Antibacterial Activity of Coumarins and Carbazole Alkaloid from Roots of Clausena anisata.

Clausena anisata is one of the medicinal plants used traditionally for treatment of parasitic infections, irritation (boils, ringworm, and eczema), flatworm infestations, influenza, abdominal cramps, and constipation. Phytochemical screening test of dichloromethane/methanol (1 : 1) roots extract revealed the presence of flavonoids, phytosterols, coumarins, phenols, alkaloids, tannins, terpenoids, and free reducing sugars and the absence of saponins. Silica gel column chromatographic separation of the dichloromethane/methanol (1 : 1) extract afforded a carbazole alkaloid derivative of heptazoli

Metronidazole-coumarin conjugates and 3-cyano-7-hydroxy-coumarin act as isoform-selective carbonic anhydrase inhibitors.

Reaction of 6-/7-hydroxycoumarin with metronidazole afforded conjugates which incorporate two interesting chemotypes which may inhibit carbonic anhydrases (CAs, EC 4.2.1.1) due to the presence of the coumarin moiety and possess radiosensitizing effects due to the presence of the nitroazole. Another dual action compound, which may act both as CA inhibitor as well as monocarboxylate transporter inhibitor, is 3-cyano-7-hydroxy-coumarin. These compounds have been investigated as inhibitors of 11 human CA isoforms. Submicromolar inhibition was observed against hCA VA, hCA VB, hCA VI, hCA VII, hCA I

Arabidopsis uses distinct coumarins and bacterial pathways for pH-adaptive iron acquisition.

Iron (Fe) limitation restricts plant growth in diverse soils, and root-associated microbes can alleviate plant Fe starvation. Whether plants integrate the edaphic environment and microbial activities into their Fe uptake strategies remains unclear. We show that bacterium-mediated alleviation of Fe deficiency in Arabidopsis functions at varying environmental pH and is taxonomically widespread among root microbiota isolates from soils with different edaphic profiles. This process is regulated by host-controlled and pH-adapted root exudation of different coumarin chemotypes. These exometabolites

Mechanistic pathways of coumarins-mediated cardioprotection: From antioxidant activity to endothelial modulation.

Coumarins are naturally occurring benzopyrones widely recognized for their antioxidant, anti-inflammatory, and vasomodulatory properties. Emerging evidence highlights their potential role in preventing and mitigating cardiovascular injury through integrated modulation of redox balance, nitric oxide (NO) signaling, and endothelial function. This review consolidates preclinical and mechanistic data derived from in vitro and in vivo studies addressing the cardioprotective effects of coumarins. Emphasis was placed on experimental models of ischemia-reperfusion, oxidative stress, and drug-induced c

[2.2]Paracyclophane-based coumarins: effective organo-photocatalysts for light-induced desulfonylation processes.

Herein, we demonstrate for the first time that coumarins derived from [2.2]paracyclophane (pCp) can act as effective organo-photocatalysts and promote the reductive cleavage of sulfonamides under light-irradiation. In the presence of these original compounds, photodesulfonylation reactions occur under mild conditions at low catalyst loadings in the presence of Hantzsch ester. Theoretical and experimental investigations are described, which elucidate the reaction mechanism and the nature of the active species involved in the photocatalytic process. This proof-of-concept study paves the way for

Toxicity evaluation of sulfamides and coumarins that efficiently inhibit human carbonic anhydrases.

Here, we report a toxicity study, conducted on zebrafish larvae, of a series of coumarin and sulfamide compounds that were previously reported as inhibitors of human (h) metalloenzymes, carbonic anhydrases (CAs, EC 4.2.1.1). Due to the high relevance of hCA inhibitors as theragnostic agents, it is of pivotal importance to address safety issues that may arise from the initial in vivo toxicological assessment using zebrafish, a relevant model for biomedical research. None of the reported compounds showed adverse phenotypic effects or tissue damage on developing zebrafish larvae after 5 days of e

Inhibition viral RNP and anti-inflammatory activity of coumarins against influenza virus.

Influenza viruses pose a severe threat to human health and a significant increase in antiviral drug-resistant among influenza viruses worldwide has been observed. Therefore, there is an urgent need to develop the new antiviral drugs, specifically from the natural products. In this study, the anti-viral and anti-inflammatory activities of coumarins against influenza A virus in vitro were investigated. One of the derivatives eleutheroside B1 showed a wide spectrum of anti- human influenza virus effect with the IC50 value of 64-125μg/ml in vitro, but it showed no effects against avian influenza v

Exploring carbonic anhydrase inhibition with multimeric coumarins displayed on a fullerene scaffold.

Carbonic anhydrases (CAs) are ubiquitous Zn metallo-enzymes that catalyze the reversible hydration/dehydration of CO2/HCO3(-). CAs are involved in many key biological processes, therefore their inhibition has become an attractive research field. Distinct families of CA inhibitors (CAIs) have been reported, most of them interacting with the Zn(II) at the active site. Some compounds such as the coumarins are hydrolyzed before binding the entrance of the active site cavity, and thus behave as "suicide" inhibitors. This study reports the first synthesis of multimeric suicide inhibitors, designed t

Hepatoprotective and TNF-α inhibitory activity of Zosima absinthifolia extracts and coumarins.

