Irradiation for Quarantine Control of Coffee Berry Borer, Hypothenemus hampei (Coleoptera: Curculionidae: Scolytinae) in Coffee and a Proposed Generic Dose for Snout Beetles (Coleoptera: Curculionoidea).

Coffee berry borer (CBB), Hypothenemus hampei (Ferrari) (Coleoptera: Curculionidae: Scolytinae), is the most serious insect pest of coffee worldwide. Green coffee used in blending and roasting is traded between countries and may be subjected to fumigation for disinfestation of CBB. For example, green coffee shipped to Hawaii from the U.S. mainland must be treated with methyl bromide. Irradiation is an alternative disinfestation treatment option. Dose-response tests were conducted with adult beetles to identify a sterilizing dose, followed by large-scale confirmatory tests with adults infesting

Characterization and bioactivities of coffee husks extract encapsulated with polyvinylpyrrolidone.

Coffee processing generates large amounts of residues of which a portion still has bioactive properties due to their richness in phenolic compounds. This study aimed to obtain a coffee husks extract (CHE) and to encapsulate it (ECHE) with polyvinylpyrrolidone using a one-step procedure of solid dispersion. The extraction and encapsulation yields were 9.1% and 92%, respectively. Thermal analyses revealed that the encapsulation increased the thermal stability of CHE and dynamic light scattering analyses showed a bimodal distribution of size with 81% of the ECHE particles measuring approximately

Coffee geographical origin rapid on-site discrimination strategy using an electrochemical fingerprinting platform by optimization of electrolyte concentration and temperature.

Coffee geographical origin discrimination is essential for quality control and market regulation. In this study, an electrochemical fingerprinting strategy was established by integrating electrochemical impedance spectroscopy (EIS), cyclic voltammetry (CV), and chronoamperometry (i-t). A copper-film electrode was selected based on EIS, scan-rate-dependent CV, and apparent double-layer capacitance analyses, while 0.01 mol/L NaOH and 35 °C were identified as the optimal operating conditions, effectively minimizing carbohydrate-derived electrochemical interference and enabling robust extraction o

Characterization of decolorized Chlorella pyrenoidosa protein foaming solution and its application in foam coffee.

Chlorella pyrenoidosa protein (CPP) possesses superior emulsification and foaming capabilities, enabling its solution to replace milk as a foaming agent in coffee products. However, its application, is constrained by the obvious pigmentation and suboptimal foam stability. To achieve this goal, ethanol decolorization and saponin (Tea saponin (Ts), Gypenoside (Gyp)) complexation were employed to prepare decolorized Chlorella pyrenoidosa protein (DCPP) foaming solution. The interfacial properties, rheological properties, and foam properties were characterized. Initially, compared with the CPP sol

Fully Automated Identification of Coffee Species and Simultaneous Quantification of Furfuryl Alcohol Using NMR Spectroscopy.

BACKGROUND: Coffee is a popular beverage with two species, Coffea canephora and C. arabica, being commercially exploited. The quality and commercial value of coffee is dependent on species and processing. C. arabica typically obtains a higher price on the market compared to C. canephora. Coffee beans undergo roasting during processing, resulting in the formation of flavor compounds including furfuryl alcohol which has been classified by the International Agency for Research on Cancer as possibly carcinogenic to humans (Group 2B). OBJECTIVE: The aim of this study was to identify coffee species

The role of dissolved cations in coffee extraction.

The flavorsome compounds in coffee beans exist in the form of aprotic charge neutral species, as well as a collection of acids and conjugate salts. The dissolution and extraction of these organic molecules is a process dependent on the dissolved mineral content of the water. It is known that different rates and compositions of coffee extraction are achieved through the control of the water "impurities", Na(+), Mg(2+), and Ca(2+), which coordinate to nucleophilic motifs in coffee. Using density functional theory, we quantify the thermodynamic binding energies of five familiar coffee-contained a

The complete and annotated mitochondrial genome of Hemileia vastatrix Race I, causal agent of coffee leaf rust.

Hemileia vastatrix is the fungal pathogen responsible for coffee leaf rust (CLR), the most economically important disease of Coffea arabica worldwide. Recently, the nuclear genome of this fungus was completely deciphered. However, the mitochondrial genome of H. vastatrix has remained undercharacterized. Here, we present the complete, circularized mitochondrial genome of H. vastatrix Race I (isolate HvRI), assembled using a hybrid approach combining PacBio HiFi long reads and BGIseq short reads. The genome is 173,525 bp in length with a GC content of 33.1% and encodes 41 functional genes, inclu

Differential accumulation of bioactive metabolites in coffee cell cultures in comparison to green coffee beans.

Conventional coffee production faces sustainability challenges. Coffee cell cultures (CC) are a promising alternative coffee raw material. This study investigated the composition of CC in comparison to conventional green coffee beans (GB) using untargeted metabolomics and lipidomics approaches. The lipid content (g/100 g) of GB was approximately four times higher than that of CC with triglycerides as the predominant lipid class identified followed by diterpenes. Additionally, differences were observed in the diterpene profiles of CC and GB. CC accumulated more thiamine and choline but lower co

Assessment of coffee endophytic yeasts for efficient mucilage removal and consistent quality improvement in coffee production.

