Response surface optimization for cellulase production from Enterococcus faecium and Stutzerimonas stutzeri isolated from Gossypium arboretum and Solanum melongena soil.

The rapid utilization of fossil fuel-based energy sources increased demand for alternate sustainable energy sources. One of the best alternate energy sources can be lignocellulosic biomass. The major constituent of lignocellulosic biomass is cellulose that can be converted into simple sugar using cellulase enzymes followed by fermentation for ethanol production. Two potential mesophilic cellulolytic bacteria, BS5 and CS7, from brinjal and cotton soil samples were screened based on high zones of hydrolysis on CMC agar plates and identified as Enterococcus faecium and Stutzerimonas stutzeri, res

Synergistic effects of biochar and cellulase on carbon emission reduction and humification during co-composting of cattle manure and sugarcane bagasse.

INTRODUCTION: The emission of carbon dioxide (CO₂) and methane (CH₄) during composting not only contributes to the greenhouse effect but also leads to carbon loss, thereby reducing humus production and compromising the quality of the final compost product. This study investigated the efficacy and underlying mechanisms of biochar and cellulase addition, individually and in combination, in mitigating emissions and enhancing compost quality within a cattle manure-sugarcane bagasse co-composting system. METHODS: By monitoring key composting parameters and combining with high-throughput sequencing

Assessing the compatibility of choline-based deep eutectic solvents for the structural stability and activity of cellulase: Enzyme sustain at high temperature.

As a new generation of 'green solvents' deep eutectic solvents (DESs) represents a promising alternative to the conventional solvents. Their environmental-benign nature and designer properties promote their utility in biocatalysis. Enzymes are marginally stable when exposed to physical/chemical disturbances. One such enzyme is cellulase which is a propitious catalyst for the depolymerization of cellulose under mild conditions. Therefore, their stability is a prerequisite condition to match demands of biorefineries. To address this issue of low stability, activity and thermal denaturation of ce

Cellulase applied to the leaves of sweet pepper (Capsicum annuum L. var. grossum) upregulates the production of salicylic and azelaic acids.

Treating the leaves of sweet pepper (Capsicum annuum L. var. grossum) with an aqueous solution of cellulase resulted in a four-fold increase in the salicylic acid level compared to a control plant. The level of endogenous azelaic acid was also elevated by the cellulase treatment. Azelaic acid has recently been reported to act as a mobile "priming" agent to arm plants against pathogenic attack. Our results are consistent with this and that the cellulase treatment enhanced the ability of sweet pepper to withstand viral attack.

Hydrophilicity Regulation of Styrene-Based Copolymers to Promote the Enzymatic Saccharification Efficiency by Dually Protecting Cellulases and Blocking Residual Lignin.

Nonproductive adsorption behavior of residual lignin on cellulase has long hindered lignocellulosic biomass enzymatic saccharification. In this study, a strategy of hydrophilicity regulation of styrene-based copolymers was developed to improve the enzymatic saccharification efficiency by specifically acting on cellulases and blocking residual lignin. Styrene-based copolymers with 2-acrylamido-2-methylpropanesulfonate (ST-co-AMPS or SA), varying in molecular weight and hydrophilicity, were synthesized. At pH 4.8 and 10 FPU/glucan cellulase dosage, adding 0.1 g/g-substrate SA8-32 (initiator dosa

Influence of Modification of Cysteine Residues in the Carbohydrate-Binding Domain on Cellulase Activity: Achieving Enhanced Performance and pH-Sensitive Recovery.

The conversion of cellulosic biomass into fermentable sugars represents a pivotal stage in the environmentally friendly and sustainable production of clean fuels and chemicals. In this paper, a new strategy to enhance enzyme performance was proposed. This strategy can achieve pH-sensitive recovery of cellulase while improving the catalytic activity and stability of the enzyme. This strategy was achieved by means of a chemical modification of the cysteine residue (-SH) of the carbohydrate-binding domain (CBD) of cellulase with a PEG-based modifier (Mal-PEG-FA). It was shown that chemical modifi

Use of spectroscopic and imaging techniques to evaluate pretreated sugarcane bagasse as a substrate for cellulase production under solid-state fermentation.

The enzymatic cocktail of cellulases is one of the most costly inputs affecting the economic viability of the biochemical route for biomass conversion into biofuels and other chemicals. Here, the influence of liquid hot water, dilute acid, alkali, and combined acid/alkali pretreatments on sugarcane bagasse (SCB) used for cellulase production was investigated by means of spectroscopic and imaging techniques. Chemical composition and structural characteristics, such as crystallinity (determined by X-ray diffraction), functional groups (Fourier transform infrared spectroscopy), and microstructure

Enhanced and Optimized Production of Cellulase Using Trichoderma harzianum through the Utilization of Pretreated Lignocellulosic Wastes: Purification, Characterization, and Industrial Applications.

In the current study, various Aspergillus and Trichoderma strains were explored to produce cellulase using a variety of agricultural wastes as cheaper substrates. Among the tested strains, Trichoderma harzianum exhibited the highest enzyme activity (84.0 FPU/mL) when grown on alkali-pretreated sugarcane bagasse after purification. Out of three substrates, pretreated sugarcane bagasse consistently resulted in higher cellulase activity (42.05 FPU/mL) as compared to wheat straw (26.5 FPU/mL) and wheat bran (16.7 FPU/mL) across all tested strains due to its high cellulose/hemicellulose content and

Characterization of a bi-functional cellulase produced by a gut bacterial resident of Rosaceae branch borer beetle, Osphranteria coerulescens (Coleoptera: Cerambycidae).

