A study of purified montmorillonite intercalated with 5-fluorouracil as drug carrier.

In the recent decade, the most useful drug for the therapy of colon cancer is 5-Fluorouracil(5-FU). It has been reported to have considerable toxicity administered by intravenous injections or via alimentary treat. Though many materials have developed for drug carrier of 5-FU, there was no clinically acceptable carrier for 5-FU till now. Montmorillonite, one of clay minerals, consists of hydrated aluminum silicates that are fined grained and usually have a large space between the layers. Isomorphous substitution of cations is common. We try to intercalate 5-FU into interlayer of montmorillonit

Intratumoral vascular damage-dependent drug permeability and anticancer effect of nano drug carriers: significant anticancer enhancement of combination therapy.

Determining how anticancer drug-induced intratumor vascular damage alters drug permeability and enhances therapeutic efficacy is crucial for optimizing cancer treatment. This study successfully quantified anticancer drug encapsulated-MnO2 (hollow PEGylated: H-MnO2-PEG) nanoparticle (NP)-induced tumor vascular damage at the microscopic level by determining the vascular surface volume (based on surface smoothing) as well as the number and volume of vascular filaments using tumor transparency imaging. Variations in the drug permeability of H-MnO2-PEG NPs encapsulating different anticancer drugs a

Extracellular Vesicles as Drug Carriers for Enzyme Replacement Therapy to Treat CLN2 Batten Disease: Optimization of Drug Administration Routes.

CLN2 Batten disease (BD) is one of a broad class of lysosomal storage disorders that is characterized by the deficiency of lysosomal enzyme, TPP1, resulting in a build-up of toxic intracellular storage material in all organs and subsequent damage. A major challenge for BD therapeutics is delivery of enzymatically active TPP1 to the brain to attenuate progressive loss of neurological functions. To accomplish this daunting task, we propose the harnessing of naturally occurring nanoparticles, extracellular vesicles (EVs). Herein, we incorporated TPP1 into EVs released by immune cells, macrophages

Recent advances in novel drug carriers and advanced administration routes for inner ear drug delivery: challenges and future perspectives.

Hearing loss (HL) affects over 1.5 billion people worldwide. The blood-labyrinth barrier (BLB) and the tympanic membrane (TM) can hinder the effectiveness of clinical treatments by preventing conventional drug delivery methods from precisely targeting the inner ear. This study aims to systematically summarise and analyse recent advances in novel drug carriers and delivery methods for inner ear drug delivery. This will provide theoretical support for the development of safe and effective HL treatment strategies. The review outlines the core characteristics of the latest invasive and non-invasiv

Two Sides of the Same Coin: Transthyretin (TTR) as a Target or Drug Carrier for Drug (Bio)conjugates.

Transthyretin (TTR) is a tetrameric protein present in plasma and cerebrospinal fluid that binds to thyroxine (T4) and retinol (vitamin A) to transport them across the blood-retina barrier and to the liver. Mutations on the TTR gene cause destabilization of the tetramer structure leading to misfolded monomers and aggregates, thus triggering several pathologies (i.e., cardiomyopathy and neurodegeneration). The stabilization of TTR tetramer architecture and the silencing of TTR gene expression represent viable therapeutic strategies for amyloidosis. Moreover, the TTR role as a delivery system us

Polydopamine/sodium alginate hydrogels with photothermal effects: a drug carrier for NIR-controlled drug release.

As a smart drug carrier, photosensitive hydrogels enable controlled drug release. Here, using polydopamine (PDA) as a photothermal agent additive, a smart PDA/SA hydrogel was constructed by ionic crosslinking of sodium alginate (SA). The PDA/SA hydrogels displayed an ideal capacity for sustained ibuprofen (IBU) release. In vitro release experiments showed that the release of IBU from PDA/SA hydrogels was affected by NIR light. The amount of IBU released from the PDA/SA hydrogel after NIR irradiation was higher than that from the SA hydrogel without NIR irradiation. The PDA/SA hydrogel effectiv

Lipid-drug-conjugate (LDC) nanoparticles as novel carrier system for the hydrophilic antitrypanosomal drug diminazenediaceturate.

The objective of the present study was to incorporate the hydrophilic drug diminazenediaceturate at a high loading into lipid nanoparticles by creating nanoparticles from lipid-drug conjugates (LDC). IR and DSC data showed that the antitrypanosomal drug diminazene is able to react with fatty acids to form water-insoluble salts like diminazenedistearate and -dioleate. The salts could be transformed into nanoparticles using high-pressure homogenization technique, established for solid lipid nanoparticles (SLN). By using polysorbate 80 as surfactant, physically stable LDC nanoparticle dispersions

A Microfluidic Platform for Evaluating the Internalization of Liposome Drug Carriers in Tumor Spheroids.

The development of in vitro models recapitulating nanoparticle transport under physiological flow conditions is of great importance for predicting the efficacy of nanoparticle drug carriers. Liposomes are extensively used for drug delivery owing to their biocompatibility and biodegradability and the ability to carry both hydrophilic and hydrophobic compounds. Here, we used a library of liposomes with various dimensions and a microfluidic platform comprising a large array of uniformly sized breast cancer spheroids to explore size-dependent liposome internalization and retention in the spheroids

The Application of Polymeric Nanoparticles as Drug Delivery Carriers to Cells in Neurodegenerative Diseases.

In spite of great advances in modern medicine, there are a few effective strategies for the treatment of neurodegenerative diseases characterised by neuron loss or degeneration. This results from complex pathogenesis of the diseases and the limited drug uptake of the brain due to the presence of blood-brain barrier. Nanoparticle-based drug delivery systems are expected to improve the drug utilisation. Polymeric nanoparticles represent promising drug delivery carriers to the brain due to their unique advantages such as good biodegradability and biocompatibility, flexibility in surface modificat

Antitumor Research Based on Drug Delivery Carriers: Reversing the Polarization of Tumor-Associated Macrophages.

The development of malignant tumors is a complex process that involves the tumor microenvironment (TME). An immunosuppressive TME presents significant challenges to current cancer therapies, serving as a key mechanism through which tumor cells evade immune detection and play a crucial role in tumor progression and metastasis. This impedes the optimal effectiveness of immunotherapeutic approaches, including cytokines, immune checkpoint inhibitors, and cancer vaccines. Tumor-associated macrophages (TAMs), a major component of tumor-infiltrating immune cells, exhibit dual functionalities: M1-like

Develop Targeted Protein Drug Carriers through a High-Throughput Screening Platform and Rational Design.

Protein-based drugs offer advantages, such as high specificity, low toxicity, and minimal side effects compared to small molecule drugs. However, delivery of proteins to target tissues or cells remains challenging due to the instability, diverse structures, charges, and molecular weights of proteins. Polymers have emerged as a leading choice for designing effective protein delivery systems, but identifying a suitable polymer for a given protein is complicated by the complexity of both proteins and polymers. To address this challenge, a fluorescence-based high-throughput screening platform call

[Research progress on the fluorescence resonance energy transfer-based polymer micelles as drug carriers].

Polymer micelles formed by self-assembly of amphiphilic polymers are widely used in drug delivery, gene delivery and biosensors, due to their special hydrophobic core/hydrophilic shell structure and nanoscale. However, the structural stability of polymer micelles can be affected strongly by environmental factors, such as temperature, pH, shear force in the blood and interaction with non-target cells, leading to degradations and drug leakage as drug carriers. Therefore, researches on the structural integrity and in vivo distribution of micelle-based carriers are very important for evaluating th
