Quantifying the effects of hemoglobin saturation on plasma strong ion difference during blood oxygenation and decarboxylation.

The strong ion difference (SID) is assessed to interpret acid-base disorders, yet measured plasma values are influenced by electrolyte redistribution across compartments. Current models focus on pH-dependent plasma-erythrocyte shifts. Here, we aim to quantify the effects of hemoglobin oxygen saturation (sO2). We induced oxygenation and decarboxylation of human venous blood (n = 20) via room-air equilibration. We modeled the contribution of ∆sO2 to plasma-erythrocyte shifts through the Haldane effect and hypothesized that combining sO2- with pH-dependent mechanisms would allow accurate predicti

Temporal dependence emerges from a carbodiimide driven acid-anhydride equilibrium coupled to a photocatalytic decarboxylation reaction.

Neurons process multiple stimuli in a temporally dependent manner, which filters noise and enables precise control over complex functions. Synthetically mimicking neuronal processes may be useful for autonomous chemical systems that are able to make their own decisions, yet such time-dependence has not found an equivalent in synthetic reaction networks. Herein, we show that carbodiimide driven equilibria between acid and anhydride can be coupled to photochemical decarboxylation from which temporal input dependence emerges. Under optimized conditions, the decarboxylation of the anhydride is muc

Transamination and oxidative decarboxylation rates of branched-chain 2-oxo acids in cultured human skin fibroblasts.

Transamination and oxidative decarboxylation of branched-chain L-amino acid derived 2-oxo acids in cultured human skin fibroblasts from normal subjects and from a patient with maple syrup urine disease (variant form) were comparatively studied in incubations with 1-14C-labeled substrates (1 mmol/liter). With normal cells, 14CO2 release ranged from about 11 to 3 nmol/90 min/mg of cell protein in the order 3-methyl-2-oxo[14C]butanoate greater than (S)-3-methyl-2-oxo[14C]pentanoate greater than 4-methyl-2-oxo[14C]pentanoate greater than (R)-3-methyl-2-oxo[14C]pentanoate. Decarboxylation of 3,4-dihydroxyphenylalanine (DOPA) by erythrocytes: a reaction promoted by methemoglobin and other ferriheme proteins.

The decarboxylation of DOPA by erythrocyte hemolysates differs from DOPA decarboxylation catalyzed by aromatic aminoacid decarboxylases that contain vitamin B(6) in several significant respects. The ability of erythrocyte hemolysates to decarboxylate DOPA is associated with interaction between DOPA and methemoglobin; the ferriheme protein is reduced and DOPA is decarboxylated, probably after oxidation to a quinone intermediate. An analogous reaction takes place between DOPA and other ferriheme proteins, such as metmyoglobin and cytochrome c. This phenomenon may be of significance in relation t

Decarboxylative carbonyl alkylative amination: A general and practical strategy for aliphatic amine synthesis.

Aliphatic amines are ubiquitous core structural motifs in bioactive molecules including natural products, pharmaceuticals, and agrochemicals, driving ongoing demand for versatile, green, and practical synthetic strategies to construct complex aliphatic amines from inexpensive, readily available, structurally diverse feedstocks. Here, we report an iron-catalyzed decarboxylative carbonyl alkylative amination of aldehydes, amines, and carboxylic acids in HFIP under mild light irradiation at room temperature, with no additional additives and only CO2 and H2O as environmentally benign by-products.

Synthesis of peptide- and drug-oligonucleotide conjugates via a photocatalyzed decarboxylative Giese reaction.

Herein, we report a photocatalyzed decarboxylative Giese reaction for the late-stage functionalization of 5'-COOH oligonucleotides. Under mild visible-light conditions, C4' radicals are generated and trapped by diverse Michael acceptors, including simple alkenes, Dha-containing peptides, and alkene-bearing drug molecules. This method enables the rapid synthesis of peptide- and drug-oligonucleotide conjugates and is applicable to dinucleotides as well as 3-4 nt oligonucleotides on a microscale (10-100 nmol).

Preassembly-Controlled Radical Recombination at Bismuth: Decarboxylative C─N Coupling with Sulfonamides.

Persistent transition metal radicals form the foundation for many metallaphotoredox protocols. Their ability to efficiently trap organic radicals and convert them into various coupling products has inspired the exploration of selective radical reactions even beyond the d-block. Radical processes involving bismuth hold great potential, but innovative strategies are required to control the reactivity of bismuth intermediates. Herein, we report preassembly as a powerful strategy to enforce a selective recombination of a bismuth(II) radical and an organic radical. As a result, an inner-sphere path

Pd-Catalyzed Decarboxylative Negishi Coupling of Zinc Aryl Carboxylates with Arylthianthrenium Salts.

We report a Pd-catalyzed decarboxylative Negishi coupling reaction for efficient biaryl synthesis from various zinc aryl carboxylates, including polyfluorobenzoates and heteroaryl carboxylates, using DMF as the solvent. This mild reaction exhibits a broad substrate scope and enables late-stage functionalization of bioactive molecules. Mechanistic studies show that DMF-assisted zinc catalyzes decarboxylation of polyfluorinated aryl carboxylates to generate arylzinc reagents in situ, which then undergo Negishi coupling catalyzed by palladium with arylthianthrenium salts to form biaryl compounds.

Chromium(II)-Catalyzed Decarboxylative Alkyl Acylation under Visible Light Irradiation.

