Transcriptomic response to compressive force simulating orthodontic tooth movement in cultured porcine epithelial cell rests of Malassez.

Mechanical stress on the periodontal ligaments, such as orthodontic force, causes the failure of homeostasis-inducing inflammation leading to inflammatory root resorption and replacement resorption. Despite these developments during orthodontic treatment, little is known about its relationship to the epithelial cell rests of Malassez (ERM) located in the periodontal ligament. To address this gap, RNA sequencing was performed on non-compression and compression forced porcine ERM groups subjected to 4.0 g/cm2 for 12 h. The results showed 529 upregulated and 273 downregulated genes with more than

Tetraspanin CD82 interaction with cholesterol promotes extracellular vesicle-mediated release of ezrin to inhibit tumour cell movement.

Tumour metastasis suppressor KAI1/CD82 inhibits tumour cell movement. As a transmembrane protein, tetraspanin CD82 bridges the interactions between membrane microdomains of lipid rafts and tetraspanin-enriched microdomains (TEMs). In this study, we found that CD82 and other tetraspanins contain cholesterol recognition/interaction amino-acid consensus (CRAC) sequences in their transmembrane domains and revealed that cholesterol binding of CD82 determines its interaction with lipid rafts but not with TEMs. Functionally, CD82 needs cholesterol binding to inhibit solitary migration, collective mig

WASP and WAVE family proteins: key molecules for rapid rearrangement of cortical actin filaments and cell movement.

Reorganization of cortical actin filaments plays critical roles in cell movement and pattern formation. Recently, the WASP and WAVE family proteins WASP and N-WASP, and WAVE1, WAVE2 and WAVE3 have been shown to regulate cortical actin filament reorganization in response to extracellular stimuli. These proteins each have a verprolin-homology (V) domain, cofilin-homology (C) domain and an acidic (A) region at the C-terminus, through which they activate the Arp2/3 complex, leading to rapid actin polymerization. N-WASP is usually present as an inactive form in which the VCA region is masked. Coope

Accelerated organelle movements can indirectly affect the motility of other organelles in plant cells.

Organelle movements in plants are driven by myosin XI motors moving along the actin filament cytoskeleton. Several experiments have suggested that indirect effects such as organelle-organelle interactions or hydrodynamic flow might be responsible for organelle movements. To test these models, we altered the motile behavior of specific organelles in Nicotiana benthamiana leaf epidermal cells and examined the effects of these perturbations on other organelles. Recruiting myosins to organelles with 'Booster' constructs resulted in significant increases of linear motions, whereas binding organelle

Excitability of Anterior Horn Cells Following Repetitive Knee Joint Movements at Different Speeds.

Introduction F-waves are compound muscle action potentials generated by the antidromic activation of alpha motor neuron axons and the backfiring of anterior horn cells in response to electrical stimulation. Several F-wave parameters are used to assess the state of alpha motor neurons. Previous studies have demonstrated that repetitive thumb movements can decrease the F/M amplitude ratio in the abductor pollicis brevis muscle of healthy individuals. This suggests that repetitive movements may be a practical and accessible self-training method for alleviating muscle hypertonia. However, it remai

Anterior Chamber XEN Gel Stent Movements: The Impact on Corneal Endothelial Cell Density.

INTRODUCTION: Although there are several examples of glaucoma drainage devices' dislocations in the literature, the movement of XEN gel stents is scarcely documented, and the impact of such displacements on the corneal endothelium remains unknown. In light of the recent global market withdrawal of the CyPass microstent over concerns with regard to endothelial cell loss, we present a case of XEN gel stent dislocation into the anterior chamber, with localized corneal damage. CASE REPORT: We describe the case of a 64-year-old female patient who underwent bilateral XEN gel stent implantation for

Purkinje cell responses during visually and vestibularly driven smooth eye movements in mice.

INTRODUCTION: An essential complement to molecular-genetic approaches for analyzing the function of the oculomotor circuitry in mice is an understanding of sensory and motor signal processing in the circuit. Although there has been extensive analysis of the signals carried by neurons in the oculomotor circuits of species, such as monkeys, rabbits and goldfish, relatively little in vivo physiology has been done in the oculomotor circuitry of mice. We analyzed the contribution of vestibular and nonvestibular signals to the responses of individual Purkinje cells in the cerebellar flocculus of mic

Embryonic development of the rat cerebellum. I. Delineation of the cerebellar primordium and early cell movements.

Short-survival and long-survival thymidine radiograms, and methacrylate-embedded tissue from normal and X-irradiated rat embryos were used to delineate the neuroepithelial source of the cerebellum and trace the earliest cell movements. The cerebellar anlage, crescent shaped, is demarcated by two ventricular landmarks, the anterior extension of the tela choroidea of the fourth ventricle and the embryonic cerebellar fissure. The cerebellar tela choroidea extends from the medullary fourth ventricle posteromedially to the lateral recess of the pontine fourth ventricle anterolaterally. The embryoni

RANKL deletion in periodontal ligament and bone lining cells blocks orthodontic tooth movement.

The bone remodeling process in response to orthodontic forces requires the activity of osteoclasts to allow teeth to move in the direction of the force applied. Receptor activator of nuclear factor-κB ligand (RANKL) is essential for this process although its cellular source in response to orthodontic forces has not been determined. Orthodontic tooth movement is considered to be an aseptic inflammatory process that is stimulated by leukocytes including T and B lymphocytes which are presumed to stimulate bone resorption. We determined whether periodontal ligament and bone lining cells were an es

Effect of phenethyl isothiocyanate on Ca2+ movement and viability in MDCK canine renal tubular cells.

