Unveiling the molecular architecture of Candida auris ribosome.

Candida auris is an emerging multidrug-resistant fungal pathogen responsible for severe healthcare-associated infections worldwide; yet, the molecular architecture of its translational apparatus remains poorly understood. Here, we present a structural and functional analysis of the C. auris ribosome. Using in-house rRNA sequencing, we found that C. auris rRNA displays substantial divergence, including the lowest 18S rRNA identity among the pathogenic Candida species examined and an unusual substitution at a functionally important nucleotide in helix h31 of the small subunit P-site region. To d

Candida auris in the neonatal intensive care unit.

Candida (a.k.a.Candidozyma) auris is a healthcare-associated yeast that has attracted attention due to its increased antimicrobial resistance and ability to spread rapidly within the healthcare environment. Infants admitted to the neonatal intensive care unit (NICU) have risk factors such as broad spectrum antibiotic exposure, prematurity and congenital heart disease that predispose them to serious infection with C. auris. Consequently, there have been global reports of C. auris outbreaks in NICUs associated with high mortality. The clinical presentation of late onset sepsis with C. auris is i

Value of qPCR in the screening strategy for Candida auris.

Candida auris has emerged as a major fungal pathogen responsible for persistent outbreaks, highlighting the importance of an effective screening strategy to control its spread in healthcare settings. Following a confirmed C. auris infection, we incorporated a specific qPCR assay into the screening process. Contact patients and environmental samples were screened weekly using culture and qPCR methods. We used these results to manage patients' cohorting, room disinfection, and new admissions in real time. The qPCR allowed us to identify 37 colonized patients and detect 47 contaminated environmen

Candida auris is emerging as a prevalent urinary pathogen.

Urinary tract infections (UTIs) are one of the most common infections, with a subgroup of these infections, catheter-associated UTIs (CAUTIs), accounting for 40% of nosocomial infections. While the majority of CAUTI pathogens are bacterial, the second most common pathogen is the fungus Candida albicans. However, in recent years, Candida auris has increasingly been isolated from urine, indicating C. auris' potential as a urinary pathogen. C. auris has rapidly emerged as a human pathogen worldwide, becoming a serious health threat. This is of great concern due to its antifungal resistance, adher

Antagonistic effect of Pseudomonas aeruginosa on Candida auris.

INTRODUCTION: Candida (Candidozyma) auris and Pseudomonas aeruginosa are frequently found in hospital environments and on medical equipment, where they commonly colonize and infect hospitalized patients, contributing to healthcare-associated infections (HAIs). Although they share similar ecological niches and may interact, the mechanisms underlying their interspecies communication remain largely unknown. METHODS: This study investigated the in vitro interaction between planktonic cells of C. auris and P. aeruginosa through co-culture experiments in various growth media, with or without iron s

Is routine screening for Candida auris necessary in ICU?

BACKGROUND AND OBJECTIVES: The capability to cause invasive infection, multi-drug resistance, and health care-associated outbreaks of Candida auris have made it a pathogen of great concern. Estimating how many patients in our intensive care unit had C. auris colonization and what characteristics put patients at risk for having Candida spp. colonization were the primary goals of the study. MATERIALS AND METHODS: Swabs from axilla and groin were collected from 229 patients getting admitted to the ICU. Samples were inoculated into CHROMagarTM Candida Plus medium. Colonies presumptively identifie

Candida auris in Dutch hospitals: are we ready for it?

BACKGROUND: Candida auris can cause nosocomial outbreaks and provides challenges concerning diagnosis, treatment, eradication and infection prevention. There are no Dutch standards or guidelines for C. auris, and current hospital practices are unknown. Therefore, we assessed whether Dutch hospitals are prepared for C. auris introduction. METHODS: An online questionnaire concerning screening, diagnostics, infection prevention and outbreaks was distributed amongst medical microbiologists and infection prevention practitioners in spring 2024. FINDINGS: Fifty-two questionnaires were processed co

Global characteristics and trends in research on Candida auris.

