Screening for Safe and Efficient Monascus Strains with Functions of Lowering Blood Lipids, Blood Glucose, and Blood Pressure.

Monascus is a fungus widely used in food fermentation. This study employed microbial technology, combined with microscopic morphological observations and ITS sequence analysis, to isolate, purify, and identify 10 strains of red yeast mold from various Monascus products. After the HPLC detection of metabolic products, the M8 strain containing the toxic substance citrinin was excluded. Using the EWM-TOPSIS model, the remaining nine safe Monascus strains were evaluated for their inhibitory activities against pancreatic lipase, α-glucosidase, α-amylase, and the angiotensin-converting enzyme. The M

Chemical response of Picea glehnii seed-epiphytic Penicillium species to Pythium vexans under in vitro competitive conditions for mycelial growth.

The potential protection of Picea glehnii seedlings from damping-off by seed-epiphytic Penicillium species was investigated. We studied the chemical response of seed-epiphytic Penicillium species (Pen. cyaneum, Pen. damascenum, and Pen. implicatum) to Pythium vexans, a damping-off fungus, in vitro. Penicillium species were cultured singly or cocultured with Pyt. vexans for 14 or 18 d, and mycelial growth, pH of culture filtrate, antifungal activity of the culture filtrate against Pyt. vexans, and the amount of antifungal compound produced by each Penicillium species, were examined. The filtrat

Microbial diversity, functional activities, and safety risks in fermented tea: a comprehensive review.

Microbial fermented teas are gaining global popularity due to their unique sensory profiles and health benefits. The quality and safety of these products are governed by complex microbial ecosystems that orchestrate the biotransformation of tea leaf components. This review addresses a critical paradox in the field: the same microbial activities that generate desirable bioactive metabolites, such as theabrownins and organic acids, also create ecological niches for mycotoxigenic fungi, posing significant health risks from contaminants like ochratoxin A, citrinin, and aflatoxins. While extensive

Mycotoxins in meat products.

Mycotoxin presence in meat products is principally due to the development of toxigenic moulds during ripening, which concretely affect the dry-cured ones. Ochratoxin A (OTA) is the most prevalent mycotoxin in dry-cured hams and fermented sausages. Apart from collecting information on the main toxigenic moulds in dry-cured meat products, the objective of this review is to compile data about the mycotoxin prevalence and the tools for their mitigation. Although no very extensive studies have been performed on the mycotoxin occurrence in such products, much research has been conducted over the las

Nephrotoxicity and Renal Pathophysiology: A Contemporary Perspective.

The kidney consists of numerous cell types organized into the nephron, which is the basic functional unit of the kidney. Any stimuli that induce loss of these cells can induce kidney damage and renal failure. The cause of renal failure can be intrinsic or extrinsic. Extrinsic causes include cardiovascular disease, obesity, diabetes, sepsis, and lung and liver failure. Intrinsic causes include glomerular nephritis, polycystic kidney disease, renal fibrosis, tubular cell death, and stones. The kidney plays a prominent role in mediating the toxicity of numerous drugs, environmental pollutants and

Retrospective analysis of the effects of a highly standardized mixture of Berberis aristata, Silybum marianum, and monacolins K and KA in patients with dyslipidemia.

BACKGROUND: Berberis aristata, because of its berberine content, and Monascus purpureus fermented rice, because of the presence of monacolins (naturally derived statins), are widely investigated food-grade ingredients used to formulate cholesterol-lowering supplements. Although they are extensively used, berberine is poorly absorbed and monacolins are poorly chemically characterized, not standardized, and possibly contaminated with toxic compounds. Silymarin is reported to enhance berberine absorption, while Monakopure™-K20 (MK-20) is a highly standardized red yeast rice containing monacolins

5-hydroxy-1-methylhydantoin (NZ-419), an endogenous antioxidant, attenuates mycotoxin-induced oxidative stress in human kidney proximal tubular epithelial cells.

BACKGROUND: Exposure to foodborne mycotoxins such as ochratoxin A (OTA) and citrinin (CIT) causes severe oxidative stress-mediated nephropathy. This study investigated the protective potential of 5-hydroxy-1-methylhydantoin (NZ-419), an endogenous creatinine metabolite, against OTA- and CIT-induced toxicity in human proximal tubular epithelial (HK-2) cells. METHODS: We evaluated the radical-scavenging activity of NZ-419 using cell-free assays, including electron spin resonance. In HK-2 cells, its effects on mycotoxin-induced cytotoxicity, reactive oxygen species (ROS) generation, mitochondria

A nationwide questionnaire study evaluated kidney injury associated with Beni-koji tablets in Japan.

