Patterns of dermal denticle loss in sharks.

As they grow, sharks both replace lost denticles and proliferate the number of denticles by developing new (de novo) denticles without prior denticle shedding. The loss and replacement of denticles has potential impacts on the energetic cost of maintaining the skin surface, the biomechanical functions of shark skin, as well as our ability to predict shark abundance from fossil denticle occurrence in sediment cores. Here, we seek to better understand patterns of denticle loss and to show how denticles are being replaced in mature sharks. We illustrate shark skin surfaces with missing denticles

Selachian tooth development: I. Histogenesis, morphogenesis, and anatomical features in Squalus acanthias.

We have determined the general features of epithelial-mesenchymal interactions during fetal and mature adult stages of odontogenesis in the selachian spiny dogfish, Squalus acanthias. The general features of odontogenesis included the formation of an extended dental lamina, bud, cap, bell, and crown stages as identified and described using serial sections from fetal as well as adult specimens and light microscopy. Fetal and adult lengths, the rostrum-caudal fin distance, were correlated with the number of tooth rows and columns present in each specimen. This positional information and terminol

Granule containing cells and fibres in the sinus venosus of elasmobranchs.

The concentrations of catecholamines in the heart chambers of elasmobranchs were measured by the fluorimetric method of Bertler et al. (1958). Noradrenaline (NA) can be detected in all the chambers, but the sinus venosus is by far the richest in NA. This can either be due to the presence of storage sites for this amine in the sinus wall, or to a transport of amine to the sinus venosus from the anterior chromaffin bodies. The sinus wall contains large numbers of "granule containing cells" and axon-like processes, both with numerous dense-core vesicles of about 1800 A diameter. The dense-core ve

Speciation analysis of mercury and lead in fish samples using liquid chromatography-inductively coupled plasma mass spectrometry.

A liquid chromatography-inductively coupled plasma mass spectrometric (LC-ICP-MS) method for lead and mercury speciation analysis was described. Sample containing ionic lead and mercury compounds was subjected to liquid chromatographic separation before injection into the direct injection high efficiency nebulizer (DIHEN, 170-AA). The species studied include inorganic lead (Pb(II)), trimethyl lead (trimethyl-Pb), triethyl lead (triethyl-Pb), inorganic mercury (Hg(II)), methyl mercury (methyl-Hg) and ethyl mercury (ethyl-Hg), which were well separated by reversed-phase liquid chromatography wit

Biochemical and physiological studies on peptides from the elasmobranch gut.

A peptide fraction has been purified from intestinal extracts of the dogfish Scyliorhinus canicula which is a powerful stimulant of rectal gland secretory activity. This peptide is distinct from elasmobranch vasoactive intestinal polypeptide (VIP). The elasmobranch VIP fraction is not active in the rectal gland assay and mammalian VIP is only an effective agonist in Squalus and not in Scyliorhinus or Raja. Preliminary characterisation of the elasmobranch VIP indicate that it has strong N-terminal similarities with mammalian VIP but has limited C-terminal comparability. It is suggested that the

Healing of corneal epithelial wounds in marine and freshwater fish.

The corneal epithelium of a fish is in direct contact with the aquatic environment and is a barrier to movement of ions and water into and through the cornea. This tissue layer is thus important in maintenance of corneal transparency. When the epithelium is wounded, its protective function is lost and corneal transparency remains compromised until the epithelial barrier is re-established. This study was undertaken to investigate the healing response of the fish cornea to epithelial abrasion. Wounds were stained with fluorescein and photographed during healing. Wound areas were measured by plan

[Fine structure of the olfactory organ of elasmobranchs (Elasmobranchii)].

The structure of the receptor cells in the olfactory organ of the rays Raja clavata and Dasyatis pastinaca and spiny dogfish Squalus acanthias is similar to that of the olfactory cells in other Vertebrates, although their tops are invariably crowned by microvilli, not flagella. In the peripheral process of these cells of Elasmobranchs and some other vertebrates represent a special type of the olfactory receptors which originated as the result of differentiation of primary olfactory cells supplied both by flagella and by microvilli. In the olfactory epithelium of Elasmobranchs, together with th

Remote nitrogen-15 isotope effects on addition of cyanide to NAD.

The reversible reaction NAD + CN(-)----NAD-CN was examined for remote secondary 15N isotope effects caused by isotopic substitution at the ring nitrogen of the nicotinamide group. These were compared with analogous effects for dehydrogenase-catalyzed reactions, since both cyanide and the hydride ion add at the N-4 position of the nicotinamide ring. The 15N effects on the rate constants for the forward and reverse processes were examined directly by conducting both the normal and isotopic reactions simultaneously under carefully controlled conditions in the sample and reference cells of a dual-

Secretion of nitrate by rectal gland of Squalus acanthias.

1. Rectal glands secrete nitrate at 30% of their capacity to secrete chloride. 2. Nitrate secretion is directly related to its concentration at constant chloride concentrations. 3. Chloride has a biphasic effect on nitrate secretion. 4. Hill coefficients at chloride < 100 mM are equal to 1, while at 100 mM indicate inhibition of nitrate by chloride. 6. Lineweaver-Burk plots at chloride < 100 indicate a single site, while at 100 mM indicate inhibition of nitrate by chloride. 7. Bumetanide inhibits nitrate secretion. 8. The data suggest that nitrate interacts with one of the two chloride sites o

What's new with the NRC certified reference material program.

The environmental CRMs currently available from NRC Ottawa comprise four natural waters for trace elements, four biological tissues for trace elements and methylmercury, three marine sediments for major and trace elements (one sediment has certified values for butyltins) and one fish tissue for organochlorines (PCBs, dioxins). Although significant effort has been expended to maintain this suite of materials, several new initiatives have also recently been completed or are in progress. The certified value for arsenobetaine in Dogfish Muscle CRM DORM-2 has been established. The certification app

[Anatomical study on the facial nerve innervating the floor of the mouth in chondrichthyes. Homology of the chorda tympani].

