Multilocus heterozygosity and coronary heart disease: nested case-control studies in men and women.

BACKGROUND: Generalized allelic heterozygosity has been proposed to improve reproductive fitness and has been associated with higher blood pressure, but its association with chronic disease is not well characterized. METHODS: Using the Affymetrix Genome-Wide Human 6.0 array, we performed whole genome scans in parallel case-control studies of coronary heart disease (CHD) nested in the Health Professionals Follow-up Study and Nurses' Health Study. We examined ~700,000 single nucleotide polymorphisms (SNPs) in 435 men with incident CHD and 878 matched controls and 435 women with incident CHD wit

Case-control studies of novel hemoglobin anomalies as differential diagnosis in sleep apnea syndrome.

PURPOSE: Pulse oximetry plays an essential role in the diagnosis of sleep apnea syndrome. We discovered two novel hemoglobin anomalies, Hb Bonn and Hb Venusberg, which initially resulted in avoidable sleep disorder examinations and therapeutic consequences due to their low oxygen saturation levels as measured by pulse oximetry. METHODS: Hematological as well as clinical chemical diagnosis was carried out. Hemoglobin anomalies were detected through electrophoresis, chromatography, spectrophotometry, and pulse oximetry as well as hemoglobin gene sequencing. RESULTS: Hb Bonn is a novel hemoglob

Including known covariates can reduce power to detect genetic effects in case-control studies.

Genome-wide association studies (GWAS) search for associations between genetic variants and disease status, typically via logistic regression. Often there are covariates, such as sex or well-established major genetic factors, that are known to affect disease susceptibility and are independent of tested genotypes at the population level. We show theoretically and with data from recent GWAS on multiple sclerosis, psoriasis and ankylosing spondylitis that inclusion of known covariates can substantially reduce power for the identification of associated variants when the disease prevalence is lower

Attributable risk in men in two French case-control studies on mesothelioma and asbestos.

Pleural mesothelioma is a primary tumor of the pleura that is mainly due to asbestos exposure. To study the relationship between mesothelioma and occupational asbestos exposure in France, two case-control studies (A and B) were conducted. A substantial difference in the attributable risk in the population (AR(p)) was observed among men: 44.5% (95% CI: [32.6-56.4]) in study A and 83.2% (95% CI: [76.8-89.6]) in study B. As different exposure assessment expert methods were used, the main objective of this work was to re-estimate the AR(p) men in two case-control studies according to a common stan

Comparing strategies for evaluation of candidate genes in case-control studies using family data.

The goal of this analysis is to compare different test strategies for genetic association in case-control studies using related individuals. The first test is the trend test that is corrected for related individuals on the basis of identity-by-descent information. The second approach is to use generalized estimating equations to adjust for the correlation between relatives, and the third is the multiple outputation method. We compare the power of these test strategies in a simulation study, and apply these methods to a candidate gene dataset of Genetic Analysis Workshop 15 from the North Ameri

Human leukocyte antigens and susceptibility to tuberculosis: a meta-analysis of case-control studies.

OBJECTIVE: To evaluate the association between human leukocyte antigens (HLA) of the major histocompatibility complex (MHC) and thoracic tuberculosis (TB) in immunocompetent adults. DESIGN: We searched Medline, Pascal, Pascal Biomed and Francis databases (all years) with the terms 'tuberculosis' and 'HLA'. Case-control studies were included that reported frequencies for the full range of antigens analysed by serological methods in healthy controls and adult patients not treated with glucocorticoids or immunosuppressive drugs, human immunodeficiency virus status negative or not reported, no de

Testicular cancer and cryptorchidism in relation to prenatal factors: case-control studies in Denmark.

To explore prenatal risk factors that are common to testicular cancer and cryptorchidism, two parallel case-control studies were conducted in Denmark. Information about characteristics of the mother, the pregnancy, and the birth were obtained from the mothers of cases and controls, using a mailed self-administered questionnaire. A maternal age above 30 years was associated with odds ratios (OR) of 1.9 (95 percent confidence interval [CI] = 1.2-3.0) for cryptorchidism and 2.0 (CI = 1.2-3.6) for testicular seminoma; the latter effect was particularly high when the boy was the first child of the

Logistic regression in case-control studies: the effect of using independent as dependent variables.

