Tinea corporis caused by Microsporum gallinae: first clinical case in Japan.

We report herein a case of tinea corporis caused by Microsporum gallinae in a 96-year-old, otherwise healthy Japanese man. The patient had a long working history as a breeder of fighting cocks, and he suffered from two erythematous macules after being bitten by a cock. M. gallinae was identified as the infectious agent based on the morphology of isolates cultured on slides and analysis of DNA sequences of the internal transcribed spacers (ITS) from ribosomal DNA from cultured isolates. The patient was successfully treated with antifungal ointments. To our knowledge, this is the first case of M

Molecular phylogeny and morphology reveal a new epiphytic species of Habenaria (Orchidaceae; Orchideae; Orchidinae) from Nepal.

Habenaria is almost cosmopolitan in distribution having predominantly terrestrial orchids, however; a remarkable epiphytic species with some unique morphological characters was collected from Nepal. We conducted a molecular phylogeny of this unusual Habenaria species using nuclear (ITS) and chloroplast (matK, rbcl) DNA sequence regions to infer its systematic position. Our molecular analyses and morphological treatment recognized this newly collected plant as an undescribed species. This species is described here which is closely related to Habenaria plurifoliata but can be distinguished by ha

Molecular evidence shows that the liver fluke Fasciola gigantica is the predominant Fasciola species in ruminants from Pakistan.

Fascioliasis is an important disease affecting livestock, with great costs to producers worldwide. It has also become a serious issue for human populations in some endemic areas as an emerging zoonotic infection. There are two Fasciola species of liver fluke responsible for this disease, which occur worldwide, Fasciola hepatica and Fasciola gigantica. Identifying these two species on the basis of adult or egg morphology requires specialist knowledge due to the similarity of characters, and may misidentify putative intermediate or hybrid forms. In this study we sequenced the internal transcribe

Ceratomyxa gracillima n. sp. (Cnidaria: Myxosporea) provides evidence of panmixia and ceratomyxid radiation in the Amazon basin.

We describe a new freshwater myxosporean species Ceratomyxa gracillima n. sp. from the gall bladder of the Amazonian catfish Brachyplatystoma rousseauxii; the first myxozoan recorded in this host. The new Ceratomyxa was described on the basis of its host, myxospore morphometry, ssrDNA and internal transcribed spacer region (ITS-1) sequences. Infected fish were sampled from geographically distant localities: the Tapajós River, Pará State, the Amazon River, Amapá State and the Solimões River, Amazonas State. Immature and mature plasmodia were slender, tapered at both ends, and exhibited vermifor

Skoulekia erythrini n. sp. (Digenea: Aporocotylidae): a parasite of Pagellus erythrinus (L.) (Perciformes: Sparidae) from the western Mediterranean with an amendment of the generic diagnosis.

A new aporocotylid, Skoulekia erythrini n. sp., is described from the heart, cephalic kidney and gill blood vessels of the common pandora Pagellus erythrinus (L.) collected from several localities of the western Mediterranean off Spain. The new species differs from the type- and only species of the genus, S. meningialis Alama-Bermejo, Montero, Raga & Holzer, 2011 in possessing a symmetrical body (vs laterally curved), short tegumental spines (3-4 vs 7-10 µm) without hooked ends, diffuse (vs conspicuous) oesophageal gland-cells, a relatively longer oesophagus, a testis that is shorter in relati

Meningoencephalitis with malacia caused by Sarcocystis calchasi in a rock pigeon in Japan.

Sarcocystis spp. cause pigeon protozoan encephalitis, a neuronal disease. A female pigeon exhibiting torticollis had a necrotic area in the cerebral hemisphere surrounded by lesions with perivascular cuffing, gliosis, granulomatous foci, and meningitis. Non-necrotic lesions were also observed in the brainstem. Intact and degenerative schizonts were observed within the neuropils and neurons in the lesions. Deoxyribonucleic acid (DNA) was extracted from paraffin-embedded brain tissues and genetically analyzed after gel electrophoresis to determine Sarcocystis spp. using specific primer sets for

New species and new genus of Pseudohaploporinae (Digenea): Pseudohaploporus pusitestis sp. n. and Parahaploporus elegantus n. g., sp. n. (Digenea: Pseudohaploporinae) from Vietnamese mullet fish.