By utilizing the stability of metal-ligand coordination and the instability of the metal-to-ligand charge transfer excited state to dissociate the ligand, the first chromium(II) decarboxylative alkyl acylation reaction under visible light irradiation is herein reported. The reaction uses α-keto acids as acyl radical precursors to react with alkyl radicals, without the need for additional photosensitizers or other additives. The elegant reaction exhibits a wide scope across α-keto acids, radical precursors, and alkanes with moderate to good yields.

[Metal-free Synthesis of α,α-Difluorocarbonyl Compounds via Decarboxylative Arylation].

Decarboxylative arylation of α,α-difluoro-β-ketoacid salts with diaryliodonium(III) salts has been developed to synthesize α-aryl-α,α-difluoromethyl ketones, which are attractive synthetic intermediates for various difluorobenzyl units. This additive-free arylation represents an alternative approach to conventional synthetic methods that rely on transition metal catalysts and/or organometallic compounds. The reaction involves sequential ligand exchange of difluoroketoacid with tosylate ligand of diaryliodonium salt, followed by decarboxylative ligand coupling. Various functional groups, includ

Catalytic enantioselective synthesis of carbocyclic and heterocyclic spiranes via a decarboxylative aldol cyclization.

The synthesis of a variety of enantioenriched 1,3-diketospiranes from the corresponding racemic allyl β-ketoesters via an interrupted asymmetric allylic alkylation is disclosed. Substrates possessing pendant aldehydes undergo decarboxylative enolate formation in the presence of a chiral Pd catalyst and subsequently participate in an enantio- and diastereoselective, intramolecular aldol reaction to furnish spirocyclic β-hydroxy ketones which may be oxidized to the corresponding enantioenriched diketospiranes. Additionally, this chemistry has been extended to α-allylcarboxy lactam substrates lea

Decarboxylative sp3 C-N coupling via dual copper and photoredox catalysis.

Over the past three decades, considerable progress has been made in the development of methods to construct sp2 carbon-nitrogen (C-N) bonds using palladium, copper or nickel catalysis1,2. However, the incorporation of alkyl substrates to form sp3 C-N bonds remains one of the major challenges in the field of cross-coupling chemistry. Here we demonstrate that the synergistic combination of copper catalysis and photoredox catalysis can provide a general platform from which to address this challenge. This cross-coupling system uses naturally abundant alkyl carboxylic acids and commercially availab

Effects of 1-S replaced and/or decarboxylated latamoxef on rabbit platelet aggregation in vitro.

Latamoxef, 1-S replaced and/or decarboxylated derivatives of latamoxef were examined for their effects on ADP-, collagen- and platelet activating factor (PAF)-induced rabbit platelet aggregation in vitro. The results were compared with those of cefotaxime, cefmetazole, carbenicillin and aspirin. Latamoxef produced a dose-dependent inhibition of platelet aggregation at concentrations over about 4 mM, and the potency was almost similar to that produced by the other beta-lactam antibiotics, although the inhibiting effect of ADP-induced aggregation was more potent for latamoxef, whereas that on co

Accelerated leucine decarboxylation in the rat brain in relation to increased blood ammonia levels during acute hepatic failure.

Leucine decarboxylation in rat brain was investigated during acute hepatic failure, induced by partial hepatectomy after carbon tetrachloride (CCl4) pretreatment of rats. These rats presented metabolic alkalosis, and had significantly higher levels of arterial blood and brain ammonia than control and CCl4-treated rats. Brain leucine decarboxylation was elevated in rats with hepatic failure. This alteration correlated with arterial blood ammonia concentrations, and probably with elevated brain ammonia levels, as brain ammonia levels were directly related to arterial blood ammonia.

Photoredox-Neutral Platform for Direct Decarboxylative Cyclopropanation via Acridine-Iron Dual Catalysis.

A photoredox-neutral approach to cyclopropanes from in situ generated non-stabilized carbenes via a new acridine-iron dual catalytic system is reported. This strategy merges the oxidative decarboxylation of bench-stable carboxylic acids with the reduction of an iron porphyrin under mild photochemical conditions. Bespoke acridine photocatalysts efficiently shuttle electrons between carboxylic acids and iron complexes, obviating the need for a sacrificial reductant. Mechanistic studies reveal that, contrary to prevailing assumptions, decarboxylation does not proceed through a concerted PCET path

Decarboxylative Cross-Acyl Coupling of Carboxylic Acids with Aldehydes Enabled by Nickel/Photoredox Catalysis.

We present a general method for accessing unsymmetrical alkyl-aryl and alkyl-alkyl ketones via nickel/photoredox-catalyzed decarboxylative cross-acyl coupling reactions between carboxylic acids and aldehydes without the need for an additional preactivation procedure. Specifically, by using the peroxide as both an oxidant and hydrogen atom transfer (HAT) reagent, we achieved the unprecedented combination of oxidative single electron transfer (SET) of carboxylates and HAT of aldehydes, in which the generated alkyl and acyl radicals were chemoselectively coupled by nickel catalysis. This method f

Decarboxylative 3-Indole Methylenation of 4-Aminocoumarin and Application in the Synthesis of Fused Polycyclic Compounds.

The decarboxylative indole methylenation of 4-aminocoumarins in the C3-site has been realized with copper promotion using 3-indolyl acetic acids as reaction partners. In addition to providing a practical method for the synthesis of indole methylene-functionalized 4-aminocoumarins, the metal-free intramolecular amination and aromatization have been achieved for the synthesis of complex pentacyclic compounds, namely, coumarin-fused α-carbolines, with broad scope and generally excellent efficiency.
