Comparative evaluation of chloroethene dechlorination to ethene by Dehalococcoides-like microorganisms.

Reductive dehalogenation of tetrachloroethene (PCE), trichloroethene (TCE), cis-1,2-dichloroethene (DCE), and vinyl chloride (VC) was examined in four cultures containing Dehalococcoides-like microorganisms. Dechlorination and growth kinetics were compared using a Monod growth-rate model for multiple electron acceptor usage with competition. Included were the Victoria mixed culture containing Dehalococcoides species strain VS (from Victoria, TX), the mixed culture KB-1/VC (from southern Ontario), the Pinellas mixed culture (from Pinellas, FL), and D. ethenogenes strain 195. All cultures, with

Genome Mining Leads to the Identification of a Stable and Promiscuous Baeyer-Villiger Monooxygenase from a Thermophilic Microorganism.

Baeyer-Villiger monooxygenases (BVMOs) are NAD(P)H-dependent flavoproteins that convert ketones to esters and lactones. While these enzymes offer an appealing alternative to traditional Baeyer-Villiger oxidations, these proteins tend to be either too unstable or exhibit too narrow of a substrate scope for implementation as industrial biocatalysts. Here, sequence similarity networks were used to search for novel BVMOs that are both stable and promiscuous. Our genome mining led to the identification of an enzyme from Chloroflexota bacterium (strain G233) dubbed ssnBVMO that exhibits i) the highe

Effects of ferrous iron supplementation on reductive dechlorination of tetrachloroethene and on methanogenic microbial community.

Chloroethenes are common soil and groundwater pollutants. Their dechlorination is impacted by environmental factors, such as the presence of metal ions. We here investigated the effect of ferrous iron on bacterial reductive dechlorination of chloroethenes and on methanogen community. Reductive dechlorination of tetrachloroethene was assayed with a groundwater sample originally containing 6.3 × 103 copies mL-1 of Dehalococcoides 16S rRNA gene and 2 mg L-1 of iron. Supplementation with 28 mg L-1 of ferrous iron enhanced the reductive dechlorination of cis-dichloroethene (cis-DCE) and vinyl chlor

Reductive dechlorination of high concentrations of chloroethenes by a Dehalococcoides mccartyi strain 11G.

Chloroethenes are common groundwater and soil contaminants due to extensive historic utilization and inappropriate discharge. The tendency for chloroethenes to become sequestered as dense non-aqueous phase liquids (DNAPL)-a point source to groundwater contamination and causing high concentrations of chloroethenes in proximal aquifers poses a great challenge for remediation of chloroethene contaminated sites. In this study, we report isolation and characterization of a Dehalococcoides mccartyi strain 11G which couples growth with reductive dechlorination of trichloroethenes (TCE), dichloroethen

Kinetics and inhibition of reductive dechlorination of trichloroethene, cis-1,2-dichloroethene and vinyl chloride in a continuously fed anaerobic biofilm reactor.

Anaerobic bioreactors containing Dehalococcoides spp. can be effective for the treatment of trichloroethene (TCE) contamination. However, reductive dehalogenation of TCE often results in partial conversion to harmless ethene, and significant production of undesired cis-1,2-dichloroethene (cis-DCE) and vinyl chloride (VC) is frequently observed. Here, a detailed modeling study was conducted focusing on the determination of biokinetic constants for the dechlorination of TCE and its reductive dechlorination intermediates cis-DCE and VC as well as any biokinetic inhibition that may exist between t

Trichloroethene and cis-1,2-dichloroethene concentration-dependent toxicity model simulates anaerobic dechlorination at high concentrations: I. batch-fed reactors.

A model was developed to describe toxicity from high concentrations of chlorinated aliphatic hydrocarbons (CAHs) on reductively dechlorinating cultures under batch-growth conditions. A reductively dechlorinating anaerobic Evanite subculture (EV-cDCE) was fed trichloroethene (TCE) and excess electron donor to accumulate cis-1,2-dichloroethene (cDCE) in batch-fed reactors. A second Point Mugu (PM) culture was also studied in the cDCE accumulating batch-fed experiment, as well as in a time- and concentration-dependent cDCE exposure experiment. Both cultures accumulated cDCE to concentrations rang

Mechanisms for enhanced lignin humification with reduced organic matter loss by goethite in biogas residue composting.

