Implementing a spinosad-based local bait station to control Bactrocera cucurbitae (Diptera: Tephritidae) in high rainfall areas of Reunion Island.

Three species of fruit flies cause serious damage to cucurbit crops on Reunion Island: Bactrocera cucurbitae (Diptera: Tephritidae) (Coquillett 1899), Dacus ciliatus (Loew 1901), and Dacus demmerezi (Bezzi 1917). To control them, a program of agroecological management of cucurbit flies has been implemented based on the application of Synéis-appât, especially spot sprays on corn borders. However, the high rainfall on Reunion Island limits the long-term efficiency of the bait; in addition, this method cannot be used for large chayote trellises, because corn borders cannot be planted around them.

Synergistic Effects of Zinc Oxide Nanoparticles and Moringa Leaf Extracts on Drought Tolerance and Productivity of Cucurbita pepo L. Under Saline Conditions.

This study investigated the combined effects of zinc oxide nanoparticles (Nano-Zn) and moringa leaf extract (MLE) on squash plants grown under water stress conditions in saline soil during 2021-2022. The research compared full irrigation (100% ETc) with water deficit conditions (60% ETc). While water deficit negatively impacted plant growth, yield, and various physiological parameters, the sequential application of Nano-Zn (at 50 or 100 mg L-1) with MLE (3%) significantly mitigated these adverse effects. The combined treatment proved more effective than individual applications, enhancing growt

Knockdown of the ZcVgR Gene Alters the Expression of Genes Related to Reproduction and Lifespan in Zeugodacus cucurbitae (Coquillett) Under Extreme Heat Conditions.

Zeugodacus cucurbitae (Coquillett) is an important migratory vegetable pest. Previous research has demonstrated that short-term high temperatures induce differential expression of the vitellogenin receptor (ZcVgR) gene, reducing the number of eggs laid and the lifespan of female Z. cucurbitae. In this paper, we used Tandem Mass Tags (TMT) quantitative proteomics and Illumina high-throughput sequencing to determine the proteomic and transcriptomic information of female Z. cucurbitae after siRNA-mediated silencing of the target gene (ZcVgR) to gain a comprehensive understanding of the molecular

Ganoderic acid D as a potential modulator of steroidogenic enzymes: Protective effects of Cucurbita maxima Duch. seed extract against nonylphenol-induced reproductive toxicity.

Nonylphenol (NP), a widespread environmental endocrine disruptor, adversely affects male reproductive health through oxidative stress and impaired steroidogenesis. Although natural products have shown protective effects against reproductive toxicity, the bioactive compounds responsible for these effects and their molecular mechanisms remain inadequately characterized. This study investigated the protective potential of Cucurbita maxima seed extract (CMSE) against NP-induced reproductive toxicity in adult male albino mice and identified key phytochemicals associated with its activity. Male mice

Schistosomicidal, antifibrotic and antioxidant effects of Cucurbita pepo L. seed oil and praziquantel combined treatment for Schistosoma mansoni infection in a mouse model.

Despite the seriousness of schistosomiasis, its treatment depends only on praziquantel (PZQ), which has begun to lose its efficacy against the emergent Schistosoma mansoni-resistant strains. Therefore, the discovery of a novel schistosomicidal drug is an urgent priority. This study was designed to evaluate treatment with Cucurbita pepo L. (pumpkin) seed oil (PSO) alone and combined with PZQ against S. mansoni in experimentally infected mice. The study involved five groups: GI was the normal control; GII was the infected control; GIII was treated with an oral dose of PZQ of 500 mg/kg/day for tw

Two heat shock cognate 70 genes involved in spermatogenesis regulate the male fertility of Zeugodacus cucurbitae, as potential targets for pest control.

The melon fly Zeugodacus cucurbitae Coquillett (Diptera: Tephritidae) is an agricultural quarantine pest threatening fruit and vegetable production. Heat shock cognate 70 (Hsc70), which is a homolog of the heat shock protein 70 (Hsp70), was first discovered in mice testes and plays an important role in spermatogenesis. In this study, we identified and cloned five Hsc70 genes from melon fly, namely ZcHsc70_1/2/3/4/5. Phylogenetic analysis showed that these proteins are closely related to Hsc70s from other Diptera insects. Spatiotemporal expression analysis showed that ZcHsc70_1 and ZcHsc70_2 ar

Genome-wide screening and expression of glutathione S-transferase genes reveal that GSTe4 contributes to sensitivity against β-cypermethrin in Zeugodacus cucurbitae.

Glutathione S-transferases (GSTs) are an essential multifunctional protein family with common detoxifying enzymes. In this study, 34 GST genes were identified from the melon fly, Zeugodacus cucurbitae, one of the most destructive pests worldwide. These GSTs include 32 cytosolic genes and two microsomal genes. Furthermore, these cytosolic GSTs were classified into six classes: 11 delta, 13 epsilon, three theta, one sigma, two zeta, and two omega. Most of these showed dynamic expression during the developmental stage, some of which showed stage-specific expression. The expression in various adul

Differential modulation of binding loop flexibility and stability by Arg50 and Arg52 in Cucurbita maxima trypsin inhibitor-V deduced by trypsin-catalyzed hydrolysis and NMR spectroscopy.

The side chains of Arg50 and Arg52 iin Cucurbita maxima trypsin inhibitor-V (CMTI-V) anchor the binding loop to the scaffold region [Cai, M., Gong, Y., Kao, J.L-F., & Krishnamoorthi, R. (1995) Biochemistry 34, 5201-5211]. The consequences of these hydrogen-bonding and electrostatic interactions on the conformational flexibility and stability of the binding loop were evaluated by trypsin-catalyzed hydrolysis of CMTI-V mutants, in which each of the arginines was individually replaced with Ala, Lys, or Gln by genetic engineering methods. All mutants exhibited significantly increased vulnerability

In vitro and in vivo evaluation of antioxidant activity, acute toxicity, and antihyperglycemic effect of individual and binary mixtures of crude methanolic extracts of Cucurbita moschata and Bidens pilosa.

BACKGROUND: Bidens pilosa (BP) and Cucurbita moschata (CM) are used to treat diabetes traditionally. Little is known scientifically about the toxicity and efficacy of the individual or binary mixtures on blood-sugar-lowering effects. Hence, this study carried out toxicity studies and examined the antidiabetic effects of these plants to support traditional knowledge. We report the phytochemical composition, total phenolic (TPC), flavonoid content (TFC), and antioxidant activity of individual and mixed methanolic BP and CM leaf extracts. We also report acute toxicity of the individual extracts.
