Catalase-peroxidase (KatG): a potential frontier in tuberculosis drug development.

Mycobacterium tuberculosis (Mtb) depends on the bifunctional enzyme catalase-peroxidase (KatG) for survival within the host. KatG exhibits both catalase and peroxidase activities, serving distinct yet critical roles. While its peroxidase activity is essential for activating the frontline tuberculosis drug isoniazid, its catalase activity protects Mtb from oxidative stress. This bifunctional enzyme is equipped with a unique, protein-derived cofactor, methionine-tyrosine-tryptophan (MYW), which enables catalase activity to efficiently disproportionate hydrogen peroxide in phagocytes. SDBS induces multiple catalase conformations in a dose-dependent manner.

Amyloid fibrils have been linked to several incurable diseases. They are long and thin fibrous proteins that self-assemble into fibrils. Small molecules can stimulate amyloid fibrillation, but the mechanism by which this happens is not well understood. This study examined how a negatively charged benzene ring containing surfactant, sodium dodecylbenzene sulphonate (SDBS), affects the fibrillation of bovine liver catalase (BLC). After SDBS treatment, BLC conformational changes were examined in vitro using turbidity, RLS kinetics, intrinsic fluorescence, ThT fluorescence, far-UV CD, and TEM. BLC

Peroxisomal abnormalities and catalase deficiency in Hutchinson-Gilford Progeria Syndrome.

Wildfire seasons have become more prolonged and intense in the western US over recent decades due to climate change and other factors. Wildfire smoke contains volatile organic compounds (VOCs) and fine particulate matter (PM2.5), which are associated with heart and lung diseases in humans and animals. The goal of this study was to assess VOCs and PM2.5 in ambient air and smoke during wildfire events on an Idaho dairy farm compared to two other Idaho cities. From 2019 to 2022, weekly passive ground-based sampling using Tenax TA sorbent tubes was conducted in three select cities of Idaho: Moscow

Overexpression of Catalase Diminishes Oxidative Cysteine Modifications of Cardiac Proteins.

Reactive protein cysteine thiolates are instrumental in redox regulation. Oxidants, such as hydrogen peroxide (H2O2), react with thiolates to form oxidative post-translational modifications, enabling physiological redox signaling. Cardiac disease and aging are associated with oxidative stress which can impair redox signaling by altering essential cysteine thiolates. We previously found that cardiac-specific overexpression of catalase (Cat), an enzyme that detoxifies excess H2O2, protected from oxidative stress and delayed cardiac aging in mice. Using redox proteomics and systems biology, we so

Atypical refsum disease with pipecolic acidemia and abnormal catalase distribution.

We describe an 18-year-old patient with psychomotor retardation and abnormally short metatarsi and metacarpals but no other signs of classic Refsum disease. Molecular analysis of the phytanoyl-coenzyme A hydroxylase gene revealed a homozygous deletion causing a frameshift. Surprisingly, L-pipecolic acid was elevated in plasma, and microscopy of the liver showed a reduced number of peroxisomes per cell and a larger average peroxisome size. These abnormal peroxisomes lacked catalase as did peroxisomes in fibroblasts of this patient. Such generalized peroxisomal abnormalities are not present in c

Developmentally related responses of maize catalase genes to salicylic acid.

The response of the maize catalase genes (Cat1, Cat2, and Cat3) to salicylic acid (SA) was examined at two distinct developmental stages: embryogenesis and germination. A unique, germination-related differential response of each maize catalase gene to various doses of SA was observed. During late embryogenesis, total catalase activity in scutella increased dramatically with 1 mM SA treatment. The accumulation of Cat2 transcript and CAT-2 isozyme protein provided the major contribution to the observed increase in total catalase activity. This increase was paralleled by the enhanced growth of ge

Evidence against specific binding of salicylic acid to plant catalase.

It was demonstrated that salicylic acid (SA) not only binds to catalase from differentiated higher plants and plant cell suspension cultures but also to those of fungi and animals. SA bound specifically to iron-containing enzymes, such as catalase, aconitase, lipoxidase and peroxidase, while not to iron-free plant enzymes. On the grounds of these experiments, the claim is further challenged that SA is a signalling compound and second messenger in plants that activates plant defense-related genes through elevated H2O2 levels by specifically inhibiting catalase activity. SA may just function as

Superoxide dismutase, glutathione peroxidase and catalase in developing rat brain.

The specific activities of Cu,Zn- and Mn-superoxide dismutases, of glutathione peroxidase and of catalase, the enzymes considered to be specifically involved in the defence of the cell against the partially reduced forms of oxygen, were determined as the function of postnatal age in the early (up to 60 days) period of rat brain development. The enzymes were assayed in the cytoplasmic fraction, in the crude mitochondrial fraction including peroxisomes, and in the mitochondria. The results show that the temporal changes of these enzymes cannot be correlated with each other, thus indicating that

Specific function of the Met-Tyr-Trp adduct radical and residues Arg-418 and Asp-137 in the atypical catalase reaction of catalase-peroxidase KatG.

Catalase activity of the dual-function heme enzyme catalase-peroxidase (KatG) depends on several structural elements, including a unique adduct formed from covalently linked side chains of three conserved amino acids (Met-255, Tyr-229, and Trp-107, Mycobacterium tuberculosis KatG numbering) (MYW). Mutagenesis, electron paramagnetic resonance, and optical stopped-flow experiments, along with calculations using density functional theory (DFT) methods revealed the basis of the requirement for a radical on the MYW-adduct, for oxyferrous heme, and for conserved residues Arg-418 and Asp-137 in the r

Catalase protects Penaeus vannamei against WSSV by regulating redox homeostasis.

