Dentinogenesis imperfecta (DI) is a hereditary disorder that affects dentin formation, leading to discolored, fragile teeth with increased susceptibility to wear and fracture. A 41-year-old woman presented with a family history of DI and significant tooth wear primarily affecting her maxillary anterior teeth due to parafunctional habits. She reported dissatisfaction with her smile, indicating that her front teeth were short and did not show when smiling. Examination revealed blue-gray teeth with craze lines characteristic of DI, receded pulp chambers, and evidence of significant tooth surface

Dentinogenesis Imperfecta (DI) is a rare genetic disorder characterized by dentin hypomineralisation. While DI is known to impair dentin's mechanical properties of the tissue and cause multiple tooth fractures, the microstructural origins of dentin fragility remain poorly understood. To address this gap,we conducted a pilot study comparing primary healthy dentin (n=4 - four incisors) and dentin affected by DI associated with a COL1A2 variant (DI type I, n=4 - two canines and two molars) using thermogravimetry and backscattered electron scanning microscopy to quantify mineralization at macrosca

PURPOSE: To demonstrate the field of application and prospects of individually modeled indirect composite restorations for the treatment of children and adolescents based on a case of dentinogenesis imperfecta. Dental malformations can affect single or multiple teeth. In most cases, direct composite fillings can be placed. However, in severe cases, these restorations may be more challenging and error-prone, especially when occlusal adjustments are necessary. Since composite materials do not require a specific lamination strength and are easy to repair, they can be applied using the indirect te

OBJECTIVE: To identify the causative mutation in a Chinese family affected with dentinogenesis imperfecta shields type II (DGI-II).
METHODS: With informed consent obtained from all participants, peripheral blood or chorionic villi samples were collected from the family members. Genomic DNA was extracted using a standard SDS-proteinase K-phenol/chloroform method. The whole coding region and exon/intron boundaries of the DSPP gene were amplified with polymerase chain reaction (PCR) and subjected to Sanger sequencing. To confirm the pathogenicity of the identified mutation, an Alu I recognition

Hydroxyapatite crystal deposition and stabilization within the collagen matrix of bone and dentin have been linked to the presence of noncollagenous proteins (NCP). Dentinogenesis imperfecta (DI), a genetic disorder of dentin mineralization, is being studied as a model for the analysis of mineralization mechanisms. A comparative study of the NCP in normal human dentin and dentinogenesis imperfecta Type II (hereditary opalescent dentin) dentin has been performed. The proteins of each tissue were extracted and separated using a variety of techniques. The calcium-binding, highly phosphorylated pr

The follow-up history and oral findings in two brothers from consanguineous parents suggest that the association of dentinogenesis imperfecta (DI), delayed tooth eruption, mild mental retardation, proportionate short stature, sensorineural hearing loss and dysmorphic facies may represent a new syndrome with autosomal recessive inheritance. Histological examination of the dentin matrix of a permanent molar from one of the siblings reveals morphological similarities with defective dentinogenesis as presenting in patients affected with Osteogenesis Imperfecta (OI), a condition caused by deficienc

OBJECTIVES: The current study aims to assess the dentin bonding performance of primary molars with dentinogenesis imperfecta type II (DGI-II). Moreover, we assess the efficacy of calcium phosphate polymer induced liquid precursor (Ca/P-PILP), a biomimetic remineralization material, in improving the resin bonding performance of DGI-II dentin. MATERIALS AND METHODS: Dentin specimens from normal and DGI-II primary molars were treated with either Ca/P-PILP or double-distilled water. Morphology of these dentin specimens were observed using scanning electron microscopy (SEM), and the calcium and pho

Dentinogenesis Imperfecta (DI) is a rare genetic disorder that disrupts the structure and mechanical integrity of dentin, often through alterations to the collagen matrix. In cases associated with Osteogenesis Imperfecta, DI arises from COL1A1 or COL1A2 variants (DI type I). The multiscale mechanisms linking alterations of the collagenic scaffold to dentin microstructural defects remain poorly understood. In this proof-of-concept study, we employed a multimodal 3D characterization framework combining biochemical assays and advanced imaging techniques to investigate the relationship between col

Teeth of seven patients from the Brandywine isolate who had dentinogenesis imperfecta (DI) type III were evaluated by clinical, radiologic, and scanning electron microscopic techniques. The deciduous and permanent teeth were opalescent, and there was marked attrition. Enamel pitting was present on some permanent teeth. Anterior open bites were found in all persons with complete permanent dentitions. Pulps of developing teeth were larger than normal during early development but rapidly became almost completely obliterated. There was increased constriction at the cementoenamel junctions. While r

