Peri-conceptional under-nutrition alters the expression of TRIM28 and ZFP57 in the endometrium and embryos during peri-implantation period in domestic pigs.

DNA methylation is maintained by the main elements of methylation complex-tripartite motif containing 28 (TRIM28) and zinc finger protein 57 (ZFP57). Previously, it was found that the activity of TRIM28 and ZFP57 determines the process of DNA methylation and preserves over-expression of genes. We hypothesized that restricted diet applied during peri-conceptional period may induce changes in the expression of methylation complex in porcine endometrium and embryos during the peri-implantation period. The aim of this study was to detect and determine the expression of TRIM28 and ZFP57 in the endo

Review and practical excursus on the comparison between traditional statics methods and Classification And Regression Tree (CART) in real-life data: Low protein diet compared to Mediterranean diet in patients with chronic kidney disease.

The Classification and Regression Tree (CART) is a supervised learning approach useful used to segment the space of and the predictors/features space into smaller homogeneous regions that are, represented in a decision tree. It computes the selection of features automatically, in contrast to traditional statistic methods. In this review, we compared CART to traditional statistics in patients who did not attempt a diet to patients who followed a low-protein diet (LPD) or the Mediterranean diet in chronic kidney disease (CKD) patients, and we analyzed them using linear regression and CART method

Beneficial responses to modified diets in treating patients with chronic kidney disease.

BACKGROUND: In rats, protein-restricted diets prolong survival from experimental chronic kidney disease (CKD). Diets for CKD patients that are restricted in protein and modified to limit the intake of phosphates and sodium not only reduce the accumulation of unexcreted metabolic products but also protect against hypertension and proteinuria. Their ability to slow progression of CKD is controversial, and this fact, plus concern about the nutritional adequacy of restricted diets, has limited this treatment strategy for CKD patients. METHODS: Expected responses of CKD patients to dietary manipul

Placental adaptations in a nonhuman primate model of gestational protein restriction.

We previously demonstrated decreased placental perfusion, reduced amniotic fluid protein content, and increased pregnancy loss in a nonhuman primate model of gestational protein restriction. Here, our objective was to link these detrimental findings with a functional placental assessment. As blood flow is critical to maternal-fetal exchange, we hypothesized that a protein-restricted diet would impair placental taurine uptake. Pregnant rhesus macaques were maintained on either control chow (CON, n = 5), a 33% protein-restricted diet (PR33, n = 5), or a 50% PR diet (PR50, n = 5) prior to and thr

Expression of P2X6, a purinergic receptor subunit, is affected by dietary zinc deficiency in rat hippocampus.

The purpose of these experiments was to determine whether dietary zinc depletion affected protein expression in the hippocampus. Eleven weanling Sprague-Dawley male rats (21 d) were fed the AIN-93G diet containing 1.5 ppm zinc and supplemented with 30 ppm of zinc in the drinking water. After 1 wk, the rats were randomly divided into three groups: control (n=3), pair fed (n=3), and zinc restricted (n=5). All groups consumed the same diet. The zinc-restricted group consumed water containing no zinc. The rats were sacrificed 3 wk later. Chelatable zinc levels in the hippocampus, as measured by N-

Role of choline deficiency in the Fatty liver phenotype of mice fed a low protein, very low carbohydrate ketogenic diet.

Though widely employed for clinical intervention in obesity, metabolic syndrome, seizure disorders and other neurodegenerative diseases, the mechanisms through which low carbohydrate ketogenic diets exert their ameliorative effects still remain to be elucidated. Rodent models have been used to identify the metabolic and physiologic alterations provoked by ketogenic diets. A commonly used rodent ketogenic diet (Bio-Serv F3666) that is very high in fat (~94% kcal), very low in carbohydrate (~1% kcal), low in protein (~5% kcal), and choline restricted (~300 mg/kg) provokes robust ketosis and weig

Protein-calorie malnutrition in squirrel monkeys: adaptive response to calorie deficiency.

