A nested multiplex polymerase chain reaction assay for the differential identification of three zooanthroponotic chlamydial strains in porcine swab samples.

Porcine chlamydial infection is an enzootic infectious disease caused by multiple members of the family Chlamydiaceae (e.g. Chlamydophila abortus, Chlamydia suis, and Chlamydophila pneumoniae). Rapid and accurate differentiation of these pathogens is critical in the control and prevention of disease. The aim of the current study was to develop a nested multiplex polymerase chain reaction (nmPCR) assay to simultaneously detect the 3 chlamydial pathogens in clinical samples. In the first round of the nmPCR, 1 pair of family-specific primers were used to amplify the 1,100 base pair (bp) fragment

Chlamydial infections.

Chlamydiae are among the most successful bacterial pathogens, and there are few branches of medicine on which chlamydial infection and its sequelae do not impinge. Chlamydia trachomatis is responsible for many million cases of blindness, pelvic inflammatory disease, urethritis, epididymitis, infertility and ectopic pregnancy annually; it also causes lymphogranuloma venereum, reactive arthritis, ophthalmia neonatorum and infantile pneumonia. C. pneumoniae is among the most common causes of community-acquired pneumonia, and recent evidence suggests that it may play a part in the pathogenesis of

Aspirin inhibits Chlamydia pneumoniae-induced nuclear factor-kappa B activation, cytokine expression, and bacterial development in human endothelial cells.

OBJECTIVE: Chlamydia pneumoniae has been associated with atherosclerosis. Infection of vascular endothelial cells with C pneumoniae increases the expression of proatherogenic cytokines mediated by nuclear factor (NF)-kappaB, a transcription factor. The present study was designed to test the effect of aspirin on C pneumoniae-induced NF-kappaB activation, interleukin expression, and bacterial development in cultured human endothelial cells. METHODS AND RESULTS: Aspirin, its metabolite salicylic acid, and 2 other unrelated NF-kappaB inhibitors showed a strong concentration-dependent inhibitory e

[Heat shock protein antibodies and other chlamydial antigens in various diseases and their occurrence in blood donors].

BACKGROUND: The chlamydial infections are very frequently considered in the causal connection with some diseases e.g. atherosclerosis or chronic joint infections. The evidence of the antibodies against the heat shock protein of chlamydial origin is not the usual part of practical serological diagnostics. The aim of this study is an attempt to identify antibodies against the heat shock protein and other antigens of chlamydiae in the sera of two groups of patients and in the sera of blood donors. METHODS AND RESULTS: The sera of patients suffering from unstable angina pectoris (NAP = 69), sera

A case of severe psittacosis pneumonia complicated by splenic infarction.

CLINICAL DATA: Chlamydia psittaci pneumonia is a community-acquired pneumonia caused by Chlamydia psittaci. While severe cases may lead to critical conditions such as respiratory failure, splenic infarction is relatively uncommon. A severe patient with Chlamydia psittaci pneumonia admitted to our hospital experienced a splenic infarction during treatment. Fortunately, the patient's situation was improved after careful treatment. Now, the patient has been discharged. Further exploration of the mechanism of concurrent splenic infarction is required. BACKGROUD: Psittacosis pneumonia, a zoonotic

Chlamydia trachomatis infections in neonates and young children.

In 1911, Lindner and colleagues identified intracytoplasmic inclusions in infants with a nongonococcal form of ophthalmia neonatorum called inclusion conjunctivitis of the newborn (ICN). Mothers of affected infants were found to have inclusions in their cervical epithelial cells, fathers of such infants had inclusions in their urethral cells, and the epidemiology of sexually transmitted chlamydial infections was revealed. Fifty years later, chlamydial isolation procedures were developed, and studies again demonstrated Chlamydia trachomatis as an etiology of ICN and the female birth canal as th

Characterization of the hypothetical protein Cpn1027, a newly identified inclusion membrane protein unique to Chlamydia pneumoniae.

