NLRP3 inflammasome activation has been implicated in the pathogenesis of human disorders, including a number of neurodegenerative diseases of the central nervous system. Pharmacological inhibition of NLRP3-mediated neuroinflammation via a brain-penetrant therapeutic agent, therefore, is an attractive target for the treatment of neurodegenerative disorders. The NLRP3 inflammasome inhibitor NT-0796 is an ester-containing prodrug that is metabolized intracellularly into its active metabolite NDT-19795 by carboxylesterase-1 (CES1). NT-0796 shows high in vitro permeability in an induced pluripotent

Human carboxylesterase 1 (hCE1) is a serine hydrolase that is distributed widely in the human liver, intestines, and various tumour tissues. It has now become a biomarker associated with diseases such as cancer. Consequently, the measurement of hCE1 levels in human serum is of significant importance for the diagnosis and assessment of these diseases. The present study involved the preparation of Ag/CQDs/ZIF-8 nanocomposites, which incorporated silver nanoparticles (Ag NPs) and carbon quantum dots (CQDs) into the ZIF-8 framework. The combination of this composite with an enzyme-linked immunosor

Carboxylesterases (CarEs) are detoxification enzymes that play important roles in xenobiotic metabolism and host adaptation in the polyphagous spider mite Tetranychus truncatus. However, the specific carboxylesterase proteins involved in host plant detoxification in Tetranychus truncatus remain poorly characterized. In this study, we investigated the activities of glutathione S-transferase (GST), carboxylesterase (CarE), and cytochrome P450 (CYP450) in T. truncatus reared on three host plants-common bean, cucumber, and jujube. CarE activity was significantly higher in mites fed on jujube, whil

Carboxylesterase 2 (CES2) is a clinically significant serine hydrolase that governs the metabolic activation and detoxification of numerous ester-containing drugs and prodrugs. Despite its pivotal role in liver and colorectal cancer progression, direct in situ imaging of CES2 activity remains challenging due to the scarcity of probes with high isoform selectivity. To address this, we herein present XM-CE, a rationally engineered near-infrared (NIR) fluorescent probe specifically optimized for the highly sensitive and selective monitoring of CES2 activity. By integrating a CES2-responsive ester

Traditional tumor treatment faces great challenge owning to inherent drawbacks. Activatable prodrugs with multi-modality therapeutic capacity are highly desired. In this consideration, a responsiveness-released multi-in-one nanoplatform, PLGA-PEG@HC, toward cervical cancer therapy was innovatively developed. Among the nanoplatform, HC was constructed by incorporating chlorambucil, a classic chemotherapy drug into a near-infrared photo- and sono-sensitizer, HCH via ester linker, which can be specifically hydrolyzed by carboxylesterase (CES). HC is scarcely fluorescent and toxic due to the cagin

Human carboxylesterase 2 (hCES2A), one of the most important serine hydrolases distributed in the small intestine and colon, plays a crucial role in the hydrolysis of various prodrugs and esters. Accumulating evidence has demonstrated that the inhibition of hCES2A effectively alleviate the side effects induced by some hCES2A-substrate drugs, including delayed diarrhea caused by the anticancer drug irinotecan. Nonetheless, there is a scarcity of selective and effective inhibitors that are suitable for irinotecan-induced delayed diarrhea. Following screening of the in-house library, the lead com

Death and toxicity after cocaine use do not correlate with cocaine blood levels. One explanation for this observation is that cocaine abusers may posses one or more of the 58 possible known mutations in the butyrylcholinesterase gene (BCHE). Butyrylcholinesterase (BChE) serves as the primary cocaine hydrolase producing a nontoxic product ecgonine methyl ester. A reduction in endogenous levels of BChE may result in increased metabolism by hepatic carboxylesterase to produce norcocaine, a toxic product. Humans have carboxylesterase in tissues but not in plasma, whereas wild-type mice have signif

Non-specific carboxylesterases (carboxylesterases) and glutathione S-transferases (GSTs) are two groups of drug metabolizing enzymes responsible for hydrolysis and glutathione conjugation of xenobiotics. This study was conducted to determine the following: (1) the distribution of carboxylesterase and GST activities in different rat liver cells, (2) the effects of vitamin A deficiency (A-) on the absolute activities and on the distribution of carboxylesterases and GSTs in rat liver. Rat livers were fractionated into parenchymal and non-parenchymal cells by means of collagenase perfusion and dif

Carboxylesterases are enzymes that hydrolyze a broad suite of endogenous and exogenous ester-containing compounds to the corresponding alcohol and carboxylic acid. These enzymes metabolize a number of therapeutics including the anti-tumor agent CPT-11, the anti-viral drug oseltamivir, and the anti-thrombogenic agent clopidogrel as well as many agrochemicals. In addition, carboxylesterases are involved in lipid homeostasis, including cholesterol metabolism and transport with a proposed role in the development of atherosclerosis. Several different scaffolds capable of inhibiting carboxylesterase

An abridged carboxylesterase E4 (CbE E4) gene was cloned from the peach-potato aphid, Myzus persicae, by reverse transcription-PCR and subcloned into the expression vector pET28b. The abridged CbE E4 gene was successfully expressed in E. coli BL21 (DE3). The recombinant CbE E4 hydrolyzed beta-naphthyl acetate and Carbaryl by 64% within 2.5 h, Malathion by 80% within 1.25 h. However, the hydrolysis of other pesticides (Dichlorovos, Parathion, Pirimicarb and Deltamethrin) was not detected.

