Morphogenesis and morphology of the brain stem nuclei of Cetacea. II. The nuclei of the accessory, vagal and glossopharyngeal nerves in baleen whales.

The development and final structure of the IXth, Xth and XIth cranial nerve nuclei are studied in ironhematoxylin -, thionin - and protargol -stained serial sections of about 50 baleen whale fetuses (blue whale, Balaenoptera musculus, and fin whale, Balaenoptera physalus ) and one adult fin whale. The nucleus ambiguus is composed of three subdivisions, oral, intermediate and caudal, the last mentioned being contiguous caudally with the dorsal motor Xth nucleus. The oral division develops as three parallel cell columns which merge into a well circumscribed solitary structure with a rostrally ex

Aspects of the fine anatomy of aphthous stomatitis.

Samples of human aphthous stomatitis were studied under the electron microscope. The epithelial cells bordering the ulcer present multivesicular bodies, pinocytotic vesicles, primary lysosomes, digestive vacuoles, and autophagic vacuoles. The golgicomplex is noticeably increased; free ribosomes and rough endoplasmic reticulum are abundant. Crystalline structures are close to the nucleus. One or two paranuclear bodies are in most of the nuclei. A frank cell injury is evident, causing a major activity of the digestive cell system; the origin and formation of lysosomes through r.e.r. and the Golg

Central projections of the lateral line and eighth nerves in the bowfin, Amia calva.

The first-order connections of the anterior and posterior lateral line nerves and of the eighth nerve were determined in the bowfin, Amia calva, using experimental degeneration and anterograde HRP transport techniques. The termination sites of these nerves define a dorsal lateralis cell column and a ventral octavus cell column. The anterior and posterior lateralis nerves distribute ipsilaterally to two medullary nuclei-nucleus medialis and nucleus caudalis. Nucleus medialis comprises the rostral two-thirds of the lateralis column and contains large, Purkinje-like cells dorsally and polygonal,

Different effects of pemphigus antibody and plasmin on the distribution of keratin intermediate filaments and desmoplakins between cultured oral and epidermal keratinocytes.

In order to clarify the molecular mechanism of blister formation in oral mucosa in pemphigus vulgaris (PV) comparing with that in epidermis, we analyzed the effects of PV serum on the distribution of keratin intermediate filaments (KIFs) and desmoplakins in oral as well as epidermal cultured keratinocytes by immunofluorescence microscopy using anti-keratin and anti-desmoplakin I/II monoclonal antibodies. After incubation with PV serum for 96 h at 37 degrees C, clusters of anti-keratin positive dots were formed around the nucleus in some of the keratinocytes from normal gingiva and soft palate

A novel nuclear trafficking module regulates the nucleocytoplasmic localization of the rabies virus interferon antagonist, P protein.

Regulated nucleocytoplasmic transport of proteins is central to cellular function and dysfunction during processes such as viral infection. Active protein trafficking into and out of the nucleus is dependent on the presence within cargo proteins of intrinsic specific modular signals for nuclear import (nuclear localization signals, NLSs) and export (nuclear export signals, NESs). Rabies virus (RabV) phospho (P) protein, which is largely responsible for antagonising the host anti-viral response, is expressed as five isoforms (P1-P5). The subcellular trafficking of these isoforms is thought to d

Cell-type specific parallel circuits in the bed nucleus of the stria terminalis and the central nucleus of the amygdala of the mouse.

The central extended amygdala (EAc) is a forebrain macrosystem which has been widely implicated in reward, fear, anxiety, and pain. Its two key structures, the lateral bed nucleus of the stria terminalis (BSTL) and the central nucleus of the amygdala (CeA), share similar mesoscale connectivity. However, it is not known whether they also share similar cell-specific neuronal circuits. We addressed this question using tract-tracing and immunofluorescence to reveal the EAc microcircuits involving two neuronal populations expressing either protein kinase C delta (PKCδ) or somatostatin (SOM). Fiber connections of the central nucleus of semicircular torus in cyprinids.

