Characterization of cyclophilin 23 as a novel factor for development and pathogenicity of Cryptosporidium parvum.

Cryptosporidium parvum (C. parvum) is a common zoonotic protozoan pathogen that can cause severe diarrhea in both humans and animals. However, the factors involved in its pathogenicity remain incompletely understood. The C. parvum genome contains nine genes that encode peptidyl-prolyl cis/trans isomerases (PPIases). Previous bioinformatics analyses have indicated that proteins within this family may be associated with the pathogenicity of C. parvum. Here, we explored the role of cyclophilin 23 (CpCyP23), a member of the PPIase family, in the development and pathogenicity of C. parvum. In this

Targeting Cyclophilin A in the Cardiac Microenvironment Preserves Heart Function and Structure in Failing Hearts.

BACKGROUND: Cardiac hypertrophy is characterized by remodeling of the myocardium, which involves alterations in the ECM (extracellular matrix) and cardiomyocyte structure. These alterations critically contribute to impaired contractility and relaxation, ultimately leading to heart failure. Emerging evidence implicates that extracellular signaling molecules are critically involved in the pathogenesis of cardiac hypertrophy and remodeling. The immunophilin CyPA (cyclophilin A) has been identified as a potential culprit. In this study, we aimed to unravel the interplay between eCyPA (extracellula

Safety of white adipose tissue activator cyclophilin A intracellular peptide (Pep19™) in healthy dogs.

Oral consumption of Pep19™, a 10 amino acid intracellular peptide, has been associated with weight loss in rodents and humans via induction of nonshivering thermogenesis. This study aimed to test its safety and tolerance in dogs. Eight healthy adult neutered university owned beagles (4 female and 4 male) were individually housed in runs and fed an extruded kibble in a quantity historically associated with weight stability. They were administered Pep19™ (5 mg/dog/day, 0.32 - 0.49 mg/kg/day) as a once daily oral dose for 28 days. Health screening, including physical examination, body weight, bod

A Pan-Cyclophilin Inhibitor, CRV431, Decreases Fibrosis and Tumor Development in Chronic Liver Disease Models.

Previous studies show that cyclophilins contribute to many pathologic processes, and cyclophilin inhibitors demonstrate therapeutic activities in many experimental models. However, no drug with cyclophilin inhibition as the primary mode of action has advanced completely through clinical development to market. In this study, we present findings on the cyclophilin inhibitor, CRV431, that highlight its potential as a drug candidate for chronic liver diseases. CRV431 was found to potently inhibit all cyclophilin isoforms tested-A, B, D, and G. Inhibitory constant or IC50 values ranged from 1 to 7

Cyclophilin A regulates HIV-1 infectivity, as demonstrated by gene targeting in human T cells.

The human immunodeficiency virus type 1 (HIV-1) Gag polyprotein binds most members of the cyclophilin family of peptidyl-prolyl isomerases. Of 15 known human cyclophilins, cyclophilin A (CypA) has been the focus of investigation because it was detected in HIV-1 virions. To determine whether CypA promotes HIV-1 replication, we deleted the gene encoding CypA (PPIA) in human CD4(+) T cells by homologous recombination. HIV-1 replication in PPIA(-/-) cells was decreased and not inhibited further by cyclosporin or gag mutations that disrupt Gag's interaction with cyclophilins, indicating that no oth

Cyclophilin A as a biomarker for the therapeutic effect of balloon angioplasty in chronic thromboembolic pulmonary hypertension.

BACKGROUND: Although cardiac troponin and natriuretic peptide have been shown to decrease after balloon pulmonary angioplasty (BPA) with improved right ventricular afterload in chronic thromboembolic pulmonary hypertension (CTEPH), biomarkers to evaluate the effects of BPA independently of heart failure status remain to be developed. METHODS: In 39 consecutive CTEPH patients including 31 who underwent BPA, we measured plasma levels of cyclophilin A (CyPA), which we demonstrated is secreted from pulmonary vascular smooth muscle cells in response to mechanical stretch and hypoxia. RESULTS: CyP

A novel cyclophilin from parasitic and free-living nematodes with a unique substrate- and drug-binding domain.

