Sensor macrophages derived from human induced pluripotent stem cells to assess pyrogenic contaminations in parenteral drugs.

Ensuring the safety of parenteral drugs before injection into patients is of utmost importance. New regulations around the globe and the need to refrain from using animals however, have highlighted the need for new cell sources to be used in next-generation bioassays to detect the entire spectrum of possible contaminating pyrogens. Given the current drawbacks of the Monocyte-Activation-Test (MAT) with respect to the use of primary peripheral blood mono-nuclear cells or the use of monocytic cell lines, we here demonstrate the manufacturing of sensor monocytes/macrophages from human induced plur

Evaluation of Closed-System Transfer Devices in Reducing Surface Contamination With Antineoplastic Drugs in Two Patient Administration Areas.

BACKGROUND: Antineoplastic drugs (ADs) are widely used in healthcare settings and dangerous to healthcare workers who may be exposed. Closed-system transfer devices (CSTDs) are engineering controls designed to reduce environmental exposure to ADs. OBJECTIVES: This study documents AD residue on surfaces in a high-volume outpatient infusion clinic and an inpatient oncology unit before and after the implementation of CSTDs to determine whether this engineering control decreased contamination with ADs. METHODS: The authors collected 60 surface wipe samples pre- and postimplementation in each set

An environmental risk assessment of contamination of lakes and rivers in Northern Ireland with pharmaceuticals, personal care products and drugs of abuse.

In this work, we present the most spatially comprehensive environmental risk assessment of 114 pharmaceuticals, personal care products and illicit drug residues in the surface waters of Northern Ireland. We sampled at 50 sites spread across five lakes and rivers. We detected 21 different substances in 137 water samples and quantified between 0.7-2967 ng L-1 (n = 834 quantifiable measurements). Within these, the top five most frequently detected compounds were the opioid painkiller tramadol (91%), the cocaine metabolite benzoylecgonine (91%, with cocaine itself at 55% frequency), caffeine (80%)

Antiviral drugs in wastewater are on the rise as emerging contaminants: A comprehensive review of spatiotemporal characteristics, removal technologies and environmental risks.

Antiviral drugs (ATVs) are widely used to treat illnesses caused by viruses. Particularly, ATVs were consumed in such large quantities during the pandemic that high concentrations were detected in wastewater and aquatic environment. Since ATVs are not fully absorbed by the human or animal body, this results in large amounts of them being discharged into the sewage through urine or feces. Most ATVs can be degraded by microbes at wastewater treatment plants (WWTPs), while some ATVs either require deep treatment to reduce concentration and toxicity. Parent and metabolites residing in effluent pos

Occupational exposure to antineoplastic drugs in hospital environments: potential risk associated with contact with cyclophosphamide- and ifosfamide-contaminated surfaces.

BACKGROUND: Cyclophosphamide (CP) and ifosfamide (IP) contaminations have been detected in hospital environments. This study was conducted to determine if there was any contamination in the spaces (floors and door handles) between the hospital exit and the antineoplastic drugs (ADs) preparation and administration units. At the same time, the authors proposed a new automation of the analytical procedure to considerably decrease the time needed for sample preparation and analysis. MATERIAL AND METHODS: To evaluate the ADs contamination of surfaces, 829 wipe tests were performed in a campaign in

Lymphocytes exposed to vegetables grown in waters contaminated by anticancer drugs: metabolome alterations and genotoxic risks for human health.

Wastewater irrigation of crops may be effective to avoid depletion (about 70%) of freshwater resources. However, the use of reclaimed waters containing persistent microcontaminants such as antineoplastic drugs is of high environmental concern. These active compounds may affect human health with potentially severe adverse effects. To better understand the impact on human health following irrigation of crops with reused contaminated waters, we exposed four edible plants, Brassica rapa, Lactuca sativa, Raphanus sativus, and Triticum durum, to two commonly used antitumoral drugs: 5-fluorouracil (5

Chlorhexidine washing in intensive care does not reduce bloodstream infections, blood culture contamination and drug-resistant microorganism acquisition: an interrupted time series analysis.