The hypothetical protein Cpn1027 was detected in the inclusion membrane of Chlamydia pneumoniae-infected cells with antibodies raised with Cpn1027 fusion proteins in an indirect immunofluorescence assay. The inclusion membrane staining by the anti-Cpn1027 antibodies co-localized with the staining of an antibody recognizing a known inclusion membrane protein designated IncA and these membrane stainings were blocked by the corresponding but not irrelevant fusion proteins. Although Cpn1027 was not predicted to be an inclusion membrane protein, it contained a bi-lobed hydrophobic domain region at

Serological studies in Chlamydia trachomatis associated pneumonia of infants.

Serological results provided evidence for the clinical diagnosis of chlamydial pneumonia in 8 out of 51 cases. Elevated chlamydia-specific IgM titres (128-2048) and IgG titres (128-2048) were recorded in 5 and 8 infants, respectively. High titres of chlamydial antibodies (512-8192) in the sera of mothers of seropositive children proved genital Chlamydia trachomatis infection.

C. pneumoniae disrupts eNOS trafficking and impairs NO production in human aortic endothelial cells.

Endothelial nitric oxide synthase (eNOS) generated NO plays a crucial physiological role in the regulation of vascular tone. eNOS is a constitutively expressed synthase whose enzymatic function is regulated by dual acylation, phosphorylation, protein-protein interaction and subcellular localization. In endothelial cells, the enzyme is primarily localized to the Golgi apparatus (GA) and the plasma membrane where it binds to caveolin-1. Upon stimulation, the enzyme is translocated from the plasma membrane to the cytoplasm where it generates NO. When activation of eNOS ceases, the majority of the

Chlamydia in the neonate.

Chlamydia trachomatis infections are the most prevalent of all sexually transmitted diseases and can be transmitted to the neonate during the birth process. Neonatal chlamydial infections include inclusion conjunctivitis and chlamydial pneumonia. These infections cause no reported mortality but are associated with a high incidence of morbidity in the newborn. Nursing care is vitally important in prevention, early detection, and management of neonates with chlamydial infections. A knowledge of Chlamydia, its risk factors, clinical manifestations, diagnosis, and treatment will assure an improved

Serotonin and melatonin, neurohormones for homeostasis, as novel inhibitors of infections by the intracellular parasite chlamydia.

OBJECTIVES: Chlamydiae are obligate intracellular bacteria, causing a variety of diseases, i.e. pneumonia, sexually transmitted disease, conjunctivitis and zoonosis. Tryptophan depletion by interferon-gamma (IFN-gamma) is the most important host defence system against chlamydial infection. Thus chlamydial tryptophan metabolism is thought to play key roles for IFN-gamma resistance, persistent infection and host/tissue tropisms. We tested tryptophan derivatives for activity against chlamydia-infected cells. METHODS: Rates of chlamydial infection and sizes of the inclusions were evaluated by in

Chlamydia pneumoniae as a respiratory pathogen.

Chlamydia pneumoniae is a recently recognized human respiratory pathogen with a unique biphasic life cycle characterized by an obligate intracellular (replicative) and an extracellular (infectious) form of the organism. C. pneumoniae is widely distributed and, via the respiratory route, infects the majority of the world's population. The majority (70%) of acute human C. pneumoniae respiratory tract infections are asymptomatic or only mildly symptomatic but a minority (30%) cause more severe respiratory illnesses including community-acquired pneumonia, bronchitis and a variety of upper airway i

Atypical pneumonias. Clinical and extrapulmonary features of Chlamydia, Mycoplasma, and Legionella infections.

Pneumonias caused by atypical organisms usually have extra-pulmonary features. Chlamydial pneumonia often starts with hoarseness and fever, and respiratory tract symptoms may not appear for days. Mycoplasmal pneumonia may manifest with ear pain and a nonproductive cough. Legionnaires' disease presents with high fevers and central nervous system and gastrointestinal abnormalities. Diagnosis of chlamydial infection is accomplished with serologic testing. Patients are unresponsive to erythromycin treatment and should be started on empirical doxycycline (Doryx, Vibramycin) therapy. The presence of

Recent advances in understanding the biology, epidemiology and control of chlamydial infections in koalas.