A highly diversified member of the cyclophilin family of peptidyl-prolyl cis-trans isomerases has been isolated from the human parasite Onchocerca volvulus (OvCYP-16). This 25-kDa cyclophilin shares 43-46% similarity to other filarial cyclophilins but does not belong to any of the groups previously defined in invertebrates or vertebrates. A homolog was also isolated from Caenorhabditis elegans (CeCYP-16). Both recombinant O. volvulus and C. elegans cyclophilins were found to possess an enzyme activity with similar substrate preference and insensitivity to cyclosporin A. They represent novel cy

Cyclophilin-a as a key paracrine factor replicating hPVSC therapeutic effects in a chemotherapy-induced ovarian damage model.

Chemotherapy-induced premature ovarian insufficiency (POI) causes significant fertility loss. We investigated whether human perivascular stem cells (hPVSC) or their secreted factor, cyclophilin-A (CYP-A), could restore ovarian function in mouse of cyclophosphamide (CP)-induced ovarian damage. Mice subjected to acute or fractionated CP regimens were treated with hPVSC or recombinant CYP-A. We assessed ovarian histology, follicle counts, and in vitro fertilization (IVF) outcomes. While ovary-to-body weight ratios recovered only in the high-dose model, both treatments significantly preserved tota

Genetic Deletion of Cyclophilin D Results in Enhanced Hypoxia Tolerance in Mice.

Hypoxic preconditioning (HPC) activates intracellular anti-hypoxia molecular defense mechanisms through short-term non-lethal repeated hypoxic stimulation, leading to the subsequent acquisition of high tolerance to lethal hypoxic damage in cells. Cyclophilin D (CypD) regulates the function of mitochondria by controlling the opening of the mitochondrial permeability transition pore. However, the mechanism of action of CypD in hypoxia and HPC is poorly understood. Here, we examined the role of CypD under HPC using both wild-type (WT) and Ppif gene knockout (KO) mice. The results showed that HPC

Structure-Based Screening Identifies Polyphenol Glucuronides as Cyclophilin D Ligands.

(1) Background: The mitochondrial permeability transition pore (mPTP) contributes to mitochondrial dysfunction under pathological conditions. Cyclophilin D (CypD), a key regulator of sustained mPTP opening, represents an important molecular target. However, systematic strategies for selecting an appropriate CypD crystal structure for virtual screening and efficiently identifying CypD-binding natural polyphenols remain limited. (2) Methods: Six CypD crystal structures were evaluated by receiver operating characteristic analysis using reported CypD inhibitors and approximately 4000 decoys. The o

Cyclophilin D reduces Ca2+ sequestration by complement 1q binding protein.

Complement 1q binding protein (C1qBP) and cyclophilin D (CypD) are mitochondrial matrix proteins; C1qBP has been implicated in many cellular processes, including the regulation of oxidative phosphorylation, and CypD is widely associated with the regulation of mitochondrial permeability transition pore (mPTP) opening. In this study, C1qBP and CypD were shown, in vitro, to form a stable protein-protein complex. CypD-C1qBP interaction was disrupted by cyclosporin A and compromised by mutations of the CypD active site residues R55 and R82. AlphaFold protein modelling revealed that the large negati

Cyclophilin D deficiency attenuates mitochondrial perturbation and ameliorates hepatic steatosis.

UNLABELLED: Physiological opening of the mitochondrial permeability transition pore (mPTP) is indispensable for maintaining mitochondrial function and cell homeostasis, but the role of the mPTP and its initial factor, cyclophilin D (CypD), in hepatic steatosis is unclear. Here, we demonstrate that excess mPTP opening is mediated by an increase of CypD expression induced hepatic mitochondrial dysfunction. Notably, such mitochondrial perturbation occurred before detectable triglyceride accumulation in the liver of high-fat diet-fed mice. Moreover, either genetic knockout or pharmacological inhib

Characterization of the Anti-HCV Activities of the New Cyclophilin Inhibitor STG-175.

