Nitrous oxide consistently attenuates thermogenic and thermoperceptual responses to repetitive cold stress in humans.

Divers are at enhanced risk of hypothermia, due to the independent action of the inspired inert gases on thermoregulation. Thus, narcosis induced by acute (≤2 h) exposure to either hyperbaric nitrogen or normobaric nitrous oxide (N2O) impairs shivering thermogenesis and accelerates body core cooling. Animal-based studies, however, have indicated that repeated and sustained N2O administration may prevent N2O-evoked hypometabolism. We, therefore, examined the effects of prolonged intermittent exposure to 30% N2O on human thermoeffector plasticity in response to moderate cold. Fourteen men partic

Using thermal stress to model aspects of disease states.

Exposure to acute heat or cold stress elicits numerous physiological responses aimed at maintaining body temperatures. Interestingly, many of the physiological responses, mediated by the cardiovascular and autonomic nervous systems, resemble aspects of, or responses to, certain disease states. The purpose of this Perspective is to highlight some of these areas in order to explore how they may help us better understand the pathophysiology underlying aspects of certain disease states. The benefits of using this human thermal stress approach are that (1) no adjustments for inherent comparative di

Heat-Shock and Cold-Shock Proteins in Energy-Based Aesthetic Medicine.

BACKGROUND: Energy-based aesthetic devices alter tissue temperature across defined spatial and temporal distributions, producing biological effects commonly described as collagen contraction, neocollagenesis, elastogenesis, adipocyte injury, or tissue tightening. These clinical labels encompass distinct processes including protein damage, cellular stress adaptation, inflammation, cell death, extracellular matrix turnover, and long-term remodeling. Heat-shock proteins (HSPs) and mammalian cold-shock proteins provide a mechanistic framework for interpreting these responses. OBJECTIVE: To evalua

Transcriptomic profiling reveals immunometabolic gene regulation during a 24-h cold exposure survival simulation in humans.

Prolonged cold exposure imposes substantial physiological and operational demands; however, the molecular mechanisms supporting human adaptation to sustained cold remain poorly defined. In this first-in-human study, we conducted genome-wide transcriptomic profiling of peripheral blood leukocytes from eight healthy adult males during a 24-h whole body cold survival simulation (7.5°C). Blood samples were collected at four standardized time points to characterize the dynamics of gene expression during continuous cold stress. Despite the clear physiological strain, the global leukocyte transcripto

Transcriptional Response of Rice Phytocystatin Family Genes to Cold Stress.

Background/Objectives: Phytocystatins are plant-specific inhibitors of papain-like cysteine proteinases that participate in plant development and responses to biotic and abiotic stresses. The roles of rice (Oryza sativa L.) phytocystatins under cold stress remain poorly understood. Xiaomagu (XMG), a cold-tolerant japonica rice landrace, provides valuable material for exploring phytocystatin transcript responses to low-temperature stress. This study characterizes 12 rice phytocystatin genes isolated from XMG and investigates their transcript profiles under cold and abscisic acid (ABA) treatment

Nitric oxide cross-links calcium signals to enhance cold tolerance via inhibiting calmodulin expression in watermelon.

Nitric oxide (NO) is a pivotal gaseous signaling molecule that plays a critical role in regulating plant tolerance to cold stress; however, the underlying mechanisms of signal transduction remain poorly elucidated. In this study, knockout of nitrate reductase 1 (ClNR1), a crucial gene for NO biosynthesis, led to reduced cold tolerance in watermelon (Citrullus lanatus), accompanied by downregulation of cycle nucleotide-gated channel (ClCNGC) 20, a key Ca2+-permeable channel gene, decreased Ca2+ influx, and upregulation of calmodulin (ClCaM) 2/5/7. Conversely, application of the NO donor sodium

Hydrogen sulfide in plant cold stress: functions, mechanisms, and challenge.

Cold stress is an environmental factor that seriously restricts the growth, production and survival of plants, and has received extensive attention in recent years. Hydrogen sulfide (H2S) is an ubiquitous gas signaling molecule, and its role in alleviating plant cold stress has become a research focus in recent years. This paper reviews for the first time the significant effect of H2S on improving plant cold resistance, which makes up for the gaps in the existing literature. In general, H2S improves plant tolerance to cold stress by activating antioxidant reaction and promoting the accumulatio

Cold stress, heart failure, and esophageal occlusion cause the death of a West Indian manatee Trichechus manatus in Alabama, USA.

Cold stress is the leading cause of mortality in West Indian manatees Trichechus manatus in the northcentral Gulf of Mexico. This report describes an adult male manatee that stranded alive with signs of cold stress in Alabama (USA) waters in January 2022 and died during the rescue intervention. Postmortem examination and histopathologic review revealed multiple contributors to death. While the animal had evidence of cold stress syndrome, there was aortic stenosis and concentric left ventricular hypertrophy as well as an esophageal foreign body (plastic bag). Main findings from blood analysis i

Characterization of a Rabbit Model of Traumatic Hemorrhagic Shock Under High-Altitude Cold Conditions.

