Physiopathological Relevance of D-Serine in the Mammalian Cochlea.

NMDA receptors (NMDARs) populate the complex between inner hair cell (IHC) and spiral ganglion neurons (SGNs) in the developing and mature cochlea. However, in the mature cochlea, activation of NMDARs is thought to mainly occur under pathological conditions such as excitotoxicity. Ototoxic drugs such as aspirin enable cochlear arachidonic-acid-sensitive NMDAR responses, and induced chronic tinnitus was blocked by local application of NMDAR antagonists into the cochlear fluids. We largely ignore if other modulators are also engaged. In the brain, D-serine is the primary physiological co-agonist

Loss of RAD6B induces degeneration of the cochlea in mice.

Presbycusis is a form of age-related hearing loss (AHL). Many studies have shown that the degeneration of various structures in the cochlea of the inner ear is related to AHL, and DNA damage is an important factor leading to the above process. As an E2 ubiquitin-conjugated enzyme, RAD6B plays an important role in DNA damage repair (DDR) through histone ubiquitination. However, the molecular mechanism is still unclear. In this study, we investigated the role of RAD6B in the morphological changes and DDR mechanisms in aging-related degeneration of the cochlea of mice. We observed that the hair c

Circadian vulnerability of cisplatin-induced ototoxicity in the cochlea.

The chemotherapeutic agent cisplatin is renowned for its ototoxic effects. While hair cells in the cochlea are established targets of cisplatin, less is known regarding the afferent synapse, which is an essential component in the faithful temporal transmission of sound. The glutamate aspartate transporter (GLAST) shields the auditory synapse from excessive glutamate release, and its loss of function increases the vulnerability to noise, salicylate, and aminoglycosides. Until now, the involvement of GLAST in cisplatin-mediated ototoxicity remains unknown. Here, we test in mice lacking GLAST the

The cochlea in skull base surgery: an anatomy study.

OBJECTIVE The object of this study was to examine the relationships of the cochlea as a guide for avoiding both cochlear damage with loss of hearing in middle fossa approaches and injury to adjacent structures in approaches directed through the cochlea. METHODS Twenty adult cadaveric middle fossae were examined using magnifications of ×3 to ×40. RESULTS The cochlea sits below the floor of the middle fossa in the area between and below the labyrinthine segment of the facial nerve and greater petrosal nerve (GPN) and adjacent to the lateral genu of the petrous carotid. Approximately one-third of

Ototoxicity of acetic acid on the guinea pig cochlea.

BACKGROUND: To evaluate the ototoxicity of acetic acid solutions. METHODS: Compound action potentials (CAPs) of the eighth nerve were measured in guinea pigs before and after the application of acetic acid in the middle ear cavity. The pH values of the acetic acid solutions were pH 3.0, 4.0, and 5.0, and the application times were 30 min, 24 h, and 1 week. RESULTS: Acetic acid solution (pH 3.0, N = 3) for 30 min caused no significant elevation in CAP threshold at 4 kHz, but a significant elevation in the threshold was noted for 8 kHz and clicks. Acetic acid solutions (pH 4.0 N = 6, 5.0 N = 5

Ototoxicity of gentian violet on the guinea pig cochlea.

PURPOSE: Gentian violet (GV) is an antimicrobial and antifungal agent that has been used widely to treat intractable discharge in the ear. The purpose of this report is to warn clinicians about the ototoxic effect of GV in the middle ear. MATERIALS AND METHODS: GV ototoxicity was evaluated by measuring compound action potentials (CAPs) in the VIIIth nerve in adult Hartley guinea pigs. The middle ear cavities of the animals were filled with GV solution (0.5% or 0.13%), and CAPs were measured after intervals of 5 and 30 minutes and 1, 2, 6, and 24 hours. After all measurements were completed, t

[Protection of the cochlea by ascorbic acid in noise trauma].

BACKGROUND: The protective effect of ascorbic acid against noise-induced hearing loss and increased nitric oxide (NO) formation after noise exposure have already been demonstrated in animal models. However, the influence of ascorbic acid on noise-induced NO production within the cochlea is still unclear. METHODS: Guinea pigs (n=48) were fed for 7 days with low [25 mg/kg bodyweight (bw)/day] and high (525 mg/kg bw/day) doses of ascorbic acid. Then half of the animals were exposed to noise (90 dB for 1 h). The hearing levels were recorded beforehand, on the 3rd and 7th days after feeding, and d

Microsurgical access for cell injection into the mammalian cochlea.

