Oxygen underpins the temperature-size relationship of protist plankton.

UNLABELLED: Temperature is known to affect organismal size, yet the underlying mechanism has been debated. Here, we develop a simple growth model of heterotrophic plankton and explain the temperature-size effect based on the demand and supply of oxygen. The model applies the growth rate-temperature relationship based on protein stability, capturing six data sets of a planktonic ciliate Urotricha. The model then computes the oxygen demand, based on the growth rate, which must be balanced by the diffusive uptake of oxygen. Because the smaller cell size favors oxygen uptake per cell volume, the p

Gorilloflasca africana n.g., n.sp., (Entodiniomorphida) from wild habituated Virunga mountain gorillas (Gorilla beringei beringei) in Rwanda.

A new entodiniomorphid ciliate species, Gorilloflasca africana n. g., n. sp. was described from the Virunga mountain gorillas, Gorilla beringei beringei, in Rwanda. It is characterized by a flask-shaped body, a long tubular vestibulum, a round frontal lobe, a large posterior cavity, an ellipsoidal or peanut-shaped macronucleus and a single contractile vacuole. G. africana has the adoral and the vestibular ciliary zones in the buccal area. The adoral ciliary zone is non-retractable, encircling the vestibular opening. The vestibular ciliary zone extends posteriorly in the vestibulum. The somatic

Molecular characterization of laao in large yellow croaker and its functional response to Cryptocaryon irritans infection.

L-amino acid oxidase (laao) is a key immune factor capable of producing reactive oxygen species (ROS) and has been demonstrated to possess significant antibacterial and immunomodulatory functions in a variety of organisms. In recent years, increasing attention has been paid to the role of laao in the immune defense of fish against Cryptocaryon irritans infection. In this study, the laao of large yellow croaker (Larimichthys crocea) was successfully cloned, and its structural and functional characteristics were systematically analyzed. The full-length ORF of large yellow croaker was 1578 bp, en

Updating specific PCR primer for detection of Cryptocaryon irritans from reared Larimichthys polyactis.

Artificial breeding of small yellow croaker (Larimichthys polyactis) was recently achieved, providing a bright future for its commercial farming. In May 2019, a disease outbreak occurred among small yellow croakers in an aquaculture farm near Xiangshan Bay, charactering by white spots spotted on the surface of fish skin, gills and fins. The parasite was preliminarily identified as Cryptocaryon irritans based on morphological feature of the parasite and the symptoms on fish. However, the previously published specific primer pairs failed to confirm the existence of C. iriitans. Six nucleotides m

l-amino acid oxidase expression profile and biochemical responses of rabbitfish (Siganus oramin) after exposure to a high dose of Cryptocaryon irritans.

Cryptocaryon irritans is an important protozoan parasite which infects almost all kinds of marine teleosts, causing heavy economic losses. In our previous studies, we found that rabbitfish (Siganus oramin) displayed high resistance to C. irritans infection, and a novel protein, l-amino acid oxidase (LAAO), was identified from the serum that was lethal to C. irritans. In this study, the rabbitfish were firstly infected with a high dose of C. irritans, then the LAAO mRNA expression pattern and the activity of three enzymes [superoxide dismutase (SOD), Na+/K+-ATPase and Ca2+/Mg2+-ATPase] were mea

Molecular cloning and expression analysis of CCL25 and its receptor CCR9s from Epinephelus coioides post Cryptocaryon irritans infection.

Among other functions, CCL25/CCR9 has an important role in regulating the trafficking of developing T cells in the thymus, and in homing memory T cells to the small intestine. The function of this chemokine-receptor complex is not well studied in fish. We identified a CCL25-like (EcCCL25, 108 aa) and two CCR9-like sequences (EcCCR9aa 373 aa; and EcCCR9b, 375 aa) from a transcriptome database of orange-spotted grouper (Epinephelus coioides). EcCCL25, EcCCR9a, and EcCCR9b shared conserved structural features with homologs from mammals and from other fish, and a consistent relationship with phylo

Grouper (Epinephelus coioides) BCR signaling pathway was involved in response against Cryptocaryon irritans infection.

