Combined Fungal Elicitor and Methyl Jasmonate Enhance Alkaloid Accumulation in Catharanthus roseus Suspension Cultures: An Integrated Molecular and Phytochemical Study.

Suspension cultures of Catharanthus roseus provide an alternative system for studying the regulation of terpenoid indole alkaloid (TIA) biosynthesis and the production of pharmaceutically important metabolites. This study evaluated the effects of the endophytic fungal elicitor RN3 (Chaetomium funicola) and methyl jasmonate (MeJA) on alkaloid accumulation and TIA pathway-associated gene expression in C. roseus suspension cultures. Co-elicitation with 1% RN3 and 150 μM MeJA produced the highest total alkaloid accumulation (14.68 mg g-1 dry weight), representing a 58.9% increase over the untreate

Chlorinated chaetoviridin-type azaphilones discovered via metabolomics: structural diversity and suppression of STAT3 signaling in gastric cancer cells.

Azaphilones are structurally diverse polyketides with broad bioactivities, among which chaetoviridins are distinguished by their chlorinated frameworks and anticancer potential. To expand the chemical space of STAT3-relevant azaphilones, a metabolomics strategy integrated with OSMAC (one strain many compounds) fermentation was applied to a deep-sea Chaetomium globosum strain. Metabolomic analysis revealed pronounced metabolic divergence, with chlorinated metabolites as key discriminants. Molecular networking further localized these features into clusters associated with chaetoviridin-type scaf

Central nervous system aspergillosis: a 20-year retrospective series.

Over the past 20 years at my institution, 71 patients with invasive necrotizing aspergillosis have been encountered; 42 have shown central nervous system (CNS) involvement by autopsy (40) or surgical biopsy (2). Most non-CNS aspergillosis patients had invasive disease confined to the lung, and only 2 with dissemination to 3 or more organs did not have spread to the CNS. In addition to the expected post-transplantation and hematologic malignancy cases, other risk groups identified included those with chronic asthma and steroid use, acquired immunodeficiency syndrome, thermal burn, hepatic failu

Bioactivity of Novel Colchicine, Colchiceine, and 10-Methylthiocolchicine Complexes with Lithium, Sodium, and Potassium Chlorides: Experimental and Theoretical Studies.

Complexes of colchicine, colchiceine, and 10-methylthiocolchicine with Li+, Na+, and K+ cations in the form of chlorides were synthesized and then subjected to spectral analysis, DFT theoretical studies, and molecular modeling. The values for water solubility and lipophilicity were also determined using various platforms; both factors are very important for determining the bioavailability of the tested compounds. These compounds were also tested for their fungicidal, herbicidal, insecticidal, and cytotoxic activities. Preliminary in silico studies showed that colchicine, colchiceine, 10-methyl

Structure and assembly of the mitochondrial membrane remodelling GTPase Mgm1.

Balanced fusion and fission are key for the proper function and physiology of mitochondria1,2. Remodelling of the mitochondrial inner membrane is mediated by the dynamin-like protein mitochondrial genome maintenance 1 (Mgm1) in fungi or the related protein optic atrophy 1 (OPA1) in animals3-5. Mgm1 is required for the preservation of mitochondrial DNA in yeast6, whereas mutations in the OPA1 gene in humans are a common cause of autosomal dominant optic atrophy-a genetic disorder that affects the optic nerve7,8. Mgm1 and OPA1 are present in mitochondria as a membrane-integral long form and a sh

Testing the effectiveness of different wash protocols to remove body surface contaminants in invertebrate food web studies.

Molecular gut content analysis via diagnostic PCR or high-throughput sequencing (metabarcoding) of consumers allows unravelling of feeding interactions in a wide range of animals. This is of particular advantage for analyzing the diet of small invertebrates living in opaque habitats such as the soil. Due to their small body size, which complicates dissection, microarthropods are subjected to whole-body DNA extraction-step before their gut content is screened for DNA of their food. This poses the problem that body surface contaminants, such as fungal spores may be incorrectly identified as inge

Enhanced pathogen identification in fungal endophthalmitis by metagenomic next-generation sequencing: a retrospective clinical evaluation.

Fungal endophthalmitis (FE) is a vision-threatening emergency that requires rapid pathogen identification. Conventional microbial culture demonstrates limited sensitivity in FE, warranting improved diagnostic approaches. We evaluated the detection performance of unbiased metagenomic next-generation sequencing (mNGS) in 31 clinically diagnosed FE cases, including 16 vitreous humor (VH) and 15 aqueous humor (AH) specimens. mNGS showed a positivity rate of 90.3% (28/31, 95% CI: 74.2%-98%), outperforming culture (9.1%, 2/22, 95% CI: 1.1%-29.2%). The positivity rates were 100% for endogenous FE and

Spectrum of central nervous system mycoses and antifungal susceptibility: a two-year retrospective analysis from a tertiary care hospital in India.