The effect of the natural compound phenethyl isothiocyanate (PEITC) on cytosolic Ca(2+) concentrations ([Ca(2+)](i)) and viability in MDCK renal cells is unknown. This study explored whether PEITC changed [Ca(2+)](i) in MDCK cells using the Ca(2+)-sensitive fluorescent dye fura-2. PEITC at 200-700 μM increased [Ca(2+)](i) in a concentration-dependent manner. The signal was reduced by removing extracellular Ca(2+). PEITC-induced Ca(2+) influx was inhibited by nifedipine, econazole, SK&F 96365 and protein kinase C modulators. In Ca(2+)-free medium, treatment with the endoplasmic reticulum Ca(2+)

Sustained and transient discharges of retinal ganglion cells during spontaneous eye movements of cat.

Discharges of 223 retinal ganglion cells during spontaneous eye movements (saccades) across a stationary grating pattern were studied in chronically prepared cats. Of these 83 showed sustained responses to local differences in luminance of the grating stripes (S-units); 84 showed transient responses to saccades and did not register local differences in luminance (T-units); and 56 showed mixed responses, i.e., transient responses to saccades and sustained firings in response to local luminance (M-units). When tested with diffuse light, 93.9% of the S-units showed either ON-sustained or OFF-sust

Feeding embryonated eggs of Heterakis papillosa to brooder chickens led to a disease of both ceca, characterized by the presence of a core consisting of fecal matter, coagulated blood, and emigrated cells from the mucosa. The walls of the ceca were thickened as a result of cell invasion and multiplication, invasion and multiplication of Amoeba meleagridis or allied parasites, and more rarely hemorrhage and edema. The respective parts played by Heterakis papillosa and the protozoa in starting the lesions and the source of the protozoa remain to be defined. The invasion of the liver by the proto

The MARCKS protein plays a critical role in phosphatidylinositol 4,5-bisphosphate metabolism and directed cell movement in vascular endothelial cells.

The MARCKS protein (myristoylated alanine-rich C kinase substrate) is an actin- and calmodulin-binding protein that is expressed in many mammalian tissues. The role of MARCKS in endothelial signaling responses is incompletely understood. We found that siRNA-mediated knockdown of MARCKS in cultured endothelial cells abrogated directed cell movement in a wound healing assay. We used biochemical and cell imaging approaches to explore the role of MARCKS in endothelial signal transduction pathways activated by insulin. Insulin treatment of vascular endothelial cells promoted the dose- and time-depe

Cell movement and the mechanism of invasiveness: a survey of the behaviour of some normal and malignant cells implanted into the developing chick wing bud.

A survey of the behaviour of a variety of normal and malignant tumours and cells has been carried out to gain insights into the mechanisms of tumour invasiveness. The tumours and cells were implanted into the developing chick wing bud, which is a loose mesenchyme bounded by ectoderm. The distribution of the grafted cells was examined histologically after one or two days. The special feature of this assay is that the behaviour of cells is tested in a 3-dimensional tissue. Cells from 3 different carcinomas, mouse lung tumour, rat bladder tumour and human breast tumour did not invade the mesenchy

CD4+ T-cell transcription factors as early predictors of phenoconversion in idiopathic rapid eye movement sleep behaviour disorder.

In this datanote we present data from 31 iRBD (idiopathic Rapid eye movement (REM) sleep Behaviour Disorder) patients followed throughout three years to assess their eventual phenoconversion to Parkinson's disease and other established neurodegenerative conditions. iRBD is a prodromal condition of neurological pathologies such as Parkinson's disease. We evaluated transcription factor mRNA levels in CD4+ T cells as predictive biomarkers of phenoconversion in iRBD, showing that among the transcription factors mRNA levels analysed, STAT1, GATA3 and FOXP3 mRNA levels may be used to predict phenoco

Cell microparticles loaded with pro-inflammatory factors and chloroquine collaborate with mechanical force to facilitate orthodontic tooth movement.

Malocclusion is regarded as a serious dental problem involving above 60 % populations worldwide. The main challenge for clinical orthodontic treatment lies in insufficient remodeling microenvironment, lack of active immune response and inadequate osteoclastic behavior, as mechanical application fails to trigger high-performance periodontal reconstruction. Regulation on macrophages is a promising method for strengthening tissue remodeling. Here, pro-inflammatory macrophage-derived microparticles loaded with chloroquine (CQ@PMMPs) are fabricated and integrated into a pH-responsive microneedle pa

Listeria monocytogenes-infected human umbilical vein endothelial cells: internalin-independent invasion, intracellular growth, movement, and host cell responses.

The interaction of Listeria monocytogenes with human umbilical vein endothelial cells was studied. We show that L. monocytogenes invades human umbilical vein endothelial cells independently of internalin A, internalin B, internalin C, and ActA. L. monocytogenes replicates efficiently inside the cells and moves intracellularly by the induction of actin polymerization. We further show that L. monocytogenes-infection of human umbilical vein endothelial cells induces interleukin-6 and interleukin-8 expression during the first 6 h of infection. The expression of MCP-1 and the adhesion molecules VCA

In Vitro Compression Model for Orthodontic Tooth Movement Modulates Human Periodontal Ligament Fibroblast Proliferation, Apoptosis and Cell Cycle.

Human Periodontal Ligament Fibroblasts (hPDLF), as part of the periodontal apparatus, modulate inflammation, regeneration and bone remodeling. Interferences are clinically manifested as attachment loss, tooth loosening and root resorption. During orthodontic tooth movement (OTM), remodeling and adaptation of the periodontium is required in order to enable tooth movement. hPDLF involvement in the early phase-OTM compression side was investigated for a 72-h period through a well-studied in vitro model. Changes in the morphology, cell proliferation and cell death were analyzed. Specific markers o