B cell antigen receptor (BCR) plays a crucial role in B cell development and antibody production. It comprises membrane immunoglobulin non-covalently associated with CD79a/CD79b heterodimer. After B cell activation, initial extracellular signals are transduced by BCR complex and amplified by two protein tyrosine kinases, LYN and SYK, which then trigger various pathways. In the present study, we cloned grouper genes for BCR accessory molecules, EcCD79a (669 bp) and EcCD79b (639 bp), as well as two protein tyrosine kinases, EcLYN (1482 bp) and EcSYK (1854 bp). Homology analysis showed that all f

Effects of a Synthetic Isoquinoline Derivative Against Ichthyophthirius multifiliis In Vivo and In Vitro in Grass Carp (Ctenopharyngodon idella).

White spot disease, caused by the parasitic Ichthyophthirius multifiliis, induces mortality exceeding 80% in intensive aquaculture systems, resulting in global annual losses of >$1 billion. Current chemical controls (e.g., formaldehyde) face environmental persistence and drug resistance challenges. This study developed a synthetic isoquinoline derivative, BHTCA (s)-2-tert-butoxycarbonyl-7-hydroxy-1,2,3,4-tetrahydroisoquinoline-3-carboxylic acid), as a biodegradable alternative against I. multifiliis in grass carp (Ctenopharyngodon idella). Antibiotic nitrofurazone drives the functional dynamics of periphytic protozoan fauna in marine environments.

The use of functional traits of a community as a method to measure its functional dynamics in response to environmental change has gained attention because trait-based approaches offer systematic opportunities to understand the interactions between species diversity and ecosystem function. However, the relationship between functional traits of periphytic protozoa and contamination of aquatic habitats with antibiotics is poorly understood. In this study, we investigated the influence of the antibiotic nitrofurazone on functional traits of marine periphytic protozoan fauna. For this purpose, the

Removal of two pathogenic scuticociliates Miamiensis avidus and Miamiensis sp. using cells or culture filtrates of the dinoflagellate Alexandrium andersonii.

Scuticociliatosis, which is caused by parasitic protistan pathogens known as scuticociliates, is one of the most serious diseases in marine aquaculture worldwide. Thus, elimination of these ciliates is a primary concern for scientists and managers in the aquaculture industry. To date, formalin and other toxic chemicals have been used as anti-scuticociliate agents, but issues regarding their secondary effects often arise. Consequently, development of safer methods is necessary. To find out a safe method of controlling scuticociliate populations in aqua-tanks or small-scale natural environments,

Species Dynamics and Gene Expression in Algal Polycultures Under Mixotrophic Cultivation in Outdoor Pilot-Scale Nordic Conditions.

Mixotrophic cultivation of microalgal polycultures can enhance biomass productivity and nutrient recovery from wastewaters compared to autotrophic systems. To better understand non-axenic polycultures under mixotrophic conditions, this study examined species composition and gene expression in a local freshwater green algal polyculture cultivated in outdoor raceway ponds in Southeast Sweden during spring and autumn. Cultures were cultivated under autotrophic conditions in leachate-KH2PO4 medium or under mixotrophic conditions with glucose or whey permeate supplementation. Community composition

Protozoan Communities and Their Contribution to Predation on E. coli in Aerobic Granular Sludge.

Protozoa contribute to water purification through predation in wastewater treatment systems. Full-scale aerobic granular sludge (AGS) reactors treating municipal wastewater contain AGS of varying sizes, with those larger than 2 mm dominating. These size fractions exhibit different sludge morphologies and microbial communities. To date, little is known about protozoan communities and their role in the removal of human-associated bacteria (like pathogens) in AGS plants, particularly across different size fractions. This study conducted uptake experiments with fluorescent Escherichia coli, as a m

Metacollinia emscheri n. sp., a novel sanguicolous apostome ciliate of freshwater amphipods (Gammarus spp.).