BACKGROUND: Health care-associated infections are a major cause of morbidity and mortality in intensive care patients. The effect of daily washing with chlorhexidine on these infections is controversial. METHODS: Single-centre, retrospective, open-label, sequential period, interrupted time series (ITS) analysis in a 31-bed tertiary referral mixed intensive care unit (ICU), comparing daily washing with water and soap (from January 2011 to August 2013) with chlorhexidine washing (from November 2013 to December 2015), after the introduction of a unit-level policy of chlorhexidine washing. Activated carbon adsorption sheets suppress volatilization of anticancer drugs excreted in urine, preventing environmental contamination and human exposure.

BackgroundAnticancer drugs excreted in urine can disperse during urination, contaminating toilets and causing secondary exposure. Activated carbon sheets adsorb urinary drugs and limit surface diffusion, but whether adsorbed drugs subsequently vaporize is unknown. We evaluated vaporization of cyclophosphamide (CPA) applied to activated carbon sheets.MethodsCPA (10 mg/2 mL) was placed on 100-cm2 activated carbon sheets or control sheets without activated carbon and incubated for 24 h in a 4-L chamber at 37 °C. Vaporization was assessed by Ames test (reverse-mutated Salmonella colony counts). CP

Proposals of guidance values for surface contamination by antineoplastic drugs based on long term monitoring in Czech and Slovak hospitals and pharmacies.

INTRODUCTION: The exposures to hazardous antineoplastic drugs (AD) represent serious risks for health care personnel but the exposure limits are not commonly established because of the no-threshold effects (genotoxic action, carcinogenicity) of many ADs. In this study, we discussed and derived practically applicable technical guidance values (TGV) suitable for management of AD risks. METHODS: The long-term monitoring of surface contamination by eight ADs was performed in pharmacies and hospitals in the Czech Republic and Slovak Republic in 2008-2021; in total 2,223 unique samples were collect

Selective adsorption and removal of drug contaminants by using an extremely stable Cu(II)-based 3D metal-organic framework.

The adsorption capacity of three representative pharmaceutical drugs and personal care products (PPCPs) viz. diclofenac sodium (DCF), chlorpromazine hydrochloride (CLF) and amodiaquin dihydrochloride (ADQ), were preliminarily studied using a water-stable Cu(II)-based metal organic framework (MOF) [Cu(BTTA)]n·2DMF (1) (H2BTTA = 1,4-bis(triazol-1-yl)terephthalic acid). We also investigated the factors influencing the adsorption such as concentration, pH, contact time, temperature and dosages. The results show that the adsorption capacity of 1 for DCF (650 mg g-1) from aqueous medium, which is hi

Beef carcass contamination by Shiga toxin-producing Escherichia coli strains in an abattoir in Brazil: characterization and resistance to antimicrobial drugs.

A survey was performed to estimate the frequency of Escherichia coli and Shiga toxin-producing E. coli (STEC) in carcasses obtained from an abattoir in Brazil between February 2006 and June 2007. A total of 216 beef carcasses were sampled at three stages of the slaughter process--preevisceration, postevisceration, and postprocessing--during the rain and dry seasons, respectively. Of the carcasses sampled, 58% were preevisceration E. coli positive, 38% were postevisceration positive, and 32% postprocessing positive. At the postprocessing stage, the isolation of E. coli was twice as high in the

Everything but the Kitchen Sink: An Analysis of Bacterial and Chemical Contaminants Found in Syringe Residue From People Who Inject Drugs.

BACKGROUND: People who inject drugs (PWID) are at high risk of severe wounds, invasive infections, and overdoses. To date, there are few data on the bacterial and chemical contaminants PWID are exposed to when using illicitly manufactured fentanyls and stimulants. METHODS: Previously used injection drug use equipment was recovered in St Louis, Missouri, by harm reduction organizations over a 12-month period. Syringe residue was analyzed for bacterial contaminants by routine culturing followed by whole genome sequencing of single bacterial isolates. Chemical adulterants in syringe residue were

Drug use disorder following early life exposure to tetrachloroethylene (PCE)-contaminated drinking water: a retrospective cohort study.

BACKGROUND: Many studies of adults with occupational exposure to solvents such as tetrachloroethylene (PCE) have shown adverse effects on cognition, mood and behavioral problems. Much less is known about neurotoxic effects in early life at lower exposure levels seen in community settings. We recently reported that illicit drug use was more frequent among adults from Cape Cod, Massachusetts who were exposed to PCE-contaminated drinking water during gestation and early childhood than their unexposed counterparts. Using newly collected data from this population-based retrospective cohort study, t
