Protective effects of diosgenin against non-alcoholic fatty liver disease through inhibiting the STING-dependent inflammatory pathway.

Growing evidence suggests that the stimulator of interferon genes (STING)-dependent inflammatory pathway is crucial in the progression of non-alcoholic fatty liver disease (NAFLD). Diosgenin (DG), a natural steroidal saponin, has demonstrated multi-pharmacological potential, such as anti-inflammatory and lipid-lowering capacities. Our previous study confirmed the protective role of DG in rat models of NAFLD. Our present study sought to further explore the protective effects of DG in NAFLD and to determine whether its mechanism involves the STING-dependent inflammatory pathway. In this research

Comparative Transcriptomic Analysis Provides Insights into PSK-α-Mediated Regulation of Diosgenin Biosynthesis in Trigonella foenum-graecum.

Diosgenin is an important steroidal sapogenin widely used for the industrial production of steroidal drugs. Phytosulfokine-α (PSK-α) is a sulfated pentapeptide hormone known to regulate plant growth, development, and stress responses. However, its role in the regulation of diosgenin biosynthesis remains unclear. Fenugreek seedlings were treated with PSK-α to investigate its effects on diosgenin biosynthesis through metabolite profiling and comparative transcriptomic analyses. PSK-α induced a dynamic response in diosgenin accumulation, characterized by an initial decrease during the early stage

Diosgenin ameliorates CLP-induced sepsis in mice via suppressing inflammatory responses and preserving intestinal mucosal barrier integrity.

BACKGROUND: Diosgenin, a well-known steroid sapogenin, has demonstrated anti-inflammatory effects; however, its therapeutic potential for sepsis remains unclear. Intestinal barrier dysfunction is a critical contributor to lethal sepsis, a systemic inflammatory response syndrome. This study investigated the protective effects of diosgenin on cecal ligation and puncture (CLP)-induced sepsis in mice, focusing on its impact on intestinal mucosal dysfunction and inflammation. Additionally, the effects of diosgenin on the expression of the endogenous antimicrobial peptide mCRAMP and the TLR4/MyD88 s

Phytofabricated ZnO-NPs mediated by Hibiscus tiliaceus leaf extract and its potential as a diosgenin delivery vehicle.

Zinc oxide nanoparticles (ZnO-NPs) have provided promising potential in the biomedical field, including the ability to overcome various health problems. Diosgenin is used to treat multiple health disorders but has very low solubility in water. Using ZnO-NPs as a diosgenin delivery vehicle was expected to increase the solubility of diosgenin, which would affect its bioavailability. This study demonstrates phytofabrication and characterization of ZnO-NPs, loading of diosgenin onto the ZnO-NPs, characterization of the product (ZnO-NPs/diosgenin), and evaluations of diosgenin release. Phytofabrica

Autophagy and mitochondrial dynamics contribute to the protective effect of diosgenin against 3-MCPD induced kidney injury.

3-Chloro-1, 2-propanediol (3-MCPD) is a widespread food contaminant with kidney as the main target organ. The exploration of ingredients as intervention strategy towards 3-MCPD induced nephrotoxicity is needed. Diosgenin (DIO) is a steroidal saponin presented in several plants and foods. Here we assessed whether DIO attenuates nephrotoxicity induced by 3-MCPD using Human embryonic kidney 293 (HEK293) cells and Sprague-Dawley (SD) rats. The results showed that DIO (2, 6, 8 μM) increased cell viability and exerted inhibitory effect on caspase 3 and caspase 9 activities. Histological examination

Diosgenin suppresses COX-2 and mPGES-1 via GR and improves LPS-induced liver injury in mouse.

Using a wild yam (Dioscorea japonica), we previously found novel anti-inflammatory and anti-carcinogenic effects via the downregulation of cyclooxygenase (COX)-2 and microsomal prostaglandin E synthase (mPGES)-1. One of the substances in wild yam is a steroidal saponin, diosgenin. We demonstrated that diosgenin suppressed COX-2 in human non-small-cell lung carcinoma A549 cells via nuclear factor-kappa B (NF-κB) translocation and the effects were reversed by a glucocorticoid receptor antagonist, RU486. In lipopolysaccharide (LPS)-induced mouse liver injury, COX-2 and mPGES-1 were induced and lo

Diosgenin presents a novel role in promoting diabetic wound healing: A mechanism involving Sirt6/Nrf2-mediated inhibition of ferroptosis.

Delayed wound healing in patients with diabetes is frequently caused by the dysfunction of human umbilical vein endothelial cells (HUVECs). Under high-glucose and high-fat (HGHF) conditions, the microenvironment induces uncontrolled lipid peroxidation and subsequent iron-dependent ferroptosis. Diosgenin (DG), a steroidal saponin extracted from herbs, including wild yam, exhibits diverse pharmacological activities. The present study aimed to investigate the role and underlying mechanism of DG in HGHF-induced ferroptosis and diabetic wound (DW) healing. A DW model was established in mice by intr