This paper deals with the results of the investigation of the facial nerves of chondrichthyes in order to consider the phylogenetic origin of the Chorda tympani in human. Six species of elasmobranchs (Chlamydoselachus anguineus, Cephaloscyllium umbratile, Squalus acanthias, Dasyatis akajei, Raja kwangtungensis and Mobura diabolus) were dissected under a stereoscopic microscope for this purpose, and the following results were obtained. Ramus palatinus and R. pre-spiracularis were observed as pre-trematic branches, while R. mandibularis externus, R. mandibularis internus and R. hyoideus originat

Dense-cored vesicle-containing components of the terminal nerve of sharks and rays.

This paper describes electron microscopic observations of dense-cored vesicle-containing axons, cell bodies, and endings of the terminal nerve in several elasmobranchs. The vesicles are found in two apparent cell types, one with a polymorphic nucleus and another with an oval nucleus. The types may correspond to cells producing one each of two neuropeptides (LHRH and FMRF-amide) that have previously been localized in the nerve. Dense-cored vesicles are found in many unmyelinated fibers in both the terminal nerve proper and its major ganglia. Some of these form complicated structures with interd

Hydrostatic pressure effects on vestibular hair cell afferents in fish and crustacea.

Following the discovery of a hydrostatic pressure sensor with no associated gas phase in the crab, and the knowledge that several systems of cells in culture show long term alterations to small changes in hydrostatic pressure, we show here that vestibular type II hair cells in a well known model system (the isolated elasmobranch labyrinth), are sensitive to hydrostatic pressure. This new finding for the vertebrate vestibular system may provide an explanation for low levels of resting activity in vertebrate hair cells and explain how fish without swim bladders sense hydrostatic cues. It could h

Methylmercury determination in biota by solid-phase microextraction matrix effect evaluation.

Aqueous-phase alkylation followed by, headspace solid-phase microextraction (SPME) for mercury speciation in biota, was developed a decade ago. Despite this, matrix effects in this technique have not yet been addressed. In this paper, the importance of these effects has been assessed and overcome by standard addition calibration. Furthermore, improvements were made in the extraction of methylmercury (MeHg) from biological matrixes by optimizing the matrix digestion procedure (temperature and digestion time) and the SPME parameters (aliquot volume of digested samples, extraction temperature and

Coupling of the respiratory rhythm in fish with activity in hypobranchial nerves and with heartbeat.

Fish have a central respiratory pattern generator (CRPG) in the brain stem that initiates activity in a series of cranial nerves innervating respiratory muscles. These nerves burst sequentially in the order of their rostrocaudal distribution in the central nervous system. When respiratory drive is high, this activity spreads caudally to occipital and anterior spinal neurons that project via the hypobranchial nerves to stimulate hypaxial muscles, causing active jaw abduction. The CRPG may also recruit the heart. Fish, like mammals, show respiratory components in the intrinsic variability of hea

Methyl mercury concentrations in edible fish and shellfish from Dunedin, and other regions around the South Island, New Zealand.

Methyl mercury (MeHg) concentrations were determined in edible fish and shellfish available in local markets in Dunedin, New Zealand. While most of the fish species were sourced in Dunedin, some specimens of fish were also collected from waters off Picton, around Stewart Island and also off-shore of the South Island in the Puysegur and Subantarctic regions. The concentrations of MeHg were analysed in 25 different fish species and shellfish (103 muscle tissue samples). Total mercury (HgT) levels were also analysed in a few (n=12) selected fish samples. Most of the Hg was in the form of MeHg (≥

Distribution and characterization of neuropeptide Y in the brain of an elasmobranch fish.

Using a specific antiserum raised against synthetic neuropeptide Y (NPY), the distribution of immunoreactivity in the brain and pituitary of the elasmobranch fish Scyliorhinus canicula has been examined with the indirect fluorescence and the peroxidase-antiperoxidase methods. The highest density of NPY-immunoreactive neurons was found in the basal telencephalon and in the hypothalamus. Numerous NPY-containing perikarya were located in the entopeduncular and the preoptic nuclei, in the nucleus lobi lateralis and in the nucleus lateralis tuberis. NPY-immunopositive fibers were observed throughou

Membrane potentials and intracellular chloride activity in the ciliary body of the shark.

We have found that membrane potential in the isolated ciliary epithelium of the shark, Squalus acanthias, is -53 mV. High extracellular potassium or ouabain (10(-5) mol X l-1) decrease the potential, and furosemide (10(-4) mol X l-1) hyperpolarizes it. There is no difference in membrane potential between the cells of the non-pigmented and pigmented layers. Intracellular chloride activity (64 mmol X l-1) was significantly higher than could be predicted from the equilibrium distribution (26 mmol X l-1) across the cell membranes. When furosemide was applied to the aqueous side of the epithelium,

Effects of acidosis and carbonic anhydrase inhibition in the elasmobranch rectal gland.

We studied the effects of acidosis and carbonic anhydrase inhibition on rectal gland fluid secretion. In the anesthetized dogfish shark, Squalus acanthias, volume expanded by a constant infusion of Ringer solution, fluid formation was halved by severe systemic metabolic and respiratory acidosis (pH approximately 7.10) and carbonic anhydrase inhibition. Mild respiratory acidosis (pH approximately 7.6) equivalent to the measured systemic pH during carbonic anhydrase inhibition was without effect. Inhibition of rectal gland enzyme produced severe glandular acidosis, which is indicated by an incre
