Critical Roles of the Direct GABAergic Pallido-cortical Pathway in Controlling Absence Seizures.

The basal ganglia (BG), serving as an intermediate bridge between the cerebral cortex and thalamus, are believed to play crucial roles in controlling absence seizure activities generated by the pathological corticothalamic system. Inspired by recent experiments, here we systematically investigate the contribution of a novel identified GABAergic pallido-cortical pathway, projecting from the globus pallidus externa (GPe) in the BG to the cerebral cortex, to the control of absence seizures. By computational modelling, we find that both increasing the activation of GPe neurons and enhancing the co

Impact of critical pathway implementation on hospital stay and costs in patients undergoing pancreaticoduodenectomy.

BACKGROUNDS/AIMS: Recent studies have shown that pancreaticoduodenectomy (PD) can be performed quite safely. Critical pathway (CP) has been one of the key tools used to achieve excellent outcomes in high-quality, high-volume centers. This study was designed to evaluate the impact of CP implementation for PD patients. METHODS: The important components of CP for PD patients include the early start of an oral diet and removal of the abdomen drain following follow-up computed tomography, with the intention of shortening the postoperative hospital stay. The study group (CP group) comprised of 88 p

The GSK3 signaling pathway is activated by cocaine and is critical for cocaine conditioned reward in mice.

The Akt - GSK3 signaling pathway has been recently implicated in psychostimulant-induced behavioral and cellular effects. Here, the ability of cocaine to regulate the activity of Akt and GSK3 was investigated by measuring the phosphorylation states of the two kinases. The anatomical specificity of the response was determined, as was the contributions of dopamine and NMDA receptors to the actions of cocaine. As GSK3 activity was found to be increased by cocaine, subsequent experiments investigated the importance of GSK3 activation in cocaine conditioned reward. Adult male CD-1 mice were injecte

Improvement in clinical outcomes in acute coronary syndromes after the implementation of a critical pathway.

Although several advances have been made in the management of acute coronary syndromes, the adoption of such measures in clinical practice has been suboptimal. The implementation of critical pathways has been suggested as a strategy to improve clinical effectiveness, although its effect is still to be demonstrated. The objective was to evaluate the impact of a critical pathway on the process of care of patients admitted with acute coronary syndromes in a teaching hospital. In a prospective cohort study, patients 30 years or older admitted to the emergency department with suspected acute corona

A Critical Review of Hydrogen Production Technologies: Comparative and Integrative Analysis From Industrial Processes to Emerging Low-Carbon Pathways.

The global transition toward low-carbon energy systems is constrained by the carbon intensity and scalability limitations of current hydrogen production technologies. Although hydrogen is widely recognized as a clean energy carrier, the absence of an integrated and comparative framework limits its system-level optimization. This review presents a critical assessment of hydrogen production technologies encompassing industrial processes, biomass, and electrochemical pathways. Conventional processes are analyzed across decarbonization strategies, while biomass-derived electrochemical pathways are

Fatty acids and physical activity are critical for β-cell mass and insulin sensitivity: Pathways to T2D and prevention.

Existing mathematical models investigating the progression of type 2 diabetes (T2D) over time primarily focus on glucose, insulin, β-cell mass, and other related factors, while often omitting fatty acids (FA) as an explicit variable-despite FA being a major energy source for the body. There exists a complex network of dynamical interactions among glucose, insulin, FA, and β-cell mass. To gain deeper insights into the metabolic dynamics and pathophysiology of T2D, it is essential to incorporate FA into such models. In this paper, we extend the classic Topp's GIβ model by explicitly incorporatin

Multi-Omics Analysis Revealed a Significant Alteration of Critical Metabolic Pathways Due to Sorafenib-Resistance in Hep3B Cell Lines.

Hepatocellular carcinoma (HCC) is the second prominent cause of cancer-associated death worldwide. Usually, HCC is diagnosed in advanced stages, wherein sorafenib, a multiple target tyrosine kinase inhibitor, is used as the first line of treatment. Unfortunately, resistance to sorafenib is usually encountered within six months of treatment. Therefore, there is a critical need to identify the underlying reasons for drug resistance. In the present study, we investigated the proteomic and metabolomics alterations accompanying sorafenib resistance in hepatocellular carcinoma Hep3B cells by employi

A spadework for integrative medicine based critical pathways for facial palsy: Web-based survey from traditional Korean medicine doctors.

Patients affected with facial palsy consult both traditional Korean medicine doctors and conventional medicine doctors. Considering that the optimal approach varies depending on the progress of the disease, there is a need for facial palsy management through integrated medical care. However, no critical pathway has been developed to manage facial palsy from an integrated medical perspective. The aims of this study were to (a) explore treatment utilization status and awareness for facial palsy; (b) understand possible traditional Korean medicine modalities; and (c) suggest interventions to be i

Structure-based simulations of the full Flock House virus capsid reveal pathways and energetics of infection-critical peptide externalization.

