Tourette syndrome: prediction of phenotypic variation in monozygotic twins by caudate nucleus D2 receptor binding.

Tourette syndrome, a chronic tic disorder with autosomal dominant inheritance, exhibits considerable phenotypic variability even within monozygotic twin pairs. The origins of this variability remain unclear. Recent findings have implicated the caudate nucleus as a locus of pathology, and pharmacological evidence supports dopaminergic involvement. Within monozygotic twins discordant for Tourette syndrome severity, differences in D2 dopamine receptor binding in the head of the caudate nucleus predicted differences in phenotypic severity (r = 0.99); this relation was not observed in putamen. Dopamine receptor blockade in nucleus accumbens or caudate nucleus differentially affects feeding induced by 8-OH-DPAT injected into dorsal or median raphe.

The 5-hydroxytryptamine (5-HT)1A receptor agonist 8-hydroxy-2-(di-n-propylamino)tetralin (8-OH-DPAT) elicits a variety of behaviours including feeding in rats. These effects are accompanied by a reduction in 5-HT neurotransmission resulting from activation of somatodendritic 5-HT receptors located in the midbrain raphe nuclei. Previous work showing that dopamine receptor antagonists attenuate 8-OH-DPAT-induced feeding indicates that a facilitation of dopamine activity, secondary to reduced 5-HT activity, is involved in the expression of this effect. Microinjection studies were conducted to exp

Altered functional connectivity of the right caudate nucleus in chronic migraine: a resting-state fMRI study.

BACKGROUND: The definitive pathogenic mechanisms underlying chronic migraine (CM) remain unclear. Mounting evidence from functional and structural magnetic resonance imaging (MRI) studies suggests that the caudate nucleus (CN) plays a role in the cognitive, sensory, and emotional integration of pain information in patients with migraine. However, evidence concerning the role played by CN in CM patients is limited. Here, we used the CN as the seed to explore patterns of functional connectivity (FC) among healthy controls (HCs), patients with episodic migraine (EM), and patients with CM. METHOD

A dysembryoplastic neuroepithelial tumor in the area of the caudate nucleus in a 57-year-old woman: case report.

OBJECTIVE: Dysembryoplastic neuroepithelial tumors (DNTs) are clinicopathologically unique tumors. They are usually located within the supratentorial cortex, most often in the temporal lobe. These lesions are frequently associated with intractable complex partial seizures in children and young adults. DNTs may also arise outside of the cerebral cortex. We report a case of a 57-year-old woman who had a DNT in the caudate nucleus. During a 7-year follow-up period, the tumor did not recur. The literature on DNTs with atypical "ectopic" localizations is summarized. CLINICAL PRESENTATION: A patien

Repetitive transcranial magnetic stimulation of the human prefrontal cortex induces dopamine release in the caudate nucleus.

Dopamine is implicated in movement, learning, and motivation, and in illnesses such as Parkinson's disease, schizophrenia, and drug addiction. Little is known about the control of dopamine release in humans, but research in experimental animals suggests that the prefrontal cortex plays an important role in regulating the release of dopamine in subcortical structures. Here we used [(11)C]raclopride and positron emission tomography to measure changes in extracellular dopamine concentration in vivo after repetitive transcranial magnetic stimulation (rTMS) of the dorsolateral prefrontal cortex in

Effect of acute and chronic fluoxetine on extracellular dopamine levels in the caudate-putamen and nucleus accumbens of rat.

Recent studies indicate that an increase in serotonergic (5-HT) activity in the nucleus accumbens (NAc) produces an increase in dopamine (DA) release, providing a possible mechanism for the involvement of DA in the therapeutic action of selective serotonin reuptake inhibitor (SSRI) antidepressants. However, acutely administered fluoxetine (2.5, 5.0, or 10.0 mg/kg, i.p.) failed to elevate extracellular levels of DA, or its metabolites in the NAc or caudate-putamen (CP). In fact, the highest dose produced a small (20%) decrease in DA levels in the NAc. Extracellular levels of the 5-HT metabolite

[The reversibility of a blockade of corticofugal pulse trains to the neurons of the caudate nucleus in cats evoked by the neurotoxin MPTP].