Previous studies have demonstrated that patients with halothane-induced hepatitis have serum antibodies that are directed against novel liver microsomal neoantigens and have suggested that these neoantigens may play an immunopathological role in development of the patients' liver damage. These investigations have further revealed that the antibodies are directed against distinct polypeptide fractions (100 kDa, 76 kDa, 59 kDa, 57 kDa, 54 kDa) that have been covalently modified by the reactive trifluoroacetyl halide metabolite of halothane. In this paper, the trifluoroacetylated (TFA) 59-kDa neo

BACKGROUND: Deoxynivalenol (DON) is among the most prevalent mycotoxins and exerts adverse effects on the liver. However, a systematic evaluation of its effects on hepatic cytochrome P450 enzymes (Cyp450) and carboxylesterases (Ces) in mice is lacking.
METHODS: Using combined in vivo and in vitro models, we investigated the impact of DON on these key enzymes. C57BL/6 mice and AML12 cells were exposed to DON. Histological and ultrastructural changes were assessed by hematoxylin and eosin staining and transmission electron microscopy. The expression and activity of target enzymes were evaluated

Insects have evolved an extremely sensitive olfactory system that is essential for a series of physiological and behavioral activities. Some carboxylesterases (CCEs) comprise a major subfamily of odorant-degrading enzymes (ODEs) playing a crucial role in odorant signal inactivation to maintain the odorant receptor sensitivity. In this study, 93 CCEs were annotated in the genome of the German cockroach Blattella germanica, a serious urban pest. Phylogenetic and digital tissue expression pattern analyses identified two antenna-enriched CCEs, BgerCCE021e3 and BgerCCE021d1, as candidate ODEs. RNA

Ulcerative colitis (UC) is associated with increased levels of inflammatory factors, which is attributed to the abnormal expression and activity of enzymes and transporters in the liver, affecting drug disposition in vivo. This study aimed to examine the impact of intestinal inflammation on the expression of hepatic carboxylesterases (CESs) in a mouse model of dextran sulfate sodium (DSS)-induced colitis. Two major CESs isoforms, CES1 and CES2, were down-regulated, accompanied by decreases in hepatic microsomal metabolism of clopidogrel and irinotecan. Meanwhile, IL-6 levels significantly incr

Flestolol, N(1,1-dimethyl-2-ureidocthyl)-2-hydroxy-3-(o-fluorobenzoyloxy++ +) propylamine, (F), is an ester containing an ultra short-acting beta blocker intended for the treatment of myocardial dysfunctions. In vitro incubation of F, procaine, chloroprocaine, and atropine with blood from different New Zealand White (NZW) rabbits resulted in a bimodal distribution (70% fast, 30% slow) of ester hydrolysis rates. Using F as a model substrate, bimodal hydrolysis rates were also observed in NZW rabbit cornea but not aqueous humor, iris-ciliary body complex and ocular tissues of pigmented rabbits.

ETHNOPHARMACOLOGICAL RELEVANCE: Metabolic dysfunction-associated steatotic liver disease (MASLD) is the most prevalent chronic liver disease globally, yet effective therapeutic options remain limited. Dysregulated lipid metabolism plays an important role in MASLD progression, making it a promising target for intervention. In Malaysia, the resin extract of Agathis dammara (Lamb.) Rich. & A.Rich. (AD) is traditionally used as a health supplement to regulate metabolism and treat inflammatory diseases. Araucarone (AO) is the most abundant monomeric component in AD. However, the therapeutic efficac

Carboxylesterase 2 (CES2) is expressed mainly in liver and intestine, but most abundantly in intestine. It hydrolyzes carboxylester, thioester, and amide bonds in many exogenous and endogenous compounds, including lipids. CES2 therefore not only plays an important role in the metabolism of many (pro-)drugs, toxins and pesticides, directly influencing pharmacology and toxicology in humans, but it is also involved in energy homeostasis, affecting lipid and glucose metabolism. In this study we investigated the pharmacological and physiological functions of CES2. We constructed Ces2 cluster knocko

Human carboxylesterase-2 (CES2) and cytochrome P450 3A4 (CYP3A4) are two major drug metabolizing enzymes that play critical roles in hydrolytic and oxidative biotransformation, respectively. They share substrates but may have opposite effect on therapeutic potential such as the metabolism of the anticancer prodrug irinotecan. Both CES2 and CYP3A4 are expressed in the liver and the gastrointestinal tract. This study was conducted to determine whether CES2 and CYP3A4 are expressed under developmental regulation and whether the regulation occurs differentially between the liver and duodenum. A la

Temephos is a major organophosphate (OP) larvicide that has been used extensively for the control of Aedes albopictus and Aedes aegypti, the major vectors for viral diseases, such as dengue fever, zika and chikungunya. Resistance to temephos has been recently detected and associated with the upregulation of carboxylesterases (CCEs) through gene amplification, in both species. Here, we expressed the CCEae3a genes which showed the most striking up-regulation in resistant Aedes strains, using the baculovirus system. All CCEae3a variants encoded functional enzymes, with high activity and preferenc
