Three kinds of corneal host cells contribute differently to corneal neovascularization.

BACKGROUND: Corneal neovascularization (angiogenesis and lymphangiogenesis) compromises corneal transparency and transplant survival, however, the molecular mechanisms of corneal host epithelial and stromal cells in neovascularization have not yet been fully elucidated. Furthermore, the contribution and mechanism of corneal host endothelial cells involved in neovascularization are largely unexplored. METHODS: Liquid chromatography-mass spectrometry, immunoblotting, and ELISA were used to screen and identify potential neovascularization-related factors in human full-thickness vascularized corn

Streptozotocin‑induced diabetic mice exhibit reduced experimental choroidal neovascularization but not corneal neovascularization.

The present study aimed to investigate the effects of diabetes mellitus (DM) on the generation of experimental corneal neovascularization (CrNV) and choroidal neovascularization (ChNV). Diabetes was induced in mice by intraperitoneal injection of streptozotocin (STZ). Experimental CrNV and ChNV were induced by alkali injury and laser photocoagulation, respectively. CrNV and ChNV were compared between the STZ‑induced diabetic mice and control mice two weeks after injury. Relative expression of angiogenic factors was quantified by reverse transcription‑quantitative polymerase chain reaction, and

The effects of leukocyte depletion on corneal neovascularization.

The role of leukocytic infiltration in the initiation and maintenance of corneal neovascularization was studied in rats. Vascular ingrowth was induced by central cauterization of the cornea with silver nitrate and assessed after intraarterial perfusion of colloidal carbon. In normal rats, the mean vascular length was 0.33 mm. at 3 days and 0.63 mm. at 4 days. Whole-body x-irradiation with 800 rads depressed peripheral blood leukocyte counts to 1% of normal, prevented infiltration of monocytes, and reduced infiltration of neutrophils, but did not alter the neovascular response at 4 days. The role of Nod1 signaling in corneal neovascularization.

PURPOSE: Corneal neovascularization (CNV) is associated with Chlamydia trachomatis. The minimal components of bacterial cell walls are recognized by nucleotide-binding oligomerization domain-containing protein (Nod), which is important for host defense--a mechanism manifested in human corneal cells. We aimed to examine whether Nod stimulation is associated with CNV. METHODS: Three groups of mice with alkali-induced CNV were topically treated with tripeptide L-Ala-γ-D-Glu-meso-diaminopimelic acid (Tri-DAP, a Nod1 agonist), muramyl dipeptide (a Nod2 agonist), or phosphate-buffered saline twice

[An investigation on morphology of experimental corneal neovascularization].

OBJECTIVE: To observe the morphologic character of the corneal neovascularization in experimental rabbits and also compare it with the morphology of that after irradiation with beta-rays. METHODS: A model of new blood vessels on the corneas of New Zealand rabbits was established with silver nitrate burning. The growth character of the neovascularization was compared between the irradiated and non-irradiated groups. The casting of the new blood vessels in different periods was made and the morphologic differences between the two groups were compared under a scanning electron microscope. RESUL

Restoration of corneal epithelial barrier function: A possible target for corneal neovascularization.

Corneal neovascularization (CoNV) is the second leading common cause of vision impairment worldwide and is a blinding pathological alteration brought on by ocular trauma, infection, and other factors. There are some limitations in the treatment of CoNV, hence it's critical to look into novel therapeutic targets. The corneal epithelial barrier, which is the initial barrier of the ocular surface, is an important structure that shields the eye from changes in the internal environment or invasion by the external environment. This study sought to collate evidence on the regulation of corneal epithe

[Management of corneal neovascularization prior to corneal transplantation: Report of 112 cases].

INTRODUCTION: The avascular nature of the cornea results from a balance between angiogenic factors and anti-angiogenic factors. Under pathological conditions, this homeostasis can be disturbed, resulting in the onset of corneal neovascularization. The purpose of our study was to report our experience in the management of corneal neovascularization prior to keratoplasty. MATERIALS AND METHODS: This is a prospective study of 112 patients with corneal neovascularization and candidates for possible corneal transplant. RESULTS: The average age of patients was 38 years, ranging from 15 to 72 years

Genome Editing VEGFA Prevents Corneal Neovascularization In Vivo.

Corneal neovascularization (CNV) is a common clinical finding seen in a range of eye diseases. Current therapeutic approaches to treat corneal angiogenesis, in which vascular endothelial growth factor (VEGF) A plays a central role, can cause a variety of adverse side effects. The technology of Clustered Regularly Interspaced Short Palindromic Repeats (CRISPR)/Cas9 can edit VEGFA gene to suppress its expression. CRISPR offers a novel opportunity to treat CNV. This study shows that depletion of VEGFA with a novel CRISPR/Cas9 system inhibits proliferation, migration, and tube formation of human u