In this study, effects of three iron (oxyhydr)oxides on the biogas residue composting, i.e., composting with goethite (CFe1), hematite (CFe2) or magnetite (CFe3), were investigated. Results showed that composting performance of CFe1 was much better than those of CFe2 and CFe3. Addition of goethite increased temperature of CFe1 and enhanced lignin humification. More than 31.49% of Fe(III) in goethite was reduced to amorphous Fe(II) during the composting, suggesting that goethite worked as electron acceptor for microbial metabolism and heat generation. The functional bacteria Chloroflexi and Act

Diversity of microbial communities potentially involved in mercury methylation in rice paddies surrounding typical mercury mining areas in China.

Mercury can be a serious hazard to human health, especially in paddy soils surrounding mining areas. In this study, mercury (Hg)-methylating microbes with the potential biomarker gene hgcA were obtained from 45 paddy soil samples in mercury mining areas in Fenghuang, Wanshan, and Xunyang. In different areas, the abundance of the hgcA gene was affected by different environmental factors, including organic matter, pH, total carbon content, total nitrogen content, and total mercury content. Phylogenetic analysis showed that hgcA microbes in paddy soils were potentially members of the phyla Proteo

Low-temperature Aquifer Thermal Energy Storage combined with in situ bioremediation of chlorinated ethenes: Pilot-scale observations and model-based interpretation.

Microbial reductive dechlorination is a key process in aquifers contaminated with chlorinated ethenes and results in a net mass reduction of organic pollutants. Biodegradation rates in the subsurface are temperature-dependent and may be enhanced by increased groundwater temperatures. This study explores the potential of combining the temperature increase from low-temperature Aquifer Thermal Energy Storage with In Situ Bioremediation (ATES-ISB). The effects of highly dynamic groundwater flow and heat transport on microbial degradation rates were examined in a contaminated aquifer based on a pil

Redox gradient shapes the abundance and diversity of mercury-methylating microorganisms along the water column of the Black Sea.

In the global context of seawater deoxygenation triggered by climate change and anthropogenic activities, changes in redox gradients impacting biogeochemical transformations of pollutants, such as mercury, become more likely. Being the largest anoxic basin worldwide, with high concentrations of the potent neurotoxic methylmercury (MeHg), the Black Sea is an ideal natural laboratory to provide new insights about the link between dissolved oxygen concentration and hgcAB gene-carrying (hgc+) microorganisms involved in the formation of MeHg. We combined geochemical and microbial approaches to asse

Influence of non-dechlorinating microbes on trichloroethene reduction based on vitamin B12 synthesis in anaerobic cultures.

In this study, the YH consortium, an ethene-producing culture, was used to evaluate the effect of vitamin B12 (VB12) on trichloroethene (TCE) dechlorination by transferring the original TCE-reducing culture with or without adding exogenous VB12. Ultra-high performance liquid chromatography - tandem mass spectrometry (UPLC-MS/MS) was applied to detect the concentrations of VB12 and its lower ligand 5,6-dimethylbenzimidazole (DMB) in the cultures. After three successive VB12 starvation cycles, the dechlorination of TCE stopped mostly at cis-dichloroethene (cDCE), and no ethene was found; methane

Application of molecular biological tools for monitoring efficiency of trichloroethylene remediation.

Trichloroethylene (TCE) is one of the most ubiquitous halogenated organic compounds of concerns of carcinogens in groundwater in Taiwan. Bioremediation has been recognized as a cost-effective approach in reducing TCE concentration. Five pilot-scale wells were constructed to monitor TCE concentrations in contaminated groundwater. With injection of EOS®, TCE was effectively degraded to 42%-93% by the end of 175 days. The biostimulation with EOS® was useful in establishing a micro-site anaerobic but with limited contribution. Dilution of the aquifer movement also caused the TCE reduction among in

Impacts of the invasive Spartina anglica on C-S-Hg cycles and Hg(II) methylating microbial communities revealed by hgcA gene analysis in intertidal sediment of the Han River estuary, Yellow Sea.

We investigated the impact of invasive vegetation on mercury cycles, and identified microorganisms directly related to Hg(II) methylation using hgcA gene in vegetated mud flats (VMF) inhabited by native Suaeda japonica (SJ) and invasive Spartina anglica (SA), and unvegetated mud flats (UMF) in Ganghwa intertidal sediments. Sulfate reduction rate (SRR) and rate constants of Hg(II) methylation (Km) and methyl-Hg demethylation (Kd) were consistently greater in VMF than in UMF, specifically 1.5, 2 and 11.7 times higher, respectively, for SA. Both Km and Kd were significantly correlated with SRR an

Enhanced resilience to oxygen exposure and toxicity of chlorinated solvents in immobilized Dehalococcoides mccartyi.