The potential use of stem cells to repair hearing loss requires surgical access to the cochlea. Here we describe a microsurgical technique for cell injection into the mouse cochlea. Green fluorescent cells (ZsGreen-MCF10A cells) were successfully injected via a lateral wall cochleostomy into the scala media, scala tympani and scala vestibuli compartments of the cochlea. The effect of surgery on auditory function was investigated with auditory brainstem responses (ABR) to click and tone stimuli. A computerised signal-to-noise ratio detection method was developed to measure ABR thresholds in con

[The effect of noise exposure on the expression of NOSmRNA in the cochlea].

OBJECTIVE: To study the effect of NO in noise-induced deafness. METHOD: The effect of noise exposure on the expression of NOSmRNA in the guinea pig cochlea was observed with NADPH-diaphorase histochemistry staining, NOSmRNA hybridization in situ and Northern blotting analysis. RESULT: NADPH-diaphorase histochemistry indicated that NOS mainly presents in the inner hair cells, outer hair cells of Corti's organ, spiral ganglion cells and the marginal cells of stria vascularis in the guinea pig cochlea; NOSmRNA hybridization in situ showed that there is positive signal in the inner hair cells, o

Musical pitch perception with electrical stimulation of the cochlea.

Studies were undertaken to investigate the ability of a user of the Nucleus multi-electrode cochlear implant to judge pitch in the context of musical intervals. The subject had qualified as a musical instrument tuner before he received his implant, and was able to judge the intervals between electrical sensations with neither training nor the guidance of familiar melodies. The procedures used were interval estimation, and interval production by the method of adjustment. The pitch of the electrical stimulation was controlled by varying the pulse repetition rate, the active electrode position, o

Systematic approach to electrode insertion in the ossified cochlea.

Ossification of the fluid spaces of the cochlea occurs often in candidates for cochlear implantation, especially children. When noted before surgery on computerized tomography, ossification previously was thought to contraindicate cochlear implantation because of possible mechanical obstruction and uncertainty about the level of function that could be achieved by stimulating an ossified cochlea. However, during the preceding 6 years, techniques have been developed that permit implantation in ossified cochleas. We present a systematic approach that has been developed to treat the three clinical

The metabolic reaction of the cochlea to unphysiological noise exposure.

Guinea pigs were exposed to continuous white noise of 120 dB SPL. The performance included a recording of CM, pO2, and metabolites in the perfusate of the perilymphatic spaces. The metabolic reactions observed following the exposure are limited. No evidence of an overstimulation or of an insufficiency of the energy metabolism in the cochlea could be found. The results suggest the metabolic processes to be secondary to the sound-dependent damage of the organ of Corti after irreversible mechanical destructions in the hair cells.

Histopathological study of the cochlea with altered perilymph metabolism.

In 41 guinea pigs an experiment was conducted to determine the effect of kanamycin on ears with or without blockage of the cochlear aqueduct. In 24 animals the cochlear aqueduct was blocked on one side, and after 1 to 21 days, kanamycin (300 mg/kg) was given daily and subcutaneously until the pinna reflex began to decrease. The remaining animals were used for controls: exposure of and trauma to the aperture of the cochlear aqueduct and kanamycin administration (8), blockage of the cochlear aqueduct only (3 plus 17 previous cases), and kanamycin administration to normal animals (6). The surviva

The influence of a chronic vitamin A deficiency on the rat cochlea.

Rats were fed a vitamin-A-free diet and the cochleas of these animals were studied under the light and electron microscope. The cochlear function was tested by means of the electrocochleography. No pathologic changes could be found in the organ of Corti; the stria vascularis and the cochleas showed normal development. The recordings of the deficient rats are identical in configuration to controls, but they are shifted to higher intensities due to an otitis media found in the tested animals. These findings suggest that vitamin A does not have an important function in the inner ear.

The frequency selectivity of the normal and pathological human cochlea.

Human AP tuning curves (tone on tone simultaneous and forward masking curves) were measured during transtympanic electrocochleography. For subjects with near normal thresholds, average Q10 dB values (simultaneous masking) were 2.3 at 2 kHz, 3.6 at 4 kHz, and 4.7 at 8 kHz. Patients with threshold elevations of more than 40 dB, resulting from sensorineural hearing loss of cochlear origin, had tuning curves less sharply tuned by a factor of 2-3, with Q10 dB values of 1-2 at 2 kHz and at 4 kHz, and 1-2.3 at 8 kHz. AP tuning curves and single fibre tuning curves (frequency threshold curves) were me
