Preclinical development of the green tea catechin, epigallocatechin gallate, as an HIV-1 therapy.

BACKGROUND: Previously, we presented evidence that at physiologic concentrations the green tea catechin, epigallocatechin gallate (EGCG), inhibited attachment of HIV-1 glycoprotein 120 to the CD4 molecule on T cells, but the downstream effects of EGCG on HIV-1 infectivity were not determined. OBJECTIVE: To evaluate the inhibition of HIV-1 infectivity by EGCG and begin preclinical development of EGCG as a possible therapy. METHODS: PBMCs, CD4(+) T cells, and macrophages were isolated from blood of HIV-1-uninfected donors. HIV-1 infectivity was assessed by an HIV-1 p24 ELISA. Cell survival was

Protective effect of (+)-catechin against gastric mucosal injury induced by ischaemia-reperfusion in rats.

Ischaemia and reperfusion are known to induce gastric lesions, predominantly due to excessive formation of reactive oxygen metabolites, adhesion of neutrophils to endothelial cells, microvascular dysfunction, gastric acid secretion, endogenous histamine and gastrin release. We have studied the effect of (+)-catechin on a gastric ulcer model involving damage to gastric injury by ischaemia- reperfusion (I/R) in rats. (+)-Catechin 50 mg kg(-1)administered orally, once daily for three days after the initiation of I/R injury showed a significant (P<0.001) anti-ulcer activity against mucosal dam- ag

Catechins as antibiofilm agents: molecular insights for virtual screening and in vitro biological evaluation.

INTRODUCTION: Bacterial Biofilms are of great concern because they are the main drivers of antimicrobial resistance. We investigated the potential of catechins against bacterial biofilms using advanced computational and in vitro models. METHODS: The HPLC profiling of Camellia sinensis extract confirmed the presence of catechins. The ADMET and drug-likeness studies were specified to comply with the optimal values. RESULTS: Molecular docking on 2UV0 indicated a moderate interaction with targets, which was further confirmed by DFT analysis (Homo-Lumo). Root mean square fluctuations and normal m

New insights into the regulatory effects of galloylated catechin on male reproduction in Camellia oleifera.

Flavonoids, particularly galloylated catechins, are known to regulate pollen tube growth, but their mode of action in woody plants remains poorly characterized. Here, we investigated the effects of epigallocatechin gallate (EGCG)-a major galloylated catechin-on Camellia oleifera pollen tubes using combined cytological and transcriptomic approaches. In vitro treatment with 100 μg mL-1 EGCG significantly reduced pollen germination and tube elongation. Confocal and fluorescence microscopy revealed that EGCG rapidly disrupted the apical Ca2+ gradient within 10 min, accompanied by disorganized acti

Effect of green tea catechins on gastric mucosal dysplasia in insulin-gastrin mice.

Green tea catechins (GTCs) have been implicated in various physiological effects, including anti-carcinogenic activities. In the present study, we evaluated the effects of GTCs specifically on the development of gastritis and pre-malignant lesions in insulin-gastrin mice. Nine-week-old male INS-GAS mice (n=38) were supplemented with GTCs for 4 and 28 weeks, and their body weights, serum gastrin levels, histopathology and pro-inflammatory cytokine levels in gastric tissue and mucosal cell proliferation were monitored. Body weights of the GTC-treated mice were significantly lower than those of t

New role of antinutritional factors, phytic acid and catechin in the treatment of CCl4 intoxication.

Both phytic acid (PA) and catechin (CA) are well known antioxidants of natural origin. They were frequently tried on experimental level as hepatoprotectants, relying only on their antioxidant properties. The present study was conducted mainly to outline the other biochemical pathways underlying the hepatotherapeutic potential of both drugs and to check a possible synergistic action if prescribed concomitantly. As both materials are frequently taken on a daily basis in food and drinks, it will be helpful to pursue their possible utility and/or to check if their value is really of medical import

Analysis of catechins in Theobroma cacao beans by cyclodextrin-modified micellar electrokinetic chromatography.