A cellulolytic bacterium was obtained from the digestive tract of Osphranteria coerulescens. The breakdown of woody and cellulosic substances by this insect may be relative in part to its symbiont bacteria. Under optimal cultural conditions the novel isolate produced 5.35U/ml cellulase after 72h. The enzyme was purified to 36 fold with a 0.59% yield and showed a specific activity of 9.0U/mg. It presented its maximum activity at 60°C and pH 5, while it was stable in a wide range of temperature from 20 to 60°C and pH from 5 to 10. The purified enzyme had a molecular weight of 42.50kDa based on S

Unlocking the endoplasmic reticulum ATP importer TrAXER as a new genetic engineering target Enhances Trichoderma reesei cellulase production and cellulose saccharification.

Trichoderma reesei has long been established as a prominent industrial platform for cellulase production. However, functionally validated targets for iteratively engineering T. reesei to boost cellulase yield remain insufficient. Herein, the T. reesei orthologue of the human endoplasmic reticulum (ER) ATP importer, designated TrAXER, was identified. TrAXER absence caused defects in fungal tip growth and resulted in a more than 95% reduction in cellulase production, together with a pronounced activation of ER stress response. Appropriate overexpression of TrAXER led to a 22%-38% increase in ext

Role of in situ surfactant modification of lignin structure and surface properties during glycerol pretreatment in modulating cellulase-lignin binding affinities.

Surfactants are effective agents for enhancing lignocellulosic pretreatment, synergistically modifying lignin with polyols to improve substrate hydrolyzability while achieving comparable delignification. However, the mechanisms underlying the multiple modifications from dual in situ surfactant/polyols grafting that passivate lignin-cellulase interactions and their core affecting factors remain unclear. Following the previously developed polyethylene glycol (PEG) and Triton-assisted pretreatment, the intrinsic correlation among lignin structures, physical barriers, and cellulase interactions wa

Cinnamomum camphora chvar. Borneol essential oil (BEO) was efficiently extracted by using pilot-plant neutral cellulase-assisted steam distillation (NCSD). Borneol, β-cadinene and α-caryophyllene were identified as major components. Bacillus subtilis was the most sensitive bacteria to BEO with the lowest minimal inhibition concentration (MIC) and minimal bactericial concentration (MBC) at 1·75 and 3·50 mg ml-1 , respectively. Antimicrobial activity of the BEO was also reasonably high against Salmonella typhimurium, Escherichia coli and Staphylococcus aureus, but not sensitive against two fungi

Modifications of the Helmintex Method: Integration of Treatments With Tween 20, Cellulase, Percoll and Auramine-O Staining for Detection of Schistosoma mansoni Eggs.

Sensitive diagnostic tools are essential to support schistosomiasis control and elimination programs, yet most long-standing parasitological and serological tests lack sufficient accuracy in low-intensity settings. The Helmintex (HTX) method, based on magnetic isolation of Schistosoma mansoni eggs from large stool volumes, is highly sensitive and serves as a reference standard for evaluating other diagnostic approaches but remains labour-intensive. We evaluated modifications aimed at reducing workload while preserving or enhancing performance. Eggs were seeded into human faeces and processed u

The development of second-generation (2G) bioethanol from lignocellulosic sources, such as sugarcane bagasse, is very important as a viable alternative to conventional fossil fuels. However, the high cost associated with enzymatic hydrolysis, which breaks down cellulose into fermentable sugars, poses a key challenge. This study focused on enhancing cellulase enzyme production by a novel, locally isolated strain, Trichoderma harzianum PP400831, using statistical optimization BBD-RSM to improve enzyme activity. Dietary fiber modification by cellulase-AP3 supplementation tailors oat digestion under elderly gastrointestinal conditions with attenuated starch hydrolysis and enhanced polyphenol bioaccessibility.

The resilient fiber matrix of oats limits nutrient release and moderates glycemic response. This study employed a dynamic in vitro stomach-intestine model to investigate how Cellulase-AP3 enzyme supplementation alters oat digestion under simulated elderly conditions. Cellulase-AP3 supplementation caused significant oat matrix disintegration by enzymatically converting insoluble fiber (54.84% to 32.18%) into soluble fiber (50.56% to 59.63%), which in turn elevated digestive fluid viscosity and delayed gastric emptying. This modified physiological environment hindered amylase action, thereby sub

Synergistic enhancement of physicochemical and immunomodulatory properties of Cyclocarya paliurus polysaccharides through co-culture solid-state fermentation with endophytic cellulase-producing fungi.

Cyclocarya paliurus (C. paliurus) polysaccharides (CP) exhibit hypoglycemic, antihypertensive, and antioxidant activities, yet their extraction efficiency and bioactivity are limited by the intact plant cell wall. Microbial fermentation, especially using endophytic fungi, offers a green strategy to degrade cell wall polysaccharides and modify their structure. In this study, two high-cellulase-producing endophytes (PZ8 and PM2) were screened from C. paliurus and used for co-culture solid-state fermentation. The fermentation significantly increased CP yield to 29.16 mg/g, reduced the molecular w