Control, low-protein, low-calorie, and low-protein-plus-low-calorie diets were fed to 37, 14, 8, and 16 infant squirrel monkeys, respectively, from ages 2-8 wk. The deficient diets were planned to prevent growth and were designed to examine the short- and long-term effects of defined nutritional restrictions on body weight, food intake, hematocrit, plasma albumin, growth hormone, and cortisol concentrations. During the restriction period, plasma albumin levels were significantly decreased in both groups with a protein restriction component: plasma cortisol levels were significantly increased o

Serum Levels of Th2-Type Immunoglobulins are Increased in Weanling Mice Subjected to Acute Wasting Protein-Energy Malnutrition.

The bulk of our knowledge on immunosuppression in malnutrition comes from the experiments done on cell-mediated immunity. However, malnutrition-induced modifications of humoral immunity have been less understood. The objective of this study was to determine the effects of acute protein-energy malnutrition on serum levels of immunoglobulins and their subclasses in murine models. Male and female C57BL/6J mice were allocated to one of the four groups: (1) zero-time control (ZC) (19 days of age); (2) ad libitum intake of a complete purified diet (control group [CG]); (3) restricted intake of the c

Additive effects of dietary protein and energy deficiencies on mandibular growth in the weanling rat.

Dietary protein restriction adversely affects mandibular growth in the weanling rat. Protein deficiency is usually accompanied by reduced food intake which, in turn, induces energy deficiency. The present study was thus designed to dissociate the effects of dietary protein and energy deficiencies on the growth of the mandible in rapidly growing rats. Four groups of Sprague-Dawley rats aged 30 days were fed a normal diet, a low-energy diet, a low protein diet, and a low-protein and low-energy diet for 20 days. Rats were sacrificed at the end of experimental period and body weight and mandibular

Multicompartment analysis of protein-restricted phenylketonuric mice reveals amino acid imbalances in brain.

BACKGROUND: The mainstay of therapy for phenylketonuria (PKU) remains dietary protein restriction. Developmental and neurocognitive outcomes for patients, however, remain suboptimal. We tested the hypothesis that mice with PKU receiving protein-restricted diets would reveal disruptions of brain amino acids that shed light on these neurocognitive deficits. METHOD: Phenylalanine hydroxylase-deficient (PKU) mice and parallel controls (both wild-type and heterozygous) were fed custom diets containing 18, 6, and 4 % protein for 3 weeks, after which tissues (brain, liver, sera) were collected for a

T cells with a quiescent phenotype (CD45RA+) are overabundant in the blood and involuted lymphoid tissues in wasting protein and energy deficiencies.

The objective of this investigation was to determine the influence of distinct forms of acute weight loss on the expression of the quiescence marker, CD45RA, by T cells in several lymphoid compartments including the blood. Male and female weanling mice, CBA/J and C57BL/6J strains, were allocated to the following groups: ad libitum intake of a complete purified diet; restricted intake of the complete diet; and ad libitum intake of an isocaloric low-protein diet. The restricted intake protocol produced weight loss through energy deficiency (marasmic-type malnutrition), whereas the low-protein di

Low-protein diet for conservative management of chronic kidney disease: a systematic review and meta-analysis of controlled trials.

BACKGROUND: Recent data pose the question whether conservative management of chronic kidney disease (CKD) by means of a low-protein diet can be a safe and effective means to avoid or defer transition to dialysis therapy without causing protein-energy wasting or cachexia. We aimed to systematically review and meta-analyse the controlled clinical trials with adequate participants in each trial, providing rigorous contemporary evidence of the impact of a low-protein diet in the management of uraemia and its complications in patients with CKD. Polyclonal B-cell response to stimulation with Escherichia coli lipopolysaccharide in dietary protein restriction.

The polyclonal B-cell response to Escherichia coli lipopolysaccharide was studied in C57BL/6 mice maintained after weaning on either a moderate protein-restricted diet with 8% casein or a normal diet. After in vitro or in vivo stimulation with the endotoxin, autoreactive and anti-hapten antibody-producing cells were quantitated by direct plaque assay, using bromelain-treated mouse erythrocytes and trinitrophenylated sheep erythrocytes as targets. Larger numbers of plaque-forming cells were generated in cultures of spleen cells from dietary-restricted than from normal mice stimulated with vario

Choline insufficiency in mothers during gestation abolishes the metabolically restrained hepatic phenotype induced by gestational protein restriction and promotes hepatic steatosis in offspring.

