Embryological Divergence and Molecular Mechanisms in Thoracic and Abdominal Aortic Aneurysms: Bridging Developmental Biology and Clinical Insights.

Aortic aneurysms are complex, predominantly asymptomatic vascular diseases with distinct incidence patterns depending on anatomical localisation. The incidence of thoracic aortic aneurysms (TAAs) has moderately increased, whereas that of abdominal aortic aneurysms has declined, primarily due to public health measures. Undiagnosed or poorly managed aneurysms are at significant risk of progression to acute aortic syndrome, with high associated mortality. The embryological origins of the aorta may have a substantial impact on its structural, cellular, and functional heterogeneity. Specifically, s

Novel Insights into the Sinoatrial Node in Single-Cell RNA Sequencing: From Developmental Biology to Physiological Function.

Normal cardiac automaticity is dependent on the pacemaker cells of the sinoatrial node (SAN). Insufficient cardiac pacemaking leads to the development of sick sinus syndrome (SSS). Since currently available pharmaceutical drugs and implantable pacemakers are only partially effective in managing SSS, there is a critical need for developing targeted mechanism-based therapies to treat SSS. SAN-like pacemaker cells (SANLPCs) are difficult to regenerate in vivo or in vitro because the genes and signaling pathways that regulate SAN development and function have not been fully elucidated. The develop

The Putative E3 Ubiquitin Ligase TEX1 Is Required for Nuclear Biology and Developmental Progression of Plasmodium berghei in the Liver.

Malaria remains a major global health burden, and the emergence of resistance to blood stage antimalarials underscores the need for new interventions targeting earlier stages of the parasite's life cycle. The pre-erythrocytic liver stage represents a critical bottleneck and an attractive target for chemotherapeutic and prophylactic interventions. In this study, we functionally characterized the putative E3 ubiquitin ligase Trophozoite Exported Protein 1 (TEX1; PBANKA_0102200) in Plasmodium berghei using gene knockout, tagging, and imaging approaches across the mosquito and liver stages. TEX1 k

Teaching the science of life: A multidisciplinary educational approach to reproductive technology debates through the lens of developmental biology.

In vitro fertilization, cryopreservation and pre-implantation genetic testing are transformative reproductive technologies offering hope for individuals facing fertility challenges. Scientifically, understanding the science of developmental biology is essential for comprehending the mechanisms and implications of these technologies. In embryological sciences, biological perspectives identify life as a series of developmental stages ranging from conception to viability, each potentially representing a different 'beginning' of life. However, the concept of ensoulment, rooted in religious and cul

A systems biology approach reveals neuronal and muscle developmental defects after chronic exposure to ionising radiation in zebrafish.

Contamination of the environment after the Chernobyl and Fukushima Daiichi nuclear power plant (NPP) disasters led to the exposure of a large number of humans and wild animals to radioactive substances. However, the sub-lethal consequences induced by these absorbed radiological doses remain understudied and the long-term biological impacts largely unknown. We assessed the biological effects of chronic exposure to ionizing radiation (IR) on embryonic development by exposing zebrafish embryo from fertilization and up to 120 hours post-fertilization (hpf) at dose rates of 0.5 mGy/h, 5 mGy/h and 5

How insights from cardiovascular developmental biology have impacted the care of infants and children with congenital heart disease.

To illustrate the impact developmental biology and genetics have already had on the clinical management of the million infants born worldwide each year with CHD, we have chosen three stories which have had particular relevance for pediatric cardiologists, cardiothoracic surgeons, cardiac anesthesiologists, and cardiac nurses. First, we show how Margaret Kirby's finding of the unexpected contribution of an ectodermal cell population - the cranial neural crest - to the aortic arch arteries and arterial pole of the embryonic avian heart provided a key impetus to the field of cardiovascular patter

Genetics and developmental biology of oesophageal atresia and tracheo-oesophageal fistula: lessons from mice relevant for paediatric surgeons.