Endophytic yeasts are commonly isolated from coffee plants, yet their capacity for mucilage removal remains underexplored. In this study, 42 pectinolytic yeast strains were isolated from coffee cherries and classified into five species. Among them, Pichia kluyveri YE7 showed superior mucilage removal and cold tolerance, achieving complete mucilage degradation within 4-12 h at 5-30 °C. Genomic analysis identified multiple genes encoding pectinases, cellulases, and hemicellulases, consistent with its enzymatic capacity. Fermentation with P. kluyveri YE7 primarily altered alkane levels in green c

Spectrophotometry and chromatography analyses combined with chemometrics tools to differentiate green coffee beans into special or traditional.

Green coffee is the hulled coffee bean, rich in chemical compounds indicative of quality before roasting, making the classification special or traditional. This work aimed to determine compounds in green coffee beans and find the differentiation of green coffee beans into special or traditional ones through chemometrics. For that, the levels of phenolic compounds, reducing, nonreducing, and total sugars were quantified by spectrophotometry: caffeine, trigonelline, 5-hydroxymethylfurfural (5-HMF), 3-hydroxybenzoic, 4-hydroxybenzoic, chlorogenic, caffeic, and nicotinic acids (NAs) by high-perfor

Newly isolated phages preying on Pseudomonas syringae pv. garcae: In vitro and ex vivo inactivation studies in coffee plant leafs.

Coffee canker, or bacterial halo blight (BHB) of coffee, is a disease caused by the phytopathogenic bacterium Pseudomonas syringae pv. garcae (Psg), having been found for the first time in 1955, in the Garça region (State of São Paulo), and which has stood out in the Brazilian coffee plantations in recent years, leading to severe economic losses that seriously affect coffee trade. The treatments available are still scarce, involving frequent spraying of coffee plantations with either copper derivatives or the antibiotic kasugamycin. However, these compounds should be avoided due to environment

Characterization of a novel GH5 β-mannanase from Hahella sp. CR1 and its synergistic role in degradation of spent coffee grounds.

An efficient enzymatic degradation of plant biomass is essential for agricultural waste valorization and biofuel production. β-Mannanase is a key lignocellulolytic enzyme that targets mannan, a major component in plant cell walls. Hahella spp. are known to produce cellulolytic enzymes, but no β-mannanase was characterized from this genus up to date. In this study, a gene encoding for β-mannanase (HcrMan1) was identified and isolated from the genome of Hahella sp. CR1. HcrMan1 encodes a multidomain enzyme comprising a GH5 subfamily 8 catalytic domain, an immunoglobulin-like (Ig) linker domain,

An aggressive nonconsumptive effect mediates pest control and multipredator interactions in a coffee agroecosystem.

Natural pest control is an alternative to pesticide use in agriculture, and may help to curb insect declines and promote crop production. Nonconsumptive interactions in natural pest control that historically have received far less attention than consumptive interactions, may have distinct impacts on pest damage suppression and may also mediate positive multipredator interactions. Additionally, when nonconsumptive effects are driven by natural enemy aggression, variation in alternative resources for enemies may impact the strength of pest control. Here we study control of the coffee berry borer

Coffee and its consumption: benefits and risks.

Coffee is the leading worldwide beverage after water and its trade exceeds US $10 billion worldwide. Controversies regarding its benefits and risks still exist as reliable evidence is becoming available supporting its health promoting potential; however, some researchers have argued about the association of coffee consumption with cardiovascular complications and cancer insurgence. The health-promoting properties of coffee are often attributed to its rich phytochemistry, including caffeine, chlorogenic acid, caffeic acid, hydroxyhydroquinone (HHQ), etc. Many research investigations, epidemiolo

Caffeine and related purine alkaloids: biosynthesis, catabolism, function and genetic engineering.

Details of the recently elucidated biosynthetic pathways of caffeine and related purine alkaloids are reviewed. The main caffeine biosynthetic pathway is a sequence consisting of xanthosine-->7-methylxanthosine-->7-methylxanthine-->theobromine-->caffeine. Genes encoding N-methyltransferases involved in three of these four reactions have been isolated and the molecular structure of N-methyltransferases investigated. Pathways for the catabolism of caffeine have also been studied, although there are currently no reports of enzymatic and genetic studies having been successfully carried out. Metabo

Volatile composition of coffee berries at different stages of ripeness and their possible attraction to the coffee berry borer Hypothenemus hampei (Coleoptera: Curculionidae).

The analysis of volatile emissions of coffee berries in different physiological states of ripeness was performed using dynamic headspace and gas chromatography/mass spectrometry analysis for Coffea arabica, var. Colombia. The composition of the volatiles emitted by coffee berries is dominated by very high levels of alcohols, mainly ethanol, in all stages of ripeness in comparison with other compounds. Overripe coffee berries have high volatile emissions and show a composition dominated mainly by esters followed by alcohols, ketones, and aldehydes. The lowest level compounds were monoterpenes.
