistance. Nanomedicine represents a transformative strategy for targeting CAFs. It leverages engineered nanomaterials to achieve precise drug delivery, improved bioavailability, and multimodal theranostic capabili

Pancreatic ductal adenocarcinomas (PDACs) are aggressive, stroma-rich tumours. They are unresponsive to treatments, and patients relapse quickly on DNA-damaging chemotherapies. PDAC stroma consists of extracellular matrix proteins (ECM), secreted by cancer-associated fibroblasts (CAFs). Here we show an unexpected link between CAF-secreted ECM proteins and enhanced DNA repair. We identify NDRG1 (N-myc downstream-regulated gene 1) as a key mediator that senses signals from the ECM via adhesion receptors and serum and glucocorticoid-activated kinase. We establish NDRG1 as a DNA repair factor that

Pancreatic ductal adenocarcinoma (PDAC) is a highly lethal malignancy that urgently needs more effective therapies. Cancer-associated fibroblasts (CAFs) contribute to the aggressive and chemo-resistant nature of the disease by creating a drug-impeding fibrotic microenvironment. We developed novel compounds, the racemate CRO-05 and its active enantiomer CRO-67, which target both pancreatic tumor and CAF cells with robust anti-cancer activity. These compounds were designed using rational medicinal chemistry based on chromans, a class of anti-cancer drugs. Their therapeutic potential and efficacy