Oesophageal atresia and tracheo-oesophageal fistula are relatively frequently occurring foregut malformations of which the aetiology and pathogenesis are poorly understood. Recent results of molecular genetic studies, in particular the use of single and compound mutant mice, have yielded a tremendous increase in the understanding of the molecular mechanisms involved in normal and abnormal foregut morphogenesis. In the introduction of this paper, we review the very early stages of normal and abnormal embryology of the foregut derivatives and the separation of the foregut into a ventral respirat

Microplastic trafficking in maternal-fetal system: a systematic review and quantitative profiling linking particle characteristics to developmental biology.

Microplastics (MPs) have been detected across multiple maternal and neonatal biological matrices, suggesting widespread exposure during critical developmental stages. However, the mechanisms shaping their distribution and accumulation within the maternal-fetal system remain poorly understood. This systematic review and quantitative synthesis provides a comprehensive quantitative profiling of MP burden across six maternal and neonatal biological matrices placenta, breast milk, endometrium, amniotic fluid, meconium, and feces. Our analysis reveals three critical insights: (1) Tissue‑specific MP

How does a tadpole know when to metamorphose? Integrating evolutionary, ecological, and developmental biology with the neuroendocrinology of amphibian metamorphosis.

Amphibian metamorphosis is an excellent model for studying how selective pressures, environmental cues, endocrine signals, and molecular mechanisms influence developmental timing. In this minireview, I discuss key questions about the timing of metamorphosis, including: when and how do larvae acquire the ability to initiate metamorphosis; what historical and current ecological factors influence the optimal timing of transformation; and what are the physiological mechanisms that translate environmental information into a developmental response? From an ecological perspective, the timing of amphi

Developmental biology of the Brazilian 'Armed' spider Phoneutria nigriventer (Keyserling, 1891): microanatomical and molecular analysis of the embryonic stages.

Phoneutria (Ctenidae) is among the most dangerous venomous spiders in Brazil. Its venom is composed of a mixture of pharmacologically active components, some of which have been quite extensively studied due to their potentiality as models for new pharmaceutical drugs. Nevertheless, literature data on the venom-producing glands are very limited. In the present study, we follow the biological development of intra-cocoon stages of Phoneutria nigriventer spiders, mainly regarding the formation of the venomous apparatus and venom production. The results showed that the venom glands of Phoneutria ar

Pristionchus pacificus: A Genetic Model System for the Study of Evolutionary Developmental Biology and the Evolution of Complex Life-History Traits.

INTRODUCTIONPristionchus pacificus is a nematode that has been established as a model system for evolutionary developmental biology. Initially, P. pacificus was used as a convenient nematode with which to compare the processes of vulva and gonad development as well as sex determination to Caenorhabditis elegans, one of the best-studied animal models. P. pacificus shares many features with C. elegans, including a short generation time, its ability to be easily cultured in the laboratory, and self-fertilization as a mode of reproduction. These features allowed forward and reverse genetic tools t

The color purple: analyzing alkaline phosphatase expression in experimentally manipulated sea urchin embryos in an undergraduate developmental biology course.

In these laboratory exercises, developed for a sophomore/junior-level undergraduate course in Developmental Biology, students explore the processes of differentiation and morphogenesis in sea urchin embryos by monitoring the spatio-temporal expression pattern of the endoderm marker, alkaline phosphatase. Once students have determined the normal alkaline phosphatase expression pattern, they are asked to treat sea urchin embryos in some way that perturbs normal morphogenesis. Their task is to discover whether the chosen treatment perturbs both morphogenesis and differentiation of the gut or only

Developmental biology and substrate preference of the quarantine pest, mango pulp weevil Sternochetus frigidus (Coleoptera: Curculionidae) under laboratory conditions.

The mango pulp weevil, Sternochetus frigidus (Coleoptera: Curculionidae), is a major monophagous and quarantine pest of mango (Mangifera indica) in tropical Asia, causing severe yield and quality losses due to its concealed feeding and development within fruit tissues. A laboratory study was conducted to investigate the developmental biology, morphometrics and substrate preference of S. frigidus using seven mango substrates: young flush, old flush, green fruit (whole and sliced), ripened fruit (whole and sliced) and stem. Experimental rearing recorded all post-ovipositional stages except the e

Resolving complex cartilage structures in developmental biology via deep learning-based automatic segmentation of X-ray computed microtomography images.

