Remineralization of dentin in vitro.

The remineralization of completely demineralized bovine dentin was studied in vitro by monitoring the velocities of fall of small particles through calcifying solutions. The density of a particle of dentin may be found from its velocity of fall through a fluid using Stokes' law. The minimum concentration product of calcium and acid phosphate ions of the solution in which remineralization would take place was 3.6 (mM)2 in the presence of 22 mM bicarbonate, pH 7.35, and ionic strength 0.1. This just above the solubility product of brushite (CaHPO4-2H2O). Incubation of decalcified dentin in a pho

Effects of Silver Diamine Nitrate and Silver Diamine Fluoride on Dentin Remineralization and Cytotoxicity to Dental Pulp Cells: An In Vitro Study.

Silver diamine nitrate (SDN) is expected to help control caries similar to silver diamine fluoride (SDF). The aim of this study was to determine the mineral precipitation in demineralized dentin and the cytotoxicity of SDN and SDF to dental pulp cells. Demineralized dentin specimens were prepared, and SDF, SDN, or water (control) was applied. The specimens were then remineralized in simulated body fluid for 2 weeks. The mineral precipitation in the specimens was examined using FTIR-ATR, SEM-EDX, and synchrotron radiation X-ray tomographic microscopy (SRXTM). Additionally, the cytotoxicity of S

Clastic cells in orthodontic treatment: Translational challenges and recent advances.

OBJECTIVES: Orthodontic treatment consists of numerous appliance activations that rely on stimulation of osteoclasts at alveolar bone sites. However, the action of osteoclast-like cells on dentin ("odontoclasts") is a pathological side effect of orthodontic treatment. The aim of this article is twofold: (a) To report preliminary results from ongoing cell culture experiments to identify unique markers of dentin resorption, and (b) To discuss our work using nanoparticle tracking analysis (NTA) and exosomes for developing biological fluid-based biopsies to monitor clastic cell activity. SETTING

Changes in the permselectivity of human teeth during caries attack.

Previous studies have shown that enamel permselectivity can influence fluid composition within caries lesions during de- and remineralization. The permselectivity of human enamel, cementum, and dentin sections was examined, in a microwell model, by measurement of the membrane potential developed by KCl diffusion while the sections were immersed in solutions simulating resting (pH = 5.6) and cariogenic plaque fluid (pH = 4.8). In a second experiment, the effects of charged compounds (phytate and Zonyl-FSC) on the tooth permselectivity were examined. The average membrane potentials (+/- SD) in "

Age-induced changes in the teeth and their attachment apparatus.

Both the hard substances and the soft tissues of the teeth and their attachment apparatus are subject to constant change. This begins immediately after eruption and continues throughout life. An exact dividing line between changes which are physiological and pathological cannot always be drawn. Enamel undergoes attrition, and in addition its mechanical characteristics alter, owing probably to changes in diffusion conditions. The age-induced changes occurring in dentine are much more obvious, the biological properties of this hard substance being fundamentally altered. The dentine of older peop

Adhesive/Dentin interface: the weak link in the composite restoration.

Results from clinical studies suggest that more than half of the 166 million dental restorations that were placed in the United States in 2005 were replacements for failed restorations. This emphasis on replacement therapy is expected to grow as dentists use composite as opposed to dental amalgam to restore moderate to large posterior lesions. Composite restorations have higher failure rates, more recurrent caries, and increased frequency of replacement as compared to amalgam. Penetration of bacterial enzymes, oral fluids, and bacteria into the crevices between the tooth and composite undermin

Mechanism of action of a desensitizing fluoride toothpaste delivering calcium and phosphate ingredients in the treatment of dental hypersensitivity. Part II: comparison with a professional treatment for tooth hypersensitivity.

Tooth hypersensitivity can occur when gum recession causes exposure of dentin. Tiny tubules, which permeate dentin, provide open passageways from the mouth to the intradental nerve in the pulpal cavity. Under such circumstances, stimuli in the mouth can cause pressure on the intradental nerve, leading to pain. Sealing the outside of the tubules with an impermeable substance can effectively treat hypersensitivity. One such clinically proven composition is a professionally applied tooth desensitizer, which has been shown to initially produce a layer of amorphous calcium phosphate (ACP) on the su

Functional recovery of demineralized dentin using a glutamic acid-modified electrospun scaffold: A multimodal in vitro characterization.

BACKGROUND: Dentin remineralization poses a significant challenge in restorative dentistry due to the complex structure and mineral-organic composition of dentin. Electrospun nanofibrous scaffolds present a biomimetic platform to facilitate mineral deposition. Functionalization of the scaffold, achieved through the incorporation of biomolecules like glutamic acid, which mimics non-collagenous proteins, may improve remineralization. OBJECTIVE: To assess the remineralization potential of a glutamic acid-loaded polycaprolactone/nanohydroxyapatite (PCL/nHA/Glu) scaffold on acid-demineralized dent
