3D Cell Culture-Based Hybrid Bioanalytical Platform for Optical Imaging Utilizing Silk Fibroin Sponges.

We present an innovative bioanalytical hybrid platform designed for the preclinical evaluation of cellular characteristics. The system combines a three-dimensional (3D) cell culture grown on an artificial extracellular matrix with a chromatography-inspired array configuration. Sponges, made from the structural protein silk fibroin, serve both as a biomimetic extracellular matrix and as a stationary phase. Silk fibroin sponges were produced in-house using a multistep process involving removal of inherent sericin proteins from raw silk fibers, followed by dissolution and dialysis to purify the f

A robust dynamic 3D liver spheroid platform for (sub)chronic toxicity assessment: Metabolic characterization of HepG2 spheroids as a case study.

In vitro assessment of (sub)chronic chemical exposure requires liver models that maintain stable functionality during long-term cultivation and whose baseline behavior is well characterized. HepG2 cells remain widely used in toxicology due to their robustness and reproducibility; however, their hepatoblastoma origin confers persistent tumor‑associated traits. Although three‑dimensional (3D) HepG2 spheroids provide greater physiological relevance than two‑dimensional (2D) cultures, their baseline behavior during prolonged dynamic cultivation remains insufficiently characterized, limiting confid

Crypt-Villus Scaffold Architecture for Bioengineering Functional Human Intestinal Epithelium.

Crypt-villus architecture in the small intestine is crucial for the structural integrity of the intestinal epithelium and maintenance of gut homeostasis. We utilized three-dimensional (3D) printing and inverse molding techniques to form three-dimensional (3D) spongy scaffold systems that resemble the intestinal crypt-villus microarchitecture. The scaffolds consist of silk fibroin protein with curved lumens with rows of protruding villi with invaginating crypts to generate the architecture. Intestinal cell (Caco-2, HT29-MTX) attachment and growth, as well as long-term culture support were demon

Large-Scale Production of Expandable Hepatoblast Organoids and Polarised Hepatocyte Organoids From hESCs Under 3D Static and Dynamic Suspension Conditions.

To date, generating viable and functional hepatocytes in large scale remains challenge. By employing 3D suspension condition with the support of low concentration Matrigel, a novel culture system was developed to generate expandable hepatoblast organoids (HB-orgs) and mature polarised hepatocyte organoids (P-hep-orgs) from human embryonic stem cells (hESCs) in both dishes and bioreactors. scRNA-seq and functional assays were used to characterise HB-orgs and P-hep-orgs. hESC-derived HB-orgs could proliferate at least for 15 passages, leading to 1012 in total cells in 4 weeks. P-hep-orgs differe

Systematic multi-parameter optimization of an injectable thermosensitive CS/β-GP/HEC hydrogel for 3D cell culture applications.

Injectable thermosensitive hydrogels have been widely investigated as minimally invasive scaffolds for three-dimensional (3D) cell culture. In this study, we systematically screened a chitosan (CS)/β-glycerophosphate (β-GP)/hydroxyethyl cellulose (HEC) ternary hydrogel system using a 2 × 2 × 2 factorial design. By evaluating gelation kinetics, injectability, and structural stability through a weighted scoring model, two superior formulations (G1 and G3) were selected for their ability to balance rapid gelation at 37 °C with reliable room-temperature injectability. Subsequent biological evaluat

Mumio and bladder cancer: unlocking its potential in 3D cell culture.

BACKGROUND: Bladder cancer remains one of the most prevalent causes of cancer-related mortality worldwide. Mumio, a naturally occurring substance with a long-standing history of use in traditional medicine, has demonstrated therapeutic potential, including anticancer properties. This study aimed to evaluate the effects of a pharmaceutical formulation of Mumio on the growth of normal (SV-HUC-1) and cancerous (T24, 5637) bladder cell lines in 3D culture. METHODS: Spheroids were generated using ultra-low attachment 96-well plates. Three concentrations of Mumio (LC₁₀, LC₅₀, LC₉₀), previously deter

Toxoplasma gondii infection inhibits myotube formation in a 3D model of chicken muscle cell culture.

BACKGROUND: Toxoplasmosis is a globally distributed zoonosis, caused mainly by the ingestion of contaminated water and food. Toxoplasma gondii is the protozoan that causes the disease and can infect humans by forming cysts in the tissues of its hosts. The parasite can also cross the transplacental barrier and infect the fetus, with consequences for embryonic development. Patients infected with T. gondii have muscle inflammation and polymyositis. Most of the knowledge regarding T. gondii infection in skeletal muscle tissue has been established from few in vivo models and 2D cell culture. Three-

Development of ZmT-PEG hydrogels through Michael addition reaction and protein self-assembly for 3D cell culture.

