Characterization of ribulose-1,5-bisphosphate carboxylase/oxygenase carrying ribulose 1,5-bisphosphate at its regulatory sites and the mechanism of interaction of this form of the enzyme with ribulose-1,5-bisphosphate-carboxylase/oxygenase activase.

Ribulose-1,5-bisphosphate carboxylase/oxygenase [Rbu(1,5)P2CO] from plant sources shows a biphasic reaction course when assayed with more than 2 mM ribulose 1,5-bisphosphate [Rbu(1,5)P2]. In the burst, Rbu(1,5)P2CO has its substrate-binding sites occupied with Rbu(1,5)P2 for the initial few minutes, then both substrate-binding and regulatory sites are occupied by Rbu(1,5)P2 in the subsequent linear phase, at physiological concentrations of Rbu(1,5)P2 [A. Yokota (1991) J. Biochem. (Tokyo) 110, 246-252]. This study attempts the characterization of spinach Rbu(1,5)P2CO carrying Rbu(1,5)P2 at the

Genome-wide identification of the peanut HD-Zip gene family and AhHDZ15 positively regulating salt and drought stress in heterologously overexpressed Arabidopsis.

Homeodomain-leucine zipper (HD-Zip) transcription factors play important roles in plant growth, development, and abiotic stress responses. However, bioinformatic analyses and functional studies of HD-Zip family in peanut are scarce. In this study, 128 AhHDZ genes were identified and classified into four subfamilies in the phylogenetic analysis. Transcriptomic data and RT-qPCR analysis indicated the expression levels of AhHDZ4 and AhHDZ15 were significantly elevated in response to 12 h of salt stress, while AhHDZ4/15/60/69/126 all showed a progressive increase over time in response to drought s

Physiological and transcriptomic responses of the microalga Isochrysis galbana during exposure to Hg(II) stress.

Heavy metal contamination, particularly mercury (Hg), represents a substantial threat to aquatic ecosystems and primary producers. In this study, we systematically examined the impacts of varying concentrations of Hg(II) on Isochrysis galbana in terms of growth, chlorophyll a content, soluble protein levels, and ultrastructure. Results demonstrated that when Hg(II) concentrations exceeded 0.2 mg/L, the growth of I. galbana was significantly inhibited. At Hg(II) concentrations below 0.2 mg/L, the activities of antioxidant enzymes such as superoxide dismutase (SOD), catalase (CAT), and peroxidas

Dissecting the Regulatory Network of Leaf Premature Senescence in Maize (Zea mays L.) Using Transcriptome Analysis of ZmELS5 Mutant.

Leaf premature senescence largely determines maize (Zea mays L.) grain yield and quality. A natural recessive premature-senescence mutant was selected from the breeding population, and near-isogenic lines were constructed using Jing24 as the recurrent parent. In the near-isogenic lines, the dominant homozygous material was wild-type (WT), and the recessive material of early leaf senescence was the premature-senescence-type ZmELS5. To identify major genes and regulatory mechanisms involved in leaf senescence, a transcriptome analysis of the ZmELS5 and WT near-isogenic lines (NILs) was performed

Effect of Thallium(I) on Growth, Nutrient Absorption, Photosynthetic Pigments, and Antioxidant Response of Dittrichia Plants.

Dittrichia plants were exposed to thallium (Tl) stress (10, 50, and 100 µM) for 7 days. The Tl toxicity altered the absorption and accumulation of other nutrients. In both the roots and the leaves, there was a decline in K, Mg, and Fe content, but an increase in Ca, Mn, and Zn. Chlorophylls decreased, as did the photosynthetic efficiency, while carotenoids increased. Oxidative stress in the roots was reflected in increased lipid peroxidation. There was more production of superoxide (O2.-), hydrogen peroxide (H2O2), and nitric oxide (NO) in the roots than in the leaves, with increases in both o

Touch, light, wounding: how anaesthetics affect plant sensing abilities.

Anaesthetics affect not only humans and animals but also plants. Plants exposed to certain anaesthetics lose their ability to respond adequately to various stimuli such as touch, injury or light. Available results indicate that anaesthetics modulate ion channel activities in plants, e.g. Ca2+ influx. The word anaesthesia means loss of sensation. Plants, as all living creatures, can also sense their environment and they are susceptible to anaesthesia. Although some anaesthetics are often known as drugs with well-defined target to their animal/human receptors, some other are promiscuous in their

In Vivo Identification of Photosystem II Light Harvesting Complexes Interacting with PHOTOSYSTEM II SUBUNIT S.

