Existence of long-lasting experience-dependent plasticity in endocrine cell networks.

Experience-dependent plasticity of cell and tissue function is critical for survival by allowing organisms to dynamically adjust physiological processes in response to changing or harsh environmental conditions. Despite the conferred evolutionary advantage, it remains unknown whether emergent experience-dependent properties are present in cell populations organized as networks within endocrine tissues involved in regulating body-wide homeostasis. Here we show, using lactation to repeatedly activate a specific endocrine cell network in situ in the mammalian pituitary, that templates of prior de

Molecular control of vascular smooth muscle cell differentiation and phenotypic plasticity.

Although the primary role of vascular smooth muscle cells (SMCs) is contraction, they exhibit extensive phenotypic diversity and plasticity during normal development, during repair of vascular injury, and in disease states including arteriosclerosis and tumour angiogenesis. Results of recent studies indicate that there are unique as well as common transcriptional regulatory mechanisms that control expression of various SMC marker genes within vascular SMC subtypes, and that these mechanisms are complex and dynamic even at the single cell level. This chapter will review recent progress in our u

Physiology, pharmacology and plasticity at the inner hair cell synaptic complex.

This report summarizes recent neuropharmacological data at the IHC afferent/efferent synaptic complex: the type of Glu receptors and transporter involved and the modulation of this fast synaptic transmission by the lateral efferents. Neuropharmacological data were obtained by coupling the recording of cochlear potentials and single unit of the auditory nerve with intra-cochlear applications of drugs (multi-barrel pipette). We also describe the IHC afferent/efferent functioning in pathological conditions. After acoustic trauma or ischemia, acute disruption of IHC-auditory dendrite synapses are

Plasticity of dorsal horn cell receptive fields after peripheral nerve regeneration.

1. The tibial and sural nerves were transected and repaired in nine adult cats. The receptive field (RF) properties of dorsal horn neurons were examined at three different intervals (5-6, 9, or 12 mo) after axotomy. The properties examined included RF location, area, and modality convergence. In some cases, discrete areas of the cell's RF were stimulated electrically while the evoked cord dorsum potentials (CDPs) and any intracellularly recorded responses were simultaneously recorded. 2. At the shortest interval following reinnervation, the somatotopic organization in the affected areas of the

From gatekeepers to regenerators: the plasticity of Paneth cells.

Paneth cells (PCs) are secretory cells in the small intestine with functions in intestinal homeostasis and innate immunity, with recent studies showing their ability to dedifferentiate and contribute to epithelial regeneration after injury. In this issue of The EMBO Journal, Yang et al, (2026) identify reactive oxygen species (ROS) as a central determinant of PC identity and uncover the Rho-family GTPase CDC42 as a critical regulator of this process.

Metabolic reprogramming and plasticity of cancer stem cells.

Numerous studies in cancer biology have provided evidence of the remarkable plasticity of cancer stem cells (CSCs). These cells play a key role in tumor initiation, metastasis, and resistance to treatment. One of the most critical features of CSCs is the epithelial-mesenchymal transition, which underlies their phenotypic plasticity. CSCs can modify their metabolic profile through interactions with cancer-associated fibroblasts, tumor-associated macrophages, and regulatory t-cells. CSCs creates a multifaceted cancerous environment that adapts to extensive changes in the secretome, variability o

Therapeutic plasticity of stem cells and allograft tolerance.

Transplantation is the treatment of choice for many diseases that result in organ failure, but its success is limited by organ rejection. Stem cell therapy has emerged in the last years as a promising strategy for the induction of tolerance after organ transplantation. Here we discuss the ability of different stem cell types, in particular mesenchymal stromal cells, neuronal stem/progenitor cells, hematopoietic stem cells and embryonic stem cells, to modulate the immune response and induce peripheral or central tolerance. These stem cells have been studied to explore tolerance induction to sev

Plasticity of bone marrow-derived stem cells.

Recently, concepts for the transplantation of hematopoietic stem cells have changed dramatically. High-dose myeloablative conditioning regimens are in the process of being replaced by immuno-suppressive regimens, ablating host myelopoiesis, and neoplastic cells by the co-transplanted donor lymphocytes. Furthermore, the presence of stem cells in the bone marrow, capable of differentiating into a variety of nonhematopoietic tissues as well as the presence of cells in other organs, capable of differentiating into hematopoietic cells, has led to the novel concept of the plasticity of stem cells de

Single-cell analysis implicates TH17-to-TH2 cell plasticity in the pathogenesis of palmoplantar pustulosis.

BACKGROUND: Palmoplantar pustulosis (PPP) is a severe inflammatory skin disorder characterized by eruptions of painful, neutrophil-filled pustules on the palms and soles. Although PPP has a profound effect on quality of life, it remains poorly understood and notoriously difficult to treat. OBJECTIVE: We sought to investigate the immune pathways that underlie the pathogenesis of PPP. METHODS: We applied bulk and single-cell RNA sequencing (RNA-Seq) methods to the analysis of skin biopsy samples and peripheral blood mononuclear cells. We validated our results by flow cytometry and immune fluor

Langerhans cell sarcoma following marginal zone lymphoma: expanding the knowledge on mature B cell plasticity.