Two new species of Pseudohaploporinae, Pseudohaploporus pusitestis sp. n. and Parahaploporus elegantus n. g., sp. n., are described from intestines of the Vietnamese mullet fish Moolgarda seheli and Osteomugil cunnesius, respectively. Pseudohaploporus pusitestis sp. n. differs from two known Pseudohaploporus species, P. vietnamensis and P. planiliza, by the absence of a diverticulate hermaphroditic duct and muscular sphincters at the proximal end of the hermaphroditic sac. Metrically, P. pusitestis sp. n. is close to P. vietnamensis and differs from this species and from P. planilizum by lower

Two new species of Gloniopsis (Hysteriales, Ascomycota) from clinical specimens: Morphological and molecular characterisation.

BACKGROUND: The coelomycetes comprise a wide range of fungal species distributed in at least three different classes of the phylum Ascomycota. These are morphologically characterised by producing their conidia inside of fruiting bodies called pycnidia or acervuli, and only a reduced number of species are able to cause human infections. However, their identification in the clinical laboratory is often difficult, due to their few morphological features or because they remain sterile. MATERIALS AND METHODS: In the present study, three isolates of coelomycetes of clinical origin were phenotypical

Characterization of the complete nuclear ribosomal DNA sequences of Eurytrema pancreaticum.

Eurytrema pancreaticum is one of the most common trematodes of cattle and sheep, and also infects humans occasionally, causing great economic losses and medical costs. In this study, the sequences of the complete nuclear ribosomal DNA (rDNA) repeat units of five E. pancreaticum individuals were determined for the first time. They were 8306-8310 bp in length, including the small subunit (18S) rDNA, internal transcribed spacer 1 (ITS1), 5.8S rDNA, internal transcribed spacer 2 (ITS2), large subunit (28S) rDNA and intergenic spacer (IGS). There were no length variations in any of the investigated

Keratitis caused by a rare fungus, Malassezia restricta.

BACKGROUND: Malassezia species rarely cause keratitis, and, thus, the clinical manifestations of Malassezia keratitis are not well known. CASE: We report the clinical findings in a 70-year-old woman who complained of pain in her left eye. Slit-lamp biomicroscopy showed a corneal ulcer with irregular infiltration, which resembled keratitis caused by filamentous fungi. KOH Parker ink stain of a corneal scraping showed mold hyphae and yeast, but cultures on Sabouraud's glucose agar plates and blood agar plates were negative. Treatment with antibacterial agents failed. OBSERVATIONS: Polymerase c

Peritonitis due to Aspergillus tamarii in peritoneal dialysis patients.

Aspergillus peritonitis is uncommon, but it is associated with high mortality and morbidity in patients undergoing peritoneal dialysis (PD). We report two cases of Aspergillus tamarii peritonitis that were initially misidentified as A. flavus by the conventional culture method. Nucleotide sequences of internal transcribed spacer regions of the ribosomal DNA gene as A. tamarii correctly identified the isolate. Despite early catheter removal and an appropriate antifunal agent, both patients had dismal outcomes. Nucleic acid sequencing offers an additional tool for better diagnosing the species w

The divergence pattern of Trichinella spiralis isolates from China revealed by mitochondrial and ribosomal markers.

Trichinella spiralis is the main etiological agent of human trichinellosis. In China, trichinellosis remains a serious food-borne parasitic zoonosis and poses a serious threat to human health. However, the genetic structure of Chinese T. spiralis population is still little known. In this study, we used three molecular markers to analyze phylogeographic structure of the Chinese T. spiralis population. A total of 11 T. spiralis isolates were collected from 10 geographical locations in mainland China. The cytochrome c-oxidase gene (COI), large subunit ribosomal DNA (mt-lsrDNA) and 5S ribosomal DN

Association of the components of soil microorganisms and the density of Schistosome-transmitting Oncomelania hupensis.

Soil microorganisms play an important role in the survival and development of soil-dwelling animals. Oncomelania hupensis, the sole intermediate host of Schistosoma japonicum, inhabitats in soil during its adult stage and is greatly influenced by the soil microenvironment. Nevertheless, the relationship between soil microorganisms and the density of O. hupensis remains poorly understood. In this study, we surveyed the density of O. hupensis and soil properties (physicochemical properties and microbial communities) in bottomlands from schistosomiasis endemic areas in the Dongting Lake and Poyan

Detection of Salmonella typhi by polymerase chain reaction.

