The effect of antihistaminics on cataleptogenic action of analgesics and haloperidol.

We investigated the effect of benzhydramine, thenalidine, mepyramine (H1 receptor antagonists) and cimetidine (H2 receptor antagonist) on cataleptogenic action of morphine, codeine, fentanyl, and haloperidol. Benzhydramine antagonized the catalepsy induced by analgesics (particularly by codeine). The antagonistic effect of thenalidine was weaker (but also very prominent for codeine catalepsy). Mepyramine did not affect the catalepsy induced by analgesics. All three H1 receptor antagonists attenuated the catalepsy induced by haloperidol. Cimetidine, injected intraventricularly, did not affect t

Nicotine potentiates the behavioral effects of haloperidol.

Nicotine potentiates the catalepsy produced by haloperidol. Furthermore, nicotine as an adjunct to haloperidol produces a remarkable improvement in motor tics in Tourette's syndrome (TS) patients. The present experiments (1) compared the ability of nicotine to potentiate the catalepsy produced by haloperidol or the selective D1 dopamine receptor antagonist SCH 23390 and (2) examined the effects of various doses of nicotine (0.1, 0.2, or 0.3 mg/kg) on haloperidol-induced (0.1, 0.2, or 0.4 mg/kg) catalepsy and locomotor hypoactivity. In the first experiment, nicotine produced a five-fold increas

Further evaluation of the tropane analogs of haloperidol.

Previous work from our labs has indicated that a tropane analog of haloperidol with potent D2 binding but designed to avoid the formation of MPP(+)-like metabolites, such as 4-(4-chlorophenyl)-1-(4-(4-fluorophenyl)-4-oxobutyl)pyridin-1-ium (BCPP(+)) still produced catalepsy, suggesting a strong role for the D2 receptor in the production of catalepsy in rats, and hence EPS in humans. This study tested the hypothesis that further modifications of the tropane analog to produce compounds with less potent binding to the D2 receptor than haloperidol, would produce less catalepsy. These tests have no

Study of the participation of the dopaminergic transmission system in the effects of two newly synthesized barbiturates.

The effects of two newly synthesized barbiturates--HB-7 (2-hydroxylamino-5-ethyl-5-sec. pentylbarbituric acid) and HB-2 (2-hydroxylamino-5-ethyl-5-propylbarbituric acid--on models of apomorphine stereotypy and haloperidol catalepsy were investigated in experiments on male rats and mice. The test drugs used were phenobarbital (PB)--in stereotypy and catalepsy, and diphenylhydantoin (DPH)--in catalepsy. HB-7 changes the stereotypy considerably: it developed faster and disappeared faster than in the control group. Phenobarbital had a weaker effect on the intensity and duration of the stereotypy.

Effects of lipopolysaccharide and interleukin-6 on cataleptic immobility and locomotor activity in mice.

Catalepsy (animal hypnosis, tonic immobility) is a type of passive defensive behavior. Its exaggerated form is a syndrome of some psychopathological disorders. Numerous neurotransmitters have impact on the regulation of catalepsy. In this paper we demonstrated the involvement of interleukin-6 (IL-6) in the mechanism of cataleptic immobility. Effects of exogenous IL-6 treatment (7.5 and 10μg/kg, i.p) or stimulation of endogenous IL-6 secretion with lipopolysaccharide (LPS) administration (50, 100 and 200μg/kg, i.p.) on catalepsy and locomotor activity were studied in adult C57BL/6 male mice. Propiverine-induced Parkinsonism: a case report and a pharmacokinetic/pharmacodynamic study in mice.

PURPOSE: We present a case report of propiverine-induced Parkinsonism. We previously reported the induction of catalepsy by amiodarone, aprindine and procaine, which possess a diethylaminomethyl moiety and demonstrated selective blockade of dopamine D2 receptors by these drugs in mice. We hypothesized that drugs possessing a diethylaminomethyl structure may generally induce Parkinsonism and/or catalepsy. METHODS: Thus, we performed a study to examine whether oxybutynin, pentoxyverine and etafenone, as well as propiverine, induce catalepsy in mice. RESULTS: The intensity of drug-induced catal

Differential reversal by scopolamine and THIP of the antistereotypic and cataleptic effects of neuroleptics.

