Mechanism of chlordiazepoxide enhancement of 4-aminobutyrate-receptor desensitization investigated with 82Br radiotracer.

Rates of desensitization of a rapidly desensitizing 4-aminobutyrate (4-Abu) receptor (t1/2 = 33 ms at saturation) were measured in a previously characterized experimental system from rat cerebral cortex. Desensitization was measured at times between 100 ms and several s. Measurements with 300-ms or 1-s 82Br- influx assays after prior incubation of the membrane with 4-Abu covered the whole 4-Abu concentration range of response. Desensitization was not affected by the low Br- concentrations (< or = 1 mM) present. The presence of the tranquilizing drug chlordiazepoxide (Librium) in the prior incu

Water intake and time course of drinking after single or repeated chlordiazepoxide injections.

Chlordiazepoxide (5.0, 10.0 or 15.0 mg/kg) was given to rats either acutely or for 9 consecutive days. Its effects were examined in a 15-min drinking test in which latency to drink, volume of water consumption and the time-course of drinking were measured. Chlordiazepoxide (10.0 mg/kg) produced the strongest stimulant effect on drinking and enhanced the frequency of occurrence of drinking both at the beginning and at the end of the test period. Chlordiazepoxide (15.0 mg/kg) delayed the onset of drinking and its peak effect was observed later than for other injection conditions. Single and repe

Acute chlordiazepoxide increases risky choice in rats and repeated administration attenuates this effect.

Risky choice is a defining feature of many psychological disorders and maladaptive behavior. There is evidence that benzodiazepines, a class of drugs that act on the GABA system, may increase risky choice. Long-term use of these medications is common, but relatively little is known about how repeated/chronic use of benzodiazepines affects risky decision making. The present study examined effects of acute and repeated chlordiazepoxide, a prototypic benzodiazepine, on probability discounting in eight male Sprague-Dawley rats. The probability-discounting task involved choices between a single foo

Respiratory Failure Requiring Mechanical Ventilation Among Patients Receiving Chlordiazepoxide for Alcohol Withdrawal Symptoms.

Benzodiazepines are a cornerstone in the treatment of alcohol withdrawal symptoms, although their use is associated with a risk of sedation leading to respiratory depression. This study describes the reasons for and frequency of Intensive Care Unit (ICU) admission, mechanical ventilation, and clinical outcomes for patients receiving symptom-triggered treatment with chlordiazepoxide for alcohol withdrawal symptoms. This retrospective quality-improvement study included admissions to a Danish general ICU after treatment with at least 200 mg chlordiazepoxide for alcohol withdrawal symptoms within

Double-blind controlled trial of bromocriptine, chlordiazepoxide and chlormethiazole for alcohol withdrawal symptoms.

Seventy-one patients undergoing withdrawal from alcohol were randomly assigned to treatment with oral bromocriptine, chlormethiazole or chlordiazepoxide. Forty-one percent had alcoholic hepatitis and/or cirrhosis. Patients were stratified into two groups: major and minor withdrawal symptoms. The latter group included a placebo tratment. Bromocriptine was ineffective in treating withdrawal symptoms, whilst chlormethiazole and chlordiazepoxide were equally effective. These findings do not support the evidence from animal and clinical studies suggesting that the disturbances in the dopaminergic s

beta-CCE and chlordiazepoxide reduce exploratory head-dipping and rearing: no mutual antagonism.

Intravenous injection of ethyl beta-carboline-3-carboxylate (beta-CCE, 2 mg/kg) significantly reduced exploratory behaviour in a holeboard and 1 & 2 mg/kg markedly reduced the number of rears made. These effects were similar to those found with chlordiazepoxide (5 mg/kg). When they were given together the two drugs neither counteracted each other's effects, nor were their combined effects additive. We suggest that beta-CCE and the benzodiazepines might both act at one class of benzodiazepine receptor, as agonists to produce the same behavioural effects. The results are also discussed with refe

Prevalence of Patients Admitted to Intensive Care After Administration of Chlordiazepoxide in the Emergency Room.

Chlordiazepoxide is effective in treating alcohol withdrawal syndrome, but it poses a risk of long-term sedation. The prevalence of this side effect and its risk factors remain uncertain. This retrospective cross-sectional study aimed to estimate both using data from Aarhus University Hospital's BI portal. We identified and manually reviewed patient records from 1 September 2019 to 31 August 2021, including the treating physicians' conclusions on ICU admissions to determine whether they were likely due to chlordiazepoxide toxicity. Chlordiazepoxide was administered to 1363 unique patients in t

Continuous evaluation of drug withdrawal in the rat using telemetry: effects of morphine and chlordiazepoxide.

INTRODUCTION: The procedures used to assess withdrawal must be sensitive and widely applicable, i.e. not specific to any particular drug class. Furthermore, the measurements should not be affected by repeat testing. METHODS: We have used implanted telemetry devices to continuously follow body temperature, locomotor activity (LMA), heart rate (HR) and mean arterial blood pressure (mean ABP) in addition to food intake and body weight gain over 20days of treatment and 8days of withdrawal. The effects of morphine (32 and 64mg/kg p.o., b.i.d.) and chlordiazepoxide (16, 32 and 64mg/kg p.o., b.i.d.)

Differential generalization to pentobarbital in rats trained to discriminate lorazepam, chlordiazepoxide, diazepam, or triazolam.

