Biochemical aspects of Parkinson's disease.

The importance of the striatal dopamine (DA) deficiency and the DA substituting property of levodopa for the pathophysiology and therapy of Parkinson's disease (PD) is reiterated. In addition, it is shown that in PD, significantly reduced DA levels are also found in the nucleus accumbens, external and internal segments of the globus pallidus, the substantia nigra reticulata, and the subthalamic nucleus. It is proposed that, in addition to the critical role played by the striatal DA loss, the DA changes in the extrastriatal nuclei of the basal ganglia are importantly involved in the pathophysio

[Bilateral asymmetric degeneration of the striatum as a cause of senile hemiballism].

We report the case of an 82-years old woman with sporadic acute hemiballism associated with bilateral striatal degeneration of non vascular origin which was most severe in the side contralateral to the hemiballism. The patient's neuropathological lesions resembled those of patients with senile chorea. Although hemiballism has never been described as an isolated manifestation of primary degeneration of the striatum, it may so occur in elderly patients with hemiballism.

Reliable criteria for the prenatal sonographic diagnosis of alobar holoprosencephaly.

A series of 10 consecutive cases of alobar holoprosencephaly is described. The disorder was diagnosed prenatally by ultrasound according to two criteria: a large central fluid collection in the fetal head, with no visible midline structures but with the presence of a mantle around the fluid collection and fusion of the thalami and corpus striatum, and sonographic abnormalities of the face, including hypotelorism, central clefts, facial asymmetry, and abnormal orbits.

Keratan sulfate proteoglycan: putative template for neuroblast migratory and axonal fascicular pathways and fetal expression in globus pallidus, thalamus, and olfactory bulb.

Keratan sulfate (KS) is a proteoglycan secreted in the fetal brain astrocytes and radial glia into extracellular parenchyma as granulofilamentous deposits. KS surrounds neurons except dendritic spines, repelling glutamatergic and facilitating GABAergic axons. The same genes are expressed in both neuroblast migration and axonal growth. This study examines timing of KS during morphogenesis of some normally developing human fetal forebrain structures. Twenty normal human fetal brains from 9-41 weeks gestational age were studied at autopsy. KS was examined by immunoreactivity in formalin-fixed par

Multiple Mechanisms of Dopamine Modulation of Neuronal Excitability and Neurotransmission in the Striatum: A Personal and Historical Perspective.

3,4-dihydroxyphenethylamine, commonly known as dopamine (DA), is a neuromodulator that fine-tunes neuronal excitability, neurotransmitter release, and the effects of other neurotransmitters on postsynaptic neurons. DA, acting on D1 and D2 receptor families, is involved in myriad functions. In the striatum, it is mainly implicated in motor control, motivation, and reward mechanisms. In the cerebral cortex it participates in attention processes, working memory, long-term memory, etc. DA overproduction or deficits lead to neuronal circuit imbalance that underlies a number of neurological and psyc

Enhanced habit formation in Tourette patients explained by shortcut modulation in a hierarchical cortico-basal ganglia model.

Devaluation protocols reveal that Tourette patients show an increased propensity to habitual behaviors as they continue to respond to devalued outcomes in a cognitive stimulus-response-outcome association task. We use a neuro-computational model of hierarchically organized cortico-basal ganglia-thalamo-cortical loops to shed more light on habit formation and its alteration in Tourette patients. In our model, habitual behavior emerges from cortico-thalamic shortcut connections, where enhanced habit formation can be linked to faster plasticity in the shortcut or to a stronger feedback from the s

Striatum and language processing: Where do we stand?

More than a century ago, Broca (1861), Wernicke (1874) and Lichteim (1885) laid the foundations for the first anatomo-functional model of language, secondarily enriched by Geschwind (1967), leading to the Broca-Wernicke-Lichteim-Geschwind model. This model included the frontal, parietal, and temporal cortices as well as a subcortical structure, which could be the striatum, whose nature and role have remained unclear. Although the emergence of language deficits in patients with striatal injury has challenged the cortical language models developed over the past 30 years, the integration of the s

Emergence of novel functions in striatal low-threshold spike interneurons (Commentary on Gazan et al., 2019).

LTSIs selectively target distal dendrites of spiny projection neurons (SPNs, top). In this article by Gazan et al., the authors performed a selective ablation of LTSIs using Cre-dependent diphtheria toxin in SST-Cre mice, injected in the striatum. They demonstrate that the ablation of LTSIs (bottom) induced a decrease in the number of distal dendritic spines on SPNs associated with an increased intrinsic excitability of these cells. Behaviorally, this is associated with a potentiation of cocaine effect on locomotion. Whether these behavioral effects are the direct result of the loss of LTSIs o

Striatal cholinergic dysfunction as a unifying theme in the pathophysiology of dystonia.

