From Follicle Cell Differentiation and Structure to Chorion Biogenesis in Insects: Cellular Mechanisms, Gene Regulation, Biochemical Composition and Structural Diversity.

Choriogenesis, the final stage of oogenesis in insects, is a highly coordinated developmental process responsible for the formation of the chorion (eggshell), a specialized multilayered extracellular matrix that protects the embryo and mediates essential physiological functions. Despite its fundamental importance for reproductive success and species survival, the mechanisms underlying chorion biogenesis remain incompletely understood across insect taxa. This review provides an updated synthesis and integrated view of choriogenesis, including cellular, molecular, biochemical, and structural per

Controlled Seeding of β-Amyloid Fibrillation Reveals Propagation of Structural Polymorphisms in Cellular Environments.

Molecular-level structural polymorphisms of β-amyloid (Aβ) aggregates in Alzheimer's disease patients are pathologically important. However, tracking the propagation and modulation of Aβ structural polymorphisms via ex vivo approaches remains challenging. The successful application of cryogenic transmission electron microscopy (cryo-TEM) in this area relies on the availability of morphologically distinct micrographs, which then enable unambiguous three-dimensional structural reconstruction of individual fibrillar polymorphs to achieve optimal resolution. As a complementary approach, solid-stat

Molecular, cellular and network mapping of brain structural deviations in patients with Post-COVID19 syndrome.

Post-COVID-19 syndrome encompasses persistent cognitive, neurological, and psychiatric symptoms following SARS-CoV-2 infection, profoundly affecting global quality of life. Clarifying the neurobiological basis of these symptoms is vital for effective therapeutic interventions. This study utilized normative modelling of brain structure ("CentileBrain") to quantify subject-level deviations in cortical thickness, surface area, and subcortical volumes among 20 patients experiencing persistent fatigue following mild COVID-19, compared to 20 matched healthy controls. Group-level analyses on deviatio

Rational design and structural insights of osthole-derived selective AKR1C3 inhibitors with cellular activity.

Aldo-keto reductase 1C3 (AKR1C3) is a key enzyme in androgen biosynthesis and a crucial driver of castration-resistant prostate cancer (CRPC), making it a promising therapeutic target. Unintended inhibition of its homologous isoforms AKR1C1 and AKR1C2 may cause off-target effects and is therefore highly undesirable. In this study, we rationally designed selective AKR1C3 inhibitors derived from the natural product osthole by optimizing the C8- and C7-substituents to target the SP1 and SP2 pockets, respectively, thereby achieving high selectivity for AKR1C3. Among the compounds synthesized, elev

Ca2+-Dependent Mitochondrial Permeability Transition Pore: Structure, Properties, and Role in Cellular Pathophysiology.

The Mitochondrial Permeability Transition pore (MPT pore) activated by Ca2+ ions is a phenomenon that has long been the subject of intense study. Cyclophilin D-dependent opening of the MPT pore in mitochondria in response to calcium overload and oxidative stress leads to swelling of the mitochondrial matrix, depolarization of the inner membrane and dysregulation of ion homeostasis. These processes are accompanied by damage to mitochondrial membranes and, ultimately, to cell death. Despite decades of research, the molecular identity of the MPT pore remains unclear. Currently, the inner membrane

Collagen XII mediated cellular and extracellular mechanisms regulate establishment of tendon structure and function.

Tendons have a uniaxially aligned structure with a hierarchical organization of collagen fibrils crucial for tendon function. Collagen XII is expressed in tendons and has been implicated in the regulation of fibrillogenesis. It is a non-fibrillar collagen belonging to the Fibril-Associated Collagens with Interrupted Triple Helices (FACIT) family. Mutations in COL12A1 cause myopathic Ehlers Danlos Syndrome with a clinical phenotype involving both joints and tendons supporting critical role(s) for collagen XII in tendon development and function. Here we demonstrate the molecular function of coll

Cellular oxygen sensing: Crystal structure of hypoxia-inducible factor prolyl hydroxylase (PHD2).

Cellular and physiological responses to changes in dioxygen levels in metazoans are mediated via the posttranslational oxidation of hypoxia-inducible transcription factor (HIF). Hydroxylation of conserved prolyl residues in the HIF-alpha subunit, catalyzed by HIF prolyl-hydroxylases (PHDs), signals for its proteasomal degradation. The requirement of the PHDs for dioxygen links changes in dioxygen levels with the transcriptional regulation of the gene array that enables the cellular response to chronic hypoxia; the PHDs thus act as an oxygen-sensing component of the HIF system, and their inhibi

Age-related structural changes in trabecular and cortical bone: cellular mechanisms and biomechanical consequences.

It is proposed that there are two structurally different forms of bone loss with different rates, cellular mechanisms, and biomechanical effects. Rapid bone loss is the result of excessive depth of osteoclastic resorption cavities. This leads in trabecular bone to perforation of structural elements, increased size of marrow cavities, and discontinuity of the bone structure, and in cortical bone to subendosteal cavitation and conversion of the inner third of the cortex to a trabecularlike structure, which then undergoes the same changes as the trabecular bone originally present. These structura

Coordinated cellular architecture reveals weakened cortical structure-behavior coupling in normal aging.