Bioactive protein-derived hydrogels are highly attractive three-dimensional (3D) platforms for in vitro cell culture. However, most protein and polypeptide hydrogels are extracted from animal tissues or chemically synthesized, with many drawbacks. Herein, we fabricated an optically transparent ZmT-PEG hydrogel via a facile one-pot strategy. The modified Z1Z2 (Zm) was obtained by introducing cysteine at the C-terminus of Z1Z2 (ZC) and inserting the RGD sequence into the low conserved (CD) loop (ZR). A Michael addition reaction occurred between Zm and 4-arm PEG-MAL, and Zm-PEG self-assembled wit

Polymer Matrix-Based 3D Culture Significantly Enhances the Differentiation and Immunomodulatory Functions of Human Adipose-Derived Stem Cells.

Human adipose-derived stem cells (hADSCs) possess immunomodulatory and multipotent properties, making them attractive candidates for regenerative therapies. However, conventional tissue culture plate (TCP)-based 2D adherent cultures or widely used ultra-low attachment (ULA) spheroid systems often compromise differentiation potential and therapeutic efficacy of hADSCs. Here, we report a synthetic polymer matrix, poly-Z, that enables the formation of hADSC spheroids with significantly enhanced biological functions. Compared to ULA or TCP culture systems, poly-Z-cultured hADSC spheroids exhibited

A floating endometrial organoid model recapitulates epithelial-stromal cell interactions in vitro.

Organoids are 3D structures in which stem, progenitor and differentiated cells spontaneously assemble into structures resembling the original tissue. Endometrial organoids, developed from tissue fragments, are genetically stable and responsive to hormone stimulation acquiring a hallow lumen, secretory activity and apico-basal polarity. However, they show some limitations in mimicking the midluteal endometrium since they lack endothelial, immune, and stromal cells, thus providing limited information about epithelial-stromal interactions. We developed a 3D-model to generate endometrial organoids

Synergizing bioprinting and 3D cell culture to enhance tissue formation in printed synthetic constructs.

Bioprinting is currently the most promising method to biofabricate complex tissuesin vitrowith the potential to transform the future of organ transplantation and drug discovery. Efforts to create such tissues are, however, almost exclusively based on animal-derived materials, such as gelatin methacryloyl, which have demonstrated efficacy in bioprinting of complex tissues. While these materials are already used in clinical applications, uncertainty about their safety still remains due to their animal origin. Alternatively, synthetic bioinks have been developed that match the printability of nat

Biological features, gene expression profile, and mechanisms of drug resistance of two- and three-dimensional hepatocellular carcinoma cell cultures.

Hepatocellular carcinoma (HCC) is a common malignant tumor with insidious onset and rapid progression. Its treatment is often difficult owing to tumor resistance. In this study, we aimed to understand the different biological characteristics, gene expression profiles, and drug resistance mechanisms of HCC cells cultured under different conditions. A conventional adherence method and a liquid overlay technique were used to prepare two- and three-dimensional cultures of Bel-7402 and 5-fluorouracil (5-Fu)-resistant Bel-7402 (Bel-7402/5-Fu) cells. Morphological characteristics were assessed via mi

Compound design of a patient-derived 3D cell culture system modelling early peritoneal endometriosis.

Peritoneal endometriosis causes pelvic pain and infertility, but the underlying mechanisms related to these symptoms are not fully understood. Endometriosis diagnosis is typically delayed; thus, patient samples are unsuitable to study early endometriosis formation in situ. We generated a 3D co-culture model of early peritoneal endometriosis using patient-derived primary cells, providing unique opportunities to examine endometriotic lesion initiation and progression. The successful assembly of a simple peritoneum layer model comprising a mesothelial monolayer, basement membrane and underlying f

Three-Dimensional Organoid-like Co-Culture of Human Endometrial Endothelial and Stromal Cells to Study Endometriosis-Associated Responses.

Three-dimensional (3D) endothelium-stromal co-cultures were established using human endometrial cells from biopsy of healthy women (n = 13) and serum samples from both healthy and endometriotic women (n = 5). For 3D construction, stromal cells were mixed with extracellular matrix components, followed by endothelial cell seeding. Morphological analysis confirmed the organization of tissue-like structures. Immunofluorescence and flow cytometry verified the expression of specific stromal and endothelial markers (Cytokeratin, Vimentin, Insulin-like growth factor-binding protein 1, and von Willebra

Recapitulating Glioma Stem Cell Niches Using 3D Spheroid Models for Glioblastoma Research.

Glioblastoma multiforme (GBM) is among the most aggressive brain cancers, and it contains glioma stem cells (GSCs) that drive tumor initiation, progression, and recurrence. These cells resist conventional therapies, contributing to high recurrence rates in GBM patients. Developing in vitro models that mimic the tumor microenvironment (TME), particularly the GSC niche, is crucial for understanding GBM growth and therapeutic resistance. Three-dimensional (3D) spheroid models provide a more physiologically relevant approach than traditional two-dimensional (2D) cultures, recapitulating key tumor