High-altitude hypoxia and cold often co-occur in traumatic hemorrhagic shock (THS), yet time-resolved models that quantify their interaction with blood loss are scarce. We established a composite-trauma THS model in New Zealand rabbits at ≈ 5,000 m equivalent altitude and 8 ± 2°C. Animals were randomized to six groups [HS (hemorrhagic shock); HS + COL (hemorrhagic shock + cold); HS+HYP (hemorrhagic shock + hypoxia); HS+HYP+COL (hemorrhagic shock + hypoxia + cold); Sham (normoxia, normothermia, no HS); HYP+COL (hypoxia + cold, no HS)]. Trauma comprised partial hepatectomy, total splenectomy, an

Ammonia borane positively regulates cold tolerance in Brassica napus via hydrogen sulfide signaling.

BACKGROUND: Cold stress adversely influences rapeseeds (Brassica napus L.) growth and yield during winter and spring seasons. Hydrogen (H2) is a potential gasotransmitter that is used to enhance tolerance against abiotic stress, including cold stress. However, convenience and stability are two crucial limiting factors upon the application of H2 in field agriculture. To explore the application of H2 in field, here we evaluated the role of ammonia borane (AB), a new candidate for a H2 donor produced by industrial chemical production, in plant cold tolerance. RESULTS: The application with AB cou

Effects of sinoaortic denervation on hemodynamic perturbations of prolonged paradoxical sleep deprivation and rapid cold stress in rats.

BACKGROUND: Sleep disturbances and aversive cold stress (CS) are cardiovascular risk factors. This study investigates how homeostatic control autonomic baroreflex influences the hemodynamic perturbations evoked by paradoxical sleep deprivation (PSD) and CS. METHODS: Conscious adult male rats were randomly divided into four groups (Sham/CON [control], Sham/PSD, sinoaortic denervation [SAD]/CON, and SAD/PSD). Spectral analysis and SAD were employed to evaluate the effects of a 72-hr PSD with 10-min CS on blood pressure variability and heart rate variability (BPV and HRV) at total power (TP) and

Cold-Induced Thermogenesis and Inflammation-Associated Cold-Seeking Behavior Are Represented by Different Dorsomedial Hypothalamic Sites: A Three-Dimensional Functional Topography Study in Conscious Rats.

In the past, we showed that large electrolytic lesions of the dorsomedial hypothalamus (DMH) promoted hypothermia in cold-exposed restrained rats, but attenuated hypothermia in rats challenged with a high dose of bacterial lipopolysaccharide (LPS) in a thermogradient apparatus. The goal of this study was to identify the thermoeffector mechanisms and DMH representation of the two phenomena and thus to understand how the same lesion could produce two opposite effects on body temperature. We found that the permissive effect of large electrolytic DMH lesions on cold-induced hypothermia was due to

Responses to sudden cold-water immersion in inexperienced swimmers following training.

BACKGROUND: When suddenly immersed in cold water, humans typically exhibit the cold shock response, although training can attenuate hyperventilation. This study extends previous findings by considering the influence of physical activity to maintain buoyancy and subsequent swimming performance. METHODS: Six inexperienced swimmers (three men and three women; mean age 22.8) received 1 wk of cold-water head-out immersions (10 x 3 min at 15 degrees C) alongside mental skills training to improve their treading water technique and to control hyperventilation upon immersion. Six inexperienced control

The brassinosteroid-responsive transcription factor CmBEEL1 through integrating ICE1 cascade and ROS homeostasis enhances cold tolerance in Chrysanthemum.

Cold stress ranks among the notable abiotic stresses that significantly hinder plant growth and geographical distribution. Brassinosteroids (BRs) have been proven to enhance the cold tolerance of plants across multiple species. However, the involvement of BR and its regulatory mechanism underlying cold tolerance in chrysanthemum has not yet been fully elucidated. In this study, we found that BR treatment alleviated cold stress-induced growth inhibition in chrysanthemum. Based on the transcriptome database of Chrysanthemum in response to cold stress, CmBEEL1, a member of the bHLH family genes,

A self-amplifying NO-H2S loop mediates melatonin-induced CBF-responsive pathway and cold tolerance in watermelon.

Melatonin is a pivotal bioactive molecule that enhances plant cold stress tolerance, but the precise mechanisms remain enigmatic. Here, we have discovered that overexpressing melatonin biosynthetic gene ClCOMT1 or applying exogenous melatonin activates the C-repeat binding factor (CBF)-responsive pathway and enhances watermelon cold tolerance. This enhancement is accompanied by elevated levels of nitric oxide (NO) and hydrogen sulfide (H2S), along with upregulation of nitrate reductase 1 (ClNR1) and L-cysteine desulfhydrase (ClLCD) genes involved in NO and H2S generation respectively.