Light is the primary energy source for photosynthetic organisms, but in excess, it can generate reactive oxygen species and lead to cell damage. Plants evolved multiple mechanisms to modulate light use efficiency depending on illumination intensity to thrive in a highly dynamic natural environment. One of the main mechanisms for protection from intense illumination is the dissipation of excess excitation energy as heat, a process called nonphotochemical quenching. In plants, nonphotochemical quenching induction depends on the generation of a pH gradient across thylakoid membranes and on the pr

Physiological and Transcriptome Responses to Combinations of Elevated CO2 and Magnesium in Arabidopsis thaliana.

The unprecedented rise in atmospheric CO2 concentration and injudicious fertilization or heterogeneous distribution of Mg in the soil warrant further research to understand the synergistic and holistic mechanisms involved in the plant growth regulation. This study investigated the influence of elevated CO2 (800 μL L(-1)) on physiological and transcriptomic profiles in Arabidopsis cultured in hydroponic media treated with 1 μM (low), 1000 μM (normal) and 10,000 μM (high) Mg2+. Following 7-d treatment, elevated CO2 increased the shoot growth and chlorophyll content under both low and normal Mg s

Differential effects of NaCl and Na2SO4 on the halophyte Prosopis strombulifera are explained by different responses of photosynthesis and metabolism.

Prosopis strombulifera (Lam.) Benth. is a halophytic shrub found in highly saline soils in Argentina, with high tolerance against NaCl but strong growth inhibition by Na2SO4. In the present study, the differences in the physiological responses caused by these salts and an iso-osmotic combination thereof on photosynthesis, mineral composition and metabolism were analyzed. Na2SO4 treated plants were the most affected by salinity, showing a significant decrease in several photosynthetic parameters. Proline and cysteine accumulated significantly in the plants in response to salt stress. These resu

Cellular regulation of iron assimilation.

Cells of plants, most microorganisms, and animals require well-defined amounts of iron for survival, replication, and differentiation. The metal is an important component of such processes as synthesis of DNA, RNA, and chlorophyll; electron transport; oxygen metabolism; and nitrogen fixation. Because of the insolubility of iron in aerobic environments at neutral and alkaline pH values, cells have had to devise specific strategies to assimilate the metal. These include (1) development of systems for reducing ferric ions to the more soluble ferrous ions at the cell surface, (2) employment of sma

Physiological-Biochemical Characteristics and Omics Landscape of Drought Tolerance in Chlorella sp. from the Taklamakan Desert of China.

Microalgae endemic to desert ecosystems possess exceptional stress-resistance traits; however, the physiological and molecular drought-adaptation mechanisms of desert-dwelling Chlorella remain largely uncharacterized. Herein, we combined physiological-biochemical assays, ultrastructural observation, transcriptomics and metabolomics to dissect drought responses in Taklamakan-desert-origin Chlorella sp. under polyethylene glycol 6000 (PEG-6000)-simulated mild (10%), moderate (20%), and severe (30%) drought stress, with mechanistic focus on moderate-drought conditions. Moderate drought repressed

Physiological and transcriptional responses to heat stress and functional analyses of PsHSPs in tree peony (Paeonia suffruticosa).

Tree peony (Paeonia suffruticosa) is a traditional Chinese flower that is not resistant to high temperatures, and the frequent sunburn during summer limits its normal growth. The lack of understanding of the molecular mechanisms in tree peony has greatly restricted the improvement of novel heat-tolerant varieties. Therefore, we treated tree peony cultivar "Yuhong" (P. suffruticosa "Yuhong") at normal (25°C) and high temperatures (40°C) and sequenced the transcriptomes, to investigate the molecular responsive mechanisms to heat stress. By comparing the transcriptomes, a total of 7,673 different

A PROMOTER::LUCIFERASE reporter system reveals key elements of the circadian regulation of Crassulacean acid metabolism (CAM) in Kalanchoë laxiflora Baker.