In anesthetised cats, the number of the caudate nucleus neurons responding to a cortical stimulation with latency less than 8.0 ms, increased within 10-12 days after the MPTP administration and then gradually recovers by the 45th-54th day after the neurotoxin administration. Dopamine seems to exert a protecting-inhibiting effect on transmission of corticofugal glutamatergic impulses travelling towards the neostriatum neurons.

Caudate nucleus neurons participate in methylphenidate function: Behavioral and neuronal recordings from freely behaving adolescent rats.

Methylphenidate (MPD) is the most commonly prescribed psychostimulant for the treatment of attention-deficit hyperactivity disorder (ADHD). MPD acts on brain structures of the reward/motivation system, including the caudate nucleus (CN). The objective of this study was to investigate the acute and chronic dose response effects of MPD on CN neurons in freely behaving adolescent rats. Semi-microelectrodes were implanted into the CN of one hundred and sixty-three adolescent male Sprague-Dawley rats. On experimental day one (ED1), each rat was injected with 0.8 ml saline followed by an injection o

Caudate nucleus atrophy as a neuroimaging feature of mild cognitive impairment induced by occupational aluminum exposure.

Occupational aluminum (Al) exposure is linked to mild cognitive impairment (MCI), yet its neuroanatomical mechanisms remain unclear. We investigated gray matter volume (GMV) alterations specifically associated with Al-induced MCI. This study enrolled 86 participants: 30 Al-exposed MCI (Al-MCI), 26 Al-exposed cognitively normal (Al-HC), and 30 non-exposed controls (Non-Al-HC). Structural MRI quantified whole-brain GMV. Plasma Al concentrations, Montreal Cognitive Assessment (MoCA), the Rey Auditory Verbal Learning Test (AVLT), and the Trail Making Test (TMT) were assessed in Al-exposed individu

Neural Representations of Post-Decision Accuracy and Reward Expectation in the Caudate Nucleus and Frontal Eye Field.

Performance monitoring that supports ongoing behavioral adjustments is often examined in the context of either choice confidence for perceptual decisions (i.e., "did I get it right?") or reward expectation for reward-based decisions (i.e., "what reward will I receive?"). However, our understanding of how the brain encodes these distinct evaluative signals remains limited because they are easily conflated, particularly in commonly used two-alternative tasks with symmetric rewards for correct choices. Previously we used a motion-discrimination task with asymmetric rewards to identify neural subs

An imaging mass spectrometry atlas of lipids in the human neurologically normal and Huntington's disease caudate nucleus.

Huntington's disease (HD) is a fatal disorder associated with germline trinucleotide repeat expansions in the HTT gene and characterised by striatal neurodegeneration. No efficacious interventions are available for HD, highlighting a major unmet medical need. The molecular mechanisms underlying HD are incompletely understood despite its monogenic aetiology. However, direct interactions between HTT and membrane lipids suggest that lipidomic perturbations may be implicated in the neuropathology of HD. In this study, we employed matrix-assisted laser desorption/ionisation imaging mass spectrometr

Hemichorea-hemiballism and anosognosia following a contralateral infarction of the caudate nucleus and anterior limb of the internal capsule.

The authors describe a patient who experienced the sudden occurrence of hemicorea-hemiballism on the left side of the body and the contemporary agnosia of it. An unenhanced CT-scan disclosed an ischemic lesion involving the head of the caudate nucleus and of the anterior limb of the internal capsule of the contralateral hemisphere. The authors discuss the possible pathophysiologic mechanisms explaining the rare association of these two symptoms and suggest that the involvement of the pyramidal tract in the internal capsule usually does not allow hyperkinesis to manifest itself.