We describe a novel sanguicolous parasitic ciliate, Metacollinia emscheri n. sp., found in the freshwater amphipods Gammarus pulex and G. fossarum. This ciliate infected 8.05 % of the amphipods collected in a German stream catchment, the Boye, a tributary of the river Emscher. The ciliate showed morphological characteristics fitting the genus Metacollinia. Different life stages of variable size occurred simultaneously in the hemocoel throughout the hosts' body. The tomont had 40-47 slightly spiraled kineties, a non-ciliated cortical band, a large macronucleus, and contractile vacuoles arranged

Evidence for three "classes" of microtubules in the interpolar space of the mitotic micronucleus of a ciliate and the participation of the nuclear envelope in conferring stability to microtubules.

Exposure of Nyctotherus ovalis to low temperatures or vinblastine caused similar reactions of "classes" of microtubules (mt) present in the mitotic micronucleus of this ciliate towards both treatments. However, differences of sensitivity between certain "classes" of mt at individual mitotic stages exist. Unlike the kinetochore mt (kmt) of most other eukaryotic cells, kmt in Nyctotherus completely disassemble after incubation at 6-8 degrees C (60 min) and most disappear after prolonged exposure to vinblastine (10(-5) M, 16 h). The depolymerization of kmt causes the collapse of the spindle and a

Characterization of E3 ubiquitin ligase neuregulin receptor degradation protein-1 (Nrdp1) in the large yellow croaker (Larimichthys crocea) and its immune responses to Cryptocaryon irritans.

Neuregulin receptor degradation protein-1 (Nrdp1) was recently identified in humans as an important immune factor responding to the challenge of virus, LPS or cytokine. Its role in fish immune defense and whether it is involved in anti-parasite immunity have not been proven yet. In this report, the full-length cDNA sequence and genomic structure of Nrdp1 in the large yellow croaker Larimichthys crocea (LcNrdp1) were identified and characterized. The full-length cDNA of LcNrdp1 was 1248bp, including a 5' untranslated region (UTR) of 32bp, a 3' UTR of 259bp and an open reading frame (ORF) of 937

Cytoplasmic streaming of symbiotic algae in the ciliate Stentor pyriformis.

Stentor pyriformis is a unicellular organism whose inherent green-algal symbionts can be utilized in evolutionary and cytological studies. The cytoplasm contains symbiotic algae and starch granules, which are in constant motion. The habitats of the ciliate S. pyriformis are restricted to a few oligotrophic ponds in Japan. This study aimed to develop a culture medium for long-term incubation based on pond water quality data and to investigate the cytoplasmic streaming of symbiotic algae and starch granules in S. pyriformis. In addition, the involvement of the cytoskeleton and motor proteins in

A safe antiparasitic extract from Psoralea corylifolia for Tetrahymeniasis control.

Tetrahymeniasis is a ciliate disease that presents significant economic challenges for the aquaculture industry. Previous research has shown promising control effects on Tetrahymena pyriformis using flavonoids from Psoralea corylifolia (P. corylifolia), but their high cost hinders practical application. This study aims to find an affordable and safe alternative antiparasitic extract derived from P. corylifolia. Initially, six different solvent extracts from P. corylifolia were compared for in vitro antiparasitic activity and toxicity, with ethyl acetate (EtOAc) extract selected for in vivo tes

Protistan epibionts affect prey selectivity patterns and vulnerability to predation in a cyclopoid copepod.

Colonisation of crustacean zooplankton with ciliate epibionts is widespread in freshwater and marine environments. However, the ecology of such association are little studied as yet. The occurrence of ciliate epibionts on copepods and the preference towards this association with different life stages of Mesocyclops were studied from winter to spring. Relative susceptibility of zooplankton species was evaluated by analysing the epibiont colonies and zooids and relate this to the surface area of the host. The maximum epibiont infestation per unit body surface area was recorded on copepodites fol

Inhibitory effect of melittin on endonuclease-like activity of centrin.

The xeroderma pigmentosum group C protein (XPC) and centrin2 are the primary initiators of global genome nucleotide excision repair (NER). Centrin, acts as a member of the EF-hand super family of calcium-binding proteins, playing roles in reconstitution of the vitro NER reaction. To understand the possible molecular and structural properties of the multiprotein process, the interactions of Euplotes octocarinatus centrin (EoCen), melittin, and DNA are described. EoCen shares a sequence identity of 66% with centrin2. Melittin possesses inverse direction hydrophobic triads-leucine-leucine-tryptop