White spot syndrome virus (WSSV) remains one of the most destructive viral pathogens in shrimp aquaculture. Reactive oxygen species (ROS) are required for innate immune signaling, but uncontrolled ROS accumulation can damage host tissues and compromise antiviral defense. Catalase (CAT) decomposes hydrogen peroxide and is therefore positioned to connect redox homeostasis with immune resilience during viral infection. Here, we cloned and characterized a CAT gene (Pv-CAT) from Penaeus vannamei and examined its role in the anti-WSSV response. Pv-CAT contained a 1563-bp open reading frame encoding

Alizarin Red perturbs skeletal patterning and biomineralization via catalase inhibition.

Alizarin Red S (AZ) is a dye that is commonly used in histological studies and textiles. We find that AZ is teratogenic to Lytechinus variegatus sea urchin larvae. Here, we demonstrate that embryos exposed to AZ have abnormal skeletal patterning and perturbed migration of skeletogenic mesenchyme cells, along with mild ectodermal abnormalities and reduced neural connectivity. Temporal transcriptomics reveal delayed development and implicate wide-reaching changes in gene expression with AZ treatment. Particle image velocimetry experiments show that AZ treatment perturbs the normal pattern of flu

PPARα regulates acyl-CoA oxidase 1 (ACOX1) but not catalase.

Peroxisomes are membranous organelles, and peroxisomal membrane protein 70 (PMP70), a peroxisome transporter, is used as a marker of peroxisomes. Like mitochondria, peroxisomes can also oxidize fatty acids and its rate limiting enzyme is acyl-CoA oxidase 1 (ACOX1). But unlike mitochondria, peroxisomes generate hydrogen peroxide (H2O2) by ACOX1, and the generated H2O2 is locally detoxified by catalase. PPARα agonist WY-14,643 induces peroxisome proliferation as indicated by the induction of PMP70, ACOX1 and catalase were also induced. However, ACOX1 was induced to a greater extent than catalase

Alizarin red perturbs skeletal patterning and biomineralization via Catalase inhibition.

Alizarin Red S (AZ) is an anthraquinone dye that is commonly used in histological studies and textiles. Exposure to AZ results in morphological perturbations in several species, including rats, frogs, and dogs; however, the mechanisms by which AZ has these effects is largely unexplored, and little is known about its effect on development. We have previously shown that AZ is teratogenic to sea urchin larvae, and that AZ was the only calcium-binding mineralization marker among five tested that perturbed skeletal patterning. Here, we further characterize these defects and demonstrate that embryos

Nitrite and nitroso compounds can serve as specific catalase inhibitors.

OBJECTIVE: We present evidence that nitrite and nitrosothiols, nitrosoamines and non-heme dinitrosyl iron complexes can reversibly inhibit catalase with equal effectiveness. METHODS: Catalase activity was evaluated by the permanganatometric and calorimetric assays. RESULTS: This inhibition is not the result of chemical transformations of these compounds to a single inhibitor, as well as it is not the result of NO release from these substances (as NO traps have no effect on the extent of inhibition). It was found that chloride and bromide in concentration above 80 mM and thiocyanate in concen

Decreased catalase activity in malformation-prone embryos of diabetic rats.

The risk for congenital malformation is increased in diabetic pregnancy. An excess of radical oxygen species (ROS) in the embryo has been suggested as a major teratogenic mechanism. We have used 2 rat strains, denoted H and U, with different catalase isoenzymes to study if the type of ROS scavenging enzyme may be of importance for the embryonic dysmorphogenesis in diabetic pregnancy. Rats were mated H x H and U x U, and about half of the females had streptozotocin-induced diabetes. Embryos were harvested from female rats on day 11 and day 20 of pregnancy. On day 11, the H embryos showed larger

Since other dysplasias are known in this syndrome, the hypertrophic cardiomyopathy in del(11p) children may also represent an abnormality of tissue development.

Role of Catalase in Oxidative Stress- and Age-Associated Degenerative Diseases.

Reactive species produced in the cell during normal cellular metabolism can chemically react with cellular biomolecules such as nucleic acids, proteins, and lipids, thereby causing their oxidative modifications leading to alterations in their compositions and potential damage to their cellular activities. Fortunately, cells have evolved several antioxidant defense mechanisms (as metabolites, vitamins, and enzymes) to neutralize or mitigate the harmful effect of reactive species and/or their byproducts. Any perturbation in the balance in the level of antioxidants and the reactive species result

Identification and characteristic analysis of the catalase gene from Locusta migratoria.

Catalase (CAT) is a ubiquitous antioxidant enzyme in almost all living organisms exposed to atmosphere, which involved in decomposing harmful hydrogen peroxide, into oxygen and water. In this study, a full-length cDNA (1524bp) encoding the catalase gene (LmCAT) from Locusta migratoria was cloned (accession number KT716445). The open reading frame of the LmCAT gene encoded 507 amino acids and shared 57.8%-97.8% amino acid identities with other insect CATs. The coding region was interrupted by 9 introns, while its promoter region contained 15 putative binding sites for 5 kinds of transcriptional
