Mutations in SAMD7 cause autosomal-recessive macular dystrophy with or without cone dysfunction.

Sterile alpha motif domain containing 7 (SAMD7) is a component of the Polycomb repressive complex 1, which inhibits transcription of many genes, including those activated by the transcription factor Cone-Rod Homeobox (CRX). Here we report bi-allelic mutations in SAMD7 as a cause of autosomal-recessive macular dystrophy with or without cone dysfunction. Four of these mutations affect splicing, while another mutation is a missense variant that alters the repressive effect of SAMD7 on CRX-dependent promoter activity, as shown by in vitro assays. Immunostaining of human retinal sections revealed t

Rod-cone dystrophy associated with a rod system electroretinogram obtained under photopic conditions.

A 14-year-old girl with a history of night blindness and poor visual acuity was examined. Cystic-appearing lesions were apparent in the fovea, and pigmentary changes were observed in the midperipheral retina. Electroretinograms (ERGs) recorded from the dark- and light-adapted eye were similar in waveform, implicit time, and approximate amplitude. Results of examination of the spectral characteristics of the light-adapted ERG demonstrated that the recordings represented activity of the rod system, despite the presence of a background field, which eliminates the response of the rod system in nor

Mutation in the auxiliary calcium-channel subunit CACNA2D4 causes autosomal recessive cone dystrophy.

Retinal signal transmission depends on the activity of high voltage-gated l-type calcium channels in photoreceptor ribbon synapses. We recently identified a truncating frameshift mutation in the Cacna2d4 gene in a spontaneous mouse mutant with profound loss of retinal signaling and an abnormal morphology of ribbon synapses in rods and cones. The Cacna2d4 gene encodes an l-type calcium-channel auxiliary subunit of the alpha (2) delta type. Mutations in its human orthologue, CACNA2D4, were not yet known to be associated with a disease. We performed mutation analyses of 34 patients who received a

Correcting photoreceptor diseases at their source: CRISPR strategies for cone-rod dystrophy and achromatopsia.

BACKGROUND: Cone-rod dystrophy (CORD) and achromatopsia (ACHM) are inherited retinal dystrophies for which conventional adeno-associated virus (AAV) gene augmentation has important limitations, particularly in autosomal-dominant gain-of-function CORD and recessive ACHM. Clustered Regularly Interspaced Short Palindromic Repeats (CRISPR)/CRISPR-associated protein 9 (Cas9) offers the potential for one-time, mutation-specific gene correction or allele ablation. This systematic review summarizes preclinical evidence on CRISPR/Cas9-based approaches for CORD and ACHM, focusing on editing efficiency,

A novel nonsense variant in REEP6 is involved in a sporadic rod-cone dystrophy case.

Rod-cone dystrophy (RCD), also called retinitis pigmentosa, is the most common form of progressive inherited retinal disorders secondary to photoreceptor degeneration. It is a genetically heterogeneous disease characterized by night blindness, followed by visual field constriction and, in most severe cases, total blindness. The aim of our study was to identify the underlying gene defect leading to severe RCD in a 60-year-old woman. The patient's DNA was investigated by targeted next generation sequencing followed by whole exome sequencing. A novel nonsense variant, c.267G>A p.(Trp89*), was ide

X linked cone-rod dystrophy, CORDX3, is caused by a mutation in the CACNA1F gene.

BACKGROUND: X linked cone-rod dystrophy (CORDX) is a recessive retinal disease characterised by progressive dysfunction of photoreceptors. It is genetically heterogeneous, showing linkage to three X chromosomal loci. CORDX1 is caused by mutations in the RPGR gene (Xp21.1), CORDX2 is located on Xq27.2-28, and we recently localised CORDX3 to Xp11.4-q13.1. We aimed to identify the causative gene behind the CORDX3 phenotype. METHODS: All 48 exons of the CACNA1F gene were screened for mutations by DNA sequencing. RNA from cultured lymphoblasts and peripheral blood activated T lymphocytes was analy

Bilateral Giant Choroidal Cavern in a Case of Rod-Cone Dystrophy: A Rare Presentation.

This report presents a case of a 22-year-old woman with bilateral giant choroidal caverns (CCs) in rod-cone dystrophy. Her best-corrected visual acuity was 20/80 in the right eye and 20/50 in the left eye. Fundus examination showed optic nerve pallor, narrowed blood vessels, and diffuse pigmentary changes. Fundus autofluorescence showed a central area of reduced autofluorescence surrounded by stippled autofluorescence. An electroretinogram showed decreased amplitude of both scotopic and photopic waves, consistent with a diagnosis of rod-cone dystrophy (retinitis pigmentosa). Optical coherence
