Avian influenza A viruses in birds of the order Psittaciformes: reports on virus isolations, transmission experiments and vaccinations and initial studies on innocuity and efficacy of oseltamivir in ovo.

Birds of the order Psittaciformes are - besides chickens, turkeys and other birds - also susceptible to infection with avian influenza A viruses (AIV) and succumb following severe disease within one week. Published data prove that various parakeets, amazons, cockatoos, African grey parrots and budgerigars (genera Barnardius, Psittacula, Cacatua, Eolophus, Amazona, Myiopsitta, Psittacus and Melopsittacus) were found dead following natural infections. Natural infections of highly pathogenic avian influenza viruses (HPAIV) of the haemagglutinin subtypes H5 and H7 cause severe disease and high rat

Disease surveillance in wild Victorian cacatuids reveals co-infection with multiple agents and detection of novel avian viruses.

Wild birds are known reservoirs of bacterial and viral pathogens, some of which have zoonotic potential. This poses a risk to both avian and human health, since spillover into domestic bird populations may occur. In Victoria, wild-caught cockatoos trapped under licence routinely enter commercial trade. The circovirus Beak and Feather Disease Virus (BFDV), herpesviruses, adenoviruses and Chlamydia psittaci have been identified as significant pathogens of parrots globally, with impacts on both aviculture and the conservation efforts of endangered species. In this study, we describe the results o

Viral vector vaccines protect cockatiels from inflammatory lesions after heterologous parrot bornavirus 2 challenge infection.

Avian bornaviruses are causative agents of proventricular dilatation disease (PDD), a chronic neurologic and often fatal disorder of psittacines including endangered species. To date no causative therapy or immunoprophylaxis is available. Our previous work has shown that viral vector vaccines can delay the course of homologous bornavirus challenge infections but failed to protect against PDD when persistent infection was not prevented. The goal of this study was to refine our avian bornavirus vaccination and infection model to better represent natural bornavirus infections in order to achieve

Cryptosporidium spp. parasitize exotic birds that are commercialized in markets, commercial aviaries, and pet shops.

The purpose of this study was to genetically characterize and phylogenetically analyze the Cryptosporidium spp. isolated from exotic birds commercialized in popular markets, commercial aviaries, and pet shops located in Rio de Janeiro, Brazil. Fecal samples from individually housed birds were collected and subjected to centrifuge-flotation technique using saturated sugar solution. DNA was isolated from Cryptosporidium positive samples, and 18S subunit rDNA was amplified and processed using nested-polymerase chain reaction (PCR). To identify the protozoan species, the PCR amplicons were used fo

Endocrine pancreatic insufficiency secondary to chronic herpesvirus pancreatitis in a cockatiel (Nymphicus hollandicus).

A cockatiel (Nymphicus hollandicus) examined because of weight loss, polydipsia, and polyuria was diagnosed with diabetes mellitus based on the presence of glucosuria and marked hyperglycemia. Medical attempts to manage the diabetes mellitus were unsuccessful, and the bird was euthanatized. Histopathologic examination of the pancreas revealed a chronic active pancreatitis with herpesviral inclusions in many of the pancreatic acinar and duct cells. Psittacid herpesvirus-1 (PsHV-1) DNA was amplified from the lesion by polymerase chain reaction. Sequencing of the amplicon showed it to be the geno

Barcoding of Giardia duodenalis isolates and derived lines from an established cryobank by a mutation scanning-based approach.

We barcoded 25 in vitro isolates (representing 92 samples) of Giardia duodenalis from humans and other animals, which have been assembled by the Upcroft team at the Queensland Institute of Medical Research over a period of almost three decades. We used mutation scanning-coupled sequencing of loci in the triosephosphate isomerase, glutamate dehydrogenase and β-giardin genes, combined with phylogenetic analysis, to genetically characterise them. Specifically, the isolates (n514) of G. duodenalis from humans from Australia (AD113; BRIS/83/HEPU/106; BRIS/87/HEPU/713; BRIS/89/HEPU/1003; BRIS/92/HEP

Development of an in Vivo Lipopolysaccharide Inflammation Model to Study the Pharmacodynamics of COX-2 Inhibitors Celecoxib, Mavacoxib, and Meloxicam in Cockatiels (Nymphicus hollandicus).

Nonsteroidal anti-inflammatory drugs (NSAIDs) are used frequently in avian medicine for their antipyretic, analgesic, and anti-inflammatory properties during surgery and for diseases that cause tissue damage and inflammation. NSAIDs inhibit cyclooxygenase (COX) enzymes, which are responsible for the induction of pyresis, pain, and inflammation. In our study, a lipopolysaccharide-induced (LPS) pyresis model was optimized using cockatiels (Nymphicus hollandicus) as subject birds (four males/three females) and validated in two females and one male, characterized by an intravenous bolus injection

Multilocus genotypic analysis of Cryptosporidium isolates from cockatiels, Japan.

Cryptosporidium is a significant pathogen in humans and animals. Cases of infection by Cryptosporidium parvum, Cryptosporidium meleagridis, and Cryptosporidium baileyi with zoonotic potential have also been reported in domestic birds, and recent studies indicate the presence of new host-adapted species or genotypes in birds. It is generally difficult to discriminate accurately among Cryptosporidium species and genotypes by light microscopy because of the morphological similarity of their oocysts. Although C. parvum and Cryptosporidium hominis are the primary Cryptosporidium species associated

Viral vector vaccines expressing nucleoprotein and phosphoprotein genes of avian bornaviruses ameliorate homologous challenge infections in cockatiels and common canaries.

Avian bornaviruses are causative agents of proventricular dilatation disease (PDD), an often fatal disease of parrots and related species (order Psittaciformes) which is widely distributed in captive psittacine populations and may affect endangered species. Here, we established a vaccination strategy employing two different well described viral vectors, namely recombinant Newcastle disease virus (NDV) and modified vaccinia virus Ankara (MVA) that were engineered to express the phosphoprotein and nucleoprotein genes of two avian bornaviruses, parrot bornavirus 4 (PaBV-4) and canary bornavirus 2