BACKGROUND AND OBJECTIVES: The frequency of central nervous system (CNS) fungal infections is rising, leading to increased mortality. These infections pose diagnostic challenges, and therapy depends on the specific fungal pathogen identified. Only a few studies from India have examined the spectrum of fungal pathogens causing CNS infections. The objective of this study was to analyze the clinical and microbiological diversity of fungal pathogens responsible for CNS infections. MATERIALS AND METHODS: This was a retrospective study conducted at a tertiary care center in India from January 2023

Identification of Dermatophyte and Nondermatophyte Molds Isolated from Animal Lesions Suspected to Dermatomycoses.

BACKGROUND: Dermatomycoses contain superficial fungal infections of keratinized layers of the body such as skin, hair, and nail that affect more than 20%-25% of people and animals worldwide. Some fungi can cause superficial infections in animals after accidental penetration and colonization on injured skin and can be transmitted to humans by exposure. The infection caused mainly by dermatophyte species and may also be caused rarely by yeasts and nondermatophytic molds. MATERIALS AND METHODS: Eighty-two skin scrapings and hair samples were collected from animals (sheep, cow, cat, camel, calf,

Sensitivity of Zea mays and Soil Microorganisms to the Toxic Effect of Chromium (VI).

Chromium is used in many settings, and hence, it can easily enter the natural environment. It exists in several oxidation states. In soil, depending on its oxidation-reduction potential, it can occur in bivalent, trivalent or hexavalent forms. Hexavalent chromium compounds are cancerogenic to humans. The aim of this study was to determine the effect of Cr(VI) on the structure of bacteria and fungi in soil, to find out how this effect is modified by humic acids and to determine the response of Zea mays to this form of chromium. A pot experiment was conducted to answer the above questions. Zea m

The occurrence of molds in patients with chronic sinusitis.

Chronic rhinosinusitis (CRS) is a common inflammatory condition of nasal and paranasal sinus mucosa. Although pathogenic bacteria were postulated as main etiological factor responsible for most cases of CRS, the involvement of molds was recently proved in some cases. The aim of the study was to conduct mycological analysis of material obtained from patients operated on due to chronic sinusitis. The study included 107 patients, 45 women and 62 men. During the surgery, a fragment of mucosa from the region of the ethmoid bulla was obtained as microbiological characteristics of this material close

Flavin-dependent halogenases from Cortinarius mushrooms for the regioselective chlorination of anthraquinone pigments.

Flavin-dependent halogenases (FDHs) are valuable catalysts for the late-stage diversification of small molecule scaffolds, modulating their biological activities and/or introducing orthogonal functionalities for regiospecific derivatization. This study broadens the repertoire of FDHs by discovering two such enzymes (CkHAL1 and CsHAL2) from Cortinarius mushrooms that chlorinate anthraquinone skeletons from the emodin family when expressed in Saccharomyces cerevisiae. As opposed to the phenolic FDH CcRadH from the ascomycete Chaetomium chiversii that can modify only neutral anthraquinones such a

Chaetoglobosin A induces apoptosis in T-24 human bladder cancer cells through oxidative stress and MAPK/PI3K-AKT-mTOR pathway.

Chaetoglobosin A (ChA) is an antitumor compound produced by Chaetomium globosum. However, the mechanism of its antitumor effect has been rarely reported. In this study, we evaluated the anti-proliferative effect of ChA on T-24 human bladder cancer cells and explored its mechanism of action. ChA was found to have a good inhibitory effect on T-24 cells by MTT assay with an IC50 value of 48.14 ± 10.25 μΜ. Moreover, it was found to have a migration inhibitory ability and a sustained proliferation inhibitory effect on tumor cells by cell aggregation assay and cell migration assay. The cells morphol

Does Organomineral Fertilizer Combined with Phosphate-Solubilizing Bacteria in Sugarcane Modulate Soil Microbial Community and Functions?

Soil bacterial and fungal communities are suitable soil ecosystem health indicators due to their sensitivity to management practices and their role in soil ecosystem processes. Here, information on composition and functions of bacterial and fungal communities were evaluated at two phenological stages of sugarcane (six and twelve months, equivalent to the most intensive vegetative stage and to final maturation, respectively) when organomineral fertilizer, combined with phosphate-solubilizing bacteria (PSB), was added into the soil. Organic compost enriched with apatite (C + A) or phosphorite (C

Metagenomics of black grains: new highlights in the understanding of eumycetoma.

BACKGROUND: Eumycetoma is a chronic subcutaneous granulomatous disease that is endemic in Sudan and other countries. It can be caused by eight different fungal orders. The gold standard diagnostic test is culture, however, culture-independent methods such as imaging, histopathological and molecular techniques can support diagnosis, especially in cases of negative cultures. METHODS: The amplicon-based internal transcribed spacer 2 metagenomic technique was used to study black grains isolated from 14 tissue biopsies from patients with mycetoma. Furthermore, mycological culture and surgical biop

Prevalence of allergic sensitization to indoor fungi in West Virginia.

Exposure to indoor fungi is of growing concern in residential and occupational environments in the United States. The purpose of this study was to determine the prevalence of sensitization to common indoor fungal species in an atopic population. We evaluated 102 patients (73 female and 29 male patients) for immunoglobulin E (IgE) reactivity to a panel of skin-prick test (SPT) reagents used for routine allergy testing. Patients also were tested for six additional fungi that are common indoor contaminants. All patients had symptoms consistent with allergic rhinitis or asthma. The presence of spe
