The First Records of Canine Babesiosis in Dogs from Dermacentor reticulatus-Free Zone in Poland.

Tick-borne microorganisms belong to important etiological agents of many infectious diseases affecting humans and animals. Among them, there are haemoprotozoans of the Babesia genus, which infect erythrocytes of a host and may cause many clinical symptoms. Canine babesiosis is an emerging tick-borne disease in Southern and Central Europe. In this study, we report two cases of symptomatic canine babesiosis caused by Babesia canis in domestic dogs from the Silesian Voivodeship, Poland, as well as the presence of Dermacentor reticulatus ticks detected on one of the Babesia-infected dogs (D. retic

Dermacentor reticulatus - a tick on its way from glacial refugia to a panmictic Eurasian population.

The ornate dog tick (Dermacentor reticulatus) shows a recently expanding geographic distribution. Knowledge on its intraspecific variability, population structure, rate of genetic diversity and divergence, including its evolution and geographic distribution, is crucial to understand its dispersal capacity. All such information would help to evaluate the potential risk of future spread of associated pathogens of medical and veterinary concern. A set of 865 D. reticulatus ticks was collected from 65 localities across 21 countries, from Portugal in the west to Kazakhstan and southern Russia in th

Identification and evaluation of midgut protein RL12 of Dermacentor silvarum interacting with Anaplasma ovis VirD4.

Anaplasma ovis, a tick-borne intra-erythrocytic Gram-negative bacterium, is a causative agent of ovine anaplasmosis. It is known that Dermacentor ticks act as biological vectors for A. ovis. VirD4 is the machine component of Type IV Secretion System of A. ovis. To better understand the pathogen-vector interaction, VirD4 was used as a bait protein for screening midgut proteins of Dermacentor silvarum via yeast two-hybrid mating assay. As a result, a ribosomal protein RL12 was identified from the midgut cDNA library of D. silvarum. For further validation, using in vitro Glutathione S-transferase

Occurrence of Francisella spp. in Dermacentor reticulatus and Ixodes ricinus ticks collected in eastern Poland.

A total of 530 questing Dermacentor reticulatus ticks and 861 questing Ixodes ricinus ticks were collected from Lublin province (eastern Poland) and examined for the presence of Francisella by PCR for 16S rRNA (rrs) and tul4 genes. Only one female D. reticulatus tick out of 530 examined (0.2%) was infected with Francisella tularensis subspecies holarctica, as determined by PCR of the rrs gene. None of 861 I. ricinus ticks were infected with F. tularensis. In contrast, the presence of Francisella-like endosymbionts (FLEs) was detected in more than half of the D. reticulatus ticks (50.4%) and 0.

Evidence for an increased geographical distribution of Dermacentor reticulatus in Germany and detection of Rickettsia sp. RpA4.

Two studies were performed to elucidate the current distribution of the tick Dermacentor reticulatus in Germany. In the first one in 2003, a total of 365 dogs from 171 sites in the states of Berlin and Brandenburg was screened for ticks, and the corresponding outdoor sites that the dogs usually visited were searched for host-seeking ticks by the flagging method. A total of 1155 ticks was removed from the dogs. The majority were Ixodes ricinus (88.5%), followed by D. reticulatus (9.1%) and I. hexagonus (2.4%). Altogether, 222 dogs carried I. ricinus (60.8%), 41 D. reticulatus (11.2%) and 15 I.

[Dermacentor ticks in France and Germany. Molecular biological differences in species, ecology and epidemiological implications].

Dermacentor reticulatus and D. marginatus, two ixodid tick species occurring in France and Germany, exhibit morphologically overlapping phenotypes. The results of sequence analyses of the hypervariable rDNA gene region ITS 2 do not support conspecifidity. As in France, D. reticulatus has to be characterized as an epidemiologically effective vector of Babesia canis in Germany as well, with the endemic area being located at Kehl/Offenburg/Lahr/Emmendingen/Freiburg in Breisgau. Ecological investigations indicate that a spreading of D. reticulatus and, as a possible consequence, that of B. canis i

County-level surveillance for the American dog tick (Dermacentor variabilis) and Rickettsia species in Kentucky.

Spotted fever group (SFG) rickettsiosis includes a group of illnesses with similar symptoms caused by Rickettsia species bacteria. While Rickettsia rickettsii, the agent of Rocky Mountain spotted fever (RMSF) is a well-known pathogen, other SFG Rickettsia spp. can infect ticks and occasionally contribute to human disease. We conducted county-level surveillance of Dermacentor variabilis ticks in Kentucky to characterize the distribution and diversity of SFG Rickettsia. Ticks were collected from the environment from 2019 to 2024 by dragging a white cloth and through our Kentucky Tick Submission

The transstadial persistence of tick-borne encephalitis virus in Dermacentor reticulatus ticks in natural conditions.

There are a number of reports regarding natural infection of Dermacentor reticulatus ticks with TBE virus; however, the transmission mode of TBE virus in this tick population has not been investigated. This study was conducted in Białowieża Primeval Forest, east Poland. Forty fully engorged nymphs of D. reticulatus were sampled from root voles (Microtus oeconomus). Ticks were kept until molting. All ticks were screened for the presence of TBE virus by nested RT-PCR. Three adult ticks were positive for infection with TBE virus. The present study for the first time demonstrates the possibility o

A systems biology approach to the characterization of stress response in Dermacentor reticulatus tick unfed larvae.