In case-control studies, cases are sampled separately from controls. In such studies the primary analysis concerns the estimation of the effect of covariables on being a case or a control. To explore causal pathways, further secondary analysis could concern the relationships among the covariables. In this paper the validity of such secondary analysis is addressed. In particular, the use of multiple logistic regression in case-control studies where the dependent variable is not the case/control indicator is explored. It is shown that only under very restrictive conditions will sample regression

A Likelihood Approach to Proper Analysis of Secondary Outcomes in Matched Case-Control Studies.

Matched case-control studies are commonly employed in epidemiological research for their convenience and efficiency. Analysis of secondary outcomes can yield valuable insights into biological pathways and help identify genetic variants of importance. Naive analysis using standard statistical methods, such as least-squares regression for quantitative traits, can be misleading because they fail to account for unequal sampling induced by the case-control design and matching. In this paper, we propose novel statistical methods that appropriately reflect the study design and sampling scheme in the

Industrial point source emissions and incident breast and lung cancers: two case-control studies.

PURPOSE: Better understanding of the environment's role in breast cancer (BC) and lung cancer (LC) incidence is crucial for population health. Growing evidence suggests environmental factors may be relevant for these cancers. We performed two case-control analyses to determine associations between industrial point source emissions, which are proxies for ambient industrial point source-based environmental exposures, and incident BC and LC. METHODS: We obtained information for 5,801 female adults with BC, 5,250 adults with LC, and 47,956 cancer-free adult controls over 2010-2023. We linked indi

Association between herpesviruses and alzheimer's disease: a meta-analysis based on case-control studies.

Herpesviruses infection has been found to be implicated in the etiology of Alzheimer's disease (AD). However, the results remain controversial. This systematic meta-analysis was aimed to evaluate the relationship between Herpesviruses infection and the risk of developing AD. Relevant literature was searched from five databases, including CNKI, PubMed, Web of Science, Embase, and Cochrane Library, to obtain case-control studies (published between the date of database establishment and February 2025; no language restrictions) that compared the Herpesviruses positivity in AD patients and healthy

Pedohebephilia and Perceived Non-coercive Childhood Sexual Experiences: Two Non-matched Case-Control Studies.

Research on the link between childhood sexual abuse experiences (CSAE) and pedohebephilia is limited by its focus on events that the respondents rate as abusive. We asked 199 German-speaking (Study 1) and 632 English-speaking (Study 2) men with and without self-reported pedohebephilia to complete the Childhood Trauma Questionnaire (CTQ) and scales to assess perceived non-coercive childhood sexual experiences with adults (PNCSE-A), and peers (PNCSE-P, only Study 2). A substantial number of participants with PNCSE-A disagreed with all items of the CTQ Sexual Abuse subscale (e.g., 35% and 26% of

Risk of Herpes Zoster and Family History: A Meta-analysis of Case-control Studies.

BACKGROUND: Herpes zoster (HZ) results from the reactivation of latent varicella zoster virus (VZV) residing in dorsal root and cranial nerve ganglia. Advanced age and dysfunctional cell-mediated immune responses are well-established risk factors for VZV reactivation. There have been recent interests in whether there is an increased risk of the disease associated with a positive family history. AIMS AND OBJECTIVES: We aimed to conduct a meta-analysis to evaluate the association between HZ infection and family history. In addition, we investigated the dose-response relationship between HZ infe

Design and validation of methods searching for risk factors in genotype case-control studies.

Accessibility of high-throughput genotyping technology allows genome-wide association studies for common complex diseases. This paper addresses two challenges commonly facing such studies: (i) searching an enormous amount of possible gene interactions and (ii) finding reproducible associations. These challenges have been traditionally addressed in statistics while here we apply computational approaches--optimization and cross-validation. A complex risk factor is modeled as a subset of single nucleotide polymorphisms (SNPs) with specified alleles and the optimization formulation asks for the on