Maternal protein restriction during gestation in rodents is widely used as a model of developmental programming; however, its metabolic effects in offspring are often modest. In humans, protein deficiency is frequently accompanied by deficiencies in essential micronutrients. This study examined whether maternal choline insufficiency, an essential micronutrient whose deficiency in adult rodents induces metabolic dysfunction-associated steatotic liver disease, modifies the metabolically restrained hepatic phenotype conferred by maternal protein restriction in offspring. Pregnant ICR mice were fe

Malnutrition during lactation changes growth hormone mRNA expression in offspring at weaning and in adulthood.

Mothers' nutrition during lactation programs growth in their offspring. We studied the contribution of the growth hormone (GH) for this programming, evaluating GH mRNA expression. Lactating dams were grouped as follows: C, control diet with 23% protein; PR, 8% protein-restricted diet; and ER, energy-restricted diet, receiving the control diet in restricted quantities of the PR group's ingestion. Some pups were killed at weaning; the others received the control diet until they were sacrificed as adults. Pituitary GH mRNA was analyzed by Northern blot analysis. At weaning, the ER and PR animals

Influence of catch up growth on spatial learning and memory in a mouse model of intrauterine growth restriction.

BACKGROUND: Intrauterine growth restriction (IUGR) and rapid postnatal weight gain or catch up growth (CUG) increase the susceptibility to metabolic syndrome during adult life. Longitudinal studies have also revealed a high incidence of learning difficulties in children with IUGR. The aim of the present study was to investigate the effect of nutrition and CUG on learning memory in an IUGR animal model. We hypothesized that synaptic protein expression and transcription, an essential mechanism for memory consolidation, might be affected by intrauterine undernutrition. METHODS: IUGR was induced

Lifelong dietary protein restriction induces denervation and skeletal muscle atrophy in mice.

As a widespread global issue, protein deficiency hinders development and optimal growth in offspring. Maternal low-protein diet influences the development of age-related diseases, including sarcopenia, by altering the epigenome and organ structure through potential increase in oxidative stress. However, the long-term effects of lactational protein restriction or postnatal lifelong protein restriction on the neuromuscular system have yet to be elucidated. Our results demonstrated that feeding a normal protein diet after lactational protein restriction did not have significant impacts on the neu

Early growth restriction leads to down regulation of protein kinase C zeta and insulin resistance in skeletal muscle.

Epidemiological studies have revealed a relationship between early growth restriction and the subsequent development of type 2 diabetes. A rat model of maternal protein restriction has been used to investigate the mechanistic basis of this relationship. This model causes insulin resistance and diabetes in adult male offspring. The aim of the present study was to determine the effect of early growth restriction on muscle insulin action in late adult life. Rats were fed either a 20% or an isocaloric 8% protein diet during pregnancy and lactation. Offspring were weaned onto a 20% protein diet and

Maternal malnutrition during lactation alters gonadotropin-releasing hormone expression in the hypothalamus of weaned male rat pups.

Gonadotropin-releasing hormone (GnRH) is the key hormone regulating reproduction. Its feedback regulation is exercised by estradiol. The early postnatal period is critical for sexual differentiation. Despite the fact that malnutrition-related reproductive suppression in rats is a well-documented phenomenon, we had no knowledge, until now, on how maternal malnutrition affects GnRH expression and estradiol serum concentrations of weaned pups. Six pregnant Wistar rats were separated into three groups at delivery with 6 pups each: control group (C) with free access to a standard diet containing 23

Effects of dietary protein and strain on the growth of broiler breeder pullets from zero to five weeks of age.

In a series of four experiments, the effects of differences in dietary protein intake on BW, skeletal growth and the weight of the Pectoralis major muscle was studied in Hubbard Standard and Hubbard Hi-Y broiler breeder pullets. In Experiment 1, pullets were fed diets with either 15, 17, or 19% CP from 1 to 21 d. There was a linear increase in BW gain and feed consumption with each increase in CP and Hubbard Standard pullets consumed significantly more feed than Hubbard Hi-Y pullets. From 21 to 34 d, pullets were subjected to feed restriction every day (ED) or every-other-day (EOD). On Day 35,
