Bioactive Chalcones from Aizoon africanum: Isolation and Cytotoxicity Against Liver and Neural Cancer Cells.

Aizoon africanum (L.) Klak (Synonym Galenia africana L.) is traditionally used for a variety of medicinal purposes; however, it has been reported to cause liver damage and severe ascites, particularly in sheep and Angora goats in the arid regions of the Western Cape. This study explores its cytotoxic properties to identify potential cytotoxic compound(s) in the plant. The methanolic extract of A. africanum was re-investigated and subjected to various chromatographic techniques, including preparative HPLC, resulting in the isolation of eight compounds (1-8). Structural elucidation was primarily

In vitro and in silico evaluation of the anti-leishmania activity of synthetic chalcones.

Leishmaniasis is a group of neglected, vector-borne infectious diseases that affect millions of people around the world. The medications available for its treatment, especially in cases of visceral leishmaniasis, are old, outdated and have serious side effects. In this work, 10 chalcones were synthesised and evaluated in vitro against promastigotes and axenic amastigotes of Leishmania infantum. Compounds CP04 and CP06 were the most promising, respectively presenting IC50 values = 13.64 ± 0.25 and 11.19 ± 0.22 µM against promastigotes, and IC50 = 18.92 ± 0.05 and 22.42 ± 0.05 µM against axenic

Synthesis, Docking Studies and Evaluation of Chalcones as Anti-Helicobacter pylori and antitumoral Agents.

Helicobacter pylori colonizes the gastric epithelium of 50 % of world population and it is the main etiological agent of human chronic gastritis, peptic ulcer, and gastric cancer. In this study, we synthesized and characterized a series of 14 chalcones and evaluated their anti-H. pylori, NO inhibition (in vitro and in silico), and AGS cells cytotoxic effects. Compounds 3b and 3h showed MIC of 8 μg/mL. We observed structure-activity relationships, mainly related to the influence of methoxy substituent at C-2 (3b) and the nitro group at C-4 (3h) in chalcone scaffold. The fourteen chalcones inhib

Disruption of endolysosomal trafficking pathways in glioma cells by methuosis-inducing indole-based chalcones.

Methuosis is a form of non-apoptotic cell death involving massive vacuolization of macropinosome-derived endocytic compartments, followed by a decline in metabolic activity and loss of membrane integrity. To explore the induction of methuosis as a potential therapeutic strategy for killing cancer cells, we have developed small molecules (indole-based chalcones) that trigger this form of cell death in glioblastoma and other cancer cell lines. Here, we report that in addition to causing fusion and expansion of macropinosome compartments, the lead compound, 3-(5-methoxy-2-methyl-1H-indol-3-yl)-1-

Synthesis and bioevaluation of substituted chalcones, coumaranones and other flavonoids as anti-HIV agents.

A series of chalcone, flavone, coumaranone and other flavonoid compounds were screened for their anti HIV-1 activity in two cell culture models using TZM-bl and PM1 cells. Within the systems evaluated, the most promising compounds contained either an α- or β-hydroxy-carbonyl motif within their structure (e.g., 8 and 9). Efficacious substituents were identified and used to design new HIV inhibitors with increased potency and lower cytotoxicity. Of the scaffolds evaluated, specific chalcones were found to provide the best balance between anti-HIV potency and low host cell toxicity. Chalcone 8l w

Synthesis and antihepatotoxic activity of some new chalcones containing 1, 4 - dioxane ring system.

Silybum marianum is a medicinal plant used widely for treating liver diseases. The silymarin, a mixture of three flavolignan isomers namely silybin (1), silydianin (2), and silychristin (3) is an active constituent of the plant. However, silybin containing 1, 4-dioxane ring is the most potent antihepatotoxic agent. In contrast, other isomers do not possess 1, 4-dioxane ring, and thus do not exhibit a significant activity. We, therefore, thought that 1, 4-dioxane ring plays an important role in displaying antihepatotoxic activity, and have prepared some chalcones containing 1, 4-dioxane ring. T

Design, Synthesis, and Evaluation of Bis-Chalcones and Bis-Flavones as Selective COX-2 Inhibitors.