In drug discrimination studies benzodiazepine-trained animals have typically responded on the drug lever when tested with barbiturates. In a recent study, greater specificity appeared to be shown when lorazepam was used as a training drug. The generality and limits of this finding were explored in the present set of experiments. The asymmetrical cross-generalization found in lorazepam- and pentobarbital-trained baboons was replicated in rats and was shown not to be a function of either lorazepam (0.1., 0.32, or 1.0 mg/kg) or pentobarbital (10 or 25 mg/kg) training dose (i.e., pentobarbital-tra

The chlordiazepoxide/pentylenetetrazol discrimination: characterization of drug interactions and homeostatic responses to drug challenges.

Rats were trained to discriminate chlordiazepoxide (CDP) from pentylenetetrazol (PTZ) in a two-lever food motivated discrimination task. Training drug doses were adjusted until subjects emitted approximately 50% of their responses on each of the two drug-appropriate levers during saline injection tests. Tests that followed injection of CDP/PTZ combinations illustrated a reciprocal antagonism between the two drugs. Saline-injection tests that followed large dose injections of CDP revealed a period of predominantly PTZ-appropriate responding that persisted after the initial period of predominant

Behavioral effects of nonbenzodiazepine anxiolytic drugs: a comparison of CGS 9896 and zopiclone with chlordiazepoxide.

Zopiclone and CGS 9896 are two nonbenzodiazepine compounds which have been shown to displace benzodiazepines from their binding sites. The present study compared the behavioral effects of these two compounds in rats with those of chlordiazepoxide. The three drugs produced dose-related increases in punished drinking as did pentobarbital and meprobamate but not PK 9084, which also acts at benzodiazepine binding sites, or buspirone. Rates of lever pressing suppressed by punishment were also increased by chlordiazepoxide and zopiclone. CGS 9896 exerted a similar although less marked effect. Lever

The effects of clozapine on shuttle-box avoidance responding in rats: comparisons with haloperidol and chlordiazepoxide.

Previous studies have shown that clozapine produces effects different from those of other antipsychotic drugs on positively reinforced responding but may give rise to similar disruptions of avoidance behavior. To investigate the actions of clozapine on avoidance responding in more detail the effects of this drug were compared with those of haloperidol and chlordiazepoxide in rats trained to avoid shock in a shuttle-box. Acute administration of all three drugs reduced avoidance responses and increased escape failures although clozapine produced this latter effect only at a high dose. With repea

Effect of combined treatment of diphenylhydantoin with clonazepam and chlordiazepoxide on the threshold for maximal electroconvulsions in mice.

The anticonvulsant effect of either clonazepam (0.2-6.4 mg/kg) or chlordiazepoxide (2.5-40 mg/kg) alone or in combination with diphenylhydantoin (4-16 mg/kg) was studied against electroconvulsions in mice. All drugs were injected intraperitoneally, diphenylhydantoin - 75 min and the benzodiazepines - 60 min before the test. Adding either clonazepam or chlordiazepoxide (in doses moderately increasing the convulsive threshold) was more effective than doubling the dose of diphenylhydantoin. Both the interaction between benzodiazepines and diphenylhydantoin at the level of the receptor for picroto

Antenatal exposure to meprobamate and chlordiazepoxide in relation to malformations, mental development, and childhood mortality.

In a follow-up study of 50,282 pregnancies (lasting at least five lunar months) and the offspring, malformations identified before the first birthday, or at death before the fourth birthday, were identified in 3248 children (6.5 per cent). A total of 1870 children exposed in utero to meprobamate or chlordiazepoxide were compared with 48,412 children who were not. No significant differences were found either overall or in specific outcomes; rates were also similar when exposures occurred during the first trimester or at other times during pregnancy. The contribution of contextual fear in the anxiolytic effect of chlordiazepoxide in the fear-potentiated startle test.

This study evaluated the extent to which a reduction in contextual fear contributes to the anxiolytic effect of benzodiazepines in the fear-potentiated startle response. To this end, chlordiazepoxide, an anxiolytic often used as positive control in preclinical drug studies, and zolpidem, known to have sedative properties and to be devoid of anxiolytic effects, were tested in two contexts: the same context as training had taken place and an alternative context. In addition, the level of muscle relaxation was assessed in a grip strength test. Chlordiazepoxide (2.5-10 mg/kg) decreased the fear-po

A randomized, double-blind comparison of lorazepam and chlordiazepoxide in patients with uncomplicated alcohol withdrawal.

OBJECTIVE: For important reasons, lorazepam (Ativan) and chlordiazepoxide (Librium) are both popular treatments for alcohol-withdrawal syndrome. Nevertheless, there is little literature directly comparing the two drugs. A formal comparison is desirable because of pharmacokinetic and other differences that could affect safety and efficacy considerations relevant to practice in developing countries. METHOD: One hundred consecutive consenting male inpatients in a state of moderately severe, uncomplicated alcohol withdrawal at screening were randomized to receive either lorazepam (8 mg/day) or ch

Chlordiazepoxide reduces the generalised anxiety, but not the direct responses, of rats exposed to cat odor.

Rats were treated for 5 days with vehicle or chlordiazepoxide (CDP, 5, 10, or 20 mg/kg/day) and then exposed to a cloth impregnated with a neutral or cat odor. Those exposed to cat odor made significantly fewer contacts with the cloth and spent less time in contact with it and more time sheltering than those exposed to the neutral odor. CDP (5 mg/kg) significantly increased the time in contact with both odor cloths, but there were no other significant effects in the cat odor group. Rats previously exposed to cat odor showed significant anxiogenic responses in the social interaction and elevate