BACKGROUND: Dermacentor reticulatus (Fabricius, 1794) is distributed in Europe and Asia where it infests and transmits disease-causing pathogens to humans, pets and other domestic and wild animals. However, despite its role as a vector of emerging or re-emerging diseases, very little information is available on the genome, transcriptome and proteome of D. reticulatus. Tick larvae are the first developmental stage to infest hosts, acquire infection and transmit pathogens that are transovarially transmitted and are exposed to extremely stressing conditions. In this study, we used a systems biolo

West-to-east differences of Babesia canis canis prevalence in Dermacentor reticulatus ticks in Slovakia.

Babesia canis canis is the most frequent causative agent of canine babesiosis in Central Europe, frequently causing severe disease. Recently, many new endemic foci of this disease have been reported from European countries. Growing incidence of canine babesiosis was recorded also in Slovakia during the last decade, from first cases in eastern Slovakia ten years ago to recent cases all over the south of the country. We have used nested PCR-RFLP method to study prevalence of B. c. canis in its natural tick vector Dermacentor reticulatus, collected at three geographically isolated lowland areas o

An arthropod defensin expressed by the hemocytes of the American dog tick, Dermacentor variabilis (Acari: Ixodidae).

Both soluble and cell-mediated components are involved in the innate immune response of arthropods. Injection of Borrelia burgdorferi, the Lyme disease agent, results in the secretion of defensin into the hemolymph of the ixodid tick, Dermacentor variabilis. The presence of the peptide is observed as early as 15 min post-challenge and remains present through 18 h post-challenge. As observed in insects and soft ticks, the transcript for defensin is detected as early as 1 h post-challenge in D. variabilis. RT-PCR resulted in an amplicon of 624 bp with a 225 bp region that translates to a 74 amin

Localization of insulin-like immunoreactivity in the synganglion of nymphal and adult Dermacentor variabilis (Acari: Ixodidae).

Immunocytochemical staining based on a peroxidase-antiperoxidase method showed neurosecretory cells (NSC) reactive to bovine insulin in five of 18 paraldehyde fuchsin-positive neurosecretory regions (NSR) in the synganglion of unfed adult Dermacentor variabilis. This is the first report of a neuropeptide in an ixodid tick. The insulin-specific immunoreactive cells included the posterior medial group of the protocerebral center, posterior group of dorsal opisthosomal center, anterior lateral group of the dorso-lateral cheliceral center, dorsal group of the frontal stomodeal center, and anterior

Dermacentor variabilis: resistance to ticks acquired by mast cell-deficient and other strains of mice.

The acquisition of resistance to ticks was monitored in mice of six different strains. Mice were subjected to repeated infestations with Dermacentor variabilis larvae, different skin sites being used for each successive infestation. In the third and fourth infestations, resistance was expressed in three strains of mice (WBB6F1-W/Wv, WBB6F1-+/+, and CFW), as demonstrated by significant reductions in percentages of larvae engorging and in mean weights of fed larvae. Both WBB6F1-W/Wv mice, which are mast cell-sufficient strain attained significantly higher levels of resistance. It is suggested th

Detection of the emerging tick-borne pathogen, Tamdy virus, in Dermacentor marginatus ticks, southeastern Türkiye.

Tamdy virus (TAMV) is an emerging tick-borne virus in the family Nairoviridae, identified as a human pathogen causing febrile illness after tick exposure. This study aimed to detect and characterise TAMV in ticks collected from non-endemic regions of Türkiye. A total of 529 ticks were sampled from six provinces and identified as Haemaphysalis parva (27%), Rhipicephalus bursa (26%), Haemaphysalis punctata (20.7%), Dermacentor marginatus (17.2%), and Ixodes ricinus (8.8%). Screening with a single-step generic PCR revealed one TAMV-positive pool out of 96. For characterisation, the S segment of T

Mixed-sex clusters on grass blades: breeding strategy of the ornate dog tick, Dermacentor reticulatus.

BACKGROUND: The ornate dog tick Dermacentor reticulatus is second only to the hard tick Ixodes ricinus in terms of importance as a vector of infectious organisms, especially of Babesia canis, the agent of canine babesiosis. Both the geographical range and local densities of D. reticulatus are steadily increasing in many regions of Europe. In the present study, we tested the hypothesis that D. reticulatus possesses an efficient breeding strategy that allows for a rapid increase in tick numbers and densities through the formation of mixed-sex clusters/aggregations while questing in the environme

Environmental sampling for Dermacentor species (Acari: Ixodidae) in southern British Columbia, Canada in May, 2022.

Traditionally, the most commonly encountered Dermacentor spp. (Acari: Ixodidae) in southern British Columbia, Canada has been Dermacentor andersoni Stiles. However, with the novel detection of Dermacentor similis Lado in southern British Columbia, environmental sampling for Dermacentor spp. is needed to provide up-to-date information on which ticks are present in this area. Drag sampling was conducted at 33 sites across southern British Columbia, covering 2000 m2 per site. Environmental and weather data were recorded at the time of sampling. The only Dermacentor spp. collected was D. andersoni

Defensin from the ornate sheep tick Dermacentor marginatus and its effect on Lyme borreliosis spirochetes.

Expression of the previously reported defensin of the tick Dermacentor marginatus (defDM) was analysed in different organs by RT-PCR. mRNA of the defDM gene was detected in the hemolymph, midgut and salivary glands. Moreover defDM was isolated from the tick hemolymph using RP-HPLC and its sequence was determined by mass spectrometry and Edman degradation. Synthetic peptide was used for determining biological activities. The results showed an anti-Gram-positive bacterial role for the defensin. As D. marginatus ticks appear not to be vectors of the Lyme disease agent of the complex Borrelia burg
