[Mechanisms of neuro-tropic disorders induced in skeletal muscles by botulinum toxin].

The relationship between activation of cell nucleus division and disturbances of electrogenesis of skeletal muscle fibers which had been paralyzed by botulinum toxin was studied in experiments on rats. On the 5th day after its intramuscular injection botulinum toxin evoked depolarization of muscle fibers and appearance of local responses in addition to break action potentials (AP) in most of the fibers. Inhibition of DNA synthesis by daily intraperitomeal injection of fluorouracil prevented the decreasing of rest membrane potentials and appearance of anode break AP.

Efferent connections of lobule IX of the posterior cerebellar cortex in the rabbit--some functional considerations.

The Purkinje cell projection from the cardiovascular region of sublobule b of the uvula (medial area of zone A) has been investigated using anterograde tracing methods in the rabbit. The importance of the integrity of the identified pathways in mediating the cardiovascular responses from the uvula has been studied in subsequent lesioning experiments. Wheat germ agglutinin-conjugated horseradish peroxidase or tritiated amino acids were microinjected into sublobule IXb. This resulted in anterogradely labelled Purkinje cell axons in both the inferior and superior cerebellar peduncle. In agreement

Anti-inflammatory activity of chloroquine and amodiaquine through p21-mediated suppression of T cell proliferation and Th1 cell differentiation.

Chloroquine (CQ) and amodiaquine (AQ) have been used for treating or preventing malaria for decades, and their application has expanded into treating inflammatory disease in humans. CQ and AQ are applicable for controlling rheumatoid arthritis, but their molecular mechanisms of anti-inflammatory activity remain to be elucidated. In this study, we examined the effects of CQ and AQ on T cell activation and T cell-mediated immune response. CQ had no significant effect on T cell numbers, but decreased the population of T cells with a high division rate. However, AQ treatment significantly increase

Direct link from the suprachiasmatic nucleus to hypothalamic neurons projecting to the spinal cord: a combined tracing study using cholera toxin subunit B and Phaseolus vulgaris-leucoagglutinin.

By combining retrograde and anterograde tracing, evidence for a bineuronal connection from the suprachiasmatic nucleus (SCN) to the intermediolateral cell column in the spinal cord (IML) was obtained. The retrograde tracer cholera toxin subunit B (ChB) was pressure-injected into the spinal cord and the anterograde tracer Phaseolus vulgaris-leucoagglutinin (PHA-L) was iontophoretically injected into the SCN. The two tracers were visualized simultaneously by a double immunohistochemical procedure. In the hypothalamus, ChB injections gave rise to retrogradely labeled cell bodies in the paraventri

Characterization of McDonald's intermediate part of the Central nucleus of the amygdala in the rat.

The actual organization of the central nucleus of the amygdala (CEA) in the rat is mostly based on cytoarchitecture and the distribution of several cell types, as described by McDonald in 1982. Four divisions were identified by this author. However, since this original work, one of these divisions, the intermediate part, has not been consistently recognized based on Nissl-stained material. In the present study, we observed that a compact condensation of retrogradely labeled cells is found in the CEA after fluorogold injection in the anterior region of the tuberal lateral hypothalamic area (LHA

The distribution of substance P-containing neurons in the cat Edinger-Westphal nucleus: relationship to efferent projection systems.

The light microscopic localization of substance P-like immunoreactivity (SPLI) was examined in the cat Edinger--Westphal complex using the peroxidase--antiperoxidase method. A high density of cell bodies and processes staining for SPLI were found in the caudal part of the Edinger--Westphal complex (EWc) capping the somatic divisions of the oculomotor nucleus. This distribution continued rostrally into the anteromedian nucleus (AM). Cells labeled with SPLI were also found arranged in a thin layer dorsally capping the oculomotor nucleus, and scattered cells were found in the periaqueductal gray

A single-nucleus atlas of ligular region uncovers brassinosteroid-driven cell fate transitions controlling leaf angle in maize.