Zosima absinthifolia (ZA) extracts and the coumarins (+)-columbianadin and (-)-deltoin were evaluated for their potential hepatoprotective and antiinflamatory effects in a CCl(4)-induced hepatotoxicity assay in rats and by the inhibition of TNF-α production on LPS-stimulated THP-1 macrophages, respectively. Both the ZA extracts and the coumarins showed hepatoprotective activity confirmed by monitoring the ALT/AST levels and by histopathological examination. The antiinflamatory activity, proved by the inhibition of TNF-α production, was found to be higher for the n-hexane root extract than for

Effect of oral antiplatelet agents on major bleeding in users of coumarins.

Treatment with vitamin K antagonists (coumarins) is associated with an increased risk of bleeding. In order to elucidate the bleeding risk of users of antiplatelet drugs among users of coumarins, we assessed the odds ratio of major bleeding associated with use of antiplatelet drugs in users of the coumarins acenocoumarol and phenprocoumon. We used data from a Dutch record linkage system, including pharmacy and linked hospitalization records for approximately two million subjects, to conduct a nested case control study in a cohort of new users of coumarins. Cases were patients who were hospital

Acidity Determination by Spectroscopic and Modeling Methods in Monohydroxy- and Dihydroxy-Substituted Coumarins.

We report the pKa values for 15 mono- and disubstituted hydroxycoumarins determined using UV-vis absorbance spectroscopy and computational methods. Six of these values, including the pKa of 5-hydroxycoumarin, are reported for the first time. While the primary focus is on the effects of hydroxyl substitution on the benzene ring, we also examine how a 4-methyl substituent may influence acidity in a subset of four coumarin pairs. The experimental results are supported by two kinds of density functional theory calculations. Time-dependent DFT (TD-DFT) models of the UV-vis spectra show strong agree

Exploring the anticancer potential and mechanisms of action of natural coumarins and isocoumarins.

Natural coumarins and isocoumarins show significant therapeutic potential against cancer in preclinical studies by targeting multiple pathways and processes. These compounds influence several critical cellular processes, such as apoptosis, autophagy, and cell cycle regulation, which are pivotal in cancer development and progression. Their capability to target multiple signalling pathways provides a strategic advantage over single-target therapies, which are often limited by drug resistance. Notably, coumarins have the potential to inhibit angiogenesis, the process through which tumours develop

Characteristic terpenylated coumarins from Ferula ferulaeoides as potential inhibitors on overactivation of microglia.

A total of 37 characteristic terpenylated coumarins (1-25), including 17 undescribed compounds (1-5, 6a/6b, 7-10, 11a/11b-13a/13b), have been isolated from the root of Ferula ferulaeoides. Meanwhile, twelve pairs of enantiomers (6a/6b, 11a/11b-15a/15b, 17a/17b, 18a/18b, 20a/20b-22a/22b, and 25a/25b) were chirally purified. The structures of these new compounds were elucidated using HRESIMS, UV, NMR, and calculated 13C NMR with a custom DP4 + analysis. The absolute configurations of all the compounds were determined for the first time using electronic circular dichroism (ECD). Then, their inhib

Enantioselective synthesis of coumarins catalyzed by an isosteviol-derived tertiary amine-squaramide catalyst.

A newly tertiary amine-squaramide organocatalyst has been successfully developed and applied into the asymmetric Michael addition of 4-hydroxycoumarin to β,γ-unsaturated α-ketoesters. The catalyst system performed well with a low catalyst loading of 1 mol% under mild reaction conditions. A series of coumarin derivatives were obtained in good to high yields (up to 97%) with high enantioselectivities (up to 96% ee).

The coumarin osthole is a non-electrophilic agonist of TRPA1.

The naturally occurring coumarin osthole has antipruritic properties, and recent reports suggest that this effect is due an inhibition or desensitization of the cation channels TRPV1 and TRPV3. Osthole was also suggested to activate TRPA1, an effect that should rather be pruritic than antipruritic. Here we characterized the effects of osthole on TRPA1 by means of ratiometric calcium imaging and patch clamp electrophysiology. In HEK 293 expressing human (h) TRPA1, osthole induced a concentration-dependent increase in intracellular calcium that was inhibited by the TRPA1-inhibitor A967079. In mo

New coumarin derivatives as carbonic anhydrase inhibitors.

In the current study, a series of 4-chloromethyl-7-hydroxy-coumarin derivatives containing imidazolium, benzimidazolium, bisbenzimidazolium and quaternary ammonium salts were synthesized, characterized and the inhibition effects of the derivatives on human carbonic anhydrases (hCA I and hCA II) were investigated as in vitro. Structures of these coumarins were confirmed by FT-IR, (1)H NMR, (13)C NMR and LC-MS analyses. Structure activity relationship study showed that 3d (IC50: 79 μM for hCA I and 88 μM for hCA II) performed higher inhibitory activity than others.

Possible interactions between the new non-steroidal antirheumatic drug Sulindac and a coumarin derivative have been investigated. 20 subjects, who required anticoagulation therapy for a variety of medical conditions, were treated for 4 consecutive weeks with Sulindac at varying dosages (200-400 mg per day). Prothrombin time, Thrombotest and bleeding time were not significantly affected by the concurrent administration of Sulindac. It can be concluded that there is no clinically important interaction between phenprocoumon and the doses of sulindac that are considered effective in rheumatic diso