Crassulacean acid metabolism (CAM) plants perform primary atmospheric CO2 fixation at night, with timekeeping by the endogenous circadian clock. Understanding of circadian coordination of CAM remains limited to rhythmic post-translational regulation of phosphoenolpyruvate carboxylase (PPC) by a specific clock-controlled protein kinase, PPCK. Here, candidate promoter regions (~3000 bp) of CAM-associated genes from Kalanchoë laxiflora Baker were coupled to a firefly luciferase reporter and stable transgenic lines of both K. laxiflora and C3 Arabidopsis thaliana were generated. In K. laxiflora, t

MgO and Zn/MgO Nanoparticles as Direct Antifungal Compounds and Elicitors Against Sclerotinia sclerotiorum in Pea.

The purpose of this study was to assess the effectiveness of two nanostructures (MgO and Zn/MgO) against Sclerotinia sclerotiorum, which causes white mold disease in peas, as direct antifungal agents or resistance inducers in pea plants. The direct antifungal activity of these nanostructures was evaluated by assessing their ability to inhibit S. sclerotiorum growth in vitro and reduce white mold severity in the greenhouse. The induction of resistance in pea plants was examined by assessing the expression of three defense-related genes using quantitative real-time PCR and measuring the phenolic

The Molecular Mechanism Regulating Flavonoid Production in Rhododendron chrysanthum Pall. Against UV-B Damage Is Mediated by RcTRP5.

Elevated levels of reactive oxygen species (ROS) are caused by ultraviolet B radiation (UV-B) stress. In response, plants strengthen their cell membranes, impeding photosynthesis. Additionally, UV-B stress initiates oxidative stress within the antioxidant defense system and alters secondary metabolism, particularly by increasing the quantity of UV-absorbing compounds such as flavonoids. The v-myb avian myeloblastosis viral oncogene homolog (MYB) transcription factor (TF) may participate in a plant's response to UV-B damage through its regulation of flavonoid biosynthesis. In this study, we dis

A new ABC half-transporter in Leishmania major is involved in resistance to antimony.

The characterization of ABCI4, a new intracellular ATP-binding cassette (ABC) half-transporter in Leishmania major, is described. We show that ABCI4 is involved in heavy metal export, thereby conferring resistance to Pentostam, to Sb(III), and to As(III) and Cd(II). Parasites overexpressing ABCI4 showed a lower mitochondrial toxic effect of antimony by decreasing reactive oxygen species production and maintained higher values of both the mitochondrial electrochemical potential and total ATP levels with respect to controls. The ABCI4 half-transporter forms homodimers as determined by a coimmuno

Significant differences of toxicity, bioconcentration and biomagnification effects of surfactants on organisms in the food chain under combined exposure from those under single exposure.

Surfactants, the primary active components in cleaning and disinfection products, are being discharged into the water environment and threatening the health of aquatic organisms. Moreover, the combined exposure of surfactants exacerbates the risks. However, the combined exposure effects such as toxicity, bioconcentration and biomagnification effects of surfactants on the organisms especially those in the food chain remain insufficiently unexplored. Therefore, the combined exposure effects of three surfactants, cetylpyridinium chloride monohydrate (CPC), didodecyldimethyl ammonium bromide (DDAB

Mechanism Study of the Protective Effects of Sodium Tanshinone IIA Sulfonate Against Atorvastatin-Induced Cerebral Hemorrhage in Zebrafish: Transcriptome Analysis.

Hemorrhage stroke is a severe vascular disease of the brain with a high mortality rate in humans. Salvia miltiorrhiza Bunge (Danshen) is a well-known Chinese Materia Medica for treating cerebral vascular and cardiovascular diseases in traditional Chinese medicine. Sodium tanshinone IIA sulfonate (STS) is a water-soluble derivative of tanshinone IIA, which is the main active ingredient of Danshen. In our previous study, we established a zebrafish model of cerebral hemorrhage and found that STS dramatically decreased both the hemorrhage rate and hemorrhage area, although the underlying mechanism

Common-Garden Transcriptomes Distinguish Leaf-Form-Associated from Site-Associated Expression in Zygophyllum xanthoxylum.

Zygophyllum xanthoxylum (Bunge) Maxim. is a dominant, deep-rooted shrub of the temperate deserts of north-western China whose leaflets range from comparatively flat and broad to slender and terete, but the molecular basis of this variation has rarely been reported. We compared three accessions from contrasting desert habitats-one flat-leaved, from a shaded high-altitude valley, and two rod-leaved, from open full-sun sites. The accessions differed in the leaflet blade but not in the petiole, the flat-leaved accession bearing shorter and much wider leaflets with a width-to-length ratio almost fo