The concept of unidirectional differentiation of the haematopoietic stem cell has been challenged after recent findings that human B cell progenitors and even mature B cells can be reprogrammed into histiocytic/dendritic cells by altering expression of lineage-associated transcription factors. The conversion of mature B cell lymphomas to Langerhans cell neoplasms is not well documented. Three previous reports have described clonally related follicular lymphoma and Langerhans cell tumours, whereas no case has been published of clonally related marginal zone lymphoma and Langerhans cell sarcoma.

Cellular Plasticity in Malignant Transformation: Mesothelial Cells.

BACKGROUND: Mesothelial proliferations range from reactive lesions to benign and malignant tumors, bearing witness to the plasticity of these cells. Their diagnosis often requires combination of morphological assessment, immunohistochemistry, and molecular testing. SUMMARY: Considerable progress has been made in recent years in all the above aspects. The separation of reactive from neoplastic mesothelial proliferations has become more robust. Mesothelioma in situ is now recognized as precursor of invasive mesothelioma and better tools exist for its differentiation from benign mimics. Differen

Plasticity mechanisms of genetically distinct Purkinje cells.

Despite its uniform appearance, the cerebellar cortex is highly heterogeneous in terms of structure, genetics and physiology. Purkinje cells (PCs), the principal and sole output neurons of the cerebellar cortex, can be categorized into multiple populations that differentially express molecular markers and display distinctive physiological features. Such features include action potential rate, but also their propensity for synaptic and intrinsic plasticity. However, the precise molecular and genetic factors that correlate with the differential physiological properties of PCs remain elusive. In

To be or not to be: endothelial cell plasticity in development, repair, and disease.

Endothelial cells display an extraordinary plasticity both during development and throughout adult life. During early development, endothelial cells assume arterial, venous, or lymphatic identity, while selected endothelial cells undergo additional fate changes to become hematopoietic progenitor, cardiac valve, and other cell types. Adult endothelial cells are some of the longest-lived cells in the body and their participation as stable components of the vascular wall is critical for the proper function of both the circulatory and lymphatic systems, yet these cells also display a remarkable ca

Mapping fibroblast plasticity across non-cancer lung diseases at single-cell resolution.

BACKGROUND: Fibroblast plasticity underlies a wide spectrum of pulmonary diseases, yet the molecular programs governing fibroblast state transitions in non-malignant contexts remain poorly understood. METHODS: We integrated 1,470,730 single-cell transcriptomes across three non-cancer lung diseases to construct a comprehensive fibroblast atlas. Integrated clustering and regulon analyses were used to delineate fibroblast subtypes, disease-associated transcriptional programs, and state transitions. ANTXR1, identified as a representative regulator, was validated by tissue immunohistochemistry and

Epithelial-mesenchymal plasticity in breast cancer: from cell states to clinical vulnerability.

Epithelial-mesenchymal transition (EMT) in breast cancer is better understood as epithelial-mesenchymal plasticity (EMP), a dynamic and reversible spectrum of epithelial, hybrid, and mesenchymal states. In this review, we discuss how EMP is deployed differently across hormone receptor-positive/HER2-negative, HER2-positive, and triple-negative/claudin-low breast cancers, with distinct implications for metastasis, immune evasion, liquid biopsy, and therapeutic resistance. We highlight the regulatory networks and microenvironmental cues that shape state transitions and emphasize that hybrid and p

LRRC55 modulates BK channels to support Purkinje cell plasticity and motor coordination.

Large-conductance Ca2+- and voltage-activated K+ (BK) channels are widely expressed, including in the brain where they shape neuronal excitability. Their physiological functions are strongly influenced by cell-type-specific auxiliary subunits. The auxiliary γ3 subunit (LRRC55) enhances BK-channel activation by shifting voltage-dependent gating toward more negative potentials; however, its protein distribution and in vivo function remain unclear. Here, we generated knock-in mice carrying a C-terminal epitope tag on endogenous LRRC55 to map its expression, and Lrrc55 knockout mice to test its fu

Cell-type Plasticity Supports Behavioral Adaptations at the Water-to-Land Interface.

Animals inhabiting aquatic or terrestrial habitats experience different constraints on their physiology and locomotion, and are exposed to fundamentally different sensory environments. Across evolutionary timescales, most species have adapted to live exclusively either in water or on land. Newts are among the vertebrates that defy this rule and split their adult lives between freshwater ponds and terrestrial habitats. In these amphibians, transitions across environments cause remarkable phenotypic plasticity in their body morphology. But whether and how the nervous system and behavior also ada

Natural Killer Cell Plasticity in Epithelial Ovarian Cancer and Their Therapeutic Implications.

Natural killer (NK) cells are key mediators of antitumor immunity; however, NK cell dysfunction in epithelial ovarian cancer should be considered not as a uniform defect, but rather as compartment-specific states that differ across the blood, ascites, primary tumor, and metastatic sites according to their local cellular interactions, soluble factors, and metabolic constraints. Peripheral blood provides an accessible systemic reference and may support immune monitoring. However, it does not fully reflect NK cell states in local or distant disease compartments. In ascites, cytokine-responsive an