A rapid and sensitive method for detection of Salmonella typhi would help in preventing the spread of outbreaks and in clinical diagnosis. In order to develop unique PCR primers to detect Salm. typhi, ribosomal RNA genes from Salm. typhi (Rawlings) were cloned in pUC18. The resulting clone was confirmed by sequencing. The cloned DNA fragment contained the 5S, part of the 23S rRNA genes and the 5S-23S spacer region (EMBL/GenBank accession No. U04734). It was expected that the 5S-23S spacer region is divergent unlike the highly conserved 23S + 5S genes. This was confirmed by comparison with the

Broadening our knowledge of the Trachelophyllidae Kent, 1881 (Litostomatea, Ciliophora): Observations on known species of three genera and a new genus and species from the Czech Republic.

Trachelopyllidae Kent, 1881 is a family of predatory litostomatean ciliates distinguished by a three-rowed dorsal brush composed of two dikinetidal rows and one monokinetid row, and one or more types of organic epicortical scales called lepidosomes. To date, the taxonomy of the nine genera comprising the family has been based, almost exclusively, on the types of scales and their combinations together with cell morphometry. Six of the nine genera are monotypic. To date, molecular data has been available from only four species representing two genera. With the aim of broadening the molecular cha

Scleroductus (Platyhelminthes: Monogenea) infecting Rhamdia spp. (Siluriformes: Heptapteridae) in Mexico, and their phylogenetic position within Gyrodactylidea.

Gyrodactylidae Cobbold, 1864 is a monophyletic family of hyperviviparous species, with 36 genera and approximately 700 species. Here, we focused on one of these genera - namely Scleroductus Jara & Cone, 1989 - characterizing its morphological variation systematically and testing its phylogenetic position in relation to other genera in the family. We collected and describe two new species of monogenean parasites of Scleroductus infecting Neotropical freshwater catfishes in Mexico. Scleroductus veracruzano n. sp. infects Guatemalan chulín, Rhamdia guatemalensis in Veracruz, while Scleroductus oa

Unroughing the cat's tongue mushrooms: Four new species of Pseudohydnum from Brazil based on morphological and molecular phylogenetic evidence.

Pseudohydnum, commonly known as cat's tongue mushrooms, is a monophyletic assemblage within Auriculariales, which encompasses species with gelatinous basidiomata, spathulate, flabellate, or shell-shaped pileus, hydnoid hymenophore, globose to ellipsoidal basidiospores, and longitudinally cruciate-septate basidia. According to the available literature, 16 species have been described in Pseudohydnum, mostly represented in temperate-boreal forests of the Northern Hemisphere. However, the limited morphological, molecular, and ecological information, especially from the Southern Hemisphere ecosyste

New record and phylogenetic assessment of Apopharynx bolodes (Braun, 1902) (Digenea: Psilostomidae), a parasite of Eurasian Coot Fulica atra Linnaeus, 1758 (Aves: Rallidae).

Phylogenetic studies of aberrant species are of considerable scientific interest because their taxonomic rank in traditional systems based on morphological characters is not infrequently overestimated. Apopharynx bolodes (Braun, ) is one of the few psilostomid digeneans devoid of the pharynx. This is considered a sufficient basis for assigning it and similar species to the subfamily Apopharynginae. We found A. bolodes in Fulica atra Linnaeus, 1758 from Belarus, described it morphologically, and genotyped it by the 28S rRNA gene and the ITS2 region. It is the first molecular data on A. bolodes

Steinernema pwaniensis n. sp., a new entomopathogenic nematode (Nematoda: Steinernematidae) from Tanzania.

A new species of entomopathogenic nematode, Steinernema pwaniensis n. sp., belonging to the glaseri group, is described from Tanzania. The infective juveniles of S. pwaniensis n. sp. are characterized by a body length of 978 μm (808-1131), distance from anterior end to excretory pore of 86 μm (80-95) and a tail length of 87 μm (75-95). The ratios a, c and E% of S. pwaniensis n. sp. are substantially lower than those of all other African 'glaseri' group members. The first-generation males of S. pwaniensis n. sp. can be distinguished by a large spicule length of 92 μm (80-97) and by the absence

Molecular and phylogenetic analysis of the filarial nematode Micipsella numidica from the hare Lepus europaeus in Italy.

The genus Micipsella comprises three species of filariae to date identified in lagomorphs only, whereas the other genera belonging to the subfamily Splendidofilariinae are described as parasites of birds, reptiles and mammals. In the present study seven specimens of Micipsella numidica (Seurat, 1917), collected from the hare Lepus europaeus in Italy, were characterized genetically by molecular amplification of the mitochondrial genes (12S rDNA; cox1) and the 5S rDNA gene spacer region.