Dystonia is a movement disorder of both genetic and non-genetic causes, which typically results in twisted posturing due to abnormal muscle contraction. Evidence from dystonia patients and animal models of dystonia indicate a crucial role for the striatal cholinergic system in the pathophysiology of dystonia. In this review, we focus on striatal circuitry and the centrality of the acetylcholine system in the function of the basal ganglia in the control of voluntary movement and ultimately clinical manifestation of movement disorders. We consider the impact of cholinergic interneurons (ChIs) on

[Micro-anatomy of collateral perforating branches of the middle cerebral artery].

The lenticulostriate arteries are the subject of a microanatomical study on 50 formalin fixed human brains. A single trunk arising from the middle cerebral artery is rarely found. In fact the branches are numerous and they are found to ramify before penetrating the anterior perforated substance. The length of the sphenoidal segment is variable and this aspect is discussed in the light of the literature data. The origin of the perforating branches is spread from the internal carotid artery to the bifurcation branches of the middle cerebral artery (M2 segment). On the other hand lenticulostriate

Electron microscopic morphometric studies on synaptic vesicles of long-term CPZ-administered rat striatum.

The effect of chronic administration of chlorpromazine (CPZ) on the striatal synaptic vesicles was quantitatively investigated with an electron microscope. Six rats, three controls and three experimentals, which received 20 mg/kg b.wt. of CPZ daily for 15 months, were sacrificed and a total of 300 axo-dendritic spine synapses (50 synapses per animal) were randomly taken and divided into three zones with a width of 160-467 nm on electron micrographs (final magnification 60,000). The first zone was adjacent to the presynaptic membrane in the region of the synaptic cleft. The second and third wer

Synaptosomal glutamate uptake declines progressively in the spinal cord of a mutant mouse with motor neuron disease.

It has been suggested that the degeneration of motor neurons in amyotrophic lateral sclerosis is a consequence of excitotoxicity resulting from a loss of synaptosomal glutamate uptake. The role of synaptosomal glutamate uptake in the pathogenesis of motor neuron disease was studied in the Mnd mouse. Glutamate uptake in spinal-cord synaptosomes declined in parallel with the onset of behavioral deficits in Mnd mice but lagged considerably behind the appearance of pathology in motor neurons. Glutamate uptake did not decline significantly in corpus striatum, and GABA uptake did not change signific

Effect of MDMA exposure during pregnancy on cell apoptosis, astroglia, and microglia activity in rat offspring striatum.

OBJECTIVES: Ecstasy is a popular recreational psychostimulant with side effects on the central nervous system. This study examined the corpus striatum tissue of adult rats that received ecstasy during the embryonic period for histological and molecular studies. MATERIALS AND METHODS: Rats were divided into control and ecstasy groups. The ecstasy group was given MDMA 15 mg/kg intraperitoneally twice daily at 8-hour intervals on days 7-15 of gestation. At the age of 15 weeks, adult offspring of both groups were examined for learning and memory study by the Morris water maze test. Then, ventral

Nigral degeneration with inclusion body formation and behavioral changes in rats after proteasomal inhibition.

OBJECTIVE: We were interested in studying nigral degeneration with inclusion body formation and behavioral changes in rats after proteasomal inhibition. METHODS: Observation of progressive behavioral and pathological changes in rats following a unilateral nigral injection of lactacystin, a selective proteasome inhibitor. RESULTS: After administration at a concentration of 10 microg (2 microl) of lactacystin, when tyrosine hydroxylase (TH) immunostaining decreased gradually in the substantia nigra pars compacta (SNc) and corpus striatum, alpha-synuclein-immunopositive inclusion appeared exten

Differential temporal and spatial post-injury alterations in cerebral cell morphology and viability.

Combination of ischemia and β-amyloid (Aβ) toxicity has been shown to simultaneously increase neuro-inflammation, endogenous Aβ deposition, and neurodegeneration. However, studies on the evolution of infarct and panorama of cellular degeneration as a synergistic or overlapping mechanism between ischemia and Aβ toxicity are lacking. Here, we compared fluorojade B (FJB) and hematoxylin and eosin (H&E) stains primarily to examine the chronology of infarct, and the viability and morphological changes in neuroglia and neurons located in different brain regions on d1, d7, and d28 post Aβ toxicity an