[Effect of DOPA and haloperidol on experimental torticollis originating in the mesencephalon and its modulation by stimulation of the caudate nucleus].

Electrolytic injury to dopaminergic neurons of the mesencephalic tegmentum in cats produced a steady experimental torticollis (ET) with contralateral turning of the head and body. DOPA potentiated and haloperidol mostly reduced ET. Analogous changes were induced by electrical stimulation of the ipsi- and contralateral caudate nucleus, respectively. Stimulation of the contralateral nucleus coupled with haloperidol administration led to a more pronounced restriction of ET. Unexpectedly, the same result was obtained after DOPA administration coupled with stimulation of the ipsi-lateral caudate nu

Topographic changes in high-affinity glutamate uptake in the cat red nucleus, substantia nigra, thalamus, and caudate nucleus after lesions of sensorimotor cortical areas.

The distribution of presumed glutamatergic projections from sensorimotor cortical areas to the red nucleus (RN), the substantia nigra (SN), the ventrolateral thalamic complex, and the caudate nucleus (CN) was investigated in the cat. For this purpose, the changes in the sodium-dependent high-affinity glutamate uptake (HAGU) rate were measured in homogenates of tissue microdissected from various parts of these subcortical structures after chronic surgical ablation of sensorimotor cortical areas. After 8 to 10 days survival, significant reductions in HAGU activity were noted in the structures st

Zinc potentiates 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine induced dopamine depletion in caudate nucleus of mice brain.

Present study describes the effect of zinc (Zn) on 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) induced dopamine depletion in mice brain. MPTP is a known neurotoxicant primarily causing marked depletion of dopamine (DA) levels in nigrostriatal dopaminergic system. Adult Male C57-mice were intraperitonially injected with 25 mg/kg MPTP in the presence or absence of zinc acetate. Twenty-four hours after treatment animals were sacrificed and DA levels were determined by high performance liquid chromatography in caudate nucleus of control and treated mice. The results showed that there was a

Resting-state functional connections with the hippocampus and with the caudate nucleus predict working memory performance in multiple sclerosis.

Working memory (WM) dysfunction is common in multiple sclerosis (MS). WM deficits also contribute to episodic memory (EM) dysfunction, a prevalent and disabling cognitive symptom of the disease. Functional connectivity between the hippocampus and caudate nucleus supports EM, but its specific influences on WM in MS are unknown. Resting-state functional connectivity (RSFC) can identify spontaneous, baseline connectivity patterns that predict cognitive impairment, thus offering clinical utility. The present study examined whether hippocampal and caudate RSFC could predict WM performance in people

Unilateral involuntary movements of the limbs with caudate nucleus restricted diffusion on magnetic resonance imaging: A case report.

Anti-leucine-rich glioma-inactivated-1 (LGI1) autoimmune encephalitis is one of the most prevalent forms of autoantibody-associated limbic encephalitis. Brain magnetic resonance imaging of patients with anti-LGI1 encephalitis often presents as fluid-attenuated inversion recovery hyperintensities of the medial temporal lobe. We report the case of a man in his early 60s who suddenly experienced involuntary movement of the right limbs. Brain magnetic resonance imaging revealed normal T1 signal and T2 hyperintensity in the left caudate nucleus, with corresponding fluid-attenuated inversion recover

Expression of osteogenic molecules in the caudate nucleus and gray matter and their potential relevance for Basal Ganglia calcification in hypoparathyroidism.

BACKGROUND: Basal ganglia calcification (BGC) is an interesting example of ectopic calcification in patients with hypoparathyroidism. Its pathogenesis and reasons for predilection of calcification at basal ganglia are not clear. OBJECTIVE: To assess the expression of osteogenesis-related molecules in the caudate nucleus and surface gray matter (an area spared from calcification) and discuss potential relevance of the results in context of BGC in idiopathic hypoparathyroidism. METHODS: Caudate nucleus and gray matter were obtained from 14 autopsies performed in accidental deaths. The mRNA exp