Normal aging compromises behavioral functions supported by cortical circuits, yet how age-related cellular reorganization relates to weakened circuit-behavior correspondence remains poorly defined. Here, we examined layer 4 (L4) of the mouse primary somatosensory barrel cortex, a thalamorecipient lamina in which dendritic organization, microglial process architecture, and whisker-dependent tactile function can be analyzed within the same sensory system. Aged mice showed impaired whisker-dependent texture discrimination. Using high-resolution three-dimensional cellular reconstructions, we quant

Spatial Compartmentalization of the Neuronal Endoplasmic Reticulum: Structural Basis of Calcium Signaling and Cellular Homeostasis.

The neuronal endoplasmic reticulum (ER) forms a continuous membrane network that extends throughout the soma, dendrites, axons, and synaptic compartments. Although traditionally viewed as a structurally continuous intracellular organelle, recent advances in volume electron microscopy, super-resolution imaging, and live-cell functional analyses have revealed that the ER is highly compartmentalized into specialized structural and molecular domains. These domains organize local Ca2+ release, store refilling, luminal Ca2+ redistribution, and inter-organelle communication through membrane contact s

Integrating rna structure and protein interactions to uncover the mechanisms of viral and cellular ires function.

BACKGROUND: RNAs fold into complex structures that critically influence gene expression. A prominent class of regulatory elements resides in the 5' untranslated region (5' UTR), where internal ribosome entry sites (IRESs) promote cap-independent translation by directly engaging the ribosome. First discovered in viral genomes, IRESs have been classified into four types according to their structural compactness and factor requirements. While viral IRESs are well studied, cellular IRESs remain poorly understood: they display limited sequence conservation, reduced structural compactness, and varia

The structural characteristics of cellular phospholipid acyl chains required for ABCA1-mediated HDL formation.

ATP-binding cassette protein A1 (ABCA1) mediates high-density lipoprotein (HDL) formation by transporting cellular cholesterol and phospholipids to apolipoprotein A-I (apoA-I). Although phospholipids serve as transport substrates for ABCA1, and the membrane constituents surrounding ABCA1, their roles in HDL formation remain unclear. Here, we elucidated the effect of the acyl chain structure of cellular phospholipids on HDL formation, particularly focusing on monounsaturated fatty acid (MUFA)-containing phosphatidylcholine (PC), the predominant phospholipid in most animal cells. PC molecules ef

Rolling into the genome: linking mutations to cellular structure through label-free holographic cytometry.

Holo-tomographic flow cytometry for label-free phenotyping of suspended acute myeloid leukemia blasts is demonstrated. A concave segmentation algorithm is applied to 3D refractive index tomograms to quantify NPM1-mutation-associated cup-like nuclear morphologies, with virtual reality visualization offering engaging immersion. The method enables population-level detection of statistically significant shifts in 3D cell morphology, originally correlating phenotype with genotype.

An all-cellulose sponge with a nanofiller-assisted hierarchical cellular structure for fruit maintaining freshness.

Cellulose sponges with compressibility and resilience are an ideal packaging material for fruits with fragile skin. Here, a soft and elastic all-cellulose sponge (CS) with a hierarchical cellular structure was fabricated, where the long molecular chain cellulose constructed major pores, the cellulose at nanoscale acted as an elastic nanofiller to fill the gaps of long molecular chain cellulose fibers and constructed minor pores. With these two kinds of pores, this structure can absorb strain hierarchically. The sponge can protect fruits from mechanical damage when dropped or repeated vibration

Secondary Structure of Influenza A Virus Genomic Segment 8 RNA Folded in a Cellular Environment.

Influenza A virus (IAV) is a member of the single-stranded RNA (ssRNA) family of viruses. The most recent global pandemic caused by the SARS-CoV-2 virus has shown the major threat that RNA viruses can pose to humanity. In comparison, influenza has an even higher pandemic potential as a result of its high rate of mutations within its relatively short (<13 kbp) genome, as well as its capability to undergo genetic reassortment. In light of this threat, and the fact that RNA structure is connected to a broad range of known biological functions, deeper investigation of viral RNA (vRNA) structures i

Endosomal structure and APP biology are not altered in a preclinical mouse cellular model of Down syndrome.

Individuals who have Down syndrome (trisomy 21) are at greatly increased risk of developing Alzheimer's disease, characterised by the accumulation in the brain of amyloid-β plaques. Amyloid-β is a product of the processing of the amyloid precursor protein, encoded by the APP gene on chromosome 21. In Down syndrome the first site of amyloid-β accumulation is within endosomes, and changes to endosome biology occur early in Alzheimer's disease. Here, we determine if primary mouse embryonic fibroblasts isolated from a mouse model of Down syndrome can be used to study endosome and APP cell biology.