Fiber connections of the presumed auditory portion of the semicircular torus or the central nucleus (TSc) as well as other central and peripheral auditory structures were studied by tract-tracing methods in carp and goldfish. Major ascending projections to the TSc originated from the descending octaval nucleus (DO) and medullary secondary octaval population (SO). Toropetal neurons of the DO were within the saccular terminal zone. A small number of toropetal DO neurons might receive inputs from other inner ear and lateral line endorgans as well. Fibers from the DO terminated in a deep zone of t

Distribution of FMRFamide-like immunoreactivity in the brain of the elasmobranch fish Scyliorhinus canicula.

The distribution of FMRFamide-like-immunoreactive peptides was investigated in the brain and pituitary of the elasmobranch fish Scyliorhinus canicula using the indirect immunofluorescence technique. FMRFamide-immunoreactive cells and fibers were mainly observed in the telencephalon and the diencephalon, while other brain structures were almost unstained. In the telencephalon, FMRFamide-like-containing neurons were seen in the caudal part of the area periventricularis pallialis, in the posterior area of the nucleus septi medialis and in the nucleus septi caudoventralis. In the diencephalon, num

[Optimization of phacoemulsification technique for Morgagnian cataract].

PURPOSE: This study was conducted to develop a new optimized phacoemulsification technique for Morgagnian cataract taking into account the anatomical and topographic parameters of the lens nucleus. MATERIAL AND METHODS: A working classification of Morgagnian cataract was developed based on the size of the nucleus: if the edge of the nucleus is visualized at the upper edge of the pupil or between the upper edge and the middle of the pupil, it was classified as an initial stage of Morgagnian cataract with a large nucleus; if the upper edge of the nucleus is visualized in the middle of the pupil

Projections from the supragenual nucleus to the lateral mammillary and dorsal tegmental nuclei.

An organism's sense of direction depends on vestibular input to thalamic and forebrain structures. The supragenual nucleus (SGN) sits at a prime location in the network to convey vestibular signals forward to update the representation of head direction (HD) with ongoing head movement. The SGN receives anatomical projections from the medial vestibular nuclei and nucleus prepositus hypoglossi and prior reports have suggested that the SGN sends projections to both the dorsal tegmental nucleus (DTN) and lateral mammillary nucleus (LMN), two midbrain nuclei crucial in generating a representation of

Central projections of the mesencephalic nucleus of the fifth nerve: an autoradiographic study.

Projections of cells of the mesencephalic nucleus of the fifth nerve (Mes V) to brainstem structures in the cat were studied by labelling Mes V cells with tritiated leucine. Before the leucine injections were made, however, large kainic acid lesions were produced in the vicinity of Mes V cells because these neurons are resistant to being killed or injured by this neurotoxin. Thus Mes V cells were selectively labelled by the leucine even though they are scattered among many other neurons. Leucine injections near Mes V cells located in the mesencephalon, which are primarily the somata of jaw mus

Dense-cored vesicle-containing components of the terminal nerve of sharks and rays.

This paper describes electron microscopic observations of dense-cored vesicle-containing axons, cell bodies, and endings of the terminal nerve in several elasmobranchs. The vesicles are found in two apparent cell types, one with a polymorphic nucleus and another with an oval nucleus. The types may correspond to cells producing one each of two neuropeptides (LHRH and FMRF-amide) that have previously been localized in the nerve. Dense-cored vesicles are found in many unmyelinated fibers in both the terminal nerve proper and its major ganglia. Some of these form complicated structures with interd

N-cadherin expression in motoneurons is directly regulated by androgens: a genetic mosaic analysis in rats.

We have recently reported that systemic androgens regulate adult N-cadherin (N-cad) expression in spinal motoneurons. However, the mechanism through which androgen mediates this effect remains undetermined. Androgen may act directly on motoneurons to regulate N-cad expression, or indirectly, via effects on androgen-sensitive afferent or efferent structures. Here, we describe a genetic mosaic investigation of this site-of-action indeterminacy. Following developmental random X chromosome inactivation, androgenized female rats heterozygous for the tfm androgen receptor mutation (X(WT)X(tfm)) are

Generation of a laminopathies-specific iPSC line EHTJUi005-A-3 with homozygous knockout of the LMNA gene by CRISPR/Cas9 technology.