This case report details the diagnosis and treatment of dentinogenesis imperfecta in a 6-year-old neutered male Labrador, presenting without concurrent osteogenesis imperfecta. Diagnostic modalities, including radiographs, CT imaging, and histopathological examination, are reviewed in conjunction with the latest literature on canine dentinogenesis imperfecta. This patient presented at a more advanced age than typically reported cases. The clinical history, as provided by referring veterinarians, documented fractured deciduous teeth with delayed exfoliation. By 10 months of age, the patient's p

We used transmission immunoelectron microscopy and polyclonal antibodies to study the reactivities of Types III and VI collagen in dentin of normal human permanent and primary teeth and in primary teeth from five patients with dentinogenesis imperfecta (DI) associated with osteogenesis imperfecta and occurring as a single trait. In the normal permanent tooth, reactivity of Type III collagen was occasional and, where present, peritubular. Staining of normal primary teeth was less occasional but still rare, whereas the abnormal dentin stained more uniformly. Atypical, non-striated fibrillar stru

An eight-year-old boy was referred for dental assessment of dentinogenesis imperfecta, a full clinical examination also revealed joint hypermobility and some features of mild osteogenesis imperfecta although he had suffered few fractures. Analysis of the collagens produced by both gingival and skin fibroblast cultures showed the synthesis and intracellular retention of an abnormal alpha 2(I) chain that migrated faster than normal on SDS-PAGE. Cyanogen bromide peptide mapping of this intracellular protein indicated a probable deletion in the N-terminal peptide alpha 2CB4. The denaturation tempe

OBJECTIVES: The aim of this study was to examine the histological appearance of dental hard tissues in primary teeth from children with DI using conventional polarised light microscopy and correlate that with 3D imaging using X-ray microtomograpy (XMT) to gain a further understanding of the dentine structure of teeth diagnosed with dentinogenesis imperfecta.
DESIGN: Undecalcified sections of primary teeth from patients diagnosed with Dentinogenesis Imperfecta Type II were examined using polarised light microscopy. XMT was employed for 3D-imaging and analysis of the dentine.
RESULTS: The pola

A case of hereditary opalescent dentin is described, which showed the same clinical and radiological features as those reported previously. However, there was no evidence of osteogenesis imperfecta. While a diagnosis of type II dentinogenesis imperfecta was therefore made, this case also had features characteristic of type III dentinogenesis imperfecta. Treatment by means of an overdenture is described.

Although the most common oral manifestation of osteogenesis imperfecta is dentinogenesis imperfecta, several authors have described jaw fractures and radiolucent bone lesions associated with this disease. We report a case of a 23-year-old white woman with osteogenesis imperfecta and dentinogenesis imperfecta who developed multiple unilocular, bilateral, radiolucent lesions of the mandible 5 years after presenting with a condylar fracture. This is the first documentation by surgical and histopathologic examination that the radiolucent lesions in this particular case represent multiple idiopathi

OBJECTIVE: The incidence of craniofacial and dental anomalies in children with the more severe nonlethal forms of osteogenesis imperfecta was evaluated.
STUDY DESIGN: The study evaluated 40 children (age range, 1-17.5 years) with types III and IV osteogenesis imperfecta. In each case, the dentition was evaluated for the presence of dentinogenesis imperfecta, attrition, and caries, as well as for radiographic appearance, dental development, and malocclusion.
RESULTS: The incidence of dentinogenesis imperfecta was greater than 80% in the primary dentition. Clinically, the color of the dentitio

BACKGROUND: Dentinogenesis imperfecta is exceptionally associated to chondrodysplasia. The aim of this work is to present four cases of such an association.
CASE REPORT: These four children (three boys; one girl) suffered from growth retardation, ligamentous hyperlaxity, scoliosis. Main features were present since the first months of life. Dentinogenesis imperfecta was more marked on the first teeth. On X-rays, all patients had short tubular bones, more pronounced at the level of the middle segment of their limbs, with irregular metaphyses. Cone-shaped epiphyses were present on the hands. Ili

We have studied 166 patients from 71 families with Sillence type I osteogenesis imperfecta (dominant inheritance and blue sclerae). We confirm earlier findings that there are two subgroups, those with and those without dentinogenesis imperfecta; each family can be allocated to one or other group. Our confidence that the two groups represent distinct disorders is increased by finding that the patients with dentinogenesis imperfecta differ not only in their dental characteristics but also in other clinical features. They have a more severe disease with a greater fracture rate and a greater likel