Red yeast rice, traditionally used in Asian cuisine and increasingly marketed as a dietary supplement for cholesterol management, has recently been linked to kidney dysfunction in Japan. In late 2023 to early 2024, multiple cases involving specific Beni-koji (red yeast rice) tablets from three different Beni-koji preparations, prompted a safety reevaluation. Although citrinin, a known nephrotoxin of red yeast rice, was not produced by the implicated strains, new safety concerns emerged. Here, we aimed to investigate the clinical, laboratory, and pathological features of affected patients with

Prevalence of mycotoxin contamination in different types of tea: a systematic review and meta-analysis.

The consumption of tea as a beverage is considerably increasing worldwide. However, some questions are raised about its safety, particularly regarding the potential contamination by mycotoxins. The objectives of this systematic review and meta-analysis were to assess the overall prevalence of mycotoxin contamination in different types of tea (black, green, dark, white and pu-erh), to compare the contamination rate between two periods (2000-2017 and 2018-2022), and to investigate the impact of country's income on this contamination. Included studies were selected from Scopus, Science Direct, We

Detection of Penicillium-Toxins in Nuts Commercialized in Italy Through LC-MS/MS Analyses.

The consumption of nuts is widespread globally and constitutes a significant component of the human diet due to its nutritional value. However, the presence of mycotoxins in food products, including nuts, is a global public health concern. Mycotoxins are toxic metabolites produced by contaminating fungi such as Aspergillus spp. and Penicillium spp., which can contaminate crops during growth, harvesting, storage, or transport. The aim of this study was to conduct monitoring for the presence of mycotoxins in nuts already on the market. Specifically, secondary metabolites produced by Penicillium

Occupational Exposure to Mycotoxins in Swine Production: Environmental and Biological Monitoring Approaches.

Swine production workers are exposed simultaneously to multiple contaminants. Occupational exposure to aflatoxin B₁ (AFB₁) in Portuguese swine production farms has already been reported. However, besides AFB₁, data regarding fungal contamination showed that exposure to other mycotoxins could be expected in this setting. The present study aimed to characterize the occupational exposure to multiple mycotoxins of swine production workers. To provide a broad view on the burden of contamination by mycotoxins and the workers' exposure, biological (urine) samples from workers (n = 25) and 38 environm

Fungi and mycotoxins during conventional and improved postharvest processing and storage of local rice in Nigeria.

This study applied a polyphasic characterization approach and LC-MS/MS analysis to elucidate fungal and mycotoxin contamination dynamics during conventional (CPRP) and improved (IPRP) postharvest processing and a 90-day storage of rice in Nigeria. Three hundred and twenty-five fungal isolates belonging to 21 genera (10 moulds, one crust fungus, 10 yeasts) and 54 species were recovered from rice across both processing chains. The CPRP chain had greater diversity of fungal genera compared to the IPRP chain. Aspergillus predominated both processing chains while yeasts occurred only in the CPRP ch

Genotoxicity of ochratoxin A and structurally related compounds in Escherichia coli strains: studies on their mode of action.

Ochratoxin A, ochratoxin alpha (its major metabolite in rodents) and seven structurally related substances were assayed for SOS DNA repair inducing activity in Escherichia coli PQ37 strain. At a concentration range of 0.1-4 mM, ochratoxin A, chloroxine, 5-chloro-8-quinolinol, 4-chloro-meta-cresol and chloroxylenol were found to induce SOS-DNA repair in the absence of an exogenous metabolic activation system. Ochratoxin B, ochratoxin alpha, 5-chlorosalicylic acid and citrinin were inactive, but all except ochratoxin alpha were cytotoxic. Thus, the presence of a chlorine at C-5 in ochratoxin A a

Nontargeted metabolomics analysis to unravel the anti-biofilm mechanism of Citrocin on Listeria monocytogenes.

Listeria monocytogenes biofilm formation is an important cause of cross-contamination in food processing. Citrinin is a potential broad-spectrum antimicrobial peptide. However, the effects of Citrocin on L. monocytogenes and its biofilm, as well as the associated mechanisms, remain to be explored. In this study, we evaluated the anti-biofilm effect of the antimicrobial peptide Citrocin on the foodborne pathogen L. monocytogenes and analyzed its anti-biofilm mechanism from the perspectives of swarming motility, extracellular polysaccharide production, and metabolite level changes. The results s

Elevated Urinary Levels of Fungal and Environmental Toxins in Patients with Pancreatic Ductal Adenocarcinoma.

BACKGROUND: Risk factors for pancreatic ductal adenocarcinoma (PDAC) include tobacco/alcohol abuse, genetic predisposition, insulin resistance, and pancreatic cysts. Despite these well-established risk factors and the screening of high-risk individuals, some people still develop PDAC. This study aims to explore a potential risk factor for PDAC by investigating the association between fungal toxins (FT) and environmental toxins (ET) and the disease. We predicted that individuals with PDAC would have higher levels of these toxins compared to healthy controls. The rationale behind this hypothesis