INTRODUCTION: Candida auris, a fungal pathogen first reported in 2009, has shown strong resistance to azole antifungal drugs and has caused severe nosocomial outbreaks. It can also form biofilms, which can colonize patients' skin and transmit to others. Despite numerous reports of C. auris isolation in various countries, many studies have reported contradictory results. METHOD: A bibliometric analysis was conducted using VOSviewer to summarize research trends and provide guidance for future research on controlling C. auris infection. The analysis revealed that the United States and the US CDC

Candida auris on Apples: Diversity and Clinical Significance.

Candida auris is a multidrug-resistant nosocomial fungal pathogen. While the marine environment was recently identified as a natural niche for C. auris, the environment(s) that might have contributed to the development and spread of antifungal resistance in C. auris remains a mystery. Because stored fruits are often treated with fungicides to prevent postharvest spoilage, we hypothesized that stored fruits could serve as a possible selective force for and a transmission reservoir of antifungal-resistant isolates of pathogenic yeasts, including C. auris. To test this hypothesis, we screened fru

Bioinformatic Identification of ABC Transporters in Candida auris.

Antifungal resistance mediated by overexpression of ABC transporters is one of the primary roadblocks to effective therapy against Candida infections. Thus, identification and characterization of the ABC transporter repertoire in Candida species are of high relevance. The method described in the chapter is based on our previously developed bioinformatic pipeline for identification of ABC proteins in Candida species. The methodology essentially involves the utilization of a hidden Markov model (HMM) profile of the nucleotide-binding domain (NBD) of ABC proteins to mine these proteins from the p

The first invasive Candida auris infection in Taiwan.

Candida auris, a multidrug resistant pathogenic yeast, has spread worldwide and caused several outbreaks in healthcare settings. Here, we report the first case of C. auris candidemia in Taiwan in a patient with a two-month history of hospitalization in Vietnam. We performed further investigation on the isolate from the present case as well as the previously reported C. auris isolate identified from a wound in 2018 in Taiwan, which was the first case reported in Taiwan. Both C. auris isolates were found to be susceptible to fluconazole, amphotericin B, and echinocandins. Additionally, mutations

[Candida auris and Mechanisms of Antifungal Drug Resistance].

Candida auris has been isolated from clinical samples in different regions and countries since it was first described in 2009. Due to the difficulties in identification; decreased susceptibility or resistance to antifungal agents; exceptional capacity to colonize and persist on surfaces; ability to survive despite standard disinfection procedures; and significant increase in the number of regions and countries with reported cases, C.auris has become a global health concern and placed among the World's ten most concerned fungi list in 2018. It is stated that 60-90% of C.auris strains are resist

Candida auris-the growing menace to global health.

A paradigm shift of candidiasis from Candida albicans to non-albicans Candida species has fundamentally increased with the advent of C. auris. C. auris, despite being a newly emerged multidrug-resistant fungal pathogen, is associated with severe invasive infections and outbreaks with high mortality rates. Initially reported from Japan in 2009, C. auris have now been found in different countries on all the continents except Antarctica. Due to its capability of nosocomial transmission and forming adherent biofilms on clinically important substrates, a high number of related hospital outbreaks ha

Candida auris: Disinfectants and Implications for Infection Control.

Candida auris is a rapidly emerging pathogen and is able to cause severe infections with high mortality rates. It is frequently misidentified in most clinical laboratories, thus requiring more specialized identification techniques. Furthermore, several clinical isolates have been found to be multidrug resistant and there is evidence of nosocomial transmission in outbreak fashion. Appropriate infection control measures will play a major role in controlling the management and spread of this pathogen. Unfortunately, there are very few data available on the effectiveness of disinfectants against C

Fatty Acid Profiling Identification Method of Emerging Fungal Pathogen Candidozyma auris (Formally Candida auris).

The species Candidozyma auris (formerly known as Candida auris) can be subdivided into four major and two minor clades. It is considered an emerging multidrug-resistant pathogen that causes invasive outbreaks around the world. Therefore, the accurate identification of this species plays an important role in combating invasion and facilitating pathogenic management. In our study an optional identification method was developed considering the possibility of using cellular fatty acids (FAs) as a taxonomic and diagnostic tool. FAs were recorded in the collected C. auris strains, and the species ch