Viral capsids are metastable assemblies whose large-scale dynamics enable critical steps in infection, including the exposure of internal protein segments that interact with host membranes. Resolving the mechanisms underlying these transitions remains challenging because they involve rare, cooperative motions distributed across entire viral particles. Here, we develop a full-capsid structure-based molecular dynamics framework to investigate infection-relevant dynamics in the T = 3 Flock House virus capsid, a model nonenveloped RNA virus. This native contact-centric coarse-grained representatio

Pathways to Crime and Antisocial Behavior: A Critical Analysis of Psychological Research and a Call for Broader Ecological Perspectives.

The United States has one of the highest rates of correctional supervision among all nations in the world, reflecting the disproportionate incarceration of racial minorities and economically disadvantaged groups. Scholars have emphasized the role of structural factors and governmental policies in long-term shifts in crime and incarceration. However, much of the psychological research on crime and antisocial behaviors has not deeply considered this broader context, focusing mostly on individual and proximal environmental risk factors. This article presents a novel synthesis of large cross-disci

Identification of core genes, critical signaling pathways, and potential drugs for countering BPA-induced hippocampal neurotoxicity in male mice.

Although the neurotoxicity of the common chemical bisphenol A (BPA) to the mouse hippocampus has been often reported, the mechanism underlying BPA-induced depression-like behavior in mice remains unclear. We evaluated BPA's role in inducing depressive-like behavior by exposing male mice to different BPA concentrations (0, 0.01, 0.1, and 1 μg/mL) and using the forced swimming test (FST) and tail suspension test (TST). We aimed to identify critical gene and anti-BPA-neurotoxicity compounds using RNA sequencing combined with bioinformatics analysis. Our results showed that 1 μg/mL BPA exposure in

Antibiotic transport requires a renewed focus on baseflow as a critical non-point source pathway.

Current research predominantly assumes that riverine antibiotic loads primarily derive from elevated surface runoff. However, the significance of baseflow is largely overlooked due to a lack of quantitative estimation and mechanistic understanding. This study analyzed the role of baseflow on transporting tetracyclines (TCs) from non-point sources in an agricultural catchment. We found that baseflow load accounted for ∼40 % (39.8 % annually and 45.8 % monthly) of the riverine load of TCs. The threshold effect of baseflow index indicates that baseflow dominates the low-level but ongoing loading

Identification of cell cycle as the critical pathway modulated by exosome-derived microRNAs in gallbladder carcinoma.

Gallbladder cancer (GBC), the most common malignancy in the biliary tract, is highly lethal malignant due to seldomly specific symptoms in the early stage of GBC. This study aimed to identify exosome-derived miRNAs mediated competing endogenous RNAs (ceRNA) participant in GBC tumorigenesis. A total of 159 differentially expressed miRNAs (DEMs) was identified as exosome-derived miRNAs, contains 34 upregulated exo-DEMs and 125 downregulated exo-DEMs based on the expression profiles in GBC clinical samples downloaded from the Gene Expression Omnibus database with the R package. Among them, 2 up-r

NF-κB Pathway as a Potential Target for Treatment of Critical Stage COVID-19 Patients.

Patients infected with SARS-CoV-2 show a wide spectrum of clinical manifestations ranging from mild febrile illness and cough up to acute respiratory distress syndrome, multiple organ failure, and death. Data from patients with severe clinical manifestations compared to patients with mild symptoms indicate that highly dysregulated exuberant inflammatory responses correlate with severity of disease and lethality. Epithelial-immune cell interactions and elevated cytokine and chemokine levels, i.e. cytokine storm, seem to play a central role in severity and lethality in COVID-19. The present pers

Defining the critical period of hedgehog pathway inhibitor-induced cranial base dysplasia in mice.

BACKGROUND: The hedgehog signaling pathway is critical for developmental patterning of the limb, craniofacial and axial skeleton. Disruption of this pathway in mice leads to a series of structural malformations, but the exact role and critical period of the Hh pathway in the early development of the cranial base have been rarely described. RESULTS: Embryos exposed to vismodegib from E7.5, E9.5, and E10.5 had a higher percentage of cranial base fenestra. The peak incidence of hypoplasia in sphenoid winglets and severe craniosynostosis in cranial base synchondroses was observed when vismodegib

Critical role for the Tsc1-mTORC1 pathway in β-cell mass in Pdx1-deficient mice.

Mutations in the pancreatic duodenal homeobox (PDX1) gene are associated with diabetes in humans. Pdx1-haploinsufficient mice also develop diabetes, but the molecular mechanism is unknown. To this end, we knocked down Pdx1 gene expression in mouse MIN6 insulinoma cells. Pdx1 suppression not only increased apoptotic cell death but also decreased cell proliferation, which was associated with a decrease in activity of mechanistic target of rapamycin complex 1 (mTORC1). We found that in Pdx1-deficient mice, tuberous sclerosis 1 (Tsc1) ablation in pancreatic β-cells restores β-cell mass, increases
