Assessing the Threat of Amphibian Chytrid Fungus in the Albertine Rift: Past, Present and Future.

Batrachochytrium dendrobatidis (Bd), the cause of chytridiomycosis, is a pathogenic fungus that is found worldwide and is a major contributor to amphibian declines and extinctions. We report results of a comprehensive effort to assess the distribution and threat of Bd in one of the Earth's most important biodiversity hotspots, the Albertine Rift in central Africa. In herpetological surveys conducted between 2010 and 2014, 1018 skin swabs from 17 amphibian genera in 39 sites across the Albertine Rift were tested for Bd by PCR. Overall, 19.5% of amphibians tested positive from all sites combined

Microbial diversity characteristics and the influence of environmental factors in a large drinking-water source.

Although the influence of environmental factors on the microbial community in water sources is crucial, it is seldom evaluated. The seasonal relationship between microbial diversity of bacteria and fungi and environmental factors was investigated in a large drinking-water reservoir using Illumina MiSeq sequencing. Forty-one bacterial phyla and nine fungal phyla were analyzed in the Qingcaosha Reservoir, Shanghai, China. The predominant bacterial phyla were Actinobacteria, Proteobacteria, Bacteroidetes, and Cyanobacteria, with the maximum relative abundance of 46%, 36.6%, 16.1%, and 14.9%, resp

A multifactor analysis of fungal and bacterial community structure in the root microbiome of mature Populus deltoides trees.

Bacterial and fungal communities associated with plant roots are central to the host health, survival and growth. However, a robust understanding of the root-microbiome and the factors that drive host associated microbial community structure have remained elusive, especially in mature perennial plants from natural settings. Here, we investigated relationships of bacterial and fungal communities in the rhizosphere and root endosphere of the riparian tree species Populus deltoides, and the influence of soil parameters, environmental properties (host phenotype and aboveground environmental settin

Chlamydia pneumoniae infection in a breeding colony of African clawed frogs (Xenopus tropicalis).

More than 90% of a breeding colony of clawed frogs (Xenopus tropicalis) imported to the United States from western Africa died in an epizootic of chlamydiosis. Chlamydial inclusions were observed by light and electron microscopy in the liver of an infected frog. Chlamydia pneumoniae was isolated in cell cultures from four frogs. A cutaneous infection by a chytridiomycete fungus observed in two frogs could have been a cofactor in the die-off.ous Diseases

The absence of zoonotic agents in invasive bullfrogs (Lithobates catesbeianus) in Belgium and The Netherlands.

Exotic invasive bullfrogs (Lithobates catesbeianus) are considered to exert a considerable negative impact on native amphibian communities. This can be due to competition and predation, but they are also a notorious source of the infectious diseases chytridiomycosis and ranavirosis, affecting amphibian populations globally. Little is known regarding their carriage of other microbial agents that might be transferred to humans or other animals. In this study we determined the occurrence of the amphibian pathogens Ranavirus and Batrachochytrium dendrobatidis and of the zoonotic agents Coxiella bu

Phylogeny of the genus Synchytrium and the development of TaqMan PCR assay for sensitive detection of Synchytrium endobioticum in soil.

Potato wart, caused by the fungal pathogen Synchytrium endobioticum, is a serious disease with the potential to cause significant economic damage. The small subunit (SSU) and internal transcribed spacer (ITS) ribosomal DNA (rDNA) were sequenced for several Synchytrium spp., showing a high rate of variability for both of these markers among the different species and monophyly of the genus within phylum Chytridiomycota. The intergenic nontranscribed spacer (IGS) of rDNA was sequenced for different pathotypes and showed no intraspecific variation within S. endobioticum, similar to the other rDNA

Regulatory Mechanism of Carbon Fractions and Microbial Metabolism on Organic Carbon Turnover in Saline-Alkali Soils.

Soil salinization is a serious threat to agricultural productivity and organic carbon cycling; however, the mechanisms by which carbon fractions and microbial metabolism regulate organic carbon sequestration remain unclear. This research investigated the effects of four sites (S1-S4) along a salinity gradient on microbial carbon metabolism, carbon fractions (particulate organic carbon, POC; mineral-associated organic carbon, MAOC), and bacterial and fungal communities in a typical saline-alkali soil. The results showed that, as the salt-alkali level increases, microbial growth, respiration, an

Cross-infection by multiple soil-borne pathogens alters the physicochemical properties and microbial community structure of tobacco rhizosphere soil.

Soil-borne diseases of tobacco roots and stems, caused by pathogens including Ralstonia solanacearum (bacterial wilt), Phytophthora parasitica var. nicotianae (black shank), and root-knot nematodes (Meloidogyne spp.), represent critical threats to tobacco cultivation. However, information regarding alterations in rhizosphere soil properties and microbial communities following co-infection by multiple soil-borne pathogens remains limited for tobacco-growing regions in Luxi County, Honghe Prefecture, Yunnan Province, China. This study therefore investigated rhizosphere soils collected from healt

The Saccharomyces killer toxin K62 is a protein of the aerolysin family.

UNLABELLED: K62 is an antifungal killer toxin produced by Saccharomyces paradoxus, encoded by a double-stranded RNA satellite. The toxin exhibits a unique antifungal activity but lacks sequence homology to other killer toxins, and its antifungal mechanism of action remains unknown. To understand the function of K62, its tertiary structure was predicted using AlphaFold, followed by molecular dynamics simulations to create high-confidence molecular models. These analyses revealed that K62 monomers closely resemble the five-beta-strand domain found in pore-forming aerolysin toxins. Models of K62

Study on soil improvement and growth-promoting effect of compound microbial agents in tomato cultivation.

Using microorganisms to improve and repair soil and assist plant growth is an effective ecological restoration method. The compound microbial inoculant prepared by mixing several microorganisms with different functions is one of the hot spots in the current research of agricultural green development and food security. In this study, four treatments were set up: no microbial agent (CK), Trichoderma harzianum agent (H), Bacillus subtilis agent (K), and compound microbial agent (K + H). The effects of single and compound microbial agents on tomato growth, rhizosphere soil physical and chemical pr

Neurochemical alterations of different cerebral regions in rats with myocardial ischemia-reperfusion injury based on proton nuclear magnetic spectroscopy analysis.

BACKGROUND: Recent studies have demonstrated a complex and dynamic neural crosstalk between the heart and brain. A heart-brain interaction has been described regarding cardiac ischemia, but the cerebral metabolic mechanisms involved are unknown. METHODS: Male Sprague Dawley rats were randomly allocated into 2 groups: those receiving myocardial ischemia-reperfusion surgery (IR group, n =10) and surgical controls (Con group, n=10). These patterns of metabolic abnormalities in different brain regions were assessed using proton magnetic resonance spectroscopy (PMRS). RESULTS: Results assessed by

WASP and SCAR are evolutionarily conserved in actin-filled pseudopod-based motility.

Diverse eukaryotic cells crawl through complex environments using distinct modes of migration. To understand the underlying mechanisms and their evolutionary relationships, we must define each mode and identify its phenotypic and molecular markers. In this study, we focus on a widely dispersed migration mode characterized by dynamic actin-filled pseudopods that we call "α-motility." Mining genomic data reveals a clear trend: only organisms with both WASP and SCAR/WAVE-activators of branched actin assembly-make actin-filled pseudopods. Although SCAR has been shown to drive pseudopod formation,