The ligular region is a critical determinant of leaf angle (LA) and plant architecture in maize, yet its developmental mechanisms remain largely unknown. In this study, we generate a comprehensive single-nucleus RNA-seq atlas of the maize ligular region, spanning from leaf primordium through the preligule band (PLB) to the mature stage. By integrating cellular and transcriptomic analyses, we reveal that brassinosteroid (BR) inhibits cell division in the adaxial division zone while promoting elongation in adaxial hypodermal cells. Consequently, BR treatment leads to ectopic emergence of the aur

GmRAV1 regulates regeneration of roots and adventitious buds by the cytokinin signaling pathway in Arabidopsis and soybean.

The division and differentiation of cells are the basis of growth and development. Cytokinin plays an active role in cell growth division and differentiation. The Related to ABI3/VP1 (RAV) family comprises transcription factors in plants and all contain both AP2- and B3-like domains. In this study, GmRAV1 (Glycine max), which belongs to the AP2/ERF transcription factor family, was isolated and functionally characterized. Subcellular localization showed that GmRAV1 was localized to the nucleus and quantitative real-time polymerase chain reaction (qRT-PCR) analysis indicated that GmRAV1 was indu

[Morphologic features of the binucleated lymphocytes for the assesment of persistent B-cell polyclonal lymphocytosis].

Observation of binucleated lymphocytes on a blood film is a cornerstone event to assess persistent polyclonal B-cell lymphocytosis diagnosis (PPBL). But seldom case reports mentioned their finding in other contexts, mainly B-cell lymphoproliferative neoplasms such chronic lymphocytic leukemia or in reactive circumstances as multiple sclerosis undergoing natalizumab treatment. We aimed to check if some particular morphologic features were more specificaly linked to PPBL, reviewing blood films of series of patients affected by PPBL or harbouring binucleated lymphocytes whatever the diagnosis was

Beta-Arrestin-2 Binds to Nucleolar Organizing Regions With P2X7 Receptor Stimulation and During Cell Division.

Nonvisual arrestins, also known as Beta-arrestins, are known for their diverse role in GPCR signal transduction. Here, we present evidence that Beta-Arrestin-2, a protein whose nuclear concentration is actively kept low, binds to Nucleolar Organizing Regions (NORs) on the metaphase chromosomes at the onset of cell division, after the disappearance of the nuclear membrane and stays bound therein in the daughter cells. After reformation of the nucleoli, it stays bound to the nucleoli until actively transported out of the reformed nucleus during interphase. This binding is unique for Beta-Arresti

Phospho-Tau Protein Expression in the Cell Cycle of SH-SY5Y Neuroblastoma Cells: A Morphological Study.

It has been reported that the main function of tau protein is to stabilize microtubules and promote the movement of organelles through the axon in neurons. In Alzheimer's disease, tau protein is the major constituent of the paired helical filament, and it undergoes post-translational modifications including hyperphosphorylation and truncation. Whether other functions of tau protein are involved in Alzheimer's disease is less clear. We used SH-SY5Y human neuroblastoma cells as an in vitro model to further study the functions of tau protein. We detected phosphorylated tau protein as small dense

G-protein-coupled receptors signalling at the cell nucleus: an emerging paradigm.

G-protein-coupled receptors (GPCRs) comprise a wide family of monomeric heptahelical glycoproteins that recognize a broad array of extracellular mediators including cationic amines, lipids, peptides, proteins, and sensory agents. Thus far, much attention has been given towards the comprehension of intracellular signaling mechanisms activated by cell membrane GPCRs, which convert extracellular hormonal stimuli into acute, non-genomic (e.g., hormone secretion, muscle contraction, and cell metabolism) and delayed, genomic biological responses (e.g., cell division, proliferation, and apoptosis). Central and peripheral expression sites of phoenixin-14 immunoreactivity in rats.