Over the past two decades, the search for safe and effective COX-2 inhibitors has spurred extensive research on flavonoids. Within this context, synthetic flavonoid dimers have emerged as a promising subclass with potential anti-inflammatory activity. To investigate whether dimerization enhances their potency and selectivity, novel A-fused bis-chalcones and A-fused bis-flavones were synthesized and evaluated for their inhibitory activity against isolated human COX-1 and COX-2 enzymes, as well as their effects on prostaglandin E2 production in whole human blood. Interestingly, the most active c

Design, Synthesis, and Evaluation of New Polyhydroxylated Bis-Chalcones as Potential COX-2 Selective Inhibitors.

Selective inhibition of COX-2 is considered one of the best strategies for treating chronic inflammatory diseases. However, the currently available options still have significant side effects due to exacerbated selectivity. Bis-chalcone derivatives have shown promising anti-inflammatory properties with reduced side effects. In this study, a family of polyhydroxylated bis-chalcones was synthesized and tested in vitro for their ability to inhibit human COX-2 and COX-1 and to assess selectivity. To further understand their mechanism of action, inhibitory kinetic analysis and in silico molecular d

A Comprehensive Review of Chalcones in Dual-Targeted Therapeutics for the Treatment of Parkinson's Disease.

INTRODUCTION: The design of novel therapeutic entities for neurodegenerative diseases, particularly Parkinson's disease, remains a major challenge in medicinal chemistry. Current treatment strategies primarily rely on symptomatic management, often involving levodopa in combination with catechol-O-methyltransferase (COMT) inhibitors to prolong its therapeutic effect. However, these approaches do not adequately address the multifactorial nature of the disease. This study aims to explore the potential of chalcone derivatives as dual-target-directed ligands capable of modulating both COMT and aden

In silico activity and effect of synthetic chalcones on Candida albicans and Candida tropicalis biofilms.

Biofilm formation is considered one of the most important virulence factors for Candida species, which presents an extracellular matrix of polymeric substances that limits the passage of antifungals, leading to fungal resistance. Therefore, the present study investigated the biofilm eradication effect of synthetic chalcones against Candida albicans and Candida tropicalis. Molecular docking studies were conducted to verify the mechanism of action of chalcones on Candida species proteins. The biofilm eradication effect was determined using crystal violet methodology to quantify biomass and Thiaz

Design and Development of Sulfonamide-Chalcones as Antiepileptic Candidates: Computational and Zebrafish-Based Validation Approaches.

The study aims to develop safer and more effective antiepileptic drugs (AEDs) by targeting the γ-aminobutyric acid type A (GABAA) receptor, an important inhibitory neurotransmitter system involved in the control of neuronal excitability and seizures. With epilepsy rates rising worldwide and current AEDs often causing serious side effects, new neurotherapeutics with better efficacy and safety are urgently needed. In this context, sulfonamides, known for their proven anticonvulsant properties, were selected as the core scaffold for the development of new sulfonamide-chalcone hybrids (compounds 1

Anti-malarial, cytotoxicity and molecular docking studies of quinolinyl chalcones as potential anti-malarial agent.

The quinolinyl chalcones series (A1-A14) were screened for antimalarial activity. According to in vitro antimalarial studies, many quinolinyl chalcones are potentially active against CQ-sensitive and resistance P. falciparum strains with no toxicity against Vero cell lines. The most active quinolinyl chalcones A4 (with IC50 0.031 μM) made a stable A4-heme complex with - 25 kcal/mole binding energy and also showed strong π-π interaction at 3.5 Å. Thus, the stable A4-heme complex formation suggested that these quinolinyl chalcones act as a blocker for heme polymerization. The docking results of