LAMIN A/C, encoded by the LMNA gene, supports the normal structure of the cell nucleus and regulates the connection between the nucleus and the cytoskeleton as a component of the nucleus envelope. The loss of expression and function of the LMNA gene would lead to the occurrence of congenital muscular dystrophy and Emery-Dreifuss muscular dystrophy which are collectively named as laminopathies. Here, we report a human induced pluripotent stem cell (iPSC) line (EHTJUi005-A-3) generated from a wild iPSC (EHTJUi005-A) with homozygous knockout of the gene LMNA through CRISPR/Cas9. This iPSC line pr

Distribution of neurons containing phenylethanolamine N-methyltransferase in medulla and hypothalamus of rat.

Neurons immunocytochemically labeled with the adrenaline-synthesizing enzyme phenylethanolamine N-methyltransferase were mapped in the brain of rat pretreated with colchicine. In medulla, immunoreactive cells in the C1 and C2 groups were distributed in a more complex manner than described previously. C1 neurons were identified in the reticular formation of ventrolateral medulla and were organized into two populations: (1) a cell column extending throughout the ventrolateral medulla, and lying ventral to the ambiguus cell group and either dorsal to the precerebellar lateral reticular nucleus or

[Dynamics of changes in a population of human fibroblasts after long-term cultivation in the stationary phase].

Defined and repeated alterations in human fibroblasts were discovered under continuous cultivation in the stationary phase. In the normal diploid fibroblast population, two cell subpopulations were distinguished by the heterochromatinization degree of the nucleus. While in the stationary phase, the number of cells with a parahaploid number of large chromocenters, regularly arranged in the nucleus, is seen increasing. It is supposed that the death of the normal diploid fibroblast culture occurs primarily at the expence of cells with heterochromatinized nuclei. Examination of strains with trisom

Projections of the ventral subiculum to the amygdala, septum, and hypothalamus: a PHAL anterograde tract-tracing study in the rat.

The projections of the ventral subiculum are organized differentially along the dorsoventral (or septotemporal) axis of this cortical field, with more ventral regions playing a particularly important role in hippocampal communication with the amygdala, bed nuclei of the stria terminalis (BST), and rostral hypothalamus. In the present study we re-examined the projection of the ventral subiculum to these regions with the Phaseolus vulgaris leucoagglutinin (PHAL) method in the rat. The results confirm and extend earlier conclusions based primarily on the autoradiographic method. Projections from

Cells of origin of pathways descending to the spinal cord in two chondrichthyans, the shark Scyliorhinus canicula and the ray Raja clavata.

The cells of origin of pathways descending to the spinal cord in the shark Scyliorhinus canicula and in the ray Raja clavata have been demonstrated by using the horseradish peroxidase (HRP) technique. Following HRP injections in the spinal cord of Scyliorhinus (fourth to sixth segment) and of Raja (15th to 20th segment) labeled neurons could be identified in the rhombencephalon, the mesencephalon, and in the diencephalon. Cells of origin of diencephalic nuclei, which project to the spinal cord, were observed in the nucleus periventricularis hypothalami and in the thalamus ventralis pars medial

Molecular Basis of Functional Effects of Phosphorylation of the C-Terminal Domain of the Rabies Virus P Protein.

The rabies virus (RABV) phosphoprotein (P protein) is expressed as several isoforms, which differ in nucleocytoplasmic localization and microtubule (MT) association, mediated by several sequences, including nuclear localization (NLS) and export (NES) sequences. This appears to underpin a functional diversity enabling multiple functions in viral replication and modulation of host biology. Mechanisms regulating trafficking are poorly defined, but phosphorylation by protein kinase C (PKC) in the P protein C-terminal domain (PCTD) regulates nuclear trafficking, mediated by PCTD-localized NLS/NES s

Bioimaging insights into structural pathways of cell-to-cell communication within the male (MGU) and female (FGU) germ units of Arabidopsis thaliana.

Cytoplasmic connections are present between cells within male and female germ units (MGU, FGU), suggesting potential structural pathways for communication. Cell-to-cell communication within the male germ unit (MGU), which consists of two sperm cells and the vegetative cell nucleus, and the female germ unit (FGU), comprising the synergids, the egg cell, and the central cell, is crucial for gamete maturation, fertilization, and early embryogenesis in angiosperms. The MGU facilitates the transport and delivery of immotile sperm cells via the elongating pollen tube to the FGU/embryo sac, which is