Phoenixin is a pleiotropic peptide involved in reproduction, anxiety and recently also implicated in the control of food intake. Besides the 20-amino acid phoenixin, the 14-amino acid phoenixin-14 also shows bioactive properties. However, the expression sites of phoenixin-14 in the brain and peripheral tissues are not yet described in detail. Therefore, a mapping of the brain and peripheral tissues from male and female Sprague-Dawley rats with a specific phoenixin-14 antibody was performed using western blot and immunohistochemistry. High density of phoenixin-14 immunoreactivity was detected i

Cell proliferation rate and nuclear morphometry in Roberts syndrome.

Roberts syndrome (RS) is a rare autosomal recessive disorder characterized primarily by symmetric reduction anomalies of all limbs, growth retardation and craniofacial abnormalities. Most RS patients are reported to present a typical abnormality of their constitutive heterochromatin, accompanied by abnormal cytological growth characteristics. We present an extremely severe case of an RS fetus, karyotypically documented, with a clinical presentation including growth deficiency, tetraphocomelia, frontal meningocele, craniofacial abnormalities and penile enlargement with hypospadias. Nuclear morp

Essential nucleus-apical pole linkage maintains division fidelity during Plasmodium progeny formation.

Plasmodium parasites, the causative agents of malaria, undergo complex replication within vertebrate and insect hosts, presenting unique opportunities for therapeutic intervention. A key challenge during these replication events, i.e., schizogony in vertebrate red blood cells and sporogony in oocysts within mosquitos, is ensuring the faithful partitioning of nuclei and organelles into the numerous daughter cells that form at once from a single parental cell. While nuclear microtubule-organizing centers, or centriolar plaques (CPs), have been hypothesized to play a central role in this process,

Disseminated lobular carcinoma - a predominantly pleomorphic lobular carcinoma of the whole breast.

Contrary to the usual distribution of lobular carcinoma in situ in a few lobules or terminal ducts, a case of disseminated lobular carcinoma is described occupying all lobules of the whole breast. The tumor developed in the left breast of a 61-year-old woman with the clinical symptoms of an increasing diffuse induration. The proliferating cells were predominantly pleomorphic with hyperchromatic nuclei and loss of cell cohesion. In 690 slides examined the tumor cells in lobules and ducts revealed various stages of proliferation with transition to invasive small cell carcinoma in 15 different ar

The human medial geniculate body.

The medial geniculate body in non-human species is divided into several parts, each with a different structure, physiological organization, and pattern of connections. Which parts of the human medial geniculate body and which types of neurons might be homologous to those of other species is unknown, and the object of the present study. The cytoarchitecture, fiber architecture, and neuronal organization of the adult human medial geniculate body were studied in Nissl, Golgi, and other preparations. Three divisions, comparable to those in other mammals, were described. The ventral division had a

Cholinergic neurons in the brain of a teleost fish (Porichthys notatus) located with a monoclonal antibody to choline acetyltransferase.

A monoclonal antibody (Ab8) to choline acetyltransferase (ChAT) was used to locate structures showing ChAT-like immunoreactivity (ChAT-IR) in the brain of a teleost fish, the midshipman (Porichthys notatus). ChAT is the synthetic enzyme for acetylcholine found in neurons using that neurotransmitter; thus ChAT-IR may be interpreted as indicating putative cholinergic activity. Robust staining is seen in all cranial nerve motor nuclei. In addition, the brainstem of Porichthys is distinguished by two other expansive ChAT-IR zones: a sonic motor nucleus, which innervates swimbladder "drum" muscles,

Previously Unidentified Histone H1-Like Protein Is Involved in Cell Division and Ribosome Biosynthesis in Toxoplasma gondii.

Chromatin dynamics can regulate all DNA-dependent processes. Access to DNA within chromatin is orchestrated mainly by histones and their posttranslational modifications (PTMs). Like other eukaryotes, the apicomplexan parasite Toxoplasma gondii encodes four canonical histones and five histone variants. In contrast, the linker histone (H1) has never been identified in apicomplexan parasites. In other eukaryotes, histone H1 compacts the chromatin by linking the nucleosome and increasing the DNA compaction. H1 is a multifunctional protein and can be involved in different steps of DNA metabolism or