Scopolamine reversed the neuroleptic-induced catalepsy and neuroleptic blockade of amphetamine-induced stereotypies in rats. The effect on catalepsy was the most pronounced. Butyrophenones were mainly sensitive to scopolamine reversal whereas the thioxanthenes cis-Z-flupentixol and piflutixol were not significantly influenced. THIP did not change the effects of cis-Z-flupentixol, fluphenazine and haloperidol. The relevance of these findings for the use of stereotypy and catalepsy tests as screening for antiscizophrenic and extrapyramidal side-effects, respectively, is discussed.

Occupancy-Guided Continuous Antipsychotic Dosing Provides a Translational Model for Evaluating Extrapyramidal Motor Liability in Mice.

The therapeutic effects of antipsychotic drugs are primarily achieved through dopamine D2 receptor antagonism. However, excessive striatal D2 receptor blockade is associated with extrapyramidal side effects (EPS) in schizophrenic patients. Clinical studies indicate a therapeutic window of approximately 60%-80% striatal D2 receptor occupancy, with higher occupancy increasing the risk of EPS. However, conventional rodent dosing paradigms rarely maintain clinically comparable receptor occupancy, limiting their translational relevance. Here, we investigated whether continuous antipsychotic adminis

[Effect of apomorphine on the wakefulness--sleep cycle of the common frog Rana temporaria].

This work considers effects of introduction into spinal lymphatic sac of dopamine agonist--apomorphine-(APO) at doses of 0.1, 1.0, 2.0 and 4.0 mg/kg body weight on the common frog wakefulness-sleep cycle (WSC). Usually the frog WSC is represented by wakefulness and three types of passive-protective behavior: by immobility states of the type of catalepsy, catatonia, and cataplexy that are characterized by high thresholds of arousal and by different (corresponding to the name) skeletal musculature tones. These immoboloty forms are considered as homologues of mammalian stress-reaction, hibernatio

Effects of the antidepressant trazodone, a 5-HT 2A/2C receptor antagonist, on dopamine-dependent behaviors in rats.

RATIONALE: 5-Hydroxytryptamine, via stimulation of 5-HT 2C receptors, exerts a tonic inhibitory influence on dopaminergic neurotransmission, whereas activation of 5-HT 2A receptors enhances stimulated DAergic neurotransmission. The antidepressant trazodone is a 5-HT 2A/2C receptor antagonist. OBJECTIVES: To evaluate the effect of trazodone treatment on behaviors dependent on the functional status of the nigrostriatal DAergic system. METHODS: The effect of pretreatment with trazodone on dexamphetamine- and apomorphine-induced oral stereotypies, on catalepsy induced by haloperidol and apomorph

A mutual prodrug ester of GABA and perphenazine exhibits antischizophrenic efficacy with diminished extrapyramidal effects.

The perphenazine and fluphenazine GABA esters 3 and 4 evaluated in rat models for antipsychotic activity displayed a significant decrease of catalepsy associated with increased prolactin blood levels. Efficacy was evaluated in the d-amphetamine-induced hyperactivity model, where perphenazine abolished hyperactivity and induced sedation and catalepsy, whereas 3 reduced hyperactivity without sedation or catalepsy. Thus, 3 (BL-1020) constitutes a prototype of novel antipsychotics possessing GABAergic activity. A phase II study is in progress.

Effects of dexfenfluramine on dopamine dependent behaviours in rats.

5-hydroxytryptamine (5-HT) inhibits the synthesis and release of dopamine (DA) from rat nigrostriatal DAergic neurons. Dexfenfluramine releases 5-HT from brain 5-HTergic neurons. The present study was undertaken to determine whether dexfenfluramine, through the released 5-HT, modulates the intensity of the behaviours dependent on the functional status of the nigrostriatal DAergic system. The effect of pretreatment with dexfenfluramine on dexamphetamine and apomorphine stereotypies of the oral movement variety and on catalepsy induced by haloperidol and small doses (0.05 and 0.1 mg/kg ip) of ap

Lack of cataleptogenic potentiation with non-imidazole H3 receptor antagonists reveals potential drug-drug interactions between imidazole-based H3 receptor antagonists and antipsychotic drugs.