The complex shape of embryonic cartilage represents a true challenge for phenotyping and basic understanding of skeletal development. X-ray computed microtomography (μCT) enables inspecting relevant tissues in all three dimensions; however, most 3D models are still created by manual segmentation, which is a time-consuming and tedious task. In this work, we utilised a convolutional neural network (CNN) to automatically segment the most complex cartilaginous system represented by the developing nasal capsule. The main challenges of this task stem from the large size of the image data (over a tho

Clustering of Schistosoma mansoni mRNA sequences and analysis of the most transcribed genes: implications in metabolism and biology of different developmental stages.

The study of the Schistosoma mansoni genome, one of the etiologic agents of human schistosomiasis, is essential for a better understanding of the biology and development of this parasite. In order to get an overview of all S. mansoni catalogued gene sequences, we performed a clustering analysis of the parasite mRNA sequences available in public databases. This was made using softwares PHRAP and CAP3. The consensus sequences, generated after the alignment of cluster constituent sequences, allowed the identification by database homology searches of the most expressed genes in the worm. We analyz

Skeletal muscle fiber type: using insights from muscle developmental biology to dissect targets for susceptibility and resistance to muscle disease.

Skeletal muscle fibers are classified into fiber types, in particular, slow twitch versus fast twitch. Muscle fiber types are generally defined by the particular myosin heavy chain isoforms that they express, but many other components contribute to a fiber's physiological characteristics. Skeletal muscle fiber type can have a profound impact on muscle diseases, including certain muscular dystrophies and sarcopenia, the aging-induced loss of muscle mass and strength. These findings suggest that some muscle diseases may be treated by shifting fiber type characteristics either from slow to fast,

Establishment of an in vitro culture system to study the developmental biology of Onchocerca volvulus with implications for anti-Onchocerca drug discovery and screening.

BACKGROUND: Infections with Onchocerca volvulus nematodes remain a threat in Sub-Saharan Africa after three decades of ivermectin mass drug administration. Despite this effort, there is still an urgent need for understanding the parasite biology especially the mating behaviour and nodule formation as well as the development of more potent drugs that can clear the developmental (L3, L4, L5) and adult stages of the parasite and inhibit parasite reproduction and behaviour. METHODOLOGY/PRINCIPAL FINDINGS: Prior to culture, freshly harvested O. volvulus L3 larvae from dissected Simulium damnosum f

Neurotoxin-Derived Optical Probes for Elucidating Molecular and Developmental Biology of Neurons and Synaptic Connections : Toxin-Derived Optical Probes for Neuroimaging.

Botulinum neurotoxins (BoNTs) and tetanus toxin (TeTX) are the deadliest biological substances that cause botulism and tetanus, respectively. Their astonishing potency and capacity to enter neurons and interfere with neurotransmitter release at presynaptic terminals have attracted much interest in experimental neurobiology and clinical research. Fused with reporter proteins or labelled with fluorophores, BoNTs and TeTX and their non-toxic fragments also offer remarkable opportunities to visualize cellular processes and functions in neurons and synaptic connections. This study presents the stat

Swimming Upstream to Understand Congenital Anomalies of the Kidney and Urinary Tract: Zebrafish Models for Developmental Biology, Disease Mechanisms, and Functional Interpretation of Genetic Variation.

Congenital anomalies of the kidney and urinary tract (CAKUT) are the leading cause of pediatric chronic kidney disease (CKD) and comprise a heterogeneous group of developmental disorders with a substantial genetic contribution. Advances in next-generation sequencing have facilitated the identification of numerous candidate genes and rare variants associated with CAKUT. However, establishing causality and defining the biological functions of implicated genes remain major challenges. Functional validation is therefore essential to bridge the gap between gene discovery and mechanistic understandi