The koala (Phascolarctos cinereus) is recognised as a threatened wildlife species in various parts of Australia. A major contributing factor to the decline and long-term viability of affected populations is disease caused by the obligate intracellular bacteria, Chlamydia. Two chlamydial species infect the koala, Chlamydia pecorum and Chlamydia pneumoniae, and have been reported in nearly all mainland koala populations. Chlamydial infections of koalas are associated with ocular infections leading to blindness and genital tract infections linked to infertility, among other serious clinical manif

Inhibitory effect of dietary phenolic compounds on Chlamydia pneumoniae in cell cultures.

Chlamydial infections are very common worldwide. All chlamydial species have a tendency to cause persistent infections, which have been associated to several chronic diseases including blinding trachoma, infertility and coronary heart disease (CHD). At present, no efficient treatment for the eradication of chronic chlamydial infections exists and, thus, new antichlamydial compounds are urgently needed. This study was designed to screen antichlamydial activity of natural flavonoids and other natural and structurally similar synthetic compounds against Chlamydia pneumoniae in human cell line (HL

Cardiovascular disease risk from prior Chlamydia pneumoniae infection can be related to certain antigens recognized in the immunoblot profile.

Chlamydia pneumoniae infection has been described as a risk factor for atherosclerosis on the basis of raised seroreactivity against complete elementary bodies among cardiovascular disease (CVD) patients. In order to identify antigens of possible pathogenetic relevance, C. pneumoniae IgG and IgA immunoblot profiles were compared for CVD patients (IgG: n = 159; IgA: n = 72) and for controls (IgG: n = 158; IgA: n = 115), all with prior C. pneumoniae infection. IgG and IgA recognition patterns were very similar, and a broad range of antigens was commonly recognized. However, statistical analysis

Heat shock protein 60 from Chlamydia pneumoniae elicits an unusual set of inflammatory responses via Toll-like receptor 2 and 4 in vivo.

Heat shock protein 60 (HSP60) from Chlamydia pneumoniae was described to trigger in vitro inflammatory and cytokine responses including TNF and IL-12p40. Although it can be found in atherosclerotic plaques of patients, the stimulatory potential of chlamydial and other HSP60 in vivo is unclear. We now report that chlamydial HSP60 fails to induce TNF expression in vivo, and significant serum levels of IL-12p40 are only found upon intraperitoneal injection of high doses of HSP60 or after intravenous application. Upon purification of chlamydial HSP60 with polymyxin B-agarose columns, its ability t

Chlamydia trachomatis in term pregnancy. isolation and serological study with a case report.

The prevalence of cervical Chlamydia trachomatis infection in healthy, symptomless full-term pregnant women (n = 92) was found to be low (1.1%). Chlamydial cervical IgA antibodies, as measured by solid-phase radioimmunoassay (RIA), were found in 5.3%, but amniotic fluid chlamydial RIA IgA antibodies were almost absent. Serum IgG antibodies were measured with indirect immunofluorescence (IF). Of the mothers 9.8% had a titer greater than or equal to 1:64, correlating closely with the figure of 10.9% for infants' cord-blood IF serology. Finally, a case report is presented. Human Infection with Chlamydia pneumoniae ST16 - Lishui City, Zhejiang Province, China, 2024.

WHAT IS ALREADY KNOWN ABOUT THIS TOPIC?: Chlamydia pneumoniae (C. pneumoniae) is an important pathogen associated with respiratory infections. In China, C. pneumoniae pneumonia is not a notifiable infectious disease and is frequently overlooked in clinical detection protocols for community-acquired pneumonia. Consequently, the prevalence and genotypic distribution of chlamydial infections remain inadequately characterized. WHAT IS ADDED BY THIS REPORT?: We investigated four patients with C. pneumoniae pneumonia in Lishui City, Zhejiang Province, China, between April and May 2024. All patients