A micellar electrokinetic chromatography (MEKC) method was developed for the quantitation of polyphenols (+)-catechin and (-)-epicatechin (catechin monomers) and the methylxanthine theobromine in Theobroma cacao beans. Owing to the poor stability of catechin monomers in alkaline conditions, a 50 mM Britton-Robinson buffer at a pH 2.50 was preferred as the background electrolyte. Under these conditions, the addition of hydroxypropyl-beta-cyclodextrin (HP-beta-CD) at a concentration of 12 mM to the SDS micellar solution (90 mM), resulted in a cyclodextrin-modified micellar electrokinetic chromat

Brewing conditions and post-brewing handling shape the catechin profile and stability of tea infusions.

This study investigated how brewing conditions and water mineralization affect catechin extraction and composition, and catechin stability during heating and acidification in green tea. Commercial green and black teas were brewed with boiling water or under manufacturer-recommended conditions. Catechins and caffeine were quantified by HPLC, with total polyphenols and DPPH radical-scavenging activity also determined. Brewing with boiling water increased catechin extraction, with a preferential increase in gallated catechin content. Increasing water mineralization reduced catechin extraction and

Toxicity and efficacy of green tea catechin derivative-based micellar nanocomplexes for anticancer protein delivery.

Toxicity towards non-tumor cells during anticancer therapy can be reduced by using nanoscale systems for anticancer drug delivery. Usually only the loaded drug has anticancer activity. Recently, micellar nanocomplexes (MNCs) comprising green tea catechin derivatives for the delivery of the anticancer proteins, such as Herceptin, have been developed. Herceptin as well as the MNCs without the drug were effective against HER2/neu-overexpressing human tumor cells and had synergistic anticancer effects in vitro and in vivo. It remained unclear which kinds of negative effects the MNCs had on tumor c

Anti-inflammatory effects of new catechin derivatives in a hapten-induced mouse contact dermatitis model.

Contact dermatitis is a common skin disease, with various treatments available to dermatologists. According to general guidelines, the first line of treatment involves topical steroids; however, this treatment has application-site restrictions in order to avoid adverse cutaneous events. Accordingly, increased demand exists for the development of new treatments. In Japan, the recent use of catechin-containing health foods and their beneficial effects has attracted attention. Indeed, several studies have examined the anticancer, anti-obesity, anti-inflammatory, and antioxidant effects of catechi

Epicatechin and catechin modulate endothelial activation induced by platelets of patients with peripheral artery disease.

Platelet activation contributes to the alteration of endothelial function, a critical initial step in atherogenesis through the production and release of prooxidant mediators. There is uncertainty about the precise role of polyphenols in interaction between platelets and endothelial cells (ECs). We aimed to investigate whether polyphenols are able to reduce endothelial activation induced by activated platelets. First, we compared platelet activation and flow-mediated dilation (FMD) in 10 healthy subjects (HS) and 10 patients with peripheral artery disease (PAD). Then, we evaluated the effect o

Catechins in insomnia-Alzheimer's disease comorbidity: A network pharmacology and molecular docking study.

The comorbidity of insomnia and Alzheimer's disease (AD) is strongly driven by the interplay between circadian rhythm disruption and immune dysfunction. Catechins are multi-target polyphenols capable of modulating both processes, yet their precise mechanism of action remains elusive. Insomnia and AD related differentialy expressed target genes were specifically sourced from circadian rhythm and immune phenotypes. Two machine learning algorithms were then applied to refine and identify the pivotal targets from this phenotype-driven target pool. Functional enrichment analyses, including kyoto en

Exploring the therapeutic potential of (+)-catechin gallate: An in vivo approach to combat liver fibrosis.

Liver fibrosis occurs as a result of the progression of chronic liver diseases, regardless of underlying causes such as hepatitis virus infection, alcohol intake, and metabolic fatty liver disorder. It is often associated with liver damage, inflammation, and cell death. In the current study, the potential of (+)-catechin gallate was checked against liver fibrosis in rat models. Carbon tetrachloride (CCl4) was administrated to induce liver fibrosis, and 0.7 mL/kg of CCl4 with olive oil was injected into rats for six weeks twice a week. The analysis of liver markers (i.e., alkaline phosphatase (