Members of Dehalococcoides mccartyi (Dhc) are strictly anaerobic and play crucial roles in the restoration of many industrial sites impacted by chlorinated solvents, such as tetrachloroethene (PCE) and trichloroethene (TCE). In situ bioremediation with Dhc involves intricate procedures intended to minimize oxygen intrusion, and achieving optimal dechlorination performance in aquifers near the dense non-aqueous phase liquids source zone is challenging. Here, we respectively embedded Dhc strain 195 and the biomass of a Dhc-containing, PCE-dechlorinating consortium in poly(vinyl alcohol)-alginate

[Characterization of Reductive Dechlorination of Chlorinated Ethylenes by Anaerobic Consortium].

Tetrachloroethylene (PCE) and trichloroethylene (TCE) are typical volatile halogenated organic compounds in groundwater that pose serious threats to the ecological environment and human health. To obtain an anaerobic microbial consortium capable of efficiently dechlorinating PCE and TCE to a non-toxic end product and to explore its potential in treating contaminated groundwater, an anaerobic microbial consortium W-1 that completely dechlorinated PCE and TCE to ethylene was obtained by repeatedly feeding PCE or TCE into the contaminated groundwater collected from an industrial site. The dechlor

Resilience of organohalide-detoxifying microbial community to oxygen stress in sewage sludge.

Organohalide pollutants are prevalent in the environment, causing harms to wildlife and human. Organohalide-respiring bacteria (OHRB) could detoxify these pollutants in anaerobic environments, but the most competent OHRB (i.e., Dehalococcoides) is susceptible to oxygen. This study reports exceptional resistance and resilience of sewage sludge microbial communities to oxygen stress for attenuation of structurally distinct organohalide pollutants, including tetrachloroethene, tetrabromobisphenol A, and polybrominated diphenyl ethers. The dehalogenation rate constant of these organohalide polluta

Managing methanogens and homoacetogens to promote reductive dechlorination of trichloroethene with direct delivery of H2 in a membrane biofilm reactor.

A study with H(2)-based membrane biofilm reactors (MBfRs) was undertaken to examine the effectiveness of direct H(2) delivery in ex-situ reductive dechlorination of chlorinated ethenes. Trichloroethene (TCE) could be reductively dechlorinated to ethene with up to 95% efficiency as long as the pH-increase effects of methanogens and homoacetogens were managed and dechlorinators were selected for during start-up by creating H(2) limitation. Based on quantitative PCR, the dominant bacterial groups in the biofilm at the end of reactor operation were Dehalococcoides, Geobacter, and homoacetogens. Py

Transcriptional expression of the tceA gene in a Dehalococcoides-containing microbial enrichment.

Dynamic changes in the transcriptional expression of the tceA gene, which encodes a trichloroethene reductive dehalogenase, were characterized in a Dehalococcoides-containing microbial enrichment culture. Expression was quantified by real-time PCR as the number of tceA transcripts per tceA gene. Expression of tceA increased 40-fold after chlorinated ethene-starved cells were exposed to trichloroethene (TCE), cis-dichloroethene (DCE), or 1,1-DCE but did not increase after exposure to tetrachloroethene or vinyl chloride. Surprisingly, tceA expression also increased 30-fold after cellular exposur

Ozone direct oxidation pretreatment and catalytic oxidation post-treatment coupled with ABMBR for landfill leachate treatment.

In order to treat the high concentration landfill leachate, ozone direct oxidation pretreatment and catalytic oxidation post-treatment coupled with anaerobic baffled membrane bioreactor (ABMBR) system was proposed in this study. For pretreatment, ozone direct oxidation could remarkably reduce UV254, 3D fluorescence peak value and fluorescence regional integration (FRI) of organic pollutants. For ABMBR treatment, the removal efficiencies of COD and ammonia nitrogen were 80.38% and 21.56%, respectively. Post-treatment included struvite precipitation, ozone catalytic oxidation and membrane biorea

Widespread microbial mercury methylation genes in the global ocean.

Methylmercury is a neurotoxin that bioaccumulates from seawater to high concentrations in marine fish, putting human and ecosystem health at risk. High methylmercury levels have been found in the oxic subsurface waters of all oceans, but only anaerobic microorganisms have been shown to efficiently produce methylmercury in anoxic environments. The microaerophilic nitrite-oxidizing bacteria Nitrospina have previously been suggested as possible mercury methylating bacteria in Antarctic sea ice. However, the microorganisms responsible for processing inorganic mercury into methylmercury in oxic sea