Since H3 receptor (H3R) antagonists/inverse agonists can improve cognitive function in animal models, they may have the potential to be used as add-on therapy in the treatment of schizophrenia, a disease with significant cognitive deficits. However, a recent study showed potentiation of haloperidol-induced catalepsy by ciproxifan, an imidazole-containing H3R antagonist/inverse agonist, suggesting there is a potential risk of exacerbating extrapyramidal symptoms (EPS) if H3R antagonists were used as adjunctive treatment [Pillot, C., Ortiz, J., Heron, A., Ridray, S., Schwartz, J.C. and Arrang, J

[Seasonal changes in the behavioral effects of neuroleptics on white rats].

Summarized results of the experiments (conducted in 1981-1984) demonstrate seasonal rhythms of some behavioural effects (catalepsy and depression of locomotor activity) of haloperidol (0.5 mg/kg) and levomepromazine (5 mg/kg) in white rats. In intact rats neuroleptics were more effective in depressing high than low motor activity. Catalepsy induced by single administration of neuroleptics was more pronounced in spring and autumn months. A certain negative correlation exists between seasonal variations of neuroleptic catalepsy and the speed of monoamine (dopamine and serotonin) metabolism in th

Effects of dextromethorphan on dopamine dependent behaviours in rats.

Dextromethorphan, a noncompetitive blocker of N-methyl-D- aspartate (NMDA) type of glutamate receptor, at 7.5-75 mg/kg, ip did not induce oral stereotypies or catalepsy and did not antagonize apomorphine stereotypy in rats. These results indicate that dextromethorphan at 7.5-75 mg/kg does not stimulate or block postsynaptic striatal D2 and D1 dopamine (DA) receptors. Pretreatment with 15 and 30 mg/kg dextromethorphan potentiated dexamphetamine stereotypy and antagonised haloperidol catalepsy. Pretreatment with 45, 60 and 75 mg/kg dextromethorphan, which release 5-hydroxytryptamine (5-HT), howe

Vinpocetine prevents haloperidol-induced cognitive and working memory deficits through attenuation of oxidative and nitrosative stress in mice.

Vinpocetine has been shown to protect against degenerative senile cerebral dysfunction via enhancement of cerebral blood flow, cognition and neuroprotective action. This study sought to investigate the protective effect of vinpocetine against haloperidol-induced catalepsy in mice. Vinpocetine (5, 10 or 20 mg/kg, p.o.) was administered 1 h after haloperidol injection for 21 consecutive days. Effect on motor coordination, depressive-like behaviour and working memory were assessed with rotarod, forced swim (FST) and Y-maze tests, respectively. Brains were collected on day 21 for biochemical estim

Effects of pentazocine on dopamine-dependent behaviours in mice.

Pentazocine, a kappa opioid receptor agonist, induced catalepsy in mice suggesting thereby that it might possess postsynaptic striatal D 2 dopamine (DA) receptor blocking activity. However, our other findings, that pentazocine pretreatment did not antagonise the cage climbing behaviour induced by the directly acting DA agonist apomorphine in mice and actually potentiated the stereotyped behaviour induced by the indirectly acting DA agonist methamphetamine in mice, indicate that pentazocine does not possess postsynaptic striatal and mesolimbic D 2 DA receptor blocking activity. Combined low calcium and lack magnesium is a risk factor for motor deficit in mice.

The populations of the Kii Peninsula in Japan and of Guam present high incidences of amyotrophic lateral sclerosis and Parkinsonism-dementia complex. It is thought that low levels of calcium (Ca) and magnesium (Mg) in the drinking water are involved in the pathogenesis of these diseases. The present study aimed to test the hypothesis that catalepsy, behavioral immobility and a Parkinsonian symptom results from functionally impaired dopaminergic neurons in mice fed low amounts of Ca and Mg (LCa/Mg). A group of mice fed a LCa/Mg diet for 6 weeks was compared to a control group on a standard diet

Antipsychotic-like vs cataleptogenic actions in mice of novel antipsychotics having D2 antagonist and 5-HT1A agonist properties.

A new generation of proven or potential antipsychotics, including aripiprazole, bifeprunox, SSR181507 and SLV313, exhibit agonist actions at serotonin 5-HT1A receptors, but little comparative data are available on their pharmacological profiles. Here, we compared in mice the in vivo antipsychotic-like vs cataleptogenic activities of these compounds with those of drugs that exhibit little interaction at 5-HT1A receptors, such as haloperidol, olanzapine and risperidone. All the drugs dose-dependently reduced apomorphine-induced climbing or sniffing and, with the exception of ziprasidone, produce