Shortened current direct-acting antiviral (DAA) therapies while less expensive, have not provided satisfactory efficacy in naïve cirrhotics, treatment experienced non-cirrhotics or even genotype-3 (GT3)-infected patients. Since DAA regimens consist of the same classes of inhibitors-NS5A (NS5Ai) and NS5B (NS5Bi) +/- NS3 (NS3i) inhibitors-it is likely that their costs will be high and will provide similar degrees of protection. Integrating drugs with distinct mechanisms of action (MoA) into DAA regimens could provide the solution for shortening the period of treatment. One such class of agents i

Study of the participation of MMP-7, EMMPRIN and cyclophilin A in the pathogenesis of periodontal disease.

BACKGROUND AND OBJECTIVE: Periodontal disease is an infectious disease resulting from the immunoinflammatory response of the host to microorganisms present in the dental biofilm which causes tissue destruction. The objective of this study was to evaluate the immunohistochemical expression of matrix metalloproteinase 7 (MMP-7), extracellular matrix metalloproteinase inducer (EMMPRIN) and cyclophilin A (CypA) in periodontal disease. DESIGN: Gingival tissue samples were divided as follows: clinically healthy gingiva (n=32), biofilm-induced gingivitis (n=28), and chronic periodontitis (n=30). His

Targeting Extracellular Cyclophilins Ameliorates Disease Progression in Experimental Biliary Atresia.

Biliary atresia (BA) is a devastating liver disease of unknown etiology affecting children generally within the first 3 months of life. The disease is manifested by inflammation and subsequent obstruction of the extrahepatic bile ducts, fibrosis and liver failure. The mechanisms responsible for disease pathogenesis are not fully understood, but a number of factors controlled by the SMAD signaling pathway have been implicated. In this study, we investigated the role of a known proinflammatory factor, extracellular cyclophilin A (CypA), in the pathogenesis of biliary atresia using the rhesus rot

Cyclophilin-D is dispensable for atrophy and mitochondrial apoptotic signalling in denervated muscle.

In the present study, we specifically determined whether the regulatory protein cyclophilin-D (CypD), and by extension opening of the permeability transition pore (PTP), is involved in the activation of mitochondria-derived apoptotic signalling previously described in skeletal muscle following loss of innervation. For this purpose, CypD-defficient (CypD-KO) mice and their littermate controls were submitted to unilateral sciatic nerve transection, and mitochondrial resistance to Ca2+-induced opening of the PTP, and muscle apoptotic signalling were investigated 14 days post-surgery. Developmental shift of cyclophilin D contribution to hypoxic-ischemic brain injury.

Cyclophilin D (CypD), a regulator of the mitochondrial membrane permeability transition pore (PTP), enhances Ca(2+)-induced mitochondrial permeabilization and cell death in the brain. However, the role of CypD in hypoxic-ischemic (HI) brain injury at different developmental ages is unknown. At postnatal day (P) 9 or P60, littermates of CypD-deficient [knock-out (KO)], wild-type (WT), and heterozygous mice were subjected to HI, and brain injury was evaluated 7 d after HI. CypD deficiency resulted in a significant reduction of HI brain injury at P60 but worsened injury at P9. After HI, caspase-d

Secreted Cyclophilin A Induces M1 and M2 Macrophage Polarization in the THP-1 Model in vitro.

Fate of monocytes that migrate into tissues is determined by various mediators at the site of inflammation. The microenvironment governs monocyte differentiation and polarization into macrophages, which acquire different surface markers and opposite functions. Secreted cyclophilin A (CypA) is a pro-inflammatory factor and a component of the monocyte humoral microenvironment. In these studies, effects of recombinant human CypA (rhCypA) on differentiation of human monocytic cells were evaluated in the THP-1 in vitro model. RhCypA induced polarization of M1 macrophages in the early stages of THP-
